Receptor-mediated endocytosis of fibroblast beta-glucuronidase by peritoneal macrophages.

beta-Glucuronidase secreted by mouse 3T3 fibroblasts in vitro was taken up into mouse peritoneal macrophages and into human fibroblasts by a process which was rapid and saturable. High concentrations of mannose-containing compounds inhibited uptake into macrophages but had no effect on uptake into fibroblasts. Mannose-6-phosphate inhibited uptake into both types of cell, reducing uptake into macrophages by 34% and abolishing uptake into fibroblasts completely at a concentration of 5 mM. Fructose-1-phosphate was almost equally as effective at inhibiting uptake into fibroblasts but had no effect

Measurement of 5'-nucleotidase in mouse peritoneal macrophages in listeriosis.

5'-Nucleotidase activity in peritoneal macrophages of mice was measured both before and after infection with Listeria monocytogenes in hosts which possessed high or low anti-listerial resistance either due to a genetically determined trait or as a result of splenectomy. Reduction in enzyme activity was directly related to the degree of infection that developed in the hosts and hence was inversely related to the level of anti-listerial resistance observed. Basal 5'-nucleotidase activity was significantly lower in noninfected hosts that possessed high anti-listerial resistance, possibly reflecti

Musculin enhances the immune function of peritoneal macrophages in mice.

INTRODUCTION: Macrophages play a key role in infection, and musculin (MSC) is closely associated with immune cells. The objective of this study was to investigate the effect of MSC on the immune function of mouse peritoneal macrophages. MATERIAL AND METHODS: Peritoneal macrophages from wild-type (WT) and MSC knockout (MSC-/-) mice were obtained and cultured for 12 hours. The cells were subsequently treated with lipopolysaccharide (LPS). After 24 hours, the peritoneal macrophages and their supernatants were collected. The changes in the adhesion index in each group were then recorded by cell c

Radiation-induced apoptosis in peritoneal resident macrophages of C3H mice.

Gamma ray-radiation induced significant apoptosis in peritoneal resident macrophages (PRMs) of C3H/HeJ (C3H) mice, but not in other strains of mice. To investigate the role of DNA damage in the apoptosis, DNA damage was quantified in PRMs by use of the alkaline single-cell gel electrophoresis (Comet) assay. No significant difference was found between C3H and C57Black/6 mice in either radiation-induced DNA damage or repair. Radiation induced apoptosis at the same levels in PRMs of p53 knockout mice and atm knockout mice as those of wild-type C3H mice; however radiation-induced apoptosis was sig

In vitro multiplication of Cryptosporidium parvum in mouse peritoneal macrophages.

Cryptosporidium parvum of bovine origin was developed in vitro in unsensitized mouse peritoneal macrophages. Macrophages growing in RPMI medium were infected with sporozoites or with oocysts, and after staining infections were studied by light microscopy. A high parasitic index was obtained with multiple infections occurring commonly. This is a simple method for the study of Cryptosporidium biology, and for in vitro assays of pharmacological activity.

Mechanisms of killing of Toxoplasma gondii by rat peritoneal macrophages.

Rats are resistant to Toxoplasma infection, and macrophages are thought to mediate this resistance. We performed a series of experiments to investigate the mechanism of the anti-Toxoplasma activity of resident rat peritoneal macrophages. Resident rat peritoneal macrophages killed more than 90% of ingested Toxoplasma gondii in vitro. This capacity was reduced progressively with the prolongation of culturing of macrophages in vitro before challenge with T. gondii. Exhaustion of the respiratory burst of macrophages with phorbol myristate acetate impaired their ability to kill and limit the replic

Infection of cultivated mouse peritoneal macrophages with lymphocytic choriomeningitis virus.

Lymphocytic choriomeningitis virus strain WE multiplied in cultivated peritoneal macrophages from unstimulated NMRI mice. As revealed by immunofluorescence technique, most cells participated in the infectious process. They were not, however, functionally altered. Morphologically, infected cells contained more cytoplasmic vacuoles, but these were also present, though less numerous, in uninfected ageing cells and in cells having phagocytosed Latex beads. As compared with the WE strain virus, the E-350 (Armstrong) strain multiplied more slowly to lower titres, but the proportion of infected cells

Peritoneal mast cell-derived IL-10 promotes peritoneal-colon tolerance by reprogramming macrophages to ameliorate colitis.

Maintaining peripheral tolerance is critical for mucosal homeostasis. The peritoneal cavity (PerC), a gut-adjacent immune niche exposed to microbial products, contains mast cells (MCs) whose regulatory role in colitis is unclear. Here we show that IL-10 derived from MCs restrains colitis by programming anti-inflammatory macrophages along the PerC-colon axis. Reconstitution of MC-deficient mice with wild-type (WT) bone marrow-derived mast cells (BMMCs), but not Il10-/- BMMCs, alleviated colitis. Mechanistically, WT MCs reduced TNF-α+ macrophages and neutrophil infiltration while increasing IL-1

The peritoneal immune niche in peritoneal dialysis-induced membrane failure: macrophages, neutrophils and extracellular trap biology.

Peritoneal dialysis (PD) is associated with progressive remodeling of the peritoneal membrane, with fibrosis, vasculopathy, and transport deterioration that culminate in ultrafiltration failure (UFF); a smaller subset of patients progresses to encapsulating peritoneal sclerosis (EPS). Myeloid-driven sterile inflammation is a central feature of this process, yet the evidence base differs sharply across the immune actors implicated. In this Perspective, we evaluate that evidence through a three-layer framework separating PD-direct data from non-PD peritoneal injury data and other-organ analogies

Development of peritoneal adhesions in macrophage depleted mice.

BACKGROUND: We present a new mouse model for the study of peritoneal adhesions using macrophage Fas-induced apoptosis (Mafia) transgenic mice expressing a Fas-FKBP construct under control of the murine c-fms promoter. Mafia mice allow systemic macrophage depletion by dimerization of Fas with a synthetic dimerizer, AP20187. Results demonstrate that macrophage depletion in Mafia mice induces peritoneal adhesion formation when the peritoneal cavity is also exposed to an irritant. The Mafia mouse model presents a reproducible, non-surgical approach for research in adhesion formation and prevention

MSCs promote the efferocytosis of large peritoneal macrophages to eliminate ferroptotic monocytes/macrophages in the injured endometria.

BACKGROUND: Endometria are one of the important components of the uterus, which is located in the peritoneal cavity. Endometrial injury usually leads to intrauterine adhesions (IUA), accompanied by inflammation and cell death. We previously reported that both the endometrial ferroptosis was increased and monocytes/macrophages were involved in endometrial injury of IUA. Large peritoneal macrophages (LPMs) are recently reported to migrate into the injured tissues and phagocytose dead cells to repair the tissues. We previously demonstrated that mesenchymal stromal cells (MSCs) had made excellent

The involvement of peritoneal GATA6+ macrophages in the pathogenesis of endometriosis.

Endometriosis is a chronic inflammatory disease that causes debilitating pelvic pain in women. Macrophages are considered to be key players in promoting disease progression, as abundant macrophages are present in ectopic lesions and elevated in the peritoneum. In the present study, we examined the role of GATA6+ peritoneal macrophages on endometriosis-associated hyperalgesia using mice with a specific myeloid deficiency of GATA6. Lesion induction induced the disappearance of TIM4hi MHCIIlo residential macrophages and the influx of increased Ly6C+ monocytes and TIM4lo MHCIIhi macrophages. The r

CD4-Positive T Cells and M2 Macrophages Dominate the Peritoneal Infiltrate of Patients with Encapsulating Peritoneal Sclerosis.

BACKGROUND: Encapsulating peritoneal sclerosis (EPS) is a severe complication of peritoneal dialysis (PD). Previously, it has been shown that infiltrating CD4-positive T cells and M2 macrophages are associated with several fibrotic conditions. Therefore, the characteristics of the peritoneal cell infiltrate in EPS may be of interest to understand EPS pathogenesis. In this study, we aim to elucidate the composition of the peritoneal cell infiltrate in EPS patients and relate the findings to clinical outcome. STUDY DESIGN, SETTING, AND PARTICIPANTS: We studied peritoneal membrane biopsies of 23

Inhibition of VEGFR1 TK Signaling in Peritoneal Macrophages Suppresses Endometriosis Development.

BACKGROUND/AIM: Endometriosis is characterized by the accumulation of immune cells in endometrial lesions and the peritoneal cavity. Macrophages contribute to the growth and neovascularization of endometriotic lesions. Vascular endothelial growth factor receptor-1 (VEGFR1) is involved in neovascularization, while peritoneal macrophages (PMs) play a critical role in endometriosis development and establishment. We examined the role of VEGFR1 signaling in PMs during endometriosis development using a murine model of ectopic endometrial transplantation. MATERIALS AND METHODS: Endometrial fragments

Postoperative adhesion formation: the role of peritoneal macrophages and targeting therapy.

BACKGROUND: Postoperative adhesion (PA) is the most common complication of abdominal or pelvic surgery, resulting in various complications, including small bowel obstruction, secondary female infertility, chronic abdominal pain and even death. Unfortunately, there are few effective methods for the prevention and treatment of it. Previous studies confirmed that coagulation response, inflammation, fibrosis and fibrinolysis played a central role in PA formation and a variety of cells, such as monocytes, macrophages, fibroblasts, neutrophils and mesothelial cells participated in this process. Inflammatory macrophages switch to CCL17-expressing phenotype and promote peritoneal fibrosis.

Peritoneal fibrosis remains a problem in kidney failure patients treated with peritoneal dialysis. Severe peritoneal fibrosis with encapsulation or encapsulating peritoneal sclerosis is devastating and life-threatening. Although submesothelial fibroblasts as the major precursor of scar-producing myofibroblasts in animal models and M2 macrophage (Mϕ)-derived chemokines in peritoneal effluents of patients before diagnosis of encapsulating peritoneal sclerosis have been identified, attenuation of peritoneal fibrosis is an unmet medical need partly because the mechanism for cross talk between Mϕs

Macrophages on the Peritoneum are involved in Gastric Cancer Peritoneal Metastasis.

Tumor-associated macrophages (TAM) have been shown to support tumor growth and progression by various mechanisms. However, the roles of TAM in gastric cancer (GC) peritoneal metastasis remain elusive. To explore the roles of macrophages in the process of GC peritoneal metastasis, we performed the present study. Samples from the primary GC tumor beds, surgical margins, peritoneal metastatic lesions and surrounding tissue, and the Pouch of Douglas, were collected, fixed by formalin, and embedded with paraffin. Immunohistochemistry staining for macrophages markers was performed. The peritoneal la

Peritoneal GATA6+ macrophages function as a portal for Staphylococcus aureus dissemination.

Essentially all Staphylococcus aureus (S. aureus) bacteria that gain access to the circulation are plucked out of the bloodstream by the intravascular macrophages of the liver - the Kupffer cells. It is also thought that these bacteria are disseminated via the bloodstream to other organs. Our data show that S. aureus inside Kupffer cells grew and escaped across the mesothelium into the peritoneal cavity and immediately infected GATA-binding factor 6-positive (GATA6+) peritoneal cavity macrophages. These macrophages provided a haven for S. aureus, thereby delaying the neutrophilic response in t

A pathogenetic role for M1 macrophages in peritoneal dialysis-associated fibrosis.

Peritoneal fibrosis (PF) is a frequent complication of peritoneal dialysis (PD) accompanied by the infiltration of inflammatory cells. Recently, the function of macrophages in an inflammatory microenvironment during PD remains unknown. This study aimed to elucidate the role of distinct macrophage phenotypes in the progression of PF through macrophage depletion in a peritoneal dialysis-induced mouse model. After injection of 200 μl liposomal clodronate (LC) at the start of instillation PD fluids (PDFs), mice were injected with 100 μL LC every 4 days after the first time injection for longer mac
