Causal relationship between hydrogen sulfide and osteoarthritis: A Mendelian randomization analysis.

Hydrogen sulfide (H2S) has been shown to alleviate bone and joint inflammation in osteoarthritis (OA). Exploring the connection between H2S and OA could reveal novel therapeutic avenues for treating this condition. This study utilized H2S as the exposure variable and OA as the outcome within a Mendelian randomization framework. Data for genome-wide association studies related to both exposure and outcome were sourced from the IEU OpenGWAS project. To estimate causal relationships, inverse variance weighted method was employed, along with supplementary analytical techniques (MR-Egger, weighted

Causal genes for osteoporosis: Mendelian randomization analysis with multilayer xQTL data.

Osteoporosis is a highly heritable metabolic bone disorder characterized by low bone mass and increased fracture risk. However, the causal genes underlying disease susceptibility remain incompletely understood. In this study, we employed a summary-data-based Mendelian randomization (SMR) framework to identify genes with potential causal effects on osteoporosis by integrating genome-wide association study summary statistics from the FinnGen consortium with multilayer molecular quantitative trait loci (xQTL) data, including expression, splicing, methylation, and protein QTLs across multiple tiss

Constipation and Parkinson disease: A 2-sample bidirectional Mendelian randomization analysis.

Patients with Parkinson disease (PD) have a higher risk of having constipation and vice versa. But so far it is not clear whether there is a causal relationship. Therefore, a 2-sample bidirectional Mendelian randomization study was performed to investigate the potential bidirectional association between constipation and PD. Independent genetic variants strongly associated with constipation were obtained from the FinnGen consortium. Data for PD were collected from the genome-wide association study summary data. We explored the causal relationship between constipation and PD using publicly avail

Vitamin K Nutritional Status and Disease Risk - A Mendelian Randomization Analysis.

Vitamin K is an essential nutrient obtained from dietary sources and commonly included in mainstream dietary supplements, playing a key role in regulating physiological functions and maintaining overall health. However, the causal nature of such associations between vitamin K nutritional status and diseases remains obscure. The existence of causality helps to prove the possibility of vitamin K becoming a biomarker for predicting susceptibility to certain diseases. To investigate this hypothesis, we conducted a literature review to identify diseases previously reported to be associated with vit

Mendelian randomization analysis unveils causal relationships between skin microbiota and osteomyelitis.

BACKGROUND: Osteomyelitis results principally from endo-/exogenous bacterial infections, whose incidence that complicating bone injuries reaches approximately 30%, with the risk of disability and teratogenicity. Skin microbiota has been found to be clinically linked to osteomyelitis, but substantial evidence is lacking. METHODS: Mendelian randomization (MR) was applied to unveil the causality between human skin microbiome and osteomyelitis. Genetic data of 1,656 skin microbiota samples were obtained from genome-wide association studies (GWAS), with the outcome of osteomyelitis from UK Biobank

Mendelian randomization analysis: causal link between hyperthyroidism/thyrotoxicosis and ovarian cancer.

BACKGROUND: The purpose of this study was to use a two-sample Mendelian randomization (MR) approach to explore the causal link between hyperthyroidism/thyrotoxicosis and ovarian cancer (OC). METHODS: By leveraging genome-wide association study (GWAS) data from publicly available databases, we identified single nucleotide polymorphisms (SNPs) representing genetic variants for both hyperthyroidism/thyrotoxicosis and OC. We employed the inverse variance weighted method (IVW) to assess the association between hyperthyroidism/thyrotoxicosis and the risk of OC. Further, we evaluated SNP heterogenei

Inflammatory bowel disease and risk for hemorrhoids: a Mendelian randomization analysis.

Observational studies have reported an association between inflammatory bowel disease (IBD), which includes Crohn's disease (CD) and ulcerative colitis (UC), and hemorrhoids (HEM). However, the presence of a causal relationship within this observed association remains to be confirmed. Consequently, we utilized the Mendelian randomization (MR) method to assess the causal effects of IBD on hemorrhoids. We validated the association between IBD and hemorrhoids in humans based on genome-wide association studies (GWAS) data. To investigate the causal relationship between IBD and hemorrhoids, we perf

Depression and NAFLD risk: A meta-analysis and Mendelian randomization study.

BACKGROUND: Both depression and nonalcoholic fatty liver disease (NAFLD) have a high global prevalence. Growing evidence suggests an association between depression and NAFLD, while the association remains unclear. Thus, in this study, we aimed to explore the effect of depression on the risk of developing NAFLD. METHODS: The meta-analysis examined the association between depression and the risk of NAFLD by including observational studies. Relevant studies were searched in PubMed, Embase, the Cochrane Library, and Web of Science. Then a two-sample Mendelian randomization (MR) analysis was perfo

Association between various cathepsins and uterine leiomyoma: A Mendelian randomization analysis.

Emerging evidence suggests a tentative association between cathepsins and uterine leiomyoma (UL). Previous investigations have predominantly focused on the role of cathepsins in the metastasis and colonization of gynecological malignancies. Still, observational studies may lead to confounding and biases. We employed a bidirectional Mendelian randomization (MR) analysis to elucidate the causative links between various cathepsins and UL. Instrumental variables (IVs) of cathepsins and UL within the European cohort were from extant genome-wide association study datasets. Sensitivity assessments wa

Causal Associations with Arterial Stiffness and Sarcopenia: A Mendelian Randomization Analysis.

Observational studies and clinical trials indicate a link between arterial stiffness (AS) and sarcopenia (SAR), yet the causal relationship between these remains unclear. The study aims to investigate the causal connection from AS to SAR by Mendelian randomization (MR). We analyzed Genome-Wide Association Studies data for AS indicators: pulse wave arterial stiffness index (PWASI) and pulse wave peak-to-peak time (PPT), and SAR indicators: low hand grip strength (LHGS), usual walking pace (UWP), moderate-to-vigorous physical activity levels (MVPA), and walk or cycle unassisted for 10 minutes. Adiposity and cancer: a Mendelian randomization analysis in the UK biobank.

BACKGROUND: Observational and Mendelian randomization (MR) studies link obesity and cancer, but it remains unclear whether these depend upon related metabolic abnormalities. METHODS: We used information from 321,472 participants in the UK biobank, including 30,561 cases of obesity-related cancer. We constructed three genetic instruments reflecting higher adiposity together with either "unfavourable" (82 SNPs), "favourable" (24 SNPs) or "neutral" metabolic profile (25 SNPs). We looked at associations with 14 types of cancer, previously suggested to be associated with obesity. RESULTS: All gen

Causality between noise pollution and Alzheimer disease: A Mendelian randomization analysis.

The role of noise pollution as a risk factor for Alzheimer disease (AD) is unclear, with observational studies yielding conflicting results susceptible to confounding and reverse causality. To clarify this relationship, we performed a 2-sample Mendelian randomization (MR) study using summary statistics from large-scale genome-wide association studies of European populations. Genetically predicted daytime and evening noise exposure was used as an instrumental variable to assess a causal effect on AD risk. The primary analysis was conducted using the inverse-variance weighted method, with weight
