Astrovirus MLB2, a New Gastroenteric Virus Associated with Meningitis and Disseminated Infection.

Next-generation sequencing has identified novel astroviruses for which a pathogenic role is not clearly defined. We identified astrovirus MLB2 infection in an immunocompetent case-patient and an immunocompromised patient who experienced diverse clinical manifestations, notably, meningitis and disseminated infection. The initial case-patient was identified by next-generation sequencing, which revealed astrovirus MLB2 RNA in cerebrospinal fluid, plasma, urine, and anal swab specimens. We then used specific real-time reverse transcription PCR to screen 943 fecal and 424 cerebrospinal fluid sample

Localization of astrovirus in experimentally infected turkeys as determined by in situ hybridization.

Twenty-one 3-day-old turkey poults from British United Turkeys of America were orally inoculated with a recently characterized astrovirus, TAstV-2, isolated from turkeys with poult enteritis and mortality syndrome. At 1, 2, 3, 4, 5, 7, and 9 days postinfection (dpi), three inoculated birds were euthanatized, and tissues (intestines, spleen, bursa, and thymus) were collected immediately into 10% neutral buffered formalin. Inoculated birds were diarrheic by 3 dpi, and frothy feces persisted throughout the experimental period. Histologically, there was only slight evidence of enteric damage, whic

[Viral diarrheas].

It is now well known that several viruses are responsible for acute diarrhoea or gastroenteritis in both children and adults. These viruses are difficult to identify since most of them cannot be isolated by stool cultures on cells. The reality of proven reinfection by some of these organisms is not always clearly understood, even though the existence of several serotypes in the same group (notably rotavirus) can be blamed, and this explains why vaccines are difficult to develop.

Quito's virome: Metagenomic analysis of viral diversity in urban streams of Ecuador's capital city.

In Quito, the microbiological contamination of surface water represents a public health problem, mainly due to the lack of sewage treatment from urban wastewater. Contaminated water contributes to the transmission of many enteric pathogens through direct consumption, agricultural and recreational use. Among the different pathogens present in urban discharges, viruses play an important role on disease, being causes of gastroenteritis, hepatitis, meningitis, respiratory infections, among others. This study analyzes the presence of viruses in highly impacted surface waters of urban rivers using n

Genetic characterization and isolation of feline astrovirus in domestic cats in Guangxi, China.

Feline astrovirus (FAstV), as an intestinal pathogen, has been widely detected worldwide, but little is known about its genetic evolutionary diversity and isolation. This study presents the comprehensive molecular epidemiological investigation of FAstV in Guangxi, China. Screening 775 feline intestinal samples revealed an FAstV prevalence of 4.25 % (33/775), with 39.4 % (13/33) of positive cases co-infected with six other major feline enteric viruses. Notably, FAstV-feline panleukopenia virus (FPV) co-infection emerged as the predominant viral combination (27.27 %, 9/33), suggesting potential

Identification of astrovirus serotypes from children treated at the Hospitals for Sick Children, London 1981-93.

An enzyme immunoassay (EIA) for astrovirus type 1 together with immune electronmicroscopy (IEM) was used to type a collection of 162 astroviruses obtained from 1981-93 from children with diarrhoea. The EIA was found to be specific for astrovirus type 1. Astrovirus types 2-4 were typed by IEM. Astrovirus type 1 was the prevalent serotype 107/125 (86%), followed by type 3 (8%), type 4 (6%) and type 2 (1%). Six samples containing astrovirus could not be typed or detected by EIA because they were coated with coproantibodies; 11 others were not identified. Virus particles could no longer be detecte

Genomic characterization of a novel human astrovirus MLB1 variant from Tanzania.

Human astroviruses (HAstV) are emerging viral pathogens responsible for gastroenteritis worldwide, but whole-genome data from Africa, including from Tanzania, remains scarce. A genomic analysis of stools of children with diarrhea was performed using nanopore sequencing. We successfully assembled HAstV-MLB1 complete genomes from two different stool samples, achieving high sequence coverage of 354.75x and 769.94x, respectively. Analyses included phylogeny, synteny, comparative genomics, and structural characterization of the capsid protein. The two Tanzanian HAstV-MLB1 strains showed high geneti

Exploratory metatranscriptomic survey of bat-associated RNA viruses in Quzhou city, China.

Bats are recognized as important reservoirs of diverse RNA viruses, yet the bat virome in Quzhou City, Zhejiang Province, remains poorly characterized. Here, we conducted an exploratory metatranscriptomic survey of bat-associated RNA viruses in this region, with particular attention to viral families that include known zoonotic members. Bats were sampled between August and October 2023 from one roost in each of six administrative areas of Quzhou City. Species identification was performed by Cytb sequencing, and pooled intestinal, lung, and other visceral tissues were subjected to high-throughp

Histopathologic and Genomic Characterization of a Novel Caprine Astrovirus Identified in a Boer Goat Kid in Illinois, United States.

Astroviruses are non-enveloped, positive-sense single-stranded RNA viruses known to infect various mammals and birds, including humans, often causing gastrointestinal disorders. In recent years, astroviruses have also been linked to neurological and respiratory diseases across several species, including ruminants, mink, deer, and other mammals. Notably, astrovirus infections in goats have been documented in countries such as Switzerland and China, where novel genotypes have been identified in fecal samples. However, their role in the context of disease remains unclear, and reports focusing sol

Gut tissue-resident memory T cells in coeliac disease.

This mini-review describes observations of the 1990ies with culturing of gluten-specific and astrovirus-specific CD4+ T cells from duodenal biopsies from subjects who presumably had a long time between the exposure to gluten or astrovirus antigens and the sampling of the biopsy. In these studies, it was also observed that antigen-specific CD4+ T cells migrated out of the gut biopsies during overnight culture. The findings are suggestive of memory T cells in tissue which are resident, but which also can be mobilised on antigen stimulation. Of note, these findings were made years before the term

Identification of sapovirus GV.2, astrovirus VA3 and novel anelloviruses in serum from patients with acute hepatitis of unknown aetiology.

Hepatitis is a general term meaning inflammation of the liver, which can be caused by a variety of viruses. However, a substantial number of cases remain with unknown aetiology. We analysed the serum of patients with clinical signs of hepatitis using a metagenomics approach to characterize their viral species composition. Four pools of patients with hepatitis without identified aetiological agents were evaluated. Additionally, one pool of patients with hepatitis E (HEV) and pools of healthy volunteers were included as controls. A high diversity of anelloviruses, including novel sequences, was

Application Research on the Triplex RT-ddPCR Assay for the Simultaneous Detection of Human Astrovirus and Sapovirus in Bivalve Shellfish.

Human astrovirus (HAstV) and sapovirus (SaV) are significant pathogens associated with acute gastroenteritis in humans. This study established a triple reverse transcription-droplet digital polymerase chain reaction (PCR) (RT-ddPCR) assay incorporating MS2 phage as a process control virus for the simultaneous quantification of HAstV and SaV. The assay was validated using 240 bivalve samples, comprising five shellfish species: Ostreidae (n = 43), Ruditapes philippinarum (n = 84), Sinonovacula constricta (n = 27), Scapharca subcrenata (n = 23), and Pectinidae (n = 63). The results indicated that

Human astrovirus infection associated with encephalitis in an immunocompetent child: a case report.

BACKGROUND: Until today, classic human astroviruses have not been associated with central nervous system infections in immunocompetent patients. CASE PRESENTATION: A 16-month-old Caucasian girl presented with repetitive generalized seizures with a 4-day history of watery diarrhea, which had already gradually improved. Initially, the prolonged seizures ceased after systemic midazolam treatment and were thought to be fever associated. However, her mental status remained altered, and after seizure recurrence, she was transferred to our pediatric intensive care unit. Seizure control was achieved
