FATP5 Is Indispensable for the Growth of Intrahepatic Cholangiocarcinoma.

UNLABELLED: Altered lipid metabolism is a common hallmark of various cancers, including intrahepatic cholangiocarcinoma (ICC), a highly lethal carcinoma that lacks effective treatment options. To elucidate the lipid metabolism changes in ICC, we coupled the expression of the firefly luciferase gene (FFL) to AKT1 (AKT-FFL) via an IRES linker, and then hydrodynamically injected mice with AKT-FFL and Notch1 intracellular cytoplasmic domain (NICD) to establish a luciferase-positive ICC model. This model not only enabled us to monitor and quantify tumor growth by injecting the mice with luciferin,

In vivo bioluminescence imaging reveals copper deficiency in a murine model of nonalcoholic fatty liver disease.

Copper is a required metal nutrient for life, but global or local alterations in its homeostasis are linked to diseases spanning genetic and metabolic disorders to cancer and neurodegeneration. Technologies that enable longitudinal in vivo monitoring of dynamic copper pools can help meet the need to study the complex interplay between copper status, health, and disease in the same living organism over time. Here, we present the synthesis, characterization, and in vivo imaging applications of Copper-Caged Luciferin-1 (CCL-1), a bioluminescent reporter for tissue-specific copper visualization in

2',3'-dideoxynucleoside triphosphates (ddNTP) and di-2',3'-dideoxynucleoside tetraphosphates (ddNp4ddN) behave differently to the corresponding NTP and Np4N counterparts as substrates of firefly luciferase, dinucleoside tetraphosphatase and phosphodiesterases.

2',3'-Dideoxynucleosides (ddN) and their derivatives are currently used as antiretroviral compounds. Their active agents are the corresponding 2',3'-dideoxynucleoside triphosphates (ddNTPs) generated inside the cell by host kinases. Dinucleoside tetraphosphates (Np4Ns) are molecules of interest in metabolic regulation; their synthesis in vitro can be catalyzed by firefly luciferase. The relative synthesis of diadenosine 5',5'''-P1,P4-tetraphosphate or adenosine(5')tetraphospho(5')adenosine (Ap4A) from ATP is about 100-fold faster than that of di-2',3'-dideoxyadenosine 5',5'''-P1,P4-tetraphosph

Aconitum alkaloids, the major components of Aconitum species, affect expression of multidrug resistance-associated protein 2 and breast cancer resistance protein by activating the Nrf2-mediated signalling pathway.

BACKGROUND: Aconitum alkaloids from Aconitum species are often used to treat arthritis and rheumatic diseases but have the drawback of high toxicity. Identifying their pharmacokinetic behaviour is important for the safe clinical application of Aconitum species. Efflux transporters (ETs), including P-glycoprotein (P-gp), multidrug resistance-associated protein 2 (MRP2), and breast cancer resistance protein (BCRP), have important functions in regulating the pharmacokinetic behaviours of drugs and in herb-herb or herb-drug interactions (HDIs). The Aconitum alkaloids regulate P-gp expression and f

Monitoring immediate-early gene expression through firefly luciferase imaging of HRS/J hairless mice.

BACKGROUND: Gene promoters fused to the firefly luciferase gene (luc) are useful for examining gene regulation in live transgenic mice and they provide unique views of functioning organs. The dynamics of gene expression in cells and tissues expressing luciferase can be observed by imaging this enzyme's bioluminescent oxidation of luciferin. Neural pathways involved in specific behaviors have been identified by localizing expression of immediate-early genes such as c-fos. A transgenic mouse line with luc controlled by the human c-fos promoter (fos::luc) has enabled gene expression imaging in br

Efficient Strategy for the Synthesis and Modification of 2-Hydroxyethylluciferin for Highly Sensitive Bioluminescence Imaging of Endogenous Hydrogen Sulfide in Cancer Cells and Nude Mice.

Bioluminescence (BL) imaging is among the most popular methods for visualizing biological processes in vitro, in live cells, and even in the whole organism. The alkylation of aminoluciferin at the 6'-amino site (2-hydroxyethyl luciferin, HE-AL) can emit more enhanced and sustained BL signals than d-luciferin and aminoluciferin, which is suitable for approaches that require long integration time such as three-dimensional animal BL imaging. Nevertheless, the drawback of HE-AL is that the synthesis procedure is complex, which leads to few BL probes based on HE-AL. In this Article, an efficient an

Denoising for high-magnification bioluminescence imaging of cells by machine learning with physics-based noise modeling of EMCCD.

Bioluminescence imaging (BLI) detects light emission from samples expressing luciferase in the presence of luciferin. Unlike fluorescence imaging, BLI does not require excitation light, thereby avoiding phototoxicity in living cells and photobleaching of labels. However, the intrinsically slow turnover of the luciferin-luciferase reaction often results in low luminescence intensity, which degrades image quality and limits practical applications, particularly in high-magnification optical microscopy of cells. In this study, we demonstrate that image denoising provides an effective strategy to o

Monitoring Py8119 breast cancer cell proliferation in direct cocultures with osteoblasts and osteoclasts.

The intricate interaction among breast cancer (BrCa) cells, stromal/osteoblastic cells, and osteoclasts in the osseous microenvironment drives a vicious cycle that accelerates both osteolysis and tumor progression. Elucidating how these bone cells directly modulate BrCa cell proliferation is of paramount importance. Due to a lack of intrinsic background of luminescence signal in the body, the luciferase/luciferin assay enables sensitive, in vivo monitoring of tumor progression. However, this technique has not yet been utilized to monitor tumor cell proliferation during direct coculture with bo

A high involvement of O2- possibly generated in inner membranes for iron-induced microsomal lipid peroxidation.

The lipid peroxidation of and the O2- generation by rat liver microsomes in the presence of NADPH or both NADPH and Fe3+ were determined by thiobarbituric acid-reacting substance formation and by chemiluminescence intensities with a cypridina luciferin analog, 2-methyl-6-(p-methoxyphenyl)-3, 7-dihydroimidazo[1,2-a]pyrazin-3-one(MCLA), as a chemiluminescence probe. Judging from the experiments with various inhibitors on the O2- generation and the lipid peroxidation, O2- generated, at intramembranous site, by cytochrome P-450 system is considered to be highly involved in the iron-induced lipid p

Validation and application of caged Z-DEVD-aminoluciferin bioluminescence for assessment of apoptosis of wild type and TLR2-deficient mice after ischemic stroke.

Programmed cell death or apoptosis is a critically important mechanism of tissue remodeling and regulates conditions such as cancer, neurodegeneration or stroke. The aim of this research article was to assess the caged Z-DEVD-aminoluciferin substrate for in vivo monitoring of apoptosis after ischemic stroke in TLR2-deficient mice and their TLR2-expressing counterparts. Postischemic inflammation is a significant contributor to ischemic injury development and apoptosis, and it is modified by the TLR2 receptor. Caged Z-DEVD-aminoluciferin is made available for bioluminescence enzymatic reaction b

A Nanobionic Light-Emitting Plant.

The engineering of living plants for visible light emission and sustainable illumination is compelling because plants possess independent energy generation and storage mechanisms and autonomous self-repair. Herein, we demonstrate a plant nanobionic approach that enables exceptional luminosity and lifetime utilizing four chemically interacting nanoparticles, including firefly luciferase conjugated silica (SNP-Luc), d-luciferin releasing poly(lactic-co-glycolic acid) (PLGA-LH2), coenzyme A functionalized chitosan (CS-CoA) and semiconductor nanocrystal phosphors for longer wavelength modulation.

Correlation between membrane-localized protons and flash-driven ATP formation in chloroplast thylakoids.

Flash-driven ATP formation by spinach chloroplast thylakoids, using the luciferin luminescence assay to detect ATP formed in single turnover flashes, was studied under conditions where a membrane protein amine buffering pool was either protonated or deprotonated before the beginning of the flash trains. The flash number for the onset of ATP formation was delayed by about 10 flashes (from 15 to about 25) when the amine pool was deprotonated as compared to the protonated state. The delay was substantially reversed again by reprotonating the pool upon application of 20-30 single-turnover flashes

Noninvasive Bioluminescence Imaging of Serum Albumins in Living Mice.

Bioluminescence (BL) is an emerging optical readout that has been extensively used in various bioassays and molecular imaging systems. In this study, we present the bioanalytical application of marine luciferins as an excellent optical indicator for noninvasive imaging of serum albumins. We synthesized 30 kinds of regioisomeric coelenterazine (CTZ) analogs and investigated their specificities for major serum proteins from various species. The results found that some of the CTZ analogs exhibited surprisingly specific optical signals upon binding with the serum albumins. These CTZ analogs showed

A new bioluminescent probe for detecting formaldehyde in real food samples.

Formaldehyde (FA) is an important type of reactive carbonyl species (RCS). The development of a rapid and efficient method to quantify FA in food and biological samples is necessary. Herein, we have developed the first bioluminescent probe FA-Fluc with homoallyl amine to cage firefly D-luciferin (Fluc), which could react almost completely with FA via aza-Cope rearrangement in 4 h to afford a turn-on Bioluminescence (BL). The detection limit of FA-Fluc was 0.047 ppm; and could detect FA in different seafood, agricultural products, and cooking processes. FA-Fluc has a good selectivity for FA, wh

Simultaneous measurement of adenosine triphosphate release and aggregation potentiates human platelet aggregation responses for some subjects, including persons with Quebec platelet disorder.

Platelet aggregometry and dense granule adenosine triphosphate (ATP) release assays are helpful to diagnose platelet disorders. Some laboratories simultaneously measure aggregation and ATP release using Chronolume® a commercial reagent containing D-luciferin, firefly luciferase and magnesium. Chronolume® can potentiate sub-maximal aggregation responses, normalising canine platelet disorder findings. We investigated if Chronolume® potentiates human platelet aggregation responses after observing discrepancies suspicious of potentiation. Among patients simultaneously tested by light transmission

Application and trend of bioluminescence imaging in metabolic syndrome research.

Bioluminescence imaging is a non-invasive technology used to visualize physiological processes in animals and is useful for studying the dynamics of metabolic syndrome. Metabolic syndrome is a broad spectrum of diseases which are rapidly increasing in prevalence, and is closely associated with obesity, type 2 diabetes, nonalcoholic fatty liver disease, and circadian rhythm disorder. To better serve metabolic syndrome research, researchers have established a variety of animal models expressing luciferase, while also committing to finding more suitable luciferase promoters and developing more ef
