Engineered Hybrid Nanovesicles Combining Macrophage Membranes and Artificial Lipids for Abdominal Aortic Aneurysm Therapy.

BACKGROUND AND AIMS: Abdominal aortic aneurysm (AAA) is a vascular condition with high mortality for which no pharmacological treatments have been approved. Targeting endothelial dysfunction as a primary disease initiator, the vascular endothelial cell (VEC)- protective compound Senkyunolide I (SEI) demonstrates therapeutic promise through robust antiapoptotic activity. Nevertheless, SEI's clinical translation faces limitations due to systemic toxicity, necessitating development of safer therapeutic alternatives. RESULTS: This study presents an engineered biomimetic nanoplatform (Lipo-MM nano

Design and In Vitro Evaluation of an Artificial Placenta Made From Hollow Fiber Membranes.

For infants born at the border of viability, care practices and morbimortality rates vary widely between centers. Trends show significant improvement, however, with increasing gestational age and weight. For periviable infants, the goal of critical care is to bridge patients to improved outcomes. Current practice involves ventilator therapy, resulting in chronic lung injuries. Research has turned to artificial uterine environments, where infants are submerged in an artificial amniotic fluid bath and provided respiratory assistance via an artificial placenta. We have developed the Preemie-Ox, a

Permeation mechanisms through artificial lipoidal membranes and effects of synthetic surfactants on xenobiotic permeability.

Through the use of permeation/lipophilicity correlations, the mechanisms of permeation of selected test compounds across artificial lipoidal membranes of the polysiloxane type, in the absence and in the presence of a nonionic surfactant (Polysorbate 80), are investigated, in order to design "in vitro" conditions and features suitable for reproducing "in vivo" intestinal absorption tests, as well as to validate some conclusions arising from "in situ" rat gut experiments about the effects of the synthetic surfactants on drug and xenobiotic absorption processes. Comparison of Cellular Monolayers and an Artificial Membrane as Absorptive Membranes in the in vitro Lipolysis-permeation Assay.

Permeation across Caco-2 cells in lipolysis-permeation setups can predict the rank order of in vivo drug exposure obtained with lipid-based formulations (LBFs). However, Caco-2 cells require a long differentiation period and do not capture all characteristics of the human small intestine. We therefore evaluated two in vitro assays with artificial lecithin-in-dodecane (LiDo) membranes and MDCK cells as absorptive membranes in the lipolysis-permeation setup. Fenofibrate-loaded LBFs were used and the results from the two assays compared to literature plasma concentrations in landrace pigs adminis

Effect of bicarbonate buffer on artificial membrane permeation of drugs.

BACKGROUND AND PURPOSE: The pH value of the small intestine is physiologically maintained by bicarbonate buffer (BCB). However, the effect of BCB on the membrane permeation of drugs has not been investigated. The purpose of this study was to investigate the effect of BCB on the passive membrane permeation of drugs. EXPERIMENTAL APPROACH: The μFlux apparatus (pION Inc.) was used for permeability measurements. To avoid a pH change of BCB, a floating lid was newly developed for μFlux. The membrane filter was coated with a 10 % soybean lecithin-decane solution. The flux measurement was performed

Descemet Membrane Endothelial Keratoplasty in Eyes with an Artificial Iris.

The purpose of this study is to report the outcome of Descemet membrane endothelial keratoplasty (DMEK) in three eyes with a complex anterior segment and an artificial iris. A retrospective chart review of three cases was performed and clinically meaningful patient characteristics, clinical events, and therapeutic interventions were outlined. A literature search was performed and the clinical course of the three cases was discussed in the context of the published literature. DMEK in the presence of an artificial iris did not match the clinical results of DMEK in uncomplicated eyes. All three e

An electro-conductive hybrid scaffold as an artificial Bruch's membrane.

Many research groups have investigated the various kinds of scaffolds to mimic the natural Bruch's membrane (BM) and support the retinal pigmented epithelial cells to form an organized cellular monolayer. While using prosthetic BM is identified as a promising treatment of age-related macular degeneration (AMD), a degenerative and progressive retinal disease, the effects of different signals such as electrical and morphological cues on the retinal pigmented epithelial (RPE) cells are still unknown. In this study, a laminated and conductive hydrogel/fiber composite scaffold by adding conductive

Effects of Hollow Fiber Membrane Oscillation on an Artificial Lung.

Gas transfer through hollow fiber membranes (HFMs) can be increased via fiber oscillation. Prior work, however, does not directly translate to present-day, full-scale artificial lungs. This in vitro study characterized the effects of HFM oscillations on oxygenation and hemolysis for a pediatric-sized HFM bundle. Effects of oscillation stroke length (2-10 mm) and frequency (1-25 Hz) on oxygen transfer were measured according to established standards. The normalized index of hemolysis was measured for select conditions. All measurements were performed at a 2.5 L min-1 blood flow rate. A lumped p

Semipermeable polyurethane membrane as an artificial skin for the premature neonate.

A thin and semipermeable polyurethane membrane adherently applied to premature neonates as an artificial skin was investigated as an atraumatic surface barrier sufficient to reduce transepidermal water loss without inhibiting natural infant skin development during the first few days of life. A sample group of 18 neonates (birth weight [mean +/- SEM] 1.39 +/- 0.12 kg, gestation [mean +/- SEM] 31 +/- 1 weeks) received two 3 X 3-cm polyurethane patches adherent over the chest and abdomen. Transepidermal water loss was measured before and after application and after membrane removal. During longit

Biocompatibility in hemodialysis: artificial membrane and human blood interactions.

Hemodialysis, a cornerstone therapy for chronic kidney disease, represented a crucial advance in the evolution of artificial organs. While its success is largely due to its efficiency in removing uremic toxins, an equally important challenge is to uphold the primum non nocere principle by minimizing the harmful effects of membrane-blood interactions. This review examines the complex mechanisms and key interactions underlying membrane biocompatibility, including complement activation, inflammation, and coagulation disturbances, paving the way for their clinical implications. We also summarize r

Optimizing tick artificial membrane feeding for Ixodes scapularis.

Artificial membrane feeding (AMF) is a powerful and versatile technique with a wide range of applications in the study of disease vectors species. Since its first description, AMF has been under constant optimization and standardization for different tick species and life stages. In the USA, Ixodes scapularis is the main vector of tick-borne zoonoses including the pathogens causing Lyme disease in humans and animals. Seeking to improve the overall fitness of I. scapularis adult females fed artificially, here, we have optimized the AMF technique, considerably enhancing attachment rate, engorgem

From Fiber Bundles to Architected Membranes: Triply Periodic Minimal Surface Architectures for Biohybrid Artificial Lungs.

Artificial lung systems rely almost exclusively on hollow fiber membrane (HFM) bundles, where gas exchange is constrained by heterogeneous flow distribution and thrombogenic blood-material interfaces. Here, we introduce an architecture-driven design framework for artificial lungs based on additively manufactured triply periodic minimal surface (TPMS) membranes. In contrast to discrete fiber bundles, TPMS membranes form continuous three-dimensional architectures that simultaneously regulate perfusion pathways, diffusion interfaces, and blood-material interactions. Computational fluid dynamics a

The Effect of Different Surfactants and Polyelectrolytes on Nano-Vesiculation of Artificial and Cellular Membranes.

Nano- and micro-sized vesicular and colloidal structures mediate cell-cell communication. They are important players in the physiology of plants, animals, and humans, and are a subject of increasing interest. We investigated the effect of three surfactants, N-cetylpyridinium chloride (CPC), sodium dodecyl sulfate (SDS), and Triton X-100 (TX100), and two anionic polyelectrolytes, sodium polystyrene sulfonate (NaPSS) and sodium polymethacrylate (NaPMA), on nanoliposomes. In addition, the effect of SDS and TX100 on selected biological membranes (erythrocytes and microalgae) was investigated. The

Does artificial rupturing of membranes in the active phase of labor enhance myometrial electrical activity?

OBJECTIVE: To determine whether artificial rupture of membranes (AROM) during active phase of labor augments uterine contractility using Electrical Uterine Myography (EUM). STUDY DESIGN: A prospective study of 31 women with term singleton pregnancy during active phase of labor. Using a non-invasive EUM technique, electrical uterine activity was recorded in the 30 min preceding AROM and in the immediate 30 min thereafter. Augmentation was defined as >5% increase in EUM index between the basal and post-AROM states, representing the mean EUM increase of the entire cohort. Low basal uterine contr

Carbonic anhydrase immobilized on hollow fiber membranes using glutaraldehyde activated chitosan for artificial lung applications.

Extracorporeal CO2 removal from circulating blood is a promising therapeutic modality for the treatment of acute respiratory failure. The enzyme carbonic anhydrase accelerates CO2 removal within gas exchange devices by locally catalyzing HCO3 (-) into gaseous CO2 within the blood. In this work, we covalently immobilized carbonic anhydrase on the surface of polypropylene hollow fiber membranes using glutaraldehyde activated chitosan tethering to amplify the density of reactive amine functional groups for enzyme immobilization. XPS and a colorimetric amine assay confirmed higher amine densities

Conductance properties of artificial lipidic membranes containing a proteolipid from Electrophorus. Response to cholinergic agents.

Previous studies have shown that a special proteolipid extract from the electric organ of Electrophorus showed high affinity binding for acetylcholine and other cholinergic agents. This proteolipid has now been incorporated into ultrathin lipidic membranes, and the membrane resistance was studied. The resistance decreased from 7.27 +/- 0.82 x 10(5) ohm cm(2) in the control membrane to 1.83 x 10(5) ohm cm(2) with addition of 72 microg/ml proteolipid. The decrease in resistance followed a potential function of order four with the proteolipid concentration in the membrane-forming solution. The pr

Membrane-targeting antimicrobial compounds have differential effects on living and artificial yeast membrane models.

The stability of the plasma membrane is crucial for cell viability and disruptions in membrane stability can significantly impact cell function. Antimicrobial compounds targeting fungal membranes are required as novel alternatives to current resistance-prone fungicides. Six antimicrobials were assessed using the yeast Saccharomyces cerevisiae in living and artificial membrane models to gain insight into their efficacy and mechanistic activity. Antimicrobial-treated yeast cultures were monitored for growth inhibition and cell membrane permeability. Liposomes prepared from yeast polar lipids wer

Membrane Formulation Impacts Vesicle-Based Artificial Cell Functions.

Engineering vesicle-based artificial cells capable of mimicking cellular processes is a central objective in synthetic biology. However, while giant unilamellar vesicles (GUVs) have become a popular artificial cell platform, the impact of fundamental design features remains obscure. Here, we assembled GUVs composed primarily of two phospholipids: POPC, a phosphatidylcholine widely employed in GUV preparation, and DOPC, a phosphatidylcholine with a lower melting temperature and higher fluidity. We found that the phospholipid mixture leads to an increase in average GUV size (compared to POPC onl
