A Bayesian joint model for mediation analysis with matrix-valued mediators.

Unscheduled treatment interruptions may lead to reduced quality of care in radiation therapy (RT). Identifying the RT prescription dose effects on the outcome of treatment interruptions, mediated through doses distributed into different organs at risk (OARs), can inform future treatment planning. The radiation exposure to OARs can be summarized by a matrix of dose-volume histograms (DVH) for each patient. Although various methods for high-dimensional mediation analysis have been proposed recently, few studies investigated how matrix-valued data can be treated as mediators. In this paper, we pr

Psychological distress associated with COVID-19 quarantine: Latent profile analysis, outcome prediction and mediation analysis.

BACKGROUND: Mental health of the population during COVID-19 quarantine could be at risk. Previous studies in short quarantines, found mood-related and anxiety symptomatology. Here we aimed to characterize the subtypes of psychological distress associated with quarantine, assess its prevalence, explore risk/protective factors, and possible mechanisms. METHODS: Online cross-sectional data (n = 4408) was collected during the Argentine quarantine, between 1st-17th April 2020 along a small replication study (n = 644). Psychological distress clusters were determined using latent profile analysis on

Gut microbiota, lipid metabolism, and PCOS: A Mendelian randomization and mediation analysis.

Polycystic ovary syndrome (PCOS) is a common reproductive and metabolic disorder. Gut microbiota (GM) and lipid metabolism are increasingly implicated in its pathogenesis. We aimed to evaluate their causal roles and the potential mediating effect of lipid metabolites on PCOS. We conducted a 2-sample Mendelian randomization using GWAS summary statistics for 211 GM taxa, lipid metabolites, and PCOS cases from the FinnGen consortium. Instrumental variables were selected at genome-wide significance (P < 5 × 10⁻⁸, linkage disequilibrium r² < 0.001). Causal effects were estimated with inverse-varian

Racial Differences in Adverse Pregnancy Outcomes and Incident Hypertension: A Mediation Analysis.

BACKGROUND: Adverse pregnancy outcomes (APOs) are associated with increased hypertension risk. Black women experience higher rates of APOs and hypertension compared with White women, but whether APOs contribute to Black-White hypertension disparities remains unknown. METHODS: The nuMoM2b-HHS (Nulliparous Pregnancy Outcomes Study: Monitoring Mothers-To-Be-Heart Health Study) prospectively recruited nulliparous pregnant people at 8 US centers (2010-2013) with follow-up 2 to 7 years postpartum (2014-2017). Race, which represents a social construct, was self-identified. The primary outcome was in

Associations between metals and anemia indicator: mixed exposure effects and mediation analysis.

Exposure to metals is an important cause of anemia. The effects of mixed metal exposure on hemoglobin have been less studied, and the existing research conclusions are inconsistent. Body mass index (BMI) is associated with both metal levels and hemoglobin and may play a mediating role in the association between metals and hemoglobin. This study selected 832 children from the National Health and Nutrition Examination Survey database and further analyzed the association between metals and hemoglobin, an indicator of anemia. We found that zinc (Zn), selenium (Se) and thallium (Tl) were associated

Respiratory muscle weakness after stroke: An exploratory mediation analysis of associated factors.

OBJECTIVE: This study had two aims: first, to compare pulmonary function and ultrasound-derived parameters between individuals with stroke and apparently healthy controls; and second, to examine whether selected pulmonary and ultrasound-derived variables mediated the association between stroke status and respiratory muscle strength. METHODS: In this cross-sectional study, 49 individuals with chronic unilateral stroke (≥6 months) and 53 apparently healthy adults were evaluated. B-mode ultrasound assessed diaphragm and transversus abdominis thickness at functional residual capacity and total lu

Knowledge, attitude, and practice toward vitiligo among vitiligo patients: a mediation analysis.

Knowing provoking factors of vitiligo progression and how to avoid them is essential to the successful self-management of vitiligo. Cultivating a positive attitude is also essential to avoid focusing on the negative aspects of the disease and to fuel proactive practice. This study aims to evaluate the knowledge, attitudes, and practices (KAP) of vitiligo patients and identify related factors. The study recruited 517 (54.55% male) adult vitiligo patients in Beijing and Nanjing. The mean knowledge, attitude, and practice scores were 5.45 ± 2.74 (possible range: 0-10), 23.15 ± 6.78 (possible rang

Discrimination, Smoking, and Cardiovascular Disease Risk: A Moderated Mediation Analysis With MESA.

BACKGROUND: Carotid intima-media thickness (cIMT) and carotid plaque are reliable indicators of cardiovascular disease risk, and research highlights that racial and ethnic minority individuals generally exhibit higher cIMT and carotid plaque than White individuals. At present, the mechanisms driving these disparities among different racial and ethnic and biological sex groups are poorly understood. METHODS AND RESULTS: Data came from the baseline examination of MESA (Multi-Ethnic Study of Atherosclerosis). A total of 6814 participants aged 45 to 84 years free of clinical cardiovascular diseas

Aetiology, ejection fraction and mortality in chronic heart failure: a mediation analysis.

OBJECTIVE: Clinical decision making in chronic heart failure (CHF) is based primarily on left ventricular ejection fraction (LVEF), and only secondarily on aetiology of the underlying disease. Our aim was to investigate the mediating role of LVEF in the relationship between aetiology and mortality. METHODS: Using data of 2056 Austrian patients with CHF (mean age 57.2 years; mean follow-up 8.8 years), effects of aetiology on LVEF and overall mortality were estimated using multivariable-adjusted linear and Cox regression models. In causal mediation analyses, we decomposed the total effect of ae

How does childhood maltreatment influence cardiovascular disease? A sequential causal mediation analysis.

BACKGROUND: Childhood maltreatment has been consistently associated with cardiovascular disease (CVD). However, the mechanisms of this relationship are not yet fully understood. We explored the relative contribution of anxiety/depression, smoking, body mass index (BMI) and inflammation (C-reactive protein, CRP) to the association between childhood maltreatment and CVD in men and women aged 40-69 years in the UK. METHODS: We used data from 40 596 men and 59 511 women from UK Biobank. To estimate the indirect effects of childhood maltreatment (physical, sexual and emotional abuse, and emotional

Tobacco control policies and smoking cessation treatment utilization: A moderated mediation analysis.

BACKGROUND: Tobacco policies, including clean indoor air laws and cigarette taxes, increase smoking cessation in part by stimulating the use of cessation treatments. We explored whether the associations between tobacco policies and treatment use varies across sociodemographic groups. METHODS: We used data from 62,165 U.S. adult participants in the 2003 and 2010/11 Tobacco Use Supplement to the Current Population Survey (TUS-CPS) who reported smoking cigarettes during the past-year. We built on prior structural equation models used to quantify the degree to which smoking cessation treatment us

Emerging Adult Risky Sexual Behavior Predicted by Parental Overprotection: Moderated Mediation Analysis.

Parental control behaviors are suggested to be related to emerging adult outcomes, such as risky sexual behavior. Although parental control behaviors are more likely to be considered protective against risky sexual behavior, the relation between parental overprotection and risky sexual behavior is unclear. Furthermore, parental overprotection may influence peer selection and prosocial behaviors in emerging adults, which may in turn influence outcomes of risky sexual behavior, as suggested by primary socialization theory. Thus, the current study investigated the relations among paternal and mat

Centrilobular emphysema and coronary artery calcification: mediation analysis in the SPIROMICS cohort.

BACKGROUND: Chronic obstructive pulmonary disease (COPD) is associated with a two-to-five fold increase in the risk of coronary artery disease independent of shared risk factors. This association is hypothesized to be mediated by systemic inflammation but this link has not been established. METHODS: We included 300 participants enrolled in the SPIROMICS cohort, 75 each of lifetime non-smokers, smokers without airflow obstruction, mild-moderate COPD, and severe-very severe COPD. We quantified emphysema and airway disease on computed tomography, characterized visual emphysema subtypes (centrilo

A Note on formulae for causal mediation analysis in an odds ratiocontext.

In a recent manuscript, VanderWeele and Vansteelandt (American Journal of Epidemiology, 2010,172:1339-1348) (hereafter VWV) build on results due to Judea Pearl on causal mediation analysis and derive simple closed-form expressions for so-called natural direct and indirect effects in an odds ratio context for a binary outcome and a continuous mediator. The expressions obtained by VWV make two key simplifying assumptions: The mediator is normally distributed with constant variance,The binary outcome is rare. Assumption A may not be appropriate in settings where, as can happen in routine epidemio
