Species-level microbiota of ticks and fleas from Marmota himalayana in the Qinghai-Tibet Plateau.

INTRODUCTION: Ticks and fleas, as blood-sucking arthropods, carry and transmit various zoonotic diseases. In the natural plague foci of China, monitoring of Yersinia pestis has been continuously conducted in Marmota himalayana and other host animals, whereas other pathogens carried by vectors are rarely concerned in the Qinghai-Tibet Plateau. METHODS: In this study, we investigated the microbiota of ticks and fleas sampling from M. himalayana in the Qinghai-Tibet Plateau, China by metataxonomics combined with metagenomic methods. RESULTS: By metataxonomic approach based on full-length 16S rD

The thyroid gland of the woodchuck, Marmota monax: a morphological study of seasonal variations in the follicular cells.

The morphology of the thyroid gland of the woodchuck, Marmota monax, was studied during the four seasons of the year. In the spring the thyroid is extremely heterogenous in appearance. Some follicular cells appear quite active. They contain a well defined Golgi apparatus, abundant large colloid droplets and pseudopodia but few, if any, apical vesicles. Other less active cells have poorly defined rough surfaced endoplasmic reticulum and lack a well developed Golgi apparatus. They do not contain apical vesicles or colloid droplets. Summer thyroids have uniformly small follicles which are lined b

Maternal F1 antibodies and cytokines in mother-neonate dog pairs in the Marmota himalayana plague focus.

In this study, we investigated the F1 antibody against Yersinia pestis in the sera of mother-neonate shepherd dog pairs in the Marmota himalayana plague focus of the Altun-Qilian Mountains, Qinghai-Tibetan Plateau, China. Seropositive shepherd dogs lived in plague-endemic regions, where marmots were infected with Y. pestis, whereas seronegative dogs lived in non-endemic regions. The neonatal F1 antibody titers positively correlated with the maternal titers within 3 months after birth, and the neonatal titers were similar to or slightly lower than the maternal titers. In the absence of reinfect

Escherichia marmotae: a multidrug-resistant opportunistic human pathogen - first clinical isolation in the Czech Republic.

The genus Escherichia comprises seven species, including Escherichia marmotae, described in 2015 from the feces of healthy Himalayan marmots (Marmota himalayana). This species exhibits atypical biochemical properties, such as negative lactose fermentation and indole production. The pathogenic potential of E. marmotae is demonstrated by its ability to invade epithelial cells and the presence of virulence genes. The first human infections with E. marmotae were recorded in Norway in 2021. This article describes the case of a 64-year-old man undergoing chemotherapy for urinary tract cancer, who wa

COMPARISON OF KETAMINE-MIDAZOLAM AND KETAMINE-MIDAZOLAM-BUTORPHANOL PREMEDICATION PRIOR TO SEVOFLURANE ANESTHESIA IN WOODCHUCKS (MARMOTA MONAX).

Cardiovascular disease is a frequent cause of death in the critically endangered Vancouver Island marmots (Marmota vancouverensis). This warrants the use of anesthetic protocols with minimal cardiovascular adverse effects. In this study, 12 adult male woodchucks (Marmota monax) were used as models for Vancouver Island marmots. The objective was to compare the physiological effects of two premedication protocols during induction and maintenance of anesthesia with sevoflurane. The two premedications were ketamine 10 mg/kg and midazolam 0.5 mg/kg (KM) or ketamine 10 mg/kg, midazolam 0.5 mg/kg, an

Hibernation does not reduce cortical bone density, area or second moments of inertia in woodchucks (Marmota monax).

Long periods of inactivity in most mammals result in bone loss that may not be completely recoverable during an individual's lifetime regardless of future activity. Prolonged inactivity is normal during hibernation, but it remains uncertain whether hibernating mammals suffer decreased bone properties after hibernation that affects survival. We test the hypothesis that relative cortical area (C(A) ), apparent density, bone area fraction (B.Ar/T.Ar), and moments of inertia do not differ between museum samples of woodchucks (Marmota monax) collected before and after hibernation. We used periphera

Whole-genome characterization and antibiotic resistance phenotype of Escherichia marmotae first isolated from Berylmys bowersi.

Escherichia marmotae was first described in 2015 as a bacterium isolated from Himalayan marmots, with recent evidence suggesting its potential to cause human diseases. This study presents the first report of E. marmotae isolation from Berylmys bowersi in Guizhou province. We conducted genetic, biochemical, and antibiotic resistance analyses on four isolates, designated as S2-2, S2-4, S2-5, and S2-6. Phylogenetic analysis based on 16S rRNA sequences revealed over 99.5% homology with E. marmotae, while pulsed-field gel electrophoresis and whole-genome sequencing confirmed genetic similarity. Gen

Metagenomics research on the gut microbiota of the Marmota himalayana of the Sanjiangyuan National Nature Reserve in Qinghai Province, China.

With the improvement of transportation and the rise of tourism on the Qinghai-Xizang Plateau, the scope of human activities has continuously expanded, increasing opportunities for contact with wildlife, also exacerbating the outbreak rate of zoonotic emerging infectious diseases. Currently, research on the gut microbiota of wildlife, especially Marmota himalayana (M. himalayana), which are reservoir hosts for plague, is scarce. In this study, we investigated the composition, function, and regional variations of the gut microbiota in M. himalayana based on the metagenomic sequencing of 45 fecal

Pasteurella multocida strains of a novel capsular serotype and lethal to Marmota himalayana on Qinghai-Tibet plateau in China.

Pasteurella multocida is a zoonotic pathogen causing serious diseases in humans and animals. Here, we report P. multocida from wildlife on China's Qinghai-Tibet plateau with a novel capsular serotype, forming a single branch on the core-genome phylogenetic tree: four strains isolated from dead Himalayan marmot (Marmota himalayana) and one genome assembled from metagenomic sequencing of a dead Woolly hare (Lepus oiostolus). Four of the strains were identified as subspecies multocida and one was septica. The mouse model showed that the challenge strain killed mice within 24 h at an infectious do

First Case Report of Human Plague Caused by Excavation, Skinning, and Eating of a Hibernating Marmot (Marmota himalayana).

INTRODUCTION: The Qinghai-Tibet Plateau is considered the most plague-heavy region in China, and skinning and eating marmots (Marmota himalayana) are understood to be the main exposure factors to plague. Yersinia pestis is relatively inactive during marmots' hibernation period. However, this case report shows plague infection risk is not reduced but rather increased during the marmot hibernation period if plague exposure is not brought under control. CASE PRESENTATION: The patient was a 45-year-old man who presented with high fever, swelling of axillary lymph nodes, and existing hand wounds o

Genomic and molecular characterisation of Escherichia marmotae from wild rodents in Qinghai-Tibet plateau as a potential pathogen.

Wildlife is a reservoir of emerging infectious diseases of humans and domestic animals. Marmota himalayana mainly resides 2800-4000 m above sea level in the Qinghai-Tibetan Plateau, and is the primary animal reservoir of plague pathogen Yersinia pestis. Recently we isolated a new species, Escherichia marmotae from the faeces of M. himalayana. In this study we characterised E. marmotae by genomic analysis and in vitro virulence testing to determine its potential as a human pathogen. We sequenced the genomes of the seven E. marmotae strains and found that they contained a plasmid that carried a

Niche modeling predictions of the potential distribution of Marmota himalayana, the host animal of plague in Yushu County of Qinghai.

BACKGROUND: After the earthquake on 14, April 2010 at Yushu in China, a plague epidemic hosted by Himalayan marmot (Marmota himalayana) became a major public health concern during the reconstruction period. A rapid assessment of the distribution of Himalayan marmot in the area was urgent. The aims of this study were to analyze the relationship between environmental factors and the distribution of burrow systems of the marmot and to predict the distribution of marmots. METHODS: Two types of marmot burrows (hibernation and temporary) in Yushu County were investigated from June to September in 2
