Lipopeptide Nanoparticles: Development of Vaccines against Hookworm Parasite.

Necator americanus (hookworm) infects over half a billion people worldwide. Anthelminthic drugs are commonly used to treat the infection; however, vaccination is a more favorable strategy to combat this parasite. We designed new B-cell peptide epitopes based on the aspartic protease of N. americanus (Na-APR-1). The peptides were conjugated to self-adjuvanting lipid core peptide (LCP) systems via stepwise solid-phase peptide synthesis (SPPS) and copper catalyst azide-alkyne cycloaddition (CuAAC) reactions. The LCP vaccine candidates were able to self-assemble into nanoparticles, were administer

Novel HIV fusion-inhibitory lipopeptides exhibit dramatically improved activity against resistant mutants.

LP-98 is a potent HIV fusion-inhibitory lipopeptide but displays reduced activity against resistant mutants. In this study, we rationally designed a group of new lipopeptides by adding different lengths of gp41 pocket-binding sequence into the N-terminus of LP-98. It was found that a single pocket-inserting residue Ile124 critically determined the anti-HIV activity, whereas incorporation of additional pocket-inserting residues Trp120 and Trp117 generated lipopeptides with greatly reduced activity. As indicated by LP-101 and LP-108, while Ile124-modified lipopeptides maintained or improved the

Super short membrane-active lipopeptides inhibiting the entry of influenza A virus.

Influenza A viruses (IAV) are significant pathogens that result in millions of human infections and impose a substantial health and economic burdens worldwide. Due to the limited anti-influenza A therapeutics available and the emergence of drug resistant viral strains, it is imperative to develop potent anti-IAV agents with different mode of action. In this study, by applying a pseudovirus based screening approach, two super short membrane-active lipopeptides of C12-KKWK and C12-OOWO were identified as effective anti-IAV agents with IC50 value of 7.30±1.57 and 8.48±0.74 mg/L against A/Puerto R

An Economical Bioprocess for Lipopeptide Production From Bacillus subtilis.

Phytopathogenic fungi such as Fusarium solani (F. solani), Rhizoctonia solani (R. solani), and Botrytis cinerea (B. cinerea) cause plant diseases, leading to significant yield losses. Lipopeptides (LPs), secondary metabolites produced by Bacillus strains, exhibit antifungal properties against these pathogens. In this study, the LP production potential of Bacillus isolates was investigated, and the best-performing Bacillus sp. was selected for the optimization of the production medium and parameters. Hemolytic activities of Bacillus isolates were screened using blood agar, revealing that all 69

Lipopeptide-Based Oral Vaccine Against Hookworm Infection.

BACKGROUND: The human hookworm, Necator americanus, is a parasite that infects almost half a billion people worldwide. Although treatment is available, vaccination is favorable to combat the spread of this parasite due to its wide distribution and continuous reinfection cycle in endemic communities. METHODS: We have designed a lipopeptide oral delivery system using a B-cell epitope derived from the aspartic protease Na-APR-1 from N americanus, attached to a T-helper epitope. Lipopeptides were self-assembled into nanoparticles or entrapped in liposomes that were electrostatically coated with a

In vivo characterization of A-192411: a novel fungicidal lipopeptide (II).

The ability of the novel antifungal cyclic hexalipopetide A-192411 to treat fungal infections in rodents is presented. Efficacy was demonstrated against Candida albicans as both prolonged survival of systemically infected mice and clearance of viable yeasts from kidneys. The efficacy of A-192411, administered intraperitoneally, was equivalent to amphotercin B at a 4-fold lower dose by weight in the systemic candidiasis models in mice, while the efficacy of A-192441 administered intravenously was equivalent to amphotercin B by weight in the Candida pyelonephritis model in rats. A-192411 also sl

Structural characterization of lipopeptide agonists for the bradykinin B2 receptor.

The conformational features of Pam-Lys(0)-Arg(1)-Pro(2)-Pro(3)-Gly(4)-Phe(5)-Ser(6)-Pro(7)-Phe(8)-Arg(9)-OH (PKD) and Pam-Gly(-1)-Lys(0)-Arg(1)-Pro(2)-Pro(3)-Gly(4)-Phe(5)-Ser(6)-Pro(7)-Phe(8)-Arg(9)-OH (PGKD), the Pam-Lys and Pam-Gly-Lys analogues of bradykinin, have been determined by high-resolution NMR in a zwitterionic lipoid environment. Radical-induced relaxation of the (1)H NMR signals was used to probe the topological orientation of the peptides with respect to the zwitterionic lipid interface. The radical-induced relaxation and molecular dynamics (MD) data indicated that the palmitic

In-vitro evaluation of cationic Lipopeptides as adjuvant candidate for DNA plasmid vaccine.

Lipopeptides with different fatty acids (palmitic, palmitoleic, stearic, oleic, and linoleic acids) conjugated to CHSPKKKRKV were synthesised by a solid-phase peptide method using the Fmoc strategy (Fmoc-SPPS) on 2-CTC (2-chlorotritylchloride) resin. The lipopeptides were purified by RP-HPLC and characterised by ToF-ESI-MS and 1D-NMR. The capability of the lipopeptide to interact with the plasmid was evaluated by DNA agarose gel electrophoresis. The particle size of the lipopeptide/DNA complexes was determined by dynamic light scattering assay and TEM analysis. The biological activities includ

Bacillus-derived lipopeptides inhibit and eradicate Candida tropicalis Biofilms: An in vitro exploration.

Candida tropicalis is a WHO-prioritised opportunistic fungal pathogen capable of causing superficial-to-systemic fungal infections and forming biofilms on both biotic and abiotic surfaces, such as tissues and medical implants. The rising incidence of fungal infections and the emergence of drug resistance underscore the urgent need for alternative, natural therapeutic agents such as antifungal macromolecules. AF4 and AF5, natural antifungal lipopeptide homologs derived from the Bacillus subtilis RLID strain, were purified to homogeneity from the cell free supernatant using semi-preparative scal

Characterization of novel HIV fusion-inhibitory lipopeptides with the M-T hook structure.

Combination antiretroviral therapy (cART) has significantly improved the survival of HIV-infected individuals, but long-term treatment can cause side-effects and drug resistance; thus, the development of new antivirals is of importance. We previously identified an M-T hook structure and accordingly designed short-peptide fusion inhibitor 2P23, which mainly targets the gp41 pocket site and displays potent, broad-spectrum anti-HIV activity. In this study, we continuingly characterized the amino acid sequences of peptide and lipopeptide-based inhibitors containing the M-T hook residues. Among a g

Lipopeptides: a novel antigen repertoire presented by major histocompatibility complex class I molecules.

Post-translationally modified peptides, such as those containing either phosphorylated or O-glycosylated serine/threonine residues, may be presented to cytotoxic T lymphocytes (CTLs) by MHC class I molecules. Most of these modified peptides are captured in the MHC class I groove in a similar manner to that for unmodified peptides. N-Myristoylated 5-mer lipopeptides have recently been identified as a novel chemical class of MHC class I-presented antigens. The rhesus classical MHC class I allele, Mamu-B*098, was found to be capable of binding N-myristoylated lipopeptides and presenting them to C

Cyclic lipopeptides from Bacillus subtilis activate distinct patterns of defence responses in grapevine.

Non-self-recognition of microorganisms partly relies on the perception of microbe-associated molecular patterns (MAMPs) and leads to the activation of an innate immune response. Bacillus subtilis produces three main families of cyclic lipopeptides (LPs), namely surfactins, iturins and fengycins. Although LPs are involved in induced systemic resistance (ISR) activation, little is known about defence responses induced by these molecules and their involvement in local resistance to fungi. Here, we showed that purified surfactin, mycosubtilin (iturin family) and plipastatin (fengycin family) are p

Assessment of mosquito larvicidal potency of cyclic lipopeptides produced by Bacillus subtilis strains.

In this study, mosquito larvicidal potency of cyclic lipopeptides (CLPs) secreted by two Bacillus subtilis strains were determined. LC50 of the crude CLPs secreted by B. subtilis DM-03 and DM-04 strains against third instar larvae of Culex quinquefasciatus was 120.0+/-5.0 and 300.0+/-8.0mg/l respectively post 24 h of treatment. Physico-chemical factors such as pH of water, incubation temperature, heating and exposure to sunlight hardly influenced the larvicidal potency of these CLPs. Present study provided the evidence that B. subtilis lipopeptides were safe to Indian major carp Labeo rohita,

Involvement of fengycin-type lipopeptides in the multifaceted biocontrol potential of Bacillus subtilis.

In this work, the potential of Bacillus subtilis strain M4 at protecting plants against fungal diseases was demonstrated in different pathosystems. We provide evidence for the role of secreted lipopeptides, and more particularly of fengycins, in the protective effect afforded by the strain against damping-off of bean seedlings caused by Pythium ultimum and against gray mold of apple in post-harvest disease. This role was demonstrated by the strong biocontrol activity of lipopeptide-enriched extracts and through the detection of inhibitory quantities of fengycins in infected tissues. Beside suc

Unlocking the potential of antimicrobial lipopeptides: a pathway to combat multidrug-resistant pathogens.

Antimicrobial lipopeptides (AMLPs) have emerged as a promising class of therapeutic agents against multidrug-resistant pathogens due to their unique amphiphilic structure and multimodal mechanisms of action. This review systematically summarizes the microbial origins and structural classification of AMLPs, elucidates their membrane-targeting and intracellular bactericidal mechanisms, and explores strategies for evading resistance. Furthermore, we analyze their structure-activity relationships (SAR) at the primary, secondary, and self-assembled structural levels, proposing optimization strategi

Relative biomembrane fusogenicities of the tumor-selective liposomes of RGDK- and CGKRK-lipopeptides.

Cancer is the second leading cause of death globally after heart diseases. Currently used highly cytotoxic anti-cancer drugs not only kill cancer cells but also often kill non-cancerous healthy body cells, causing adverse side effects. Efforts are now being directed towards developing tumor-selective chemotherapy. Tumor/tumor endothelial cell selective peptide ligands are being covalently grafted onto the exo-surfaces of drug carriers such as liposomes, polymers, etc. A number of prior studies used conjugation of tumor/tumor endothelial cell-selective RGDK- or CGKRK-peptide ligands on the oute

Effect of self-assembly on antimicrobial activity of double-chain short cationic lipopeptides.

Short cationic antimicrobial lipopeptides with surfactant-like structure are promising antibiotic candidates that preferentially target microbial membranes. Therefore, we focused our study on double-chain lipopeptides, (C10-16)2Dab-KKK-NH2 and (C10-16)2Dap-KKK-NH2, where Dab and Dap are 2,4-diaminobutyric and 2,3-diaminopropionic acids, respectively. We tried to answer a question how the self-assembly behaviour affects biological activities of the tested compounds. The subject compounds were synthesized by solid-phase method and screened for their antimicrobial and haemolytic activities. Cytot

Biocontrol of the wheat pathogen Zymoseptoria tritici using cyclic lipopeptides from Bacillus subtilis.

Innovation toward ecofriendly plant protection products compatible with sustainable agriculture and healthy food is today strongly encouraged. Here, we assessed the biocontrol activity of three cyclic lipopeptides from Bacillus subtilis (mycosubtilin, M; surfactin, S; fengycin, F) and two mixtures (M + S and M + S + F) on wheat against Zymoseptoria tritici, the main pathogen on this crop. Foliar application of these biomolecules at a 100-mg L-1 concentration on the wheat cultivars Dinosor and Alixan, 2 days before fungal inoculation, provided significant reductions of disease severity. Lipopeptides as the antifungal and antibacterial agents: applications in food safety and therapeutics.

A lot of crops are destroyed by the phytopathogens such as fungi, bacteria, and yeast leading to economic losses to the farmers. Members of the Bacillus genus are considered as the factories for the production of biologically active molecules that are potential inhibitors of growth of phytopathogens. Plant diseases constitute an emerging threat to global food security. Many of the currently available antimicrobial agents for agriculture are highly toxic and nonbiodegradable and thus cause extended environmental pollution. Moreover, an increasing number of phytopathogens have developed resistan

A Toll-like receptor 2-integrin beta3 complex senses bacterial lipopeptides via vitronectin.

Toll-like receptor 2 (TLR2) initiates inflammation in response to bacterial lipopeptide (BLP). However, the molecular mechanisms enabling the detection of BLP by TLR2 are unknown. Here we investigated the interaction of BLP with human serum proteins and identified vitronectin as a BLP-recognition molecule. Vitronectin and its receptor, integrin beta(3), were required for BLP-induced TLR2-mediated activation of human monocytes. Furthermore, monocytes from patients with Glanzmann thrombasthenia, which lack integrin beta(3), were completely unresponsive to BLP. In addition, integrin beta(3) forme
