Volatile organic compounds (VOCs) are valuable compounds in diverse microbial interactions with certain biotic stimuli, yet pivotal roles of any VOCs derived by the rice blast fungus, Magnaporthe oryzae, in intra- and interspecies communications have been poorly understood. Therefore, in this study, we investigated that how certain VOCs by M. oryzae affect the fungal growth of the same species and other species. Using a partitioned I-plate assays, the result showed that pre-inoculated fungal colony (sender) significantly suppressed the late arriving colony (receiver) in a time- and mediadepend

Sexual reproduction of the ascomycetous fungus Magnaporthe oryzae is reported to be regulated by illuminance and temperature, but their temporal interaction during perithecial production remains unclear. Here, we performed full-factorial condition transfer assays (illuminance: low/0 lx, medium/1000 lx, or high/5000 lx; temperature: low/19 °C or high/25 °C) using a pair of high-fertility M. oryzae isolates collected from green bristlegrass to characterize sequential and combinatorial effects of illuminance and temperature on perithecial production. We found that perithecial production showed a

BACKGROUND: The rice blast fungus, Magnaporthe oryzae, poses a serious threat to global rice production. Cell cycle and polarized growth are two essential processes for host infection of M. oryzae; however, the association of them remains largely unknown.
RESULTS: MoHym1, a homolog of yeast RAM (regulation of Ace2 and morphogenesis) network component, was identified as a key regulator for fungal development and pathogenicity in M. oryzae. Deletion of MoHYM1 resulted in severe defects in vegetative growth, conidiation, polarized growth, cell wall integrity, surface hydrophobicity, and secretio

Rice blast, caused by the filamentous fungus Magnaporthe oryzae, is a serious disease of rice. In this study, by using homologous recombination, gene knockout mutants of ΔMokmt1 (a histone methyltransferase) and ΔMocul4 (Cullin4 subunit of an E3 ubiquitin ligase) were generated. Phenotypic analysis revealed that both MoKmt1 and MoCul4 are essential for fungal growth, development, and pathogenicity. Compared to the wild-type strain, the ΔMokmt1 mutant exhibited a slower growth rate (11.6% reduction) and significantly decreased virulence. Western blot analysis confirmed a significant decrease of

Blast, caused by the fungus Magnaporthe oryzae, is one of the most devastating diseases of rice worldwide. Phenylalanine ammonia lyase (PAL) is a key enzyme in the phenylpropanoid pathway, which leads to the biosynthesis of defense-related phytohormone salicylic acid (SA) and flavonoid-type phytoalexins sakuranetin and naringenin. However, the roles and biochemical features of individual rice PALs in defense responses to pathogens remain unclear. Here, we report that rice OsPAL06, which can catalyze the formation of trans-cinnamate using l-phenylalanine, is involved in rice root-M. oryzae inte

The effector proteins secreted by a pathogen not only promote virulence and infection of the pathogen, but also trigger plant defense response. Therefore, these proteins could be used as important genetic resources for transgenic improvement of plant disease resistance. Magnaporthe oryzae systemic defense trigger 1 (MoSDT1) is an effector protein. In this study, we compared the agronomic traits and blast disease resistance between wild type (WT) and MoSDT1 overexpressing lines in rice. Under control conditions, MoSDT1 transgenic lines increased the number of tillers without affecting kernel mo

Cystathionine β-synthase (CBS) domain containing proteins (CDCPs) constitute a big family in plants and some members in this family have been implicated in a variety of biological processes, but the precise functions and the underlying mechanism of the majority of this family in plant immunity remain to be elucidated. In the present study, a CBS domain containing protein gene, OsCBSX3, is functionally characterized in rice resistance against Magnaporthe oryzae (M. oryzae). By quantitative real-time PCR, transcripts of OsCBSX3 are up-regulated significantly by inoculation of M. oryzae and the e

Rice blast disease, caused by the fungus Magnaporthe oryzae, poses a severe threat to rice production, particularly in Asia where rice is a staple food. Concerns over fungicide resistance and environmental impact have sparked interest in exploring natural fungicides as potential alternatives. This study aimed to identify highly potent natural fungicides against M. oryzae to combat rice blast disease, using advanced molecular dynamics techniques. Four key proteins (CATALASE PEROXIDASES 2, HYBRID PKS-NRPS SYNTHETASE TAS1, MANGANESE LIPOXYGENASE, and PRE-MRNA-SPLICING FACTOR CEF1) involved in M.

Plant-pathogenic fungi threaten both crop yield and quality. This, together with the increasing resistance of plant-pathogenic fungi to existing fungicides, underscores the need to develop environmentally sound fungicides with novel modes of action. In this study, we systematically evaluated the antifungal activity of plumbagin and its inhibition of dihydroorotase (DHOase), a key enzyme in the de novo pyrimidine biosynthesis pathway of major plant-pathogenic fungi. Our results showed that plumbagin significantly inhibited DHOase activity in Magnaporthe oryzae, Fusarium graminearum and Fusarium

Biological control strategies are increasingly being explored as sustainable alternatives for managing rice blast disease caused by Magnaporthe oryzae. In this study, we characterized three Bacillus pumilus isolates (DC01, DC09, and DC13) and evaluated their antifungal and plant-beneficial properties against M. oryzae. Whole-genome sequencing revealed multiple biosynthetic gene clusters associated with the production of antimicrobial metabolites. All three isolates inhibited fungal growth in dual-culture assays, whereas heat-stable diffusible antifungal activity was primarily associated with t

Pathogen attack can increase plant levels of reactive oxygen species (ROS), which act as signaling molecules to activate plant defense mechanisms. Elucidating these processes is crucial for understanding redox signaling pathways in plant defense responses. Using an iodo-tandem mass tag (TMT)-based quantitative proteomics approach, we mapped 3362 oxidized cysteine sites in 2275 proteins in rice leaves. Oxidized proteins were involved in gene expression, peptide biosynthetic processes, stress responses, ROS metabolic processes, and translation pathways. Magnaporthe oryzae infection led to increa

Identification of infection process and defense response during M. oryzae infecting Acuce. Magnaporthe oryzae is a destructive rice pathogen. Recent studies have focused on the initial infectious stage, with a few studies conducted to elucidate the characteristics of the late infectious stages. This study aims to decipher the characteristics at different stages (biotrophic, biotrophy-necrotrophy switch (BNS), and necrotrophic) between the interaction of two M. oryzae-rice combinations and investigate the resistance mechanisms of rice to M. oryzae using cytological and molecular methods. We examined the frequency of and variation in eight avirulence (AVR) genes from 840 Magnaporthe oryzae field isolates collected from the Southern United States relative to rice resistance (R) genes deployed from 1959 to 2017. Little coding or noncoding variation was detected in AVR-Pi9, AVR-Pib, ACE1 and AVR-Pizt, which correspond to R genes not used in the United States. In the United States, AVR-Pik, corresponding to rice R gene Pi-k alleles deployed at varying levels since the late 1960s, was present in 37% of the isolates over six decades. AVR-Pita1, corresponding to Pi-ta deployed in the

The rice blast fungus Magnaporthe oryzae senses plant surface cues, such as cutin monomers, to initiate appressorium formation. Although cutin monomers are known to trigger the cAMP-protein kinase A signaling pathway, their fungal receptor has remained unknown. Here, we identify the adenylate cyclase Mac1 as a direct receptor for cutin monomers in M. oryzae. A 32-amino acid segment (residues 424 to 455) in Mac1's N terminus is specifically required for its binding to and activation by cutin monomers. Strikingly, Mac1's subcellular localization dictates its functional response to cutin monomers

Nitric oxide (NO) is a key signaling molecule and is known to modulate diverse physiological processes, including defense responses against pathogens. However, the molecular mechanism underlying NO-induced plant immunity remains largely elusive. Here, we investigated the dynamics of NO biosynthesis and its downstream signaling through S-nitrosoproteome analysis of two rice cultivars, Dongjin (DJ, resistant) and Nipponbare (NIP, susceptible), in response to Magnaporthe oryzae, the causative agent of the rice blast disease. M. oryzae inoculation triggered relatively higher nitric oxide synthase

Rice is one of the most important crops worldwide, and it is a model for molecular studies of monocotyledonous species, particularly for understanding the molecular mechanisms of plant disease resistance. Jasmonic acid (JA) is an important plant hormone involved in rice-pathogen interactions. In addition, JA-induced volatiles are known to be involved in the rice defense system regulated by JA signaling. In this study, we isolated a JA-induced terpene synthase from rice, and found that it produces two sesquiterpenes; β-elemene and β-bisabolene. Furthermore, β-elemene exhibited significant antif

Phytopathogenic fungi secrete effector proteins to promote virulence. The MAX (Magnaporthe Avrs and ToxB-like) effectors form a structurally conserved family despite significant sequence diversity. AVR-Pia, a MAX effector from the rice blast fungus Magnaporthe oryzae, is recognised by the model rice nucleotide-binding leucine-rich repeat (NLR) receptor pair OsRGA4/OsRGA5 via direct binding to a heavy metal-associated (HMA) integrated domain (ID) in OsRGA5. While the structural basis of AVR-Pia recognition is well defined, the role of this effector in promoting virulence has remained elusive. H

Rice blast disease caused by Magnaporthe oryzae poses a severe threat to rice production. To counteract M. oryzae, plants synthesize jasmonate (JA) and lignin, two primary defense-related metabolites, to initiate defense programs. However, the mechanism through which M. oryzae modulates JA- and lignin-mediated plant immunity remains unclear. In this study, a novel M. oryzae effector, MoBys1, was identified as being involved in pathogenesis. Knockout of MoBys1 in M. oryzae significantly reduced its infection ability. Conversely, overexpression of MoBys1 in rice impaired the rice defense respons
