In dialyzed squid axons oxidative stress inhibits the Na+/Ca2+ exchanger by impairing the Cai2+-regulatory site.

The Na(+)/Ca(2+) exchanger, a major mechanism by which cells extrude calcium, is involved in several physiological and physiopathological interactions. In this work we have used the dialyzed squid giant axon to study the effects of two oxidants, SIN-1-buffered peroxynitrite and hydrogen peroxide (H(2)O(2)), on the Na(+)/Ca(2+) exchanger in the absence and presence of MgATP upregulation. The results show that oxidative stress induced by peroxynitrite and hydrogen peroxide inhibits the Na(+)/Ca(2+) exchanger by impairing the intracellular Ca(2+) (Ca(i)(2+))-regulatory sites, leaving unharmed the

Mobility and transport of magnesium in squid giant axons.

1. The Mg content of axons obtained from Loligo forbesi averaged 6.4 +/- 0.8 m-mole/kg axoplasm.2. A small patch of radioactive (28)Mg injected into an axon broadened considerably. A similar patch of (45)Ca showed hardly any broadening. The self-diffusion coefficient of Mg in axoplasm is about 2 x 10(-6) cm(2)/sec which is at least twenty times greater than that of Ca.3. Under the influence of an applied electric field Mg migrated towards the cathode. Its mobility was about half of that of Mg in free solution. This suggests that the concentration of ionized Mg in squid axoplasm is between 2 an

High-Throughput DNA Sequencing Reveals Gastric Content Composition and Inter-Specific Variation in Pampus Fishes.

Pampus species play a significant role in offshore food webs and are regarded as representative economically important fish. Owing to variations in their geographical distribution, distinct differences in feeding habits exist among pomfret species. However, the presence of an esophageal sac in pomfrets complicates accurate identification of their dietary composition through conventional stomach content analysis. Consequently, research on the feeding ecology of pomfrets remains limited and imprecise. This study employed environmental DNA (eDNA) high-throughput sequencing technology to analyze t

Establishment of a dexamethasone-induced zebrafish skeletal muscle atrophy model and exploration of its mechanisms.

BACKGROUND: Skeletal muscle atrophy is one of the main side effects of high-dose or continuous use of glucocorticoids (such as dexamethasone). However, there are limited studies on dexamethasone-induced skeletal muscle atrophy in zebrafish and even fewer explorations of the underlying molecular mechanisms. This study aimed to construct a model of dexamethasone-induced skeletal muscle atrophy in zebrafish and to investigate the molecular mechanisms. METHODS: Zebrafish soaked in 0.01 % dexamethasone solution for 10 days. Loli Track (Denmark) and Loligo Swimming Respirometer were used to observe

Wild fish and seafood species in the western Mediterranean Sea with low safe mercury concentrations.

A total of 1345 specimens belonging to 58 different species of wild fish and seafood from the western Mediterranean Sea were analyzed to assess total mercury levels and to estimate which species meet the EU recommendations for human consumption (0.5 μg g-1 ww) in all cases. All fish species were caught off the Mediterranean coasts and intended for human consumption. All specimens were collected from local markets located in Spain, Italy and France that sell fish caught by local fishermen (Eivissa, Menorca, Mallorca, Alacant, L'Ampolla, Ametlla de Mar, Marseille, Genoa, Civitavecchia, Alghero)

Mercury in the tissues of five cephalopods species: First data on the nervous system.

Mercury (Hg), one of the elements most toxic to biota, accumulates within organisms throughout their lifespan and biomagnifies along trophic chain. Due to their key role in marine systems, cephalopods constitute a major vector of Hg in predators. Further, they grow rapidly and display complex behaviours, which can be altered by neurotoxic Hg. This study investigated Hg concentrations within 81 cephalopod specimens sampled in the Bay of Biscay, which belonged to five species: Eledone cirrhosa, Sepia officinalis, Loligo vulgaris, Todaropsis eblanae and Illex coindetii. Hg concentrations were mea

Loss of popdc3 Impairs Mitochondrial Function and Causes Skeletal Muscle Atrophy and Reduced Swimming Ability in Zebrafish.

BACKGROUND: The Popeye domain containing 3 (POPDC3) protein is essential for the maintenance of skeletal muscle homeostasis. POPDC3 is a pathogenic variant gene of limb-girdle muscular dystrophy (LGMD), and its variants lead to LGMDR26. At the animal level, zebrafish larvae with popdc3 mutations develop tail curls and muscle atrophy. However, the mechanism of skeletal muscle atrophy induced by POPDC3 variants/loss remains unclear. METHODS: Eight-month-old male WT and popdc3 mKO zebrafish were used for this research. Loli Track (Denwmark) and Loligo Swimming Respirometer were used to observe t

Pharmacological induction of rhythmical activity and plateau action potentials in unmyelinated axons.

The physiological function of the axon is to conduct short all-or-none action potentials from their site of initiation (usually the cell body) to the synapse. To ensure this function, both passive and active biophysical properties of the axons are tuned very precisely, especially the voltage-dependent ionic conductances to sodium and potassium. Under normal conditions, axons are not spontaneously active. Minor modifications of their ionic micro-environment or slight changes in the membrane properties are however sufficient to induce rhythmical activity and modify the time course of the action

Восстановление ТРНС последнего универсального общего пастора (Общества) на пути к восстановлению системы перевода Древних.

Реконструирование ансестральной последовательности (АСР) было использовано для изучения эволюции переводческой системы путем определения последовательности tRNA в "Last Archaeal Common Ancestor" (LACA), "Last Bacterial Common Ancestor" (LBCA) и "Last Universal Common Ansancestor" (LUCA), которые в наших предыдущих исследованиях назывались "Общество". Составное филогенетическое дерево, взятое из реконструированных последовательности ТРНА, выявило топологию, отличную от ранее выведенных аминоачил-тРНА синтетазы (АРС). Такая непоследовательность может отражать изменения в структурных особенностях

Biomaterials from the sea: Future building blocks for biomedical applications.

Marine resources have tremendous potential for developing high-value biomaterials. The last decade has seen an increasing number of biomaterials that originate from marine organisms. This field is rapidly evolving. Marine biomaterials experience several periods of discovery and development ranging from coralline bone graft to polysaccharide-based biomaterials. The latter are represented by chitin and chitosan, marine-derived collagen, and composites of different organisms of marine origin. The diversity of marine natural products, their properties and applications are discussed thoroughly in t
