Mimicking nature: Models of lipid membranes created with the Langmuir monolayer technique: A review of lipid membrane composition.

The Langmuir monolayer technique has proven to be an effective method for constructing lipid-based models of cell membranes. Compared to other artificial membrane systems, such as liposomes or supported lipid bilayers, it offers a relatively simple and versatile approach to reconstructing lipid components of biological membranes and systematically modifying their composition. The technique allows precise control over experimental parameters such as surface pressure and temperature, which influence the physical state and organization of lipid monolayers. Lipid monolayer models are widely used t

Role of membrane organization and membrane domains in endocytic lipid trafficking.

Lipid compositions vary greatly among organelles, and specific sorting mechanisms are required to establish and maintain these distinct compositions. In this review, we discuss how the biophysical properties of the membrane bilayer and the chemistry of individual lipid molecules play a role in the intracellular trafficking of the lipids themselves, as well as influencing the trafficking of transmembrane proteins. The large diversity of lipid head groups and acyl chains lead to a variety of weak interactions, such as ionic and hydrogen bonding at the lipid/water interfacial region, hydrophobic

Neuronal sphingolipidoses: Membrane lipids and sphingolipid activator proteins regulate lysosomal sphingolipid catabolism.

Glycosphingolipids and sphingolipids of cellular plasma membranes (PMs) reach luminal intra-lysosomal vesicles (LVs) for degradation mainly by pathways of endocytosis. After a sorting and maturation process (e.g. degradation of sphingomyelin (SM) and secretion of cholesterol), sphingolipids of the LVs are digested by soluble enzymes with the help of activator (lipid binding and transfer) proteins. Inherited defects of lipid-cleaving enzymes and lipid binding and transfer proteins cause manifold and fatal, often neurodegenerative diseases. The review summarizes recent findings on the regulation

Hyperbaric nitrogen and pentobarbital on synaptosomal membrane lipids and free fatty acids.

Nitrogen at high pressures and anesthetics increase lipid monolayer surface pressure and in turn modulates monolayer associated lipolytic enzyme activity that could alter membrane lipids. We tested the hypothesis that nitrogen at pressures of 5 and 10 megapascals (MPa) and pentobarbital induce alterations in synaptosomal membrane phospholipid and free fatty acid (FFA). Rat cortical synaptosomes in Krebs-Henseleit buffer were placed in steel chambers and incubated for four hours at 37 degrees C: at 5 or 10 MPa of O2/balance N2; at one 0.1 MPa on room air, and with 10 mg pentobarbital. Modulation of the activities of membrane enzymes by cereal grain resorcinolic lipids.

Resorcinolic lipids, amphiphilic compounds from cereal grains show strong effects upon the activity of membrane enzymes. The concentrations for 50% inhibition of erythrocyte membrane acetylcholinesterase were in the range of 18-90 microM and were dependent on the length of the aliphatic side chain of the homologue and on the modification of hydroxyl groups in the benzene ring. Sulfonation of OH groups resulted in a drastic decrease of the inhibitory potency. The effect of resorcinolic lipids on the activity of Ca2+(calmodulin)-ATPase was the opposite. Up to concentrations of 50 microM alk(en)y

Oxidative damage to membrane lipids in the thyroid - no differences between sexes.

It has long been observed that thyroid diseases are more prevalent in women than in men. However, there are limited experimental data demonstrating mechanisms by which sex differences in thyroid diseases may occur and exact molecular mechanisms involved are still far from clear. The aim of the study was to evaluate if there are sex differences concerning oxidative damage to membrane lipids in thyroid homogenates in response to Fenton reaction substrates, i.e., Fe2+ and/or H2O2, and, additionally, in response to potentially protective agent, i.e., melatonin. Homogenates of male or female thyroi

Membrane lipids define small extracellular vesicle subtypes secreted by mesenchymal stromal cells.

The therapeutic efficacy of mesenchymal stromal cells (MSCs), multipotent progenitor cells, is attributed to small (50-200 nm) extracellular vesicles (EVs). The presence of a lipid membrane differentiates exosomes and EVs from other macromolecules. Analysis of this lipid membrane revealed three distinct small MSC EV subtypes, each with a differential affinity for cholera toxin B chain (CTB), annexin V (AV), and Shiga toxin B chain (ST) that bind GM1 ganglioside, phosphatidylserine, and globotriaosylceramide, respectively. Similar EV subtypes are also found in biologic fluids and are independen

Lipids regulate the hydrolysis of membrane bound glucosylceramide by lysosomal β-glucocerebrosidase.

Glucosylceramide (GlcCer) is the primary storage lipid in the lysosomes of Gaucher patients and a secondary one in Niemann-Pick disease types A, B, and C. The regulatory roles of lipids on the hydrolysis of membrane bound GlcCer by lysosomal β-glucocerebrosidase (GBA1) was probed using a detergent-free liposomal assay. The degradation rarely occurs at uncharged liposomal surfaces in the absence of saposin (Sap) C. However, anionic lipids stimulate GlcCer hydrolysis at low pH by up to 1,000-fold depending on the nature and position of the negative charges in their head groups while cationic lip

Erythrocyte membrane lipids and serum selenium in post-viral and alcoholic cirrhosis.

Erythrocyte-membrane fatty acid composition and cholesterol content were evaluated along with serum selenium in 33 patients with liver cirrhosis and in 40 normal subjects. Thirteen patients were suffering from post-viral (group V) and 20 from alcoholic (group A) cirrhosis. The aim of the study was to elucidate whether membrane lipid abnormalities in cirrhosis were linked to the aetiology of the disease or whether they were the results of the cirrhotic process itself. The patients presented a significant increase in membrane cholesterol, palmitic acid (C16:0) and saturated fatty acids (SFA), an

The potent effect of mycolactone on lipid membranes.

Mycolactone is a lipid-like endotoxin synthesized by an environmental human pathogen, Mycobacterium ulcerans, the causal agent of Buruli ulcer disease. Mycolactone has pleiotropic effects on fundamental cellular processes (cell adhesion, cell death and inflammation). Various cellular targets of mycolactone have been identified and a literature survey revealed that most of these targets are membrane receptors residing in ordered plasma membrane nanodomains, within which their functionalities can be modulated. We investigated the capacity of mycolactone to interact with membranes, to evaluate it

Fluorescent labeling of membrane lipid rafts.

Lipid rafts are structures within the plasma membrane that have a different lipid composition than the bulk membrane, specifically being enriched in certain lipids. Lipid rafts are believed to have physiologically important functions. Commercially available kits have been developed (Invitrogen/Molecular Probes) for labeling lipid rafts using a cholera toxin subunit that has been fluorescently tagged with one of several Alexa Fluor dyes. As described in this protocol, cells are labeled while in suspension, after which they can be mounted for imaging.

Magnetic micromotors crossing lipid membranes.

Nano/micromotors are self-propelled particles that show enhanced motion upon being triggered by a stimulus. Their use in nanomedicine has been widely explored, with special focus on imaging or drug delivery. However, a thorough understanding of the requirements for more efficient locomotion is still lacking. In this paper, we assembled magnetically propelled motors of different sizes (i.e., 0.5, 1 and 4 μm) and surface chemistries (positive charge or PEGylated) and assessed their motion in the presence of giant unilamellar lipid vesicles (GUVs) of varying compositions (zwitterionic, negatively

NTA-Ni lipids affect membrane phase behavior and fluorescence of lipid-like dyes.

The nitrilotriacetic acid group coordinated with nickel, NTA(Ni), is the standard chelator for purifying fusion proteins containing poly-histidine tags. The development of lipids with NTA(Ni) headgroups has enabled the specific attachment of His-tagged peptides and proteins to model membranes. Recently, these lipids have become essential for the study of surface-mediated protein phase separation and membrane interaction with biomolecular condensates. While this approach is gaining popularity, the intrinsic effects of NTA(Ni) lipids on the properties of the host membrane have been very little s

Relation between low erythrocyte acetylcholinesterase activity and membrane lipids in paroxysmal nocturnal haemoglobinuria.

Acetylcholinesterase of intact erythrocytes, their ghost and salt soluble extracts obtained from patients with paroxysmal nocturnal haemoglobinuria (PNH) does not differ from normal with respect to Km values for acetylthiocholine, and Ki values for phenyltrimethylammonium iodide. However, the enzyme from PNH sources has lower V max values than normal, has different thermal stability from normal, has less distinctive transition temperature in the Arrhenius plots, and is less subject to inhibition by stearic acid. These results and that from comparison of activation of deoxycholate-extracted enz

Study of red cell membrane lipids in glucose-6-phosphate dehydrogenase deficiency anemia.

Red cell membrane lipids and plasma lipids were studied in 20 patients with G.6.P.D. deficiency, during acute hemolysis and 45-60 days later-20 normal controls were similarly investigated. All lipids in G-6-P-D deficient cells were found to be diminished during the attack of hemolysis and to return back with the exception of lecithin to normal after recovery. Plasma cholesterol and phospholipids were markedly diminished during acute hemolysis, but slightly reduced on recovery.

Membrane lipids and sodium pumps of cattle and crocodiles: an experimental test of the membrane pacemaker theory of metabolism.

The influence of membrane lipid composition on the molecular activity of a major membrane protein (the sodium pump) was examined as a test of the membrane pacemaker theory of metabolism. Microsomal membranes from the kidneys of cattle (Bos taurus) and crocodiles (Crocodylus porosus) were found to possess similar sodium pump concentrations, but cattle membranes showed a four- to fivefold higher enzyme (Na(+)-K(+)-ATPase) activity when measured at 37 degrees C. The molecular activity of the sodium pumps (ATP/min) from both species was fully recoverable when delipidated pumps were reconstituted w

Promoting plant protoplast fusion via membrane decoration with cell-penetrating peptide-conjugated lipids.

Protoplast fusion is a powerful plant cell engineering approach that enables the formation of new hybrids and facilitates genetic exchange across species barriers. However, conventional fusion techniques, such as polyethylene glycol (PEG)-induced fusion and electrofusion, often have low efficiency and poor reproducibility. In this study, we used Tat peptide-conjugated PEG-lipids (Tat-PEG-lipids) with different alkyl chain lengths (C9, C12, and C14) to examine the effect of PEG-lipid conjugates on fusion efficiency in rice (Oryza sativa L. cv. Nipponbare) protoplasts. Initially, we used rhodami

Membrane lipids and their degradation compounds control GM2 catabolism at intralysosomal luminal vesicles.

The catabolism of ganglioside GM2 is dependent on three gene products. Mutations in any of these genes result in a different type of GM2 gangliosidosis (Tay-Sachs disease, Sandhoff disease, and the B1 and AB variants of GM2 gangliosidosis), with GM2 as the major lysosomal storage compound. GM2 is also a secondary storage compound in lysosomal storage diseases such as Niemann-Pick disease types A-C, with primary storage of SM in type A and cholesterol in types B and C, respectively. The reconstitution of GM2 catabolism at liposomal surfaces carrying GM2 revealed that incorporating lipids into t
