CYP2U1: a cardiac metabolic switch alleviating obesity cardiomyopathy via lipid metabolic reprogramming.

Cytochrome P450 2U1 (CYP2U1) modulates lipid metabolism, yet its role in obesity-induced cardiomyopathy remains unknown. Here, we demonstrate that cardiac-specific overexpression of CYP2U1 safeguards against high-fat diet (HFD)-induced myocardial dysfunction. Mice received tail-vein injection of adeno-associated virus serotype 9 (AAV9) carrying CYP2U1 under the cardiac troponin T promoter (AAV9-cTnT-CYP2U1) and were maintained on a 16-week HFD. Cardiac-specific CYP2U1 overexpression was associated with better preservation of systolic function and with reduced myocardial hypertrophy, interstiti

Metabolic dysfunction-associated steatotic liver disease: Mechanisms, metabolic reprogramming, and therapeutic insights.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a progressive metabolic disorder that is pathologically characterized by abnormal lipid deposition in the liver and metabolic inflammation. The current clinical management of MASLD largely involves generalized lifestyle modifications including diet and broad-spectrum metabolic interventions such as insulin sensitizers. These approaches often yield suboptimal outcomes because of poor long-term adherence, heterogeneous patient responses, and limited efficacy in advanced disease stages. Crucially, they fail to address disease-spe

Immune cell metabolic reprogramming mediates ICI therapy resistance.

Immune checkpoint inhibitors (ICIs) have revolutionized the treatment of advanced malignancies; however, their efficacy in solid tumors remains limited by therapy resistance. This resistance arises from the metabolic reprogramming of immune cells in the tumor microenvironment (TME), a metabolic 'cage' where immune and cancer cells compete for nutrients. Immune effector cells succumb to metabolic exhaustion amid nutrient competition, whereas immunosuppressive cells augment inhibitory functions via metabolic adaptation, collectively mediating tumor immune evasion. This review systematically deli

Metabolic reprogramming of the immune response in periodontitis.

OBJECTIVE: Periodontitis, a highly prevalent chronic inflammatory disease, is characterized by the progressive destruction of the tooth-supporting tissues, ultimately causing tooth loss. In recent years, the emerging field of immunometabolism has revealed that immune cell function is tightly regulated by intracellular metabolic pathways that govern cellular activation, differentiation, and effector responses. Therefore, the aim of this review was to synthesize current evidence on the role of immunometabolism in the pathogenesis of periodontitis, with a particular focus on how metabolic reprogr

Amino acid metabolic reprogramming in tumor metastatic colonization.

Metastasis is considered as the major cause of cancer death. Cancer cells can be released from primary tumors into the circulation and then colonize in distant organs. How cancer cells acquire the ability to colonize in distant organs has always been the focus of tumor biology. To enable survival and growth in the new environment, metastases commonly reprogram their metabolic states and therefore display different metabolic properties and preferences compared with the primary lesions. For different microenvironments in various colonization sites, cancer cells must transfer to specific metaboli

Metabolic Reprogramming in Adipose Tissue During Cancer Cachexia.

Cancer cachexia is a disorder of energy balance characterized by the wasting of adipose tissue and skeletal muscle resulting in severe weight loss with profound influence on morbidity and mortality. Treatment options for cancer cachexia are still limited. This multifactorial syndrome is associated with changes in several metabolic pathways in adipose tissue which is affected early in the course of cachexia. Adipose depots are involved in energy storage and consumption as well as endocrine functions. In this mini review, we discuss the metabolic reprogramming in all three types of adipose tissu

Pathophysiological Integration of Metabolic Reprogramming in Breast Cancer.

Metabolic changes that facilitate tumor growth are one of the hallmarks of cancer. The triggers of these metabolic changes are located in the tumor parenchymal cells, where oncogenic mutations induce an imperative need to proliferate and cause tumor initiation and progression. Cancer cells undergo significant metabolic reorganization during disease progression that is tailored to their energy demands and fluctuating environmental conditions. Oxidative stress plays an essential role as a trigger under such conditions. These metabolic changes are the consequence of the interaction between tumor

IFI16 Impacts Metabolic Reprogramming during Human Cytomegalovirus Infection.

Cellular lipid metabolism plays a pivotal role in human cytomegalovirus (HCMV) infection, as increased lipogenesis in HCMV-infected cells favors the envelopment of newly synthesized viral particles. As all cells are equipped with restriction factors (RFs) able to exert a protective effect against invading pathogens, we asked whether a similar defense mechanism would also be in place to preserve the metabolic compartment from HCMV infection. Here, we show that gamma interferon (IFN-γ)-inducible protein 16 (IFI16), an RF able to block HCMV DNA synthesis, can also counteract HCMV-mediated metabol

Metabolic Reprogramming and Immune Evasion in Nasopharyngeal Carcinoma.

Nasopharyngeal carcinoma (NPC) is a malignant tumor of the nasopharynx mainly characterized by geographic distribution and EBV infection. Metabolic reprogramming, one of the cancer hallmarks, has been frequently reported in NPCs to adapt to internal energy demands and external environmental pressures. Inevitably, the metabolic reprogramming within the tumor cell will lead to a decreased pH value and diverse nutritional supplements in the tumor-infiltrating micro-environment incorporating immune cells, fibroblasts, and endothelial cells. Accumulated evidence indicates that metabolic reprogrammi

Metabolic Reprogramming in Immune Response and Tissue Inflammation.

Innate and adaptive immunity participate in and regulate numerous human diseases. Increasing evidence implies that metabolic reprogramming mediates immune cell functional changes during immune responses. In this review, we present and discuss our current understanding of metabolic regulation in different immune cells and their subsets in response to pathological stimuli. An interactive biochemical and molecular model was established to characterize metabolic reprogramming and their functional implication in anti-inflammatory, immune resolution, and proinflammatory responses. We summarize 2 maj

Quantitative proteomics suggests metabolic reprogramming during ETHE1 deficiency.

Deficiency of mitochondrial sulfur dioxygenase (ETHE1) causes the severe metabolic disorder ethylmalonic encephalopathy, which is characterized by early-onset encephalopathy and defective cytochrome C oxidase because of hydrogen sulfide accumulation. Although the severe systemic consequences of the disorder are becoming clear, the molecular effects are not well defined. Therefore, for further elucidating the effects of ETHE1-deficiency, we performed a large scale quantitative proteomics study on liver tissue from ETHE1-deficient mice. Our results demonstrated a clear link between ETHE1-deficie

Iron Stress Reprograms Enterocyte Metabolism.

Objectives: This study utilized IPEC-J2, a neonatal pig jejunum-derived cell line, to assess how iron deficiency (ID) and excess (IE) alter enterocyte metabolism and the transcription of inflammatory markers. Methods: Cells were treated with deferiprone (DFP) or ferric ammonium citrate (FAC) to induce ID or IE, respectively. The study evaluated: (1) transcriptional changes in iron-regulatory genes over 96 h under ID or IE; (2) the interaction between iron imbalance and lipopolysaccharide (LPS) exposure on mRNA expression of inflammation markers and iron transporters; and (3) cellular metabolic

Melatonin-mediated metabolic reprogramming and stress adaptation in plants.

Melatonin acts as a multifunctional regulator of plant stress adaptation by reprogramming primary and secondary metabolism and coordinating ROS, nitric oxide, and phytohormone signalling. Integrating metabolomics with transcriptomics and proteomics provides new insights into these mechanisms and highlights melatonin as a promising strategy for enhancing crop resilience and productivity under diverse environmental stresses. Plants, being sessile, are continuously exposed to diverse environmental challenges, which disrupt cellular homeostasis and reduce crop productivity. To survive under such c

Amino acid metabolic reprogramming: future prospects for cholangiocarcinoma therapy.

Cholangiocarcinoma (CCA) is a highly heterogeneous disease with a poor prognosis and a 5-year survival rate of less than 20% due to late diagnosis and limited therapeutic options, and the current problems in the treatment of CCA can be mainly attributed to the low rate of early diagnosis, the limited availability of targeted drugs, and the gradual increase in chemoresistance. Metabolic reprogramming in CCA causes the accumulation of large amounts of lactic acid and glycolytic intermediates, exacerbating hypoxia and the formation of an acidic environment at the tumor site, which further reduces

Glucose metabolic reprogramming: mechanisms and therapeutic implications in neuroinflammation.

Microglia play dual and context-dependent roles in the central nervous system, contributing both to the maintenance of brain homeostasis and the propagation of neuroinflammatory responses. Under pathological conditions, microglia undergo profound glycolytic reprogramming, characterized by a shift from oxidative phosphorylation to enhanced aerobic glycolysis. This review focuses on the glucose-glycolysis-lactate metabolic axis and its pivotal role in microglial immunometabolism. We elucidated how key glycolytic enzymes (e.g., HK2, PKM2) and metabolites (e.g., lactate, pyruvate, ATP) regulate mi

Research Progress of Metabolic Reprogramming in Childhood Acute Leukemia.

Pediatric acute leukemia (AL), encompassing both acute lymphoblastic leukemia (ALL) and acute myeloid leukemia (AML), represents a highly aggressive malignancy that poses a substantial threat to child health. Among its defining hallmarks, metabolic reprogramming has emerged as a critical determinant of disease initiation, progression, therapeutic resistance, and relapse. Leukemic cells reshape their metabolic pathways to meet the survival demands of rapid proliferation. This review provides a comprehensive synthesis of recent advances on the characteristic mechanisms of metabolic reprogramming

Physiological and metabolic reprogramming under stress combination in plants.

In nature, plants are rarely challenged by a single, isolated, environmental stress factor. Instead, they continuously experience a dynamic range of abiotic and biotic constraints, generating conditions of multifactorial stress combination (MFSC). Driven by anthropogenic activities, many stressors are increasing in their frequency, intensity, and duration, exposing plants to complex conditions that go well beyond two interacting factors. A recent conceptual advance is the recognition that plant responses to MFSC cannot be predicted by simply summing up plant responses of the different individu

The GATA1 N terminus coordinates metabolic reprogramming in erythropoiesis.

Mutations in GATA1 that cause skipping of exon 2, which encodes the N terminus, are associated with the myeloid leukemia of Down syndrome and Diamond-Blackfan anemia (DBA). To elucidate the molecular function of this N-terminal region, we used single-cell RNA sequencing (scRNA-seq) on fetal liver cells from Gata1-mutant embryos that express only the short isoform of GATA1 (GATA1s) lacking the N terminus of full-length GATA1 (GATA1FL). scRNA-seq revealed defects in erythropoiesis and aberrant upregulation of glycolytic genes, including PKM, which encodes pyruvate kinase to catalyze the final an
