M6P-modified solid lipid nanoparticles loaded with matrine for the treatment of fibrotic liver.

Liver fibrosis is a key pathological process shared by the progression of various chronic liver diseases. Treatment of liver fibrosis can effectively block the occurrence and development of hepatic cirrhosis or even carcinoma. Currently, there is no effective drug delivery vehicle for curing liver fibrosis. In this study, we designed matrine (MT)-loaded mannose 6-phosphate (M6P) modified human serum albumin (HSA) conjugated solid lipid nanoparticles (SLN), named M6P-HSA-MT-SLN for treatment of hepatic fibrosis. We demonstrated that M6P-HSA-MT-SLN exhibited controlled and sustained release prop

Matrine exhibits antiviral activity in a PRRSV/PCV2 co-infected mouse model.

BACKGROUND: PRRSV and PCV2 co-infection is very common in swine industry which results in huge economic losses worldwide. Although vaccination is used to prevent viral diseases, immunosuppression induced by PRRSV and PCV2 leads to vaccine failure. PURPOSE: Our previous results have demonstrated that Matrine possess antiviral activities against PRRSV/PCV2 co-infection in vitro. This study aims to establish a PRRSV/PCV2 co-infected KM mouse model and evaluate the antiviral activities of Matrine against PRRSV/PCV2 co-infection. STUDY DESIGN: A total of 144 KM mice were randomly divided into six

Subcellular Localization of Matrin 3 Containing Mutations Associated with ALS and Distal Myopathy.

BACKGROUND: Mutations in Matrin 3 [MATR3], an RNA- and DNA-binding protein normally localized to the nucleus, have been linked to amyotrophic lateral sclerosis (ALS) and distal myopathies. In the present study, we have used transient transfection of cultured cell lines to examine the impact of different disease-causing mutations on the localization of Matrin 3 within cells. RESULTS: Using CHO and human H4 neuroglioma cell models, we find that ALS/myopathy mutations do not produce profound changes in the localization of the protein. Although we did observe variable levels of Matrin 3 in the cy

Design, synthesis, and biological evaluation of X19: a matrine pyrazole derivatives therapeutic candidate.

A series of novel matrine-pyrazole derivatives were designed and synthesized through structural modification of the natural alkaloid matrine to enhance its antitumor efficacy. Among these derivatives, compound X19 was identified as the most potent candidate through comprehensive in vitro screening, demonstrating superior anti-proliferative activity against human hepatocellular carcinoma Huh-7 cells with an IC50 value of 5.92 μM. Mechanistic investigations revealed that X19 suppresses tumor growth through induction of apoptosis, G0/G1 phase cell cycle arrest, and dose-dependent inhibition of th

Exosome-mimetic vesicles derived from fibroblasts carrying matrine for wound healing.

BACKGROUND: Chronic skin wounds are a leading cause of hospital admissions and reduced life expectancy among older people and individuals with diabetes. Delayed wound healing is often attributed to a series of cellular abnormalities. Matrine, a well-studied component found in Sophora flavescens, is recognized for its anti-inflammatory effects. However, its impact on wound healing still remains uncertain. This study aims to explore the potential of matrine in promoting wound healing. METHODS: In this study, we utilized gradient extrusion to produce fibroblast-derived exosome-mimetic vesicles a

Design, synthesis and biological evaluation of matrine-dithiocarbamate hybrids as potential anticancer agents.

A series of novel matrine-dithiocarbamate (DTC) hybrids were efficiently synthesised from matrine through a three-step sequence involving basic hydrolysis, esterification, and DTC formation. They were evaluated for their in vitro cytotoxic potency on several human cancer and normal cells. All matrine-DTC hybrids were much more toxic against the human hepatoma cell line HepG2 than the parent matrine. Hybrid 4l (IC50 = 31.39 μM) was the most potent compound against HepG2 cells, being 156- and 3-fold more toxic than matrine (IC50 > 4900 μM) and the reference vincristine (VCR, IC50 = 93.67 μM), re

Matrine promotes mitochondrial biosynthesis and reduces oxidative stress in experimental optic neuritis.

Optic neuritis (ON), characterized by inflammation of the optic nerve and apoptosis of retinal ganglion cells (RGCs), is one of the leading causes of blindness in patients. Given that RGC, as an energy-intensive cell, is vulnerable to mitochondrial dysfunction, improving mitochondrial function and reducing oxidative stress could protect these cells. Matrine (MAT), an alkaloid derived from Sophora flavescens, has been shown to regulate immunity and protect neurons in experimental autoimmune encephalomyelitis (EAE), an animal model of multiple sclerosis and ON. However, the protective mechanism

Matrine treatment reduces retinal ganglion cell apoptosis in experimental optic neuritis.

Inflammatory demyelination and axonal injury of the optic nerve are hallmarks of optic neuritis (ON), which often occurs in multiple sclerosis and is a major cause of visual disturbance in young adults. Although a high dose of corticosteroids can promote visual recovery, it cannot prevent permanent neuronal damage. Novel and effective therapies are thus required. Given the recently defined capacity of matrine (MAT), a quinolizidine alkaloid derived from the herb Radix Sophorae flavescens, in immunomodulation and neuroprotection, we tested in this study the effect of matrine on rats with experi

RNA-recognition motif in Matrin-3 mediates neurodegeneration through interaction with hnRNPM.

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is an adult-onset, fatal neurodegenerative disease characterized by progressive loss of upper and lower motor neurons. While pathogenic mutations in the DNA/RNA-binding protein Matrin-3 (MATR3) are linked to ALS and distal myopathy, the molecular mechanisms underlying MATR3-mediated neuromuscular degeneration remain unclear. METHODS: We generated Drosophila lines with transgenic insertion of human MATR3 wildtype, disease-associated variants F115C and S85C, and deletion variants in functional domains, ΔRRM1, ΔRRM2, ΔZNF1 and ΔZNF2. Matrine prevents bone loss in ovariectomized mice by inhibiting RANKL-induced osteoclastogenesis.

Osteoporosis is a metabolic bone disease characterized by decreased bone density and strength due to excessive loss of bone protein and mineral content. The imbalance between osteogenesis by osteoblasts and osteoclastogenesis by osteoclasts contributes to the pathogenesis of postmenopausal osteoporosis. Estrogen withdrawal leads to increased levels of proinflammatory cytokines. Overactivated osteoclasts by inflammation play a vital role in the imbalance. Matrine is an alkaloid found in plants from the Sophora genus with various pharmacological effects, including anti-inflammatory activity. Her

Deep eutectic solvents based on matrine and menthol: A novel and safe transdermal penetration enhancer.

Transdermal drug delivery has attracted extensive attention due to its ability to bypass the first-pass metabolism, maintain plasma drug concentrations, and improve patient compliance. However, the transdermal delivery of hydrophilic drugs is still quite challenging. This study aims to develop a novel deep eutectic solvent (DES) composed of matrine (MT) from sophora flavescens Aiton and menthol (Men) from Mentha canadensis to enhance the transdermal delivery of hydrophilic drugs. Geniposide (GS) was selected as the model drug. Combining multiple experimental techniques and theoretical calculat

Platelet membrane-camouflaged matrine biomimetic nanoparticles for efficient acute lung injury therapy.

Sepsis frequently progresses to acute lung injury (ALI) and acute respiratory distress syndrome (ARDS), which are associated with high mortality. Current treatments face limitations due to resistance and toxicity. Matrine, a component of traditional Chinese medicine, exhibits anti-inflammatory properties but suffers from poor selectivity and potential toxicity. To address these challenges, this study developed platelet membrane-coated matrine nanoparticles (PM@Mat-NPs) for targeted ALI treatment. The PM@Mat-NPs, characterized by a particle size of ∼500 nm, zeta potential of -58 mV, and encapsu

Matrine induces cardiotoxicity by promoting ferroptosis through the Nrf2 antioxidant system in H9c2 cells.

Matrine (MT) has shown promising efficacy in various cancers and chronic hepatitis; however, its clinical application is limited because of its side effects. Our previous studies have indicated that MT can induce severe hepatotoxicity and nephrotoxicity. The current study aimed to investigate its cardiotoxicity and potential underlying mechanisms in H9c2 cells. Our results showed that MT induced H9c2 cell death and disrupted the cellular membrane integrity. Moreover, MT decreased glutathione (GSH) and cysteine (Cys) levels, and increased Fe2+, lipid peroxidation, reactive oxygen species (ROS),

Matrine relieved DHAV-1-induced hepatocyte excessive interferon and pyroptosis by activating mitophagy.

Duck hepatitis A virus type 1 (DHAV-1) is a significant pathogen affecting ducklings, capable of causing rapid mortality and adversely impacting the development of the duck industry. Matrine, the primary active ingredient in various Chinese herbal medicines, has demonstrated antiviral and anti-inflammatory properties. Nevertheless, the effects and mechanisms of action of matrine against DHAV-1 infection remain unclear. This research investigates the effects of matrine on DHAV-1 infection and elucidates the mechanisms involved. We found that matrine mitigated the excessive retinoic acid-inducib
