Energetic stability states in mast cell activation syndrome: operationalizing reserve, pressure, and threshold collapse.

Mast cell activation syndrome (MCAS) is characterized by pronounced heterogeneity, fluctuating severity, delayed post-perturbation crashes analogous to those documented in post-exertional malaise, and highly variable clinical trajectories that are poorly explained by mediator-centric diagnostic approaches. Patients with similar symptom profiles or laboratory findings often follow divergent courses, while resting biomarkers frequently fail to correlate with functional capacity or prognosis. Beyond serving as markers of activation, mast-cell mediators influence neural, vascular, epithelial, immu

Boxcar Imaging FCS Reveals Membrane Raft Stabilization Kinetics in Antigen-Stimulated Mast Cells.

Antigen (Ag) crosslinking of immunoglobulin E-receptor (IgE-FcεRI) complexes in mast cells and consequent coupling with Lyn tyrosine kinase in the plasma membrane inner leaflet stimulates transmembrane signaling to initiate allergic and inflammatory responses. As established previously, this coupling requires formation of liquid-ordered (Lo)-like regions (aka "rafts") around the nano-clustered receptors to facilitate lipid-based partitioning of Lyn via its membrane anchor, followed by receptor phosphorylation mediated by protein-protein interactions. Imaging fluorescence correlation spectrosco

Magnesium and Zinc Dose-Dependently Stabilize Rat Peritoneal Mast Cells and Enhance the Effects of Adrenaline.

BACKGROUND/AIMS: Magnesium and zinc are vital trace elements found in numerous foods and dietary supplements. In addition to their antioxidant, anticancer, antibacterial, and anti-inflammatory effects, clinical research has suggested that they possess anti-allergic properties. METHODS: Using differential-interference contrast (DIC) microscopy, we examined the effects of magnesium chloride (MgCl2) and zinc chloride (ZnCl2) on rat peritoneal mast cell degranulation. We also examined their effects in conjunction with adrenaline, the first-choice drug for anaphylaxis treatment. RESULTS: Both MgC

Anti-Allergic Drugs Tranilast and Ketotifen Dose-Dependently Exert Mast Cell-Stabilizing Properties.

BACKGROUND: Anti-allergic drugs, such as tranilast and ketotifen, inhibit the release of chemokines from mast cells. However, we know little about their direct effects on the exocytotic process of mast cells. Since exocytosis in mast cells can be monitored electrophysiologically by changes in the whole-cell membrane capacitance (Cm), the absence of such changes by these drugs indicates their mast cell-stabilizing properties. METHODS: Employing the standard patch-clamp whole-cell recording technique in rat peritoneal mast cells, we examined the effects of tranilast and ketotifen on the Cm duri

Resveratrol efficiently improves pulmonary function via stabilizing mast cells in a rat intestinal injury model.

BACKGROUND: Intestinal ischemia/reperfusion (IIR) leads to acute lung injury (ALI) distally by aggravating pulmonary oxidative stress. Resveratrol is effective in attenuating ALI through its antioxidant capacity. This study aimed to determine the effects of resveratrol on IIR-induced ALI and to explore the role of mast cells (MCs) activation in a rat model of IIR. METHODS: Adult Sprague-Dawley rats were subjected to IIR by occluding the superior mesenteric artery for 60min followed by 4-hour reperfusion. Resveratrol was intraperitoneally injected at a dose of 15mg/kg for 5days before IIR. MCs

Pharmaceutical stabilization of mast cells attenuates experimental atherogenesis in low-density lipoprotein receptor-deficient mice.

Mast cells (MCs) contribute to atherogenesis by releasing pro-inflammatory mediators to activate vascular cells and other inflammatory cells. This study examined whether MC activation or stabilization affects diet-induced atherosclerosis in low-density lipoprotein receptor-deficient (Ldlr(-/-)) mice. When Ldlr(-/-) mice consumed an atherogenic diet for 3 or 6 months, MC activation with compound 48/80 (C48/80) increased aortic arch intima and total lesion areas, and plasma total cholesterol, LDL, and triglyceride levels, whereas MC stabilization with cromolyn reduced these parameters. Antisecretory effect of DS-4574, a mast cell stabilizer with peptidoleukotriene antagonism, on gastric acid secretion in the pig.

The antisecretory effect of DS-4574, a mast cell stabilizer with peptidoleukotriene antagonism, on the hypersecretion of gastric acid stimulated by several secretagogues was examined in the pig. Goettingen miniature pigs with chronic gastric fistula were used. Intramuscular injection of carbachol (60 micrograms/kg), tetragastrin (50 micrograms/kg) or histamine (200 micrograms/kg)-induced gastric acid hypersecretion. Intraduodenal administration of DS-4574 (10 and 20 mg/kg) significantly inhibited both the hypersecretion induced by carbachol and that by tetragastrin. On the other hand, DS-4574

Mast cell stabilizer, an anti-allergic drug, reduces ventricular arrhythmia risk via modulation of neuroimmune interaction.

Mast cells (MCs) are important intermediates between the nervous and immune systems. The cardiac autonomic nervous system (CANS) crucially modulates cardiac electrophysiology and arrhythmogenesis, but whether and how MC-CANS neuroimmune interaction influences arrhythmia remain unclear. Our clinical data showed a close relationship between serum levels of MC markers and CANS activity, and then we use mast cell stabilizers (MCSs) to alter this MC-CANS communication. MCSs, which are well-known anti-allergic agents, could reduce the risk of ventricular arrhythmia (VA) after myocardial infarction (

Zirconium-Doped Ceria Oxide Nanozymes Alleviate Allergic Rhinitis by Attenuating Oxidative Stress and Stabilizing Mast Cells.

BACKGROUND: Oxidative stress is fundamental to the pathogenesis and progression of allergic rhinitis (AR) during mast cell (MC) degranulation. However, effective therapeutic strategies that target both oxidative stress and the degranulation process remain limited. We developed ultrasmall mPEG (2000)-PE-modified zirconium-doped cerium oxide nanoparticles (PEG-CZ NPs) with broad-spectrum reactive oxygen/nitrogen species (ROS/RNS) scavenging and intrinsic phosphatase-like activity and investigated their prophylactic efficacy in an ovalbumin (OVA)-induced mouse model of AR. The Interaction of Innate and Adaptive Immunity and Stabilization of Mast Cell Activation in Management of Infusion Related Reactions in Patients with Fabry Disease.

Fabry disease (FD) is an X-linked lysosomal disorder caused by mutations in GLA gene resulting in lack of or faulty α-galactosidase A (α-GalA) enzyme. Enzyme replacement therapy (ERT) with recombinant human α-GalA enzyme (agalsidase) is the standard treatment option for FD. Infusion-related reactions (IRRs), with symptoms ranging from rigors, to fever, pain, vomiting, angioedema and diarrhea, are often seen due to immune response against the exogenous enzyme. To elucidate the mechanisms causing the IRRs in FD, eight patients who developed IRRs were investigated. All, except one, tested negativ

The mast cell stabilizing activity of Chaga mushroom critical for its therapeutic effect on food allergy is derived from inotodiol.

While an anti-allergic effect of Chaga mushroom (Inonotus obliquus) has been indicated, its therapeutic effect on allergy and immunoregulatory mechanisms and chemical constituents directly responsible for that are hardly known. We examined the effect of 70% ethanol extract of Chaga mushroom (EE) and its dichloromethane (DF) and aqueous (AF) fractions using a mouse model of chicken ovalbumin (cOVA)-induced food allergy, and found that only EE and DF ameliorated allergy symptoms to a significant extent. The in vivo mast cell-stabilizing activity was also found only in EE and DF whereas the activ

Effect of DS-4574, a novel peptidoleukotriene antagonist with mast cell stabilizing action, on acute gastric lesions and gastric secretion in rats.

DS-4574 is a peptidoleukotriene antagonist with mast cell stabilizing activity. In the present study, we studied the effects of this compound on gastric secretion and various acute gastric lesions in rats. Intraduodenal administration of DS-4574 at doses of 5 to 10 mg/kg significantly and dose-dependently inhibited gastric acid secretion in pylorus-ligated rats, but a further increase in the dose up to 50 mg/kg did not cause any further inhibition.
