Use of plate-wash samples to monitor the fates of culturable bacteria in mercury- and trichloroethylene-contaminated soils.

With the ultimate aim of developing bioremediation technology that use the optimum bacterial community for each pollutant, we performed polymerase chain reaction (PCR)-denaturing gradient gel electrophoresis (DGGE) and phylogenetic analysis and identified communities of culturable bacteria in HgCl(2)- and trichloroethylene (TCE)-contaminated soil microcosms. PCR-DGGE band patterns were similar at 0 and 1 ppm HgCl(2), but changes in specific bands occurred at 10 ppm HgCl(2). Band patterns appearing at 10 and 100 ppm TCE were very different from those at 0 ppm. Phylogenetic analysis showed four

Subsurface drainage enhances the cadmium enrichment efficiency of flax-sesame crop rotation by inhibiting the immobilization effect of iron oxides.

Flax-sesame crop rotation is an effective phytoremediation method for cadmium (Cd)-contaminated farmland. To investigate the effect of drainage on the uptake efficiency of soil Cd by the crops, field experiments were conducted to analyze the migration of Cd, iron (Fe), and manganese (Mn), and the soil microbial community. Compared with conventional drainage, the deep furrow treatment increased the Cd2 + accumulated in the flax and sesame organs by up to 73.9 % and decreased the soil Cd2+ content by 4.7 %-7.7 %. The accumulation of Fe2+/Fe3+ and Mn2+ showed the same trend as that of Cd. In the

Organic fertilizer improves soil fertility and restores the bacterial community after 1,3-dichloropropene fumigation.

Although fumigants can effectively control soil-borne diseases they are typically harmful to beneficial microorganisms unless methods are developed to encourage their survival after fumigation. The soil fumigant 1,3-dichloropropene (1,3-D) is widely used because of its effective management of pathogenic nematodes and weeds. After fumigation with 1,3-D, Bacillus subtilis and Trichoderma harzianum fertilizer (either singularly or together) or humic acid were added to soil that had been used to produce tomatoes under continuous production for >20 years. We evaluated changes to the soil's physicoc

Intratumoral Microbiome of Metastatic Pancreatic Ductal Adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) remains one of the most lethal oncological diseases, with a 5-year survival rate of approximately 13%-among the lowest in oncology. Poor survival is driven by aggressive tumor progression and metastasis, which may be influenced by the tumor microbiome. This study aimed to evaluate the role of microbiome in PDAC progression and metastasis. First, we assessed the microbial composition of control samples (surface swabs, empty paraffin, extraction controls, and sequencing controls) and removed contaminant taxa. Overall bacterial biomass was extremely low, wi

Effects of AMF on tobacco growth in continuous cropping soils: impacts on soil chemical properties and rhizosphere microbial diversity.

INTRODUCTION: Continuous tobacco cropping leads to reduced nutrient bioavailability, severe autotoxicity, and disrupts the rhizosphere microbial balance, ultimately reducing leaf yield and quality. Inoculation with arbuscular mycorrhizal fungi (AMF) can promote plant growth by enhancing nutrient uptake and modulating soil microbial communities. METHODS: This study investigated the mechanisms underlying growth inhibition in consecutively cropped soils and evaluated the potential of AMF to alleviate such cropping stress through a pot experiment. RESULTS: Compared with non-inoculated plants, AM

Efficient immobilization of enzyme on covalent organo-framework for remediation of pyrene-contaminated soil and degradation mechanism.

Bioremediation of polycyclic aromatic hydrocarbons (PAHs) using immobilized enzymes has garnered significant interest due to its cost-effectiveness, stability, and efficiency. In this regard enzyme laccase have been extensively used for the remediation of organic contaminants in aqueous solutions. However, the use of a single and/or free enzyme may not show better results due to its rapid degradation and loss of activity. Moreover, the use of immobilized enzymes for remediating specific PAH compounds in soil remains underexplored. Therefore, the aim of the present study was to prepare laccase

Multi-omics reveals manure-borne doxycycline and fragmented oversized microplastics co-disrupt pak choi growth and amplify antibiotic resistance in agroecosystems.

The intensification of livestock farming and plastic consumption has led to widespread co-contamination of agricultural soils with veterinary antibiotics (e.g., doxycycline, DOX) and microplastics (MPs). This study employed an integrative multi-omics approach (transcriptomics, metabolomics, microbiome analysis, qPCR) to investigate the synergistic effects of fragmented oversized microplastics (OMPs; 5-20 mm) and manure-borne DOX on pak choi growth and antibiotic resistance gene (ARG) dissemination in a realistic rhizobox system simulating manure-amended soil. We observed that co-exposure to DO

Weed control, maize (Zea mays L.) metabolism, and rhizosphere microbiome of hollow mesoporous silica nanoparticles loaded with S-metolachlor and ZnO quantum dots.

BACKGROUND: Conventional pesticide formulations employ large amounts of organic solvents, raising concerns regarding their environmental contamination, pesticide residues, and potential toxicity to non-target organisms. Hollow mesoporous silica nanoparticles (HMSNs) are non-toxic to both animals and plants, making them a suitable candidate for pesticide loading and delivery studies. RESULTS: HMSNs with an average particle size of 165 nm were synthesized using a self-template method. Fluorescein isothiocyanate was successfully conjugated to the HMSNs to enable the investigation of their transp

Evolution of beta-lactam biosynthesis genes and recruitment of trans-acting factors.

Penicillins and cephalosporins belong chemically to the group of beta-lactam antibiotics. The formation of hydrophobic penicillins has been reported in fungi only, notably Penicillium chrysogenum and Emericella nidulans, whereas the hydrophilic cephalosporins are produced by both fungi, e.g., Acremonium chrysogenum (cephalosporin C), and bacteria. The producing bacteria include Gram-negatives and Gram-positives, e.g. Lysobacter lactamdurans (cephabacins) and Streptomyces clavuligerus (cephamycin C), respectively. For a long time the evolutionary origin of beta-lactam biosynthesis genes in fung

Identification of the rhizospheric microbe and metabolites that led by the continuous cropping of ramie (Boehmeria nivea L. Gaud).

Continuous cropping lowers the production and quality of ramie (Boehmeria nivea L. Gaud). This study aimed to reveal the metagenomic and metabolomic changes between the healthy- and obstacle-plant after a long period of continuous cropping. After 10 years of continuous cropping, ramie planted in some portions of the land exhibited weak growth and low yield (Obstacle-group), whereas, ramie planted in the other portion of the land grew healthy (Health-group). We collected rhizosphere soil and root samples from which measurements of soil chemical and plant physiochemical properties were taken. Al

Mercapto-Functionalized Carbon Dots Reduce Cadmium Accumulation in Wheat Grains via Plant-Soil-Microbial Regulation.

Sustainable strategies to reduce Cd accumulation in wheat grown on Cd-contaminated alkaline soils remain limited. Here, we developed mercapto-functionalized carbon dots (SH-CDs) from Salvia miltiorrhiza biomass via a solvent-free ball milling approach with 3-mercaptopropyltrimethoxysilane. SH-CDs exhibited superior Cd2+ adsorption capacity (35.9 mg·g-1), primarily attributed to the strong affinity of surface-bound mercapto (-SH) groups, as confirmed by DFT calculations. SH-CDs applied via soil irrigation (800 mg·L-1) and foliar spraying (1 mg·mL-1) effectively reduced Cd content in wheat grain

Response of bacterial community metabolites to bacterial wilt caused by Ralstonia solanacearum: a multi-omics analysis.

The soil microbial community plays a critical role in promoting robust plant growth and serves as an effective defence mechanism against root pathogens. Current research has focused on unravelling the compositions and functions of diverse microbial taxa in plant rhizospheres invaded by Ralstonia solanacearum, however, the specific mechanisms by which key microbial groups with distinct functions exert their effects remain unclear. In this study, we employed a combination of amplicon sequencing and metabolomics analysis to investigate the principal metabolic mechanisms of key microbial taxa in p

Selenium and Bacillus proteolyticus SES synergistically enhanced ryegrass to remediate Cu-Cd-Cr contaminated soil.

Heavy metal compound contaminated soil is an ecological threat, and soil containing copper (Cu), cadmium (Cd) and chromium (Cr) simultaneously is widely distributed. The application of phytoremediation in heavy metal combined contamination is still limited. In this study, to explore whether and how exogenous selenium (Se) and Bacillus proteolyticus SES enhance the remediation of combined Cu-Cd-Cr contaminated soil by ryegrass, pot experiments were carried out. Se alone or in combination with B. proteolyticus SES treatment increased the removal rates of heavy metals in the rhizosphere soil by 1

Dynamics of soil bacterial communities in response to repeated application of manure containing sulfadiazine.

Large amounts of manure have been applied to arable soils as fertilizer worldwide. Manure is often contaminated with veterinary antibiotics which enter the soil together with antibiotic resistant bacteria. However, little information is available regarding the main responders of bacterial communities in soil affected by repeated inputs of antibiotics via manure. In this study, a microcosm experiment was performed with two concentrations of the antibiotic sulfadiazine (SDZ) which were applied together with manure at three different time points over a period of 133 days. Samples were taken 3 and

Biosynthesized selenium nanoparticles increase soybean resistance to root rot by recruiting beneficial microbes and reprogramming host metabolism.

BACKGROUND: Soybean root rot, primarily caused by Fusarium species, is a devastating soil-borne disease that severely threatens global soybean production. Conventional chemical controls are problematic due to their environmental impact and the risk of fostering pathogen resistance. There is therefore an urgent need to develop sustainable and eco-friendly alternative strategies. RESULTS: This study demonstrated that selenium nanoparticles (SeNPs) biosynthesized by Bacillus subtilis ZY56 significantly increased soybean resistance to F. oxysporum. Characterization confirmed that the obtained SeN
