VpSTS29/STS2 enhances fungal tolerance in grapevine through a positive feedback loop.

Accumulation of stilbene phytoalexins stimulates resistance mechanisms against the grapevine fungus Uncinula necator. However, the defensive mechanisms triggered by stilbene synthase (STS) genes, remain largely unknown. Here, we report the function and molecular mechanism of the stilbene synthase gene VpSTS29/STS2 from Vitis pseudoreticulata in the regulation of plant responses to powdery mildew. Stilbene synthesis occurred mainly in root tips and mesophyll cells of transgenic grapevines via transport through the vascular bundles. Overexpression of VpSTS29/STS2 in Vitis vinifera increased the

Complex plastome structural variation caused by intermolecular and intramolecular recombination accounts for leaf variegation in Heptapleurum ellipticum.

Plastome variation, including single spontaneous nucleotide substitutions and single insertions/deletions, is the major source of leaf variegation in plants. Additionally, one recent study has showed that a simple plastome structural variation, which is induced by one pair of small inverted repeats, can also result in leaf variegation. Here we show a complex plastome structural variation caused by intermolecular and intramolecular recombination across three pairs of small inverted repeats accounts for leaf variegation in a widely cultivated shrub Heptapleurum ellipticum (Araliaceae). This plas

Plant Tissue Cultures.

Plant tissue cultures are an efficient system to study cell wall biosynthesis in living cells in vivo. Tissue cultures also provide cells and culture medium from which enzymes and cell wall polymers can easily be separated for further studies. Tissue cultures with tracheary element differentiation or extracellular lignin formation have provided useful information related to several aspects of xylem and lignin formation. In this chapter, methods for nutrient medium preparation and callus culture initiation and its maintenance as well as those for protoplast isolation and viability observation a

A Method to Quantify Relative Stomatal, Mesophyll, and Biochemical Limitations to C3 Leaf Photosynthesis in a Specific Growth Environment.

Determining relative limitations of stomatal, mesophyll, and biochemical components (ls, lm, and lb, respectively) to photosynthetic rate (A) can help understand leaf ecophysiology for specific growth conditions. The differential method has been widely applied to estimate the relative limitations in C3 plants, first for comparing two contrasting growth conditions, but also increasingly for a single condition. In addition to the common practical issue that estimating mesophyll conductance is sensitive to measurement noise, the method has a theoretical feature when applied to photosynthesis for

Leaf diffusional capacity largely contributes to the reduced photosynthesis in rice plants under magnesium deficiency.

Numerous studies have clarified the impacts of magnesium (Mg) on leaf photosynthesis from the perspectives of protein synthesis, enzymes activation and carbohydrate partitioning. However, it still remains largely unknown how stomatal and mesophyll conductances (gs and gm, respectively) are regulated by Mg. In the present study, leaf gas exchanges, leaf hydraulic parameters, leaf structural traits and cell wall composition were examined in rice plants grown under high and low Mg treatments to elucidate the impacts of Mg on gs and gm. Our results showed that reduction of leaf photosynthesis unde

In vivo reorganization of the actin cytoskeleton in leaves of Nicotiana tabacum L. transformed with plastin-GFP. Correlation with light-activated chloroplast responses.

BACKGROUND: The actin cytoskeleton is involved in the responses of plants to environmental signals. Actin bundles play the role of tracks in chloroplast movements activated by light. Chloroplasts redistribute in response to blue light in the mesophyll cells of Nicotiana tabacum. The aim of this work was to study the relationship between chloroplast responses and the organization of actin cytoskeleton in living tobacco cells. Chloroplast movements were measured photometrically as changes in light transmission through the leaves. The actin cytoskeleton, labeled with plastin-GFP, was visualised b

The AcLFYL1-AcCSE module enhances kiwifruit resistance to Pseudomonas syringae pv. actinidiae by regulating cell density and lignin biosynthesis.

Canker disease caused by Pseudomonas syringae pv. actinidiae (Psa) is a severe bacterial infection threatening global kiwifruit production. Psa causes lignin degradation, cell wall rupture, leaf wilting, and canker formation on branches and trunks, often leading to plant death. The plant cell wall serves as a structural barrier against pathogens, with its thickness, composition, and cell density influencing disease resistance. Comparative studies between resistant germplasms Actinidia eriantha "Maohuaxiong" (A. eriantha 'MHX') and Actinidia latifolia "Kuoye" (A. latifolia 'KY') and susceptible

Foliar-applied manganese and phosphorus in deficient barley: Linking absorption pathways and leaf nutrient status.

Foliar fertilization delivers essential nutrients directly to plant tissues, reducing excessive soil fertilizer applications that can lead to eutrophication following nutrient leaching. Foliar nutrient absorption is a dynamic process affected by leaf surface structure and composition, plant nutrient status, and ion physicochemical properties. We applied multiple methods to study the foliar absorption behaviors of manganese (Mn) and phosphorus (P) in nutrient-deficient spring barley (Hordeum vulgare) at two growth stages. Nutrient-specific chlorophyll a fluorescence assays were used to visualiz

Albinism in plants - far beyond the loss of chlorophyll: Structural and physiological aspects of wild-type and albino royal poinciana (Delonix regia) seedlings.

The partial or complete loss of chlorophylls, or albinism, is a rare phenomenon in plants. In the present study, we provide the first report of the occurrence in albino Delonix regia seedlings and describe the morpho-physiological changes associated with albinism. Wild-type (WT) and albino seedlings were characterized. Leaflets samples were processed following common procedures for analysis with light, scanning and transmission electron microscopy. The chlorophyll a fluorescence parameters and the carbohydrate, lipid and soluble protein content were also determined in leaf and cotyledon sample

Aspects of programmed cell death during early senescence of barley leaves: possible role of nitric oxide.

Leaf senescence is a highly coordinated process which involves programmed cell death (PCD). Early stages of leaf senescence occurring during normal leaf ontogenesis, but not triggered by stress factors, are less well known. In this study, we correlated condensation of chromatin and nuclear DNA (nDNA) fragmentation, two main features of PCD during early senescence in barley leaves, with the appearance of nitric oxide (NO) within leaf tissue. With the help of the alkaline version of the comet assay, together with measurements of nDNA fluorescence intensity, we performed a detailed analysis of th

Growth Temperature Alters the Thermal Sensitivity of Mesophyll Conductance in Wild and Cultivated Tomato.

Mesophyll conductance (gm) is a key component of internal CO2 diffusion and can limit photosynthesis in C3 leaves. Temperature responses of gm are commonly treated as intrinsic properties of the leaf, yet it remains unclear whether they acclimate to growth conditions. Here, we tested whether growth temperature alters the thermal sensitivity of gm and whether this differs between wild and cultivated tomato. We compared the wild species Solanum pennellii and the cultivated Solanum lycopersicum cv. M82 grown under four temperature regimes (16°C/10°C to 34°C/28°C, day/night). Using combined gas ex

Hydrogen sulfide-mitigated salinity stress impact in sunflower seedlings was associated with improved photosynthesis performance and osmoregulation.

BACKGROUND: Salinity is one major abiotic stress affecting photosynthesis, plant growth, and development, resulting in low-input crops. Although photosynthesis underlies the substantial productivity and biomass storage of crop yield, the response of the sunflower photosynthetic machinery to salinity imposition and how H2S mitigates the salinity-induced photosynthetic injury remains largely unclear. Seed priming with 0.5 mM NaHS, as a donor of H2S, was adopted to analyze this issue under NaCl stress. Primed and nonprime seeds were established in nonsaline soil irrigated with tape water for 14 d

Loss of conductance between mesophyll symplasm and intercellular air spaces explains nonstomatal control of transpiration.

The conventional assumption is that stomatal conductance ([Formula: see text]) dominates the regulation of water and carbon dioxide fluxes between leaves and the atmosphere. Here, a nanoreporter of water status at the mesophyll cell surface and local xylem within intact maize leaves documents significant undersaturation of water vapor in the outside-xylem zone (OXZ) and a large loss of conductance of this zone ([Formula: see text]) at moderate xylem water stress, without stomatal closure or turgor loss. The ratio of the resistances [Formula: see text] serves as a predictive phenotype of unders

Intracellular localization of CA1P and CA1P phosphatase activity in leaves of Phaseolus vulgaris L.

CA1P and CA1P phosphatase occur in the chloroplasts of leaf mesophyll cells of many species. However, whether either may occur exclusively in the chloroplast has not yet been established. To examine their intracellular distribution, mature, dark-or light-treated leaves of Phaseolus vulgaris were frozen, lyophilized and then centrifuged in density gradients of heptane and tetrachloroethylene. After gradient fractionation, both CA1P and CA1P phosphatase activity co-segregated with chloroplast material. Distribution analyses using sub-cellular compartment markers indicated that both CA1P and CA1P

The decline in leaf mesophyll conductance is derived from anatomical traits under multiple stresses of water, salt and high temperature in cotton.

Climate change-driven extreme abiotic environmental stresses, for instance water stress, salt stress, and high temperature, prominently threaten agricultural crop development and yield. Although we have extensive knowledge regarding how plants react to each of these single stresses, we have limited understanding of how the combination of multiple factors (i.e., multiple stresses) have impact on leaf CO2 diffusion and photosynthesis. This study examined the response of leaf mesophyll conductance (gm) and its consequences for photosynthesis and biomass in flower and boll stages of cotton under t

An efficient method for tissue-specific protoplast isolation suitable for single-cell RNA sequencing and transient gene expression analysis in saffron (Crocus sativus L.).

BACKGROUND: Protoplasts have been widely utilized in tissue culture, single-cell RNA sequencing, and transient gene expression analyses. However, the primary challenge in their preparation lies in the efficient removal of rigid cell walls to obtain high-quality plant cells. The aim of this study is to establish an effective method for protoplast isolation from saffron (Crocus sativus L.), thereby accelerating advances in molecular biology research in this species. RESULT: Tissue-specific protoplasts were successfully isolated from apical buds, petals, leaves, and roots of saffron, with yields

Effects of HgCl(2) on CO(2) dependence of leaf photosynthesis: evidence indicating involvement of aquaporins in CO(2) diffusion across the plasma membrane.

Experiments were conducted to examine whether mercury-sensitive aquaporins facilitate photosynthetic CO(2) diffusion across the plasma membrane of leaf mesophyll cells. Discs without abaxial epidermes from Vicia faba leaflets were treated with HgCl(2), an inhibitor of aquaporins. Hydraulic conductivity of the plasma membrane of these discs, measured as the weight loss of the discs in the 1 M sorbitol solution, was inhibited by sub-mM concentrations of HgCl(2) by 70 to 80%. Photosynthetic CO(2) fixation was also inhibited by the HgCl(2) treatment in a similar concentration range. When 0.3 mM Hg

Sulfur Compounds in Regulation of Stomatal Movement.

Sulfur, widely present in the soil and atmosphere, is one of the essential elements for plants. Sulfate is a dominant form of sulfur in soils taken up by plant roots. In addition to the assimilation into sulfur compounds essential for plant growth and development, it has been reported recently that sulfate as well as other sulfur containing compounds can also induce stomatal movement. Here, we first summarized the uptake and transport of sulfate and atmospheric sulfur, including H2O and SO2, and then, focused on the effects of inorganic and organic sulfur on stomatal movement. We concluded all

Hydrogen sulphide improves adaptation of Zea mays seedlings to iron deficiency.

Hydrogen sulphide (H2S) is emerging as a potential molecule involved in physiological regulation in plants. However, whether H2S regulates iron-shortage responses in plants is largely unknown. Here, the role of H2S in modulating iron availability in maize (Zea mays L. cv Canner) seedlings grown in iron-deficient culture solution is reported. The main results are as follows: Firstly, NaHS, a donor of H2S, completely prevented leaf interveinal chlorosis in maize seedlings grown in iron-deficient culture solution. Secondly, electron micrographs of mesophyll cells from iron-deficient maize seedlin
