VLDL lipidomics reveals hepatocellular lipidome changes in metabolic dysfunction-associated steatotic liver disease.

BACKGROUND: The production of VLDL is one of the primary mechanisms through which liver cells regulate intracellular lipid homeostasis. We hypothesize that the disease characteristics of metabolic dysfunction-associated steatotic liver disease (MASLD) differentially impact VLDL lipid composition. This study comprehensively examines the relationship between VLDL-lipidome and MASLD histology and disease-associated genetics, aiming to define MASLD-related VLDL changes. METHODS: We performed untargeted lipidomics on serum VLDL particles in a cohort of biopsy-proven MASLD patients to examine the r

Comparison of the capillary and venous blood plasma lipidomes: validation of self-collected blood for plasma lipidomics.

Venipuncture of the upper extremities is commonly used to collect blood for plasma lipidomics. However, self-administered blood collection devices such as the Tasso+™ system for capillary blood sampling and plasma separation are convenient and enable frequent sampling without a clinical blood draw. The purpose of this study is to validate Tasso+ sampling for plasma lipidomics by comparing the venous blood and Tasso+-sampled capillary blood plasma lipidomes. Lipids are proven or putative biomarkers of human health and disease and indicators of nutritional and toxicological status. Because excha

Alterations in the Plasma Lipidome of Adult Women With Bipolar Disorder: A Mass Spectrometry-Based Lipidomics Research.

Lipidomics has become a pivotal tool in biomarker discovery for the diagnosis of psychiatric illnesses. However, the composition and quantitative analysis of peripheral lipids in female patients with bipolar disorder (BD) have been poorly addressed. In this study, plasma samples from 24 female patients with BD and 30 healthy controls (HCs) were analyzed by comprehensive lipid profiling and quantitative validation based on liquid chromatography-mass spectrometry. Clinical characteristics and a correlation between the level of lipid molecules and clinical symptoms were also observed. We found th

LC-MS/MS-Based Untargeted Lipidomics Using MS-DIAL.

Mass spectrometry (MS) has become the cornerstone of metabolomics and lipidomics because of its high sensitivity, specificity, and ability to analyze complex biological samples. However, the data complexity necessitates the use of robust computational tools. MS-DIAL is a versatile and user-friendly software for untargeted metabolomics and lipidomic workflows. It was launched as a universal program for untargeted metabolomics and supports multiple instruments (GC/MS, GC/MS/MS, LC/MS, and LC/MS/MS) and MS vendors (Agilent, Bruker, LECO, Sciex, Shimadzu, Thermo, and Waters). Common data formats s

The Neurolipid Atlas: a lipidomics resource for neurodegenerative diseases.

Lipid alterations in the brain have been implicated in many neurodegenerative diseases. To facilitate comparative lipidomic research across brain diseases, we establish a data common named the Neurolipid Atlas that we prepopulated with isogenic induced pluripotent stem cell (iPS cell)-derived lipidomics data for different brain diseases. Additionally, the resource contains lipidomics data of human and mouse brain tissue. Leveraging multiple datasets, we demonstrate that iPS cell-derived neurons, microglia and astrocytes exhibit distinct lipid profiles that recapitulate in vivo lipotypes. Notab

Lipidomics and in vitro azole resistance in Candida albicans.

We have shown earlier that fluconazole (FLC) stress induces global changes in the lipidome of Candida albicans in clinically adapted isolates. However, several laboratories have developed adapted in vitro FLC resistant strains of C. albicans to study azole resistance mechanisms. This study aimed to identify the lipid changes associated with FLC resistance in these in vitro adapted isolates. Using comparative lipidomics and principal component and discriminant analyses, we observed gradual changes in several lipid classes and molecular species upon FLC exposure of in vitro resistant C. albicans

Integrated lipidomics in the secreted phospholipase A(2) biology.

Mammalian genomes encode genes for more than 30 phospholipase A(2)s (PLA(2)s) or related enzymes, which are subdivided into several subgroups based on their structures, catalytic mechanisms, localizations and evolutionary relationships. More than one third of the PLA(2) enzymes belong to the secreted PLA(2) (sPLA(2)) family, which consists of low-molecular-weight, Ca(2+)-requiring extracellular enzymes, with a His-Asp catalytic dyad. Individual sPLA(2) isoforms exhibit unique tissue and cellular localizations and enzymatic properties, suggesting their distinct pathophysiological roles. Recent

DIATAGeR: Triacylglycerol annotation of data-independent acquisition based lipidomics.

BACKGROUND: Triacylglycerols (TGs) are the most abundant lipids in the human body and the primary source of energy storage. TGs are comprised of three fatty acyls with various lengths and double bond composition, complicating structural annotation when performing lipidomics by LCMS. Data-independent acquisition (DIA) based lipidomics enables a continuous and unbiased acquisition of all TGs, creating the potential for more comprehensive TG analysis. However, TG identification in DIA lipidomics data is challenging due to the difficulty analyzing multiplexed tandem mass spectra (MS2). RESULTS: I

A platelet lipidomics signature in patients with COVID-19.

Ischemic cardiovascular and venous thromboembolic events are a frequent cause of death in severe COVID-19 patients. Platelet activation plays a key role in these complications, however platelet lipidomics have not been studied yet. The aim of our pilot investigation was to perform a preliminary study of platelet lipidomics in COVID-19 patients compared to healthy subjects. Lipid extraction and identification of ultrapurified platelets from eight hospitalized COVID-19 patients and eight age- and sex-matched healthy controls showed a lipidomic pattern almost completely separating COVID-19 patien

[Research progress on biomarkers for endometriosis based on lipidomics].

The pathogenesis of endometriosis is not well understood at the moment, and the lack of effective biomarkers often leads to delayed diagnosis of the disease. Lipidomics provides a new approach for the diagnosis and prediction of endometriosis. Sphingomyelin, phosphatidylcholine and phosphatidylserine in peripheral blood, endometrial fluid, peritoneal fluid and follicular fluid have good diagnostic value for endometriosis and disease classification; the lipid metabolites in the eutopic endometrium tissue are expected to be biomarkers of early endometriosis; and the lipid metabolites in peripher

Comprehensive Lipoprotein Characterization Using Lipidomics Analysis of Human Plasma.

Lipoproteins are responsible for the transport of lipids and other nutrients in the circulation and therefore play an important role in lipid metabolism and dyslipidemia. They have also been linked to multiple metabolic disorders including cardiovascular disease, and thus understanding their lipid composition is of crucial importance. Characterization of lipoproteins is a challenging task due to their heterogeneity. In particular, their fractionation is often laborious and time-consuming, making large sets of clinical samples difficult to analyze. We developed and validated lipidomics analysis

Lipidome remodeling in primary biliary cholangitis.

Primary biliary cholangitis (PBC) is a chronic, cholestatic liver disease characterized by progressive destruction of intrahepatic bile ducts, impaired bile flow, and complex disturbances in bile acid and lipid metabolism. PBC is associated with a distinctive dyslipidemic profile, marked by elevated total cholesterol, accumulation of lipoprotein X, and paradoxically high HDL levels. At the molecular level, dysregulation of bile acid-activated nuclear receptors - particularly farnesoid X receptor (FXR) and peroxisome proliferator-activated receptors (PPARs) - contributes to abnormalities in cho
