The consolidation theory states that with the passage of time the engram of a recent learning experience grows increasingly resistant to disruption by amnesic treatment. The time required to reach complete resistance ("consolidation time") is a controversial issue; current estimates range from 10(1) to 10(5) seconds. The present study suggests a parsimonious interpretation of the divergence, namely, that weak amnesic treatments fail to block memory consolidation but do slow its rate, so that post-treatment consolidation inflates the retention scores measured 24 hours later and leads to variabl

The formation of memory representations requires the transformation of perceptual information into stable traces via short-term consolidation. In this study, we measured the time-course of forming color-object associations in working memory (WM) and in long-term memory (LTM), and examined whether a single consolidation process supports both memory systems. Across three experiments, we estimated consolidation time-accuracy curves in WM and LTM by varying the free time to consolidate the information from 300 to 8,050 ms in a continuous reproduction task. We found that both WM and LTM showed a co

The inhibitory avoidance (IA) task is a paradigm widely used to investigate the molecular and cellular mechanisms involved in the formation of long-term memory of aversive experiences. In this review, we discuss studies on different brain structures in rats associated with memory consolidation, such as the hippocampus, striatum, and amygdala, as well as some cortical areas, including the insular, cingulate, entorhinal, parietal and prefrontal cortex. These studies have shown that IA training triggers the release of neurotransmitters, hormones, growth factors, etc., that activate intracellular

Cation-independent mannose-6-phosphate receptor, also called insulin-like growth factor two receptor (CIM6P/IGF2R), plays important roles in growth and development, but is also extensively expressed in the mature nervous system, particularly in the hippocampus, where its functions are largely unknown. One of its major ligands, IGF2, is critical for long-term memory formation and strengthening. Using CIM6P/IGF2R inhibition in rats and neuron-specific knockdown in mice, here we show that hippocampal CIM6P/IGF2R is necessary for hippocampus-dependent memory consolidation, but dispensable for lear

We examined whether retrograde amnesia would be more likely for object discriminations learned an hour before hippocampal damage than object discriminations learned days before. Specifically, rats were trained on two object-discrimination problems 72 h before surgery and another discrimination problem and the reversal of one of the previously learned problems 1 h before surgery. Importantly, novel procedures that minimized overtraining on the object discriminations were used to increase the possibility of the lesions causing amnesia. After either receiving sham or neurotoxic-induced hippocampa

Young rats (16-30 days), trained on a one-trial passive avoidance task, were subsequently submitted to the amnesic agent hypothermia 0, 5, 10, 20 or 30 min later, and tested for retention 6 h after training. While hypothermia administered 10 min after training was no longer effective in 30-day rats, it still induced a significant retrograde amnesia in younger animals. In 16- and 18-day rats, administration of hypothermia 20 min after training could still produce a slight but statistically non-significant amnesia. Additionally, exposition to a reinstatement treatment was found to alleviate the

System consolidation transforms temporary hippocampal representation of memory into long-term storage in cortex. The underlying neural substrate, however, remains enigmatic. Here, we tracked the spatiotemporal evolution of hippocampus (HPC)-cortex local field potentials and single-neuron spikes in behaving animals during fear memory formation. During learning, HPC fast gamma exhibited a progressive phase shift relative to prefrontal cortex (PFC) theta oscillations, with gamma power aligning to progressively later phases of the PFC theta cycle. Strikingly, a related phase-shifted coupling patte

STUDY OBJECTIVES: Disrupted nighttime sleep is common in pediatric narcolepsy type 1, yet its cognitive impact is unknown. As N2 sleep spindles are necessary for sleep-dependent memory consolidation, we hypothesized that narcolepsy type 1 impairs memory consolidation via N2 sleep fragmentation and N2 sleep spindle alterations.
METHODS: We trained 28 pediatric narcolepsy type 1 participants and 27 healthy controls (HCs) on a spatial declarative memory task before a nocturnal in-lab polysomnogram and then gave them a cued recall test upon awakening in the morning. We extracted wake and sleep st

It is uncertain whether sleep preferentially consolidates emotional over neutral material. Some studies suggest that sleep enhances emotional memory (i.e., that there are large differences in strength of memory for valenced material compared to neutral material after a sleep-filled interval, but that this difference is smaller after a wake-filled interval). Others find no such effect. We attempted to resolve this uncertainty by conducting a meta-analysis that compared valenced to neutral material after both sleep- and wake-filled delays. Standard search strategies identified 31 studies (contai

We report that a peripheral injection of Escherichia coli produces both anterograde and retrograde amnesia in 24 month old, but not 3 month old rats for memories that depend on the hippocampus, that is, memory of context, contextual fear, and place learning. The anterograde effect was restricted to measures of long-term memory. Short-term memory was not affected, nor did E. coli produce amnesia for auditory-cue fear conditioning. There were no age related effects on memory in vehicle-treated rats. In addition to these age-related cognitive effects of E.coli, we report that it produced a marked

Despite abundant evidence that systemic administration of adrenergic drugs and hormones can produce retrograde memory enhancement, the literature contains no clear demonstration that postlearning systemic administration of adrenergic antagonists produces retrograde amnesia. Here we demonstrate retrograde amnesia for a stressful learning task (a spatial water maze) with systemic administration of the beta-adrenergic antagonist propranolol (5 mg/kg). The amnesic effect of the drug depended on the degree of learning in the subjects: Propranolol caused a robust retrograde amnesia in "good learners

This study provides evidence from intracarotid amobarbital tests (IAT) in patients with epilepsy that complete suppression of the electrophysiological activity of the left language-dominant hemisphere through left IAT does not impair memory for verbal information given 1 min or immediately before the injection of the barbiturate. Although language functions were completely disrupted and patients were unable to encode new information during the left IAT, pre/post memory was as good as in the right IAT. This lack of retrograde amnesia leads to the conclusion that a relatively stable representati

Memory reactivation during hippocampal ripples is critical for sleep-dependent memory consolidation. However, the molecular-to-circuit mechanism enabling sleep-predominant ripple generation remains unresolved. Here, we discovered that a sparse population of GABAergic interneurons in the mouse hippocampus specializes in gating ripple generation and memory consolidation. While all known genetically labeled hippocampal interneurons were highly active during wakefulness and rapid-eye-movement (REM) sleep, genetic targeting of M2 acetylcholine receptor (Chrm2)-expressing neurons isolated a specific

Fear generalization is a symptom of anxiety-related disorders, including acute stress disorder and post-traumatic stress disorder. Using a contextual fear conditioning paradigm, we found that mice exposed to a similar neutral context but not a different neutral context soon after training showed fear generalization immediately after contextual fear memory consolidation (i.e., 6 h after training). This fear generalization was reflected by a change not only in the total amount but also the pattern of freezing between conditioned and generalized contexts. These results provide insight into the fa

Transcranial direct current stimulation (tDCS) is a promising tool for modulation of learning and memory, allowing to transiently change cortical excitability of specific brain regions with physiological and behavioral outcomes. A detailed exploration of factors that can moderate tDCS effects on episodic long-term memory (LTM) is of high interest due to the clinical potential for patients with traumatic or pathological memory deficits and with cognitive impairments. This commentary discusses findings by Marián et al. (2018) recently published in Cortex within a broad context of brain stimulati

Brain-derived neurotrophic factor (BDNF) is a crucial regulator of neuronal survival and neuroplasticity in the central nervous system (CNS). As a result, there has been a growing interest in the role of BDNF in neuropsychiatric disorders associated with neurodegeneration, including depression and dementia. However, until now, BDNF-targeting therapies have yielded disappointing results. BDNF is thought to exert its beneficial effects on synaptic and neuronal plasticity mainly through binding to the tyrosine kinase B (TrkB) receptor. Recently, 7,8-dihydroxyflavone (7,8-DHF) was identified as th

Previous studies in the auditory cortex of Mongolian gerbils on discrimination learning of the direction of frequency-modulated tones (FMs) revealed that long-term memory formation involves activation of the dopaminergic system, activity of the protein kinase mammalian target of rapamycin (mTOR), and protein synthesis. This led to the hypothesis that the dopaminergic system might modulate memory formation via regulation of mTOR, which is implicated in translational control. Here, we report that the D1/D5 dopamine receptor agonist SKF-38393 substantially improved gerbils' FM discrimination lear

The amygdala, specifically its basolateral nucleus (BLA), is a critical site integrating neuromodulatory influences on memory consolidation in other brain areas. Almost 20 years ago, we reported the first direct evidence that BLA activity is required for modulatory interventions in the entorhinal cortex (EC) to affect memory consolidation (Roesler, Roozendaal, and McGaugh, 2002). Since then, significant advances have been made in our understanding of how the EC participates in memory. For example, the characterization of grid cells specialized in processing spatial information in the medial EC

The present study examined mechanisms underlying memory deficits in Parkinson's disease (PD) and their associations with brain structural metrics. Nineteen PD patients and twenty-two matched controls underwent two memory experiments. In Experiment 1 (delayed memory task), subjects were asked to remember an array of colored rectangles with varying memory set sizes (Low-Load (2 items), Low-Load (relevant 2 items) with Distractor (irrelevant 3 items), and High-Load (5 items)). After a 7 s delay period, they reported whether the orientation of any relevant figures had changed (test period). In Exp
