Are Bordetella bronchiseptica Siphoviruses (Genus Vojvodinavirus) Appropriate for Phage Therapy-Bacterial Allies or Foes?

Bordetella bronchiseptica is a respiratory animal pathogen that shows growing resistance to commonly used antibiotics, which has necessitated the examination of new antimicrobials, including bacteriophages. In this study, we examined the previously isolated and partially characterized B. bronchiseptica siphoviruses of the genus Vojvodinavirus (LK3, CN1, CN2, FP1 and MW2) for their ability to inhibit bacterial growth and biofilm, and we examined other therapeutically important properties through genomic analysis and lysogeny experiments. The phages inhibited bacterial growth at a low multiplici

Phage VP882 possesses quorum-sensing-driven lytic induction and stress-mediated host growth suppression mechanisms.

Vibriophage VP882 launches its lytic cascade upon detection of a quorum-sensing autoinducer produced by its bacterial host. This capability enables the phage to maximize transmission by transitioning from lysogeny to lysis only at high host cell density, when abundant host cells are present to infect. Here, we show that two different pathways can be triggered upon prophage induction. When the prophage is induced via quorum sensing, Qtip, an anti-repressor, inhibits the cI lysogeny maintenance protein, driving expression of the lysis genes. The phage can also launch a cascade that causes host g

[Identification of Vibrio mimicus bacteriophages].

Lysogeny was studied in Vibrio mimicus; the indicator V. cholerae El Tor strain was selected to identify phages. New V. mimicus phages were obtained and identified, which had a morphological similarity and an antigen affinity for morphological group I cholerae phages. Phage differentiation revealed that morphological group I V. mimicus phages showed certain differences manifested as their lytic activity against V. cholerae strain 1322-69 of serovar 37 while this property was absent in cholerae phages.

Reply to Abedon, S.T. Dual-Receptor Recognition, Lysis Inhibition, Endolysin Release, and Reaction-Diffusion as Alternative Explanations. Comment on "Rojero et al. Bypassing Evolution of Bacterial Resistance to Phages: The Example of Hyper-Aggressive Phage 0524phi7-1. Int. J. Mol. Sci. 2025, 26, 2914".

In this manuscript, we isolate and characterize a phage that displays what we call anti-bacterial hyper-aggressive behavior. This behavior appears ideal for phage therapy of bacterial disease. It includes (1) formation of semi-turbid zones that subsequently clear, (2) formation of miniature satellite plaques, which probably constitute the foundation of the semi-turbid zones, (3) multi-day enlargement of both circular plaques and cleared semi-turbid zones, and (4) non-formation of phage-resistant host colonies. We emphasize the following key details in our response. (1) The semi-turbid zones ar

Control of λ Lysogenic Escherichia coli Cells by Synthetic λ Phage Carrying cIantisense.

Enterobacterial phage λ is a temperate phage that infects Escherichia coli and has a lytic-lysogenic life cycle. CI, a λ repressor, regulates the expression of lytic transcripts and acts as a major genetic switch that determines the lysogenic state. To manipulate the genome of phage λ, the CRISPR-Cas9 genome editing system was constructed in lysogenic E. coli MG1655 cells. For instance, we successfully changed cI857 to cIWT in the phage genome through Cas9-mediated single-nucleotide editing. A lytic phage was prepared by introducing an amber mutation in the middle of the cI gene, but it could

A recombinant lysogenic bacteriophage inhibits Salmonella virulence.

The gut environment includes an abundance of chemicals emanating from the host and the microbiota. To colonize animals, Salmonella uses gut chemicals as locational cues to ensure expression of energy-intensive virulence factors only when their production is necessary. cis-2-hexadecenoic acid (c2HDA), a member of the diffusible signal factor family of quorum-sensing signals, potently represses virulence-gene expression by Salmonella. Here, we report the construction and use of a recombinant bacteriophage that can establish lysogeny within Salmonella and induce it to produce c2HDA, thus repressi

Temperate bacteriophage SapYZUs7 alters Staphylococcus aureus fitness balance by regulating expression of phage resistance, virulence and antimicrobial resistance gene.

Temperate bacteriophages are crucial for maintaining the pathogenicity and fitness of S. aureus, which also show promise as a biocontrol agent for S. aureus. However, the fitness benefit and cost of lysogeny by S. aureus temperate phages and their underlying mechanisms remain unexplored. In this study, phage resistance, virulence, antimicrobial resistance (AMR), transcriptome, and metabolome of phage SapYZUs7 lysogenic and non-lysogenic S. aureus strains were compared. Whole-genome analysis revealed that SapYZUs7 harbouring smaII associated with a single-protein MazF-like antiphage system coul

Omnipresent Allies: The Role of Temperate Phages in Microbial Adaptation Across Ecosystems.

Understanding the extraordinary environmental adaptability of prokaryotes is crucial for manipulating microbial communities, as their adaptive mechanisms drive community dynamics, resilience and functional responses to interventions like bioremediation. Microbial adaptation to the environment is shaped not only by their intrinsic characteristics but also by interactions with other microorganisms. Among them, temperate phages, which reside alongside cellular microorganisms across diverse ecosystems, are emerging as key players in microbial adaptation. This review delves into the contribution of

Anti-CRISPR proteins trigger a burst of CRISPR-Cas9 expression that enhances phage defense.

CRISPR-Cas immune systems provide bacteria with adaptive immunity against bacteriophages, but they are often transcriptionally repressed to mitigate auto-immunity. In some cases, CRISPR-Cas expression increases in response to a phage infection, but the mechanisms of induction are largely unknown, and it is unclear whether induction occurs strongly and quickly enough to benefit the bacterial host. In S. pyogenes, Cas9 is both an immune effector and auto-repressor of CRISPR-Cas expression. Here, we show that phage-encoded anti-CRISPR proteins relieve Cas9 auto-repression and trigger a rapid incr

Isolation, Characterization, Genome Analysis and Host Resistance Development of Two Novel Lastavirus Phages Active against Pandrug-Resistant Klebsiella pneumoniae.

Klebsiella pneumoniae is a global health threat and bacteriophages are a potential solution in combating pandrug-resistant K. pneumoniae infections. Two lytic phages, LASTA and SJM3, active against several pandrug-resistant, nosocomial strains of K. pneumoniae were isolated and characterized. Their host range is narrow and latent period is particularly long; however, their lysogenic nature was refuted using both bioinformatic and experimental approaches. Genome sequence analysis clustered them with only two other phages into the new genus Lastavirus. Genomes of LASTA and SJM3 differ in only 13

Bacteriophage-encoded 24B_1 molecule resembles herpesviral microRNAs and plays a crucial role in the development of both the virus and its host.

The 24B_1 small non-coding RNA molecule has been identified in Escherichia coli after induction of Shiga toxin-converting bacteriophage Φ24B. In this work, we focused on its direct role during phage and bacterial host development. We observed that in many aspects, this phage sRNA resembles herpesviral microRNAs. Similar to microRNAs, the mature 24B_1 is a short molecule, consisting of just 20 nucleotides. It is generated by cleaving the 80-nt long precursor transcript, and likely it undergoes a multi-step maturation process in which the Hfq protein plays an important role, as confirmed by demo

[The level of spontaneous phage production and sensitivity to melioidosis phages of museum cultures of Pseudomonas pseudomallei].

The phage-producing activity and sensitivity of museum cultures of a melioidosis agent to melioidosis phages have been studied. Most cultures show spontaneous phage-production, though its level differs in strains. Some of the melioidosis cultures demonstrate lack of phages. The fact that these strains have different sensitivity to chloroform lysate of cultures producing phages indicates their possible lysogenic state. Electron microscopy has shown the presence of at least tow morphological types of phages in P. pseudomallei C-141. These data confirm existence of polysogeny in the melioidosis a

Prophage ϕSA169 Enhances Vancomycin Persistence in Methicillin-Resistant Staphylococcus aureus (MRSA).

Background: Persistent methicillin-resistant Staphylococcus aureus (MRSA) endovascular infections present a significant clinical therapeutic challenge. Prophages are increasingly recognized as important genetic factors influencing the pathogenicity of S. aureus, yet their role in antibiotic persistence in MRSA remains underexplored. Our previous work demonstrated that prophage ϕSA169 promotes vancomycin (VAN) persistence in an experimental model of endocarditis caused by MRSA strains with a clonal complex (CC) 45 genetic background. However, it is unknown whether this persistence-promoting eff

Genomic and phenotypic characterization of Klebsiella pneumoniae phage KP Ø1: a novel lytic Slopekvirus targeting uropathogenic multidrug-resistant Klebsiella pneumoniae.

The rise of multidrug-resistant (MDR) uropathogenic gram-negative bacteria (GNB) necessitates the development of alternative therapeutic strategies. This study aimed to isolate, phenotypically characterize, and perform whole-genome sequencing of the bacteriophage demonstrating the broadest host range against MDR uropathogens. Fifty MDR GNB isolates were screened for lytic phages. The most promising candidate, Klebsiella pneumoniae phage KP Ø1, was characterized using plaque assay, Transmission Electron Microscopy (TEM), and pH/thermal stability testing. Genomic characterization was performed v

Temperate phages limit their own propagation under spatial constraint in biofilms on chitin.

Biofilm growth and phage exposure are common to diverse bacterial species. Studying phage-host interaction and population dynamics in biofilms with cellular resolution remains a significant challenge, especially when attempting to recapitulate the natural environments that microbes occupy. Here, we study the population dynamics of phage K139 lysogenized and nonlysogenized Vibrio cholerae when growing in biofilms on the surface of chitin particles in seawater, replicating key features of V. cholerae ecology in the marine environment. We find that lysogenized V. cholerae, via spontaneous lytic i

Genetic Engineering of Lysogenic-Lytic Switch Genes Improves Burkholderia Phage Killing Efficacy.

Burkholderia pseudomallei, the causative agent of melioidosis, presents significant challenges in both treatment and environmental decontamination. Bacteriophages, or phages, are increasingly being explored as potential diagnostic, therapeutic, and biocontrol agents against this bacterial pathogen. Our recent investigation has shown that most B. pseudomallei genomes contained prophage(s) associated with specific tRNA gene loci, prompting us to explore these detectable prophages as sources of temperate phages for further applications. The Role of hlb-Converting Bacteriophages in Staphylococcus aureus Host Adaption.

As an opportunistic pathogen of humans and animals, Staphylococcus aureus asymptomatically colonizes the nasal cavity but is also a leading cause of life-threatening acute and chronic infections. The evolution of S. aureus resulting from short- and long-term adaptation to diverse hosts is tightly associated with mobile genetic elements. S. aureus strains can carry up to four temperate phages, many of which possess accessory genes encoding staphylococcal virulence factors. More than 90% of human nasal isolates of S. aureus have been shown to carry Sa3int phages, whereas invasive S. aureus isola

Characterization of the Complete Genome of Arthrobacter globiformis Cluster FF Bacteriophage QuinnAvery.

Phage QuinnAvery produces turbid plaques in Arthrobacter globiformis B-2979 lawns, displays a siphovirus morphology, and is clustered with FF actinobacteriophages. The 43,177 bp genome of QuinnAvery is predicted to contain a tRNA-Arg(TCT) gene and 69 protein-coding genes. Homologs of a subset of QuinnAvery's predicted genes have putative roles in viral structure, particle assembly, host cell lysis, and lysogeny. QuinnAvery's genome is predicted to contain two tyrosine integrases, similar to many other cluster FF phages. However, the area immediately surrounding the integration cassette in Quin
