Comparison of structural variants in the whole genome sequences of two Medicago truncatula ecotypes: Jemalong A17 and R108.

BACKGROUND: Structural variants (SVs) constitute a large proportion of the genomic variation that results in phenotypic variation in plants. However, they are still a largely unexplored feature in most plant genomes. Here, we present the whole-genome landscape of SVs between two model legume Medicago truncatula ecotypes-Jemalong A17 and R108- that have been extensively used in various legume biology studies. RESULTS: To catalogue SVs, we first resolved the previously published R108 genome assembly (R108 v1.0) to chromosome-scale using 124 × Hi-C data, resulting in a high-quality genome assemb

Medicago truncatula Gaertn. as a model for understanding the mechanism of growth promotion by bacteria from rhizosphere and nodules of alfalfa.

The present study showed all the 16 strains isolated and identified from the alfalfa rhizosphere and nodules, and registered in GenBank, to be good candidates for targeted use in studies addressing the rather weak known mechanism of plant growth promotion, including that of Medicago truncatula, a molecular crop model. Based on physiological, biochemical and molecular analysis, the 16 isolates obtained were ascribed to the following five families: Bacillaceae, Rhizobiaceae, Xantomonadaceae, Enterobacteriaceae and Pseudomonadaceae, within which 9 genera and 16 species were identified. All these

NIN-like protein 2 regulation of nitrite reductase mediates vacuole integrity in Medicago truncatula nodules under high nitrate.

The NIN-like protein 2 (NLP2) transcription factor is highly expressed in the infected cells of the N2 fixation zone of mature nodules in Medicago truncatula. In these cells, NLP2 directly activates the expression of leghemoglobin genes and is required for the full expression of nitrite reductase (NiR). Histological examination of nodules formed under different nitrate regimes revealed that the infected cells of nlp2 display abnormal vacuole morphology under high nitrate (5 mm KNO3). This phenotype is associated with starch accumulation, higher expression of starch biosynthesis genes, and incr

Negative regulation of seed germination in Medicago truncatula by the lncRNA MtCIR1 under salt stress is associated with alternative splicing.

Seed germination, an initial stage of the plant life cycle, is sensitive to salt stress. Phytohormones such as abscisic acid (ABA) and gibberellins (GA) are involved in the suppression of seed germination by salt stress. Despite identification of numerous salt stress-responsive long noncoding RNAs (lncRNAs) in different vascular plants, the mechanisms by which lncRNAs regulate seed germination under salt stress at transcriptional and post-transcriptional levels are being deciphered. In the present study, we identified an intergenic lncRNA from Medicago truncatula, referred to as MtCIR1, and fu

The MYB transcription factor Emission of Methyl Anthranilate 1 stimulates emission of methyl anthranilate from Medicago truncatula hairy roots.

Plants respond to herbivore or pathogen attacks by activating specific defense programs that include the production of bioactive specialized metabolites to eliminate or deter the attackers. Volatiles play an important role in the interaction of a plant with its environment. Through transcript profiling of jasmonate-elicited Medicago truncatula cells, we identified Emission of Methyl Anthranilate (EMA) 1, a MYB transcription factor that is involved in the emission of the volatile compound methyl anthranilate when expressed in M. truncatula hairy roots, giving them a fruity scent. Aphid infestation differently affects the defences of nitrate-fed and nitrogen-fixing Medicago truncatula and alters symbiotic nitrogen fixation.

Legumes can meet their nitrogen requirements through root nodule symbiosis, which could also trigger plant systemic resistance against pests. The pea aphid Acyrthosiphon pisum, a legume pest, can harbour different facultative symbionts (FS) influencing various traits of their hosts. It is therefore worth determining if and how the symbionts of the plant and the aphid modulate their interaction. We used different pea aphid lines without FS or with a single one (Hamiltonella defensa, Regiella insecticola, Serratia symbiotica) to infest Medicago truncatula plants inoculated with Sinorhizobium mel

Proteomics analysis of Medicago truncatula response to infection by the phytopathogenic bacterium Ralstonia solanacearum points to jasmonate and salicylate defence pathways.

The infection of the model legume Medicago truncatula with Ralstonia solanacearum GMI1000 gives rise to bacterial wilt disease via colonisation of roots. The root and leaf responses to early infection (1 and 3 days post infection) were characterised to investigate the molecular mechanisms of plant resistance or susceptibility. A proteomics approach based on pools of susceptible and resistant recombinant inbred lines was used to specifically target the mechanisms for tolerance. Differential abundances were evidenced for proteins involved in defence (e.g., PR5, PR10, or Kunitz protease inhibitor

Rhizophagus irregularis modulates neighbour-primed defences in Medicago truncatula against foliar pathogens via common mycelial network-mediated inter-plant signals.

Arbuscular mycorrhizal fungi (AMF) are ubiquitous root symbionts that form common mycelial networks (CMN), linking multiple plants underground. CMN are thought to contribute to information exchange between plants and to neighbour-primed defences, but the transcriptomic and metabolomic responses of receiver plants connected through CMN remain poorly understood. In addition, the confounding effects of hyphal damage caused by CMN disconnection have not been clearly resolved. To assess the contribution of CMN integrity to neighbour-primed plant defences, we used the model AMF Rhizophagus irregular

LncRNA MtCIR2 Negatively Regulates Response of Medicago truncatula Seedlings to Salt Stress by Targeting SOS2-Dependent Na Efflux via H2A Ubiquitination.

Increasing evidence indicates that long non-coding RNAs (lncRNAs) play a regulatory role in plant responses to environmental stress. However, the epigenetic mechanisms behind the regulation of salt stress by lncRNAs remain largely elusive. We previously discovered that the lncRNA MtCIR2 of Medicago trnucatula negatively regulated seed germination under salt stress by altering endogenous ABA and GA concentrations via histone ubiquitination. Here we evaluated the roles of MtCIR2 in regulating response of M. truncatula seedlings to salt stress. We found that over-expression and mutation of MtCIR2

Biochemical and molecular assessment of the superoxide dismutase (SOD) antioxidant enzyme regulation within Medicago truncatula root in response to iron deficiency.

Iron (Fe) deficiency is a major nutritional stress affecting plant growth and metabolism. This study was conducted on three Medicago truncatula genotypes (TN8.20 and A17 as tolerant and T1.11 as a sensible genotype) cultivated in optimal and Fe-deficient conditions. Assessment of Fe deficiency effects was performed on some physiological and biochemical parameters with a particular focus on superoxide dismutase (SOD) activities and genes expression in roots. Our data showed that the sensitive genotype TN1.11 was more affected by Fe starvation compared to A17 and TN8.20. Overall, the relatively

The Compact Root Architecture 2 systemic pathway is required for the repression of cytokinins and miR399 accumulation in Medicago truncatula N-limited plants.

Legume plants can acquire mineral nitrogen (N) either through their roots or via a symbiotic interaction with N-fixing rhizobia bacteria housed in root nodules. To identify shoot-to-root systemic signals acting in Medicago truncatula plants at N deficit or N satiety, plants were grown in a split-root experimental design in which either high or low N was provided to half of the root system, allowing the analysis of systemic pathways independently of any local N response. Among the plant hormone families analyzed, the cytokinin trans-zeatin accumulated in plants at N satiety. Cytokinin applicati

Comparison of systemic and local interactions between the arbuscular mycorrhizal fungus Funneliformis mosseae and the root pathogen Aphanomyces euteiches in Medicago truncatula.

It has been shown in a number of pathosystems that arbuscular mycorrhizal (AM) fungi confer resistance against root pathogens, including in interactions between Medicago truncatula and the root rot-causing oomycete Aphanomyces euteiches. For the current study of these interactions, a split root system was established for plant marker gene analysis in order to study systemic defense responses and to compare them with local interactions in conventional pot cultures. It turned out, however, that split root systems and pot cultures were in different physiological stages. Genes for pathogenesis-rel

Nodulation Induces Systemic Resistance of Medicago truncatula and Pisum sativum Against Erysiphe pisi and Primes for Powdery Mildew-Triggered Salicylic Acid Accumulation.

Plants encounter beneficial and detrimental microorganisms both above- and belowground and the health status of the plant depends on the composition of this pan-microbiome. Beneficial microorganisms contribute to plant nutrition or systemically or locally protect plants against pathogens, thus facilitating adaptation to a variety of environments. Induced systemic resistance, caused by root-associated microbes, manifests as aboveground resistance against necrotrophic pathogens and is mediated by jasmonic acid/ethylene-dependent signaling. By contrast, systemic acquired resistance relies on sali

Bacterial Compound N,N-Dimethylhexadecylamine Modulates Expression of Iron Deficiency and Defense Response Genes in Medicago truncatula Independently of the Jasmonic Acid Pathway.

Plants face a variety of biotic and abiotic stresses including attack by microbial phytopathogens and nutrient deficiencies. Some bacterial volatile organic compounds (VOCs) activate defense and iron-deficiency responses in plants. To establish a relationship between defense and iron deficiency through VOCs, we identified key genes in the defense and iron-deprivation responses of the legume model Medicago truncatula and evaluated the effect of the rhizobacterial VOC N,N-dimethylhexadecylamine (DMHDA) on the gene expression in these pathways by RT-qPCR. DMHDA increased M. truncatula growth 1.5-

Medicago truncatula Molybdate Transporter type 1 (MtMOT1.3) is a plasma membrane molybdenum transporter required for nitrogenase activity in root nodules under molybdenum deficiency.

Molybdenum, as a component of the iron-molybdenum cofactor of nitrogenase, is essential for symbiotic nitrogen fixation. This nutrient has to be provided by the host plant through molybdate transporters. Members of the molybdate transporter family Molybdate Transporter type 1 (MOT1) were identified in the model legume Medicago truncatula and their expression in nodules was determined. Yeast toxicity assays, confocal microscopy, and phenotypical characterization of a Transposable Element from Nicotiana tabacum (Tnt1) insertional mutant line were carried out in the one M. truncatula MOT1 family

The arbuscular mycorrhizal fungus Rhizophagus irregularis MUCL 41833 increases the phosphorus uptake and biomass of Medicago truncatula, a benzo[a]pyrene-tolerant plant species.

The accumulation of phosphorus (P) in plants increases their biomass and resistance/tolerance to organic pollutants. Both characteristics are mandatory for the utilization of plants in phytoremediation. Arbuscular mycorrhizal (AM) fungi improve plant P nutrition, and thus growth. However, only a few studies have focused on the dynamics of inorganic P (Pi) uptake in AM fungal-colonized plants in the presence of organic pollutants. Indeed, most of the results so far were obtained after harvesting the plants, thus by evaluating P concentration and content at a single time point. Here, we investig

Comparative genome-wide analysis of CAD (Cinnamyl Alcohol Dehydrogenase) gene family in Medicago truncatula and Lotus japonicus and their expression profiles in response to various abiotic abiotic stresses.

The CAD (Cinnamyl Alcohol Dehydrogenase) gene family is a key determinant for lignin biosynthesis in plants. In legumes, CAD enzymes are involved in the development of vascular tissues such as xylem and Casparian strip and they contribute to the production of antimicrobial and antifungal compounds. Thereby, it offers defense against pathogens and pests. Despite their biological significance, a comparative genome-wide analysis of the CAD gene family in Medicago truncatula and Lotus japonicus has not been explored. Therefore, we conducted a comparative genome-wide study to investigate the charac

The Medicago genome provides insight into the evolution of rhizobial symbioses.

Legumes (Fabaceae or Leguminosae) are unique among cultivated plants for their ability to carry out endosymbiotic nitrogen fixation with rhizobial bacteria, a process that takes place in a specialized structure known as the nodule. Legumes belong to one of the two main groups of eurosids, the Fabidae, which includes most species capable of endosymbiotic nitrogen fixation. Legumes comprise several evolutionary lineages derived from a common ancestor 60 million years ago (Myr ago). Papilionoids are the largest clade, dating nearly to the origin of legumes and containing most cultivated species.

Cleaning the Medicago Microarray Database to Improve Gene Function Analysis.

Transcriptomics studies have been facilitated by the development of microarray and RNA-Seq technologies, with thousands of expression datasets available for many species. However, the quality of data can be highly variable, making the combined analysis of different datasets difficult and unreliable. Most of the microarray data for Medicago truncatula, the barrel medic, have been stored and made publicly accessible on the web database Medicago truncatula Gene Expression atlas (MtGEA). The aim of this work is to ameliorate the quality of the MtGEA database through a general method based on logic

Inoculation with arbuscular mycorrhizal fungi does not confer tolerance of pasture legumes to per- and poly-fluoroalkyl substance-contaminated soil.

Environmental contamination due to per- and poly-fluoroalkyl substances (PFAS) is a growing concern globally, as they do not easily degrade and their toxicity causes chronic health issues in humans. When present in water and soil, PFAS bioaccumulate in crops and pastures, and are subsequently taken up by livestock and humans. This study examined how arbuscular mycorrhizal (AM) fungi interact with PFAS in soil, and the subsequent effects on the growth of three pasture legume crops (Medicago truncatula, Medicago sativa and Trifolium subterraneum). To do this, we used a sterilised soil spiked wit
