Island-specific evolution of a sex-primed autosome in a sexual planarian.

The sexual strain of the planarian Schmidtea mediterranea, indigenous to Tunisia and several Mediterranean islands, is a hermaphrodite1,2. Here we isolate individual chromosomes and use sequencing, Hi-C3,4 and linkage mapping to assemble a chromosome-scale genome reference. The linkage map reveals an extremely low rate of recombination on chromosome 1. We confirm suppression of recombination on chromosome 1 by genotyping individual sperm cells and oocytes. We show that previously identified genomic regions that maintain heterozygosity even after prolonged inbreeding make up essentially all of

Ephrin Signaling Patterns Sensory Neurons During Tissue Homeostasis in Planarians.

Eph and ephrin genes encode receptor-ligand pairs that mediate contact-dependent cell signaling and are essential for nervous system development. However, less is known about the role of Ephrin signaling during adult tissue homeostasis and regeneration. Here, we investigated the role of Ephrin signaling in neural patterning in the planarian Schmidtea mediterranea . We discovered that RNAi against the Eph receptor EphR1 led to striking ectopic expression of the mechanosensory neuron markers pkd1L-2 and hmcn-1-L, without obvious disruption of the overall architecture of the central nervous syste

Development of a cold-adapted enzymatic cascade system for in situ value-added conversion of milk.

The global prevalence of lactose intolerance presents a major nutritional challenge, severely limiting milk consumption. In this study, a cold-adapted enzymatic cascade was developed for in-situ lactose-to-lactulose conversion. Molecular dynamics-based screening identified a novel cellobiose 2-epimerase (CE), Papo-CE, exhibiting high cold activity and rapid inactivation above 70 °C. Subsequently, semi-rational engineering of a cold-adapted Marinomonas mediterranea D-mannose isomerase (MmMI) was conducted, which resulted in a mutant M1 (MmMI/F242N/G375W/P373T) with 4.2-fold increase in isomeriz

A Schmidtea nova (Benazzi, 1982)-like Population in SW Romania (Platyhelminthes, Tricladida, Dugesiidae), a Morphological Characterization.

This study has begun as exploratory research on freshwater planarians from southwestern Romania for faunal purposes. Specimens were sampled and processed for standard histology (paraffin wax embedding, horizontal and sagittal serial sections, and Haematoxylin-Eosin staining). The histological analysis provided evidence for a population belonging to the genus Schmidtea. The morphological and histological characteristics were compared with those of all other Schmidtea species. The histological slides reveal numerous diagnostic characters for Schmidtea nova: the presence of two nipples on the cou

A bone morphogenetic protein homologue in the parasitic flatworm, Schistosoma mansoni.

Members of the bone morphogenetic protein (BMP) subfamily of cytokines control many aspects of metazoan development including patterning and organogenesis. Despite the recognition that schistosomes possess key components of a BMP signaling pathway, a BMP-like ligand in the parasitic flatworm Schistosoma mansoni remained elusive. Here, we describe the cloning and characterisation of an S. mansoni BMP (SmBMP). SmBMP is most closely related to BMP homologues from the free-living flatworms Schmidtea mediterranea and Dugesia japonica, with 51% and 47% identity at the amino acid level, respectively.

Morphogenic Effect of Exogenous Glucocorticoid Hormones in the Girardia tigrina Planarian (Turbellaria, Tricladida).

We have studied the effect of two glucocorticoid hormones: hydrocortisone and its synthetic analogue methylprednisolone on the regeneration activity of head and tail blastema of the Girardia tigrina planarian. The regeneration activity was studied in head and tail blastema formed after resection by means of lifetime computer morphometry and immunohistochemical labeling of neoblasts. The search for orthologous proteins-glucocorticoid receptors (hydrocortisone) was performed using the SmedGD database of the Schmidtea mediterranea planarian. The results indicate that both hormones influence the r

Independent and parallel lateral transfer of DNA transposons in tetrapod genomes.

In animals, the mode of transmission of transposable elements is generally vertical. However, recent studies have suggested that lateral transfer has occurred repeatedly in several distantly related tetrapod lineages, including mammals. Using transposons extracted from the genome of the lizard Anolis carolinensis as probes, we identified four novel families of hAT transposons that share extremely high similarity with elements in other genomes including several mammalian lineages (primates, chiropters, marsupials), one amphibian and one flatworm, the planarian Schmidtea mediterranea. The discon

A chromosome-level genome of the lethal tapeworm Sparganum proliferum, with candidate genomic safe harbours for functional genetics.

Sparganum proliferum is an enigmatic, highly proliferative cestode that causes fatal sparganosis. We present a 681 Mb chromosome-level genome assembled from Oxford Nanopore, PacBio HiFi, Illumina, and Hi-C data, yielding nine chromosome-length scaffolds consistent with 2n = 18. The assembly exhibits strong intra-chromosomal Hi-C contact signals and a repeat content of 55.8%, dominated by LINEs. We annotated 29 231 protein-coding genes, including 6316 transposable element-associated loci, with BUSCO completeness scores of 91% (eukaryota_odb10) and 73% (metazoa_odb10). Disrupting quorum sensing alters social interactions in Chromobacterium violaceum.

Quorum sensing (QS) is a communication system used by bacteria to coordinate a wide panel of biological functions in a cell density-dependent manner. The Gram-negative Chromobacterium violaceum has previously been shown to use an acyl-homoserine lactone (AHL)-based QS to regulate various behaviors, including the production of proteases, hydrogen cyanide, or antimicrobial compounds such as violacein. By using combined metabolomic and proteomic approaches, we demonstrated that QS modulates the production of antimicrobial and toxic compounds in C. violaceum ATCC 12472. We provided the first evide

A novel pathogenic variant of CFAP221 is a cause of a mild form of primary ciliary dyskinesia.

Primary ciliary dyskinesia (PCD) is a heritable disease caused by the dysfunction of motile cilia, with a highly heterogeneous genetic background. CFAP221 (Cilia and Flagella Associated Protein 221) is one of the genes, whose role in the PCD pathogenesis requires further evidence. Using whole-exome sequencing we found a novel, homozygous protein-truncating variant in CFAP221 in a Polish PCD patient. To better understand the effect of the identified pathogenic variant on motile cilia structure and function, the proband's nasal epithelium was examined using immunofluorescence, high-speed videomi

In Vivo Expression of Chicken Gut Anaerobes Identifies Carbohydrate- or Amino Acid-Utilising, Motile or Type VI Secretion System-Expressing Bacteria.

Complex gut microbiota increases chickens' resistance to enteric pathogens. However, the principles of this phenomenon are not understood in detail. One of the possibilities for how to decipher the role of gut microbiota in chickens' resistance to enteric pathogens is to systematically characterise the gene expression of individual gut microbiota members colonising the chicken caecum. To reach this aim, newly hatched chicks were inoculated with bacterial species whose whole genomic sequence was known. Total protein purified from the chicken caecum was analysed by mass spectrometry, and the obt

The stability of m6A-marked transcripts is linked to cell identity in planarians.

N 6-methyladenosine (m6A) is a prevalent internal modification of eukaryotic mRNA that influences transcript fate, including mRNA stability and cell-type-specific gene expression. However, the mechanisms underlying m6A-mediated regulation remain poorly understood in many systems, including the highly regenerative planarian Schmidtea mediterranea. To address this, we generated a high-confidence atlas of ∼72 200 m6A sites across the planarian transcriptome using multiplexed direct RNA sequencing. The m6A sites follow a DRAYW consensus motif and are highly enriched near stop codons while being la

Re-evaluation of the taxonomic status of taxa recognized within the expanded concept of Sphincteristomum Oshmarin, Mamaev & Parukhin, 1961 and the related Lobatotrema Manter, 1963 (Digenea: Megaperidae).

The taxonomic status of Sphincteristomum Oshmarin, Mamaev & Parukhin, 1961 (sensu lato) and Lobatotrema Manter, 1963 is controversial due to overlap/confusion in distinctive diagnostic characteristics for each genus and morphological/allometric ambiguity among some of their representatives and/or within records of the same species. To address these in-depth, morphological descriptions, molecular characterizations, and species delimitation analyses were conducted using a combination of comparative morphology, molecular phylogeny, multivariate analyses, and host-parasite data. Following a compre

Double Gamers-Can Modified Natural Regulators of Higher Plants Act as Antagonists against Phytopathogens? The Case of Jasmonic Acid Derivatives.

As key players in biotic stress response of plants, jasmonic acid (JA) and its derivatives cover a specific and prominent role in pathogens-mediated signaling and hence are promising candidates for a sustainable management of phytopathogenic fungi. Recently, JA directed antimicrobial effects on plant pathogens has been suggested, supporting the theory of oxylipins as double gamers in plant-pathogen interaction. Based on these premises, six derivatives (dihydrojasmone and cis-jasmone, two thiosemicarbazonic derivatives and their corresponding complexes with copper) have been evaluated against 1

Microscopic overview of crinoid regeneration.

Crinoids are well known for their striking regenerative potential and can rapidly and completely regenerate arms lost following self-induced or traumatic amputation. Thus they provide a valuable experimental model for investigation of the regenerative process from the macroscopic to the molecular level. In these last years we have studied in detail the overall process of arm regeneration in the comatulid Antedon mediterranea. This phenomenon can be described on the whole as a typical blastemal regeneration in which new structures develop from migratory pluripotential, actively proliferating ce

Host plant specificity and multi-level infestation patterns of scale insects in semi-arid forests of North Africa.

Scale insects (Hemiptera: Coccomorpha) are important sap-feeding herbivores that influence plant physiology and contribute to forest ecosystem functioning. Despite their ecological importance, their host specificity and infestation dynamics remain insufficiently documented in Mediterranean semi-arid forests. This study investigated the distribution and ecological specialization of scale insects in Belezma National Park (Algeria), focusing on host tree species, plant communities, and infested plant parts. Significant variations (p = 0.006) in infestation rates were observed among host tree spec

Exploring the Microbial Flora Associated with Penaeidae Shrimp Habitats Along the Northeastern Mediterranean Coast of Egypt.

Shrimp is one of Egypt’s most widely consumed and economically valuable seafoods; however, it is vulnerable to various microorganisms. This study identifies the bacterial and yeast communities associated with wild and farmed Penaeidae shrimp from El-Deeba Triangle (Lake Manzala) along Egypt’s Mediterranean coast. Seasonal samples (2022–2023) of shrimp and surrounding water were collected from three sites in triplicate and processed for microbial isolation using baiting and direct plating on multiple culture media. Isolates were identified based on macro- and microscopic morphology and further

Chromosome-level genome assembly of the nematophagous flatworm Luticola nematophagus: revealing molecular adaptations for predation and its biocontrol potential against nematode diseases.

BACKGROUND: The soil-dwelling Rhabdocoela flatworm, Luticola nematophagus, recently identified as a specialized predator of plant-parasitic nematodes such as Meloidogyne incognita, demonstrates considerable potential as a biocontrol agent for agricultural nematode diseases. This study presents the first chromosome-level genome assembly of L. nematophagus, achieved by integrating PacBio HiFi long-read sequencing with Hi-C chromatin conformation capture. RESULTS: The assembled genome is approximately 899.92 Mb with a scaffold N50 of 231.83 Mb, comprising 50.08% repetitive sequences and 45,225 pr

Melastatin subfamily Transient Receptor Potential channels support spermatogenesis in planarian flatworms.

The Transient Receptor Potential superfamily of proteins (TRPs) form cation channels that are abundant in animal sensory systems. Amongst TRPs, the Melastatin-related subfamily (TRPMs) is composed of members that respond to temperature, pH, sex hormones, and various other stimuli. Some TRPMs exhibit enriched expression in gonads of vertebrate and invertebrate species, but their contributions to germline development remain to be determined. We identified twenty-one potential TRPMs in the planarian flatworm Schmidtea mediterranea and analyzed their anatomical distribution of expression by whole-
