Effects of non-iron metalloporphyrins on growth and gene expression of Porphyromonas gingivalis.

The oral anaerobic bacterium Porphyromonas gingivalis, which is implicated as an important pathogen for chronic periodontitis, requires heme for its growth. Non-iron metalloporphyrins, In-PPIX and Ga-PPIX, were examined for antibacterial effects on P. gingivalis. Both In-PPIX and Ga-PPIX caused retardation of P. gingivalis growth in a dose-dependent fashion. Microarray and qPCR analyses revealed that In-PPIX treatment upregulated the expression of several genes encoding proteins including ClpB and ClpC, which are members of the Clp (caseinolytic protease, Hsp100) family, and aRNR, aRNR-activat

Structure of malaria pigment and related propanoate-linked metalloporphyrin dimers.

Malaria pigment, the heme detoxification product of malaria's invasion, digestion, and growth inside mammalian red blood cells, is an insoluble phase of iron(III)protoporphyrin-IX. Even though its structure was determined in 2000 by powder X-ray diffraction, significant questions remain about its formation and possible interaction with quinoline antimalarial drugs. A recent structural study, also with X-ray powder diffraction, has reconfirmed that the material isolated from the parasite is isostructural with its synthetic equivalent. It was recently suggested that other isomers may also be for

Role of metalloporphyrin core metals in the mediated reductive dechlorination of tetrachloroethylene.

A promising approach to abiotically dechlorinate a variety of chlorinated organic contaminants under reducing conditions is to utilize porphyrins or other tetrapyrrole macrocycles as electron transfer mediators/shuttles for catalyzing their reduction. In this study, various experimental approaches were used to elucidate the role of porphyrin core metals in the reductive dechlorination of tetrachloroethylene (PCE). The importance of specific core metals for the reactivity of a porphyrin and its mediated reaction mechanisms was demonstrated by inserting different metals into metallo tetrakis (N-

Effects of a series of metalloporphyrins on adrenal, testicular and thyroid function in rats.

We have extended our earlier studies [Pharmacology 1986;34:9-16] on the effects of certain synthetic heme analogues and cobalt chloride (CoCl2) on endocrine functions mediated by the hypothalamic-pituitary axis to examine specifically the ability of Sn-protoporphyrin (SnPP) and Sn-mesoporphyrin (SnMP) to perturb adrenal, testicular and thyroid function since there is interest in the use of Sn(tin)-porphyrins in the treatment of hyperbilirubinemia of the newborn. SnPP and SnMP when administered to adult male rats did not alter serum corticosterone, testosterone, thyroxine or triiodothyronine le

Selection of metalloporphyrin heme oxygenase inhibitors based on potency and photoreactivity.

The heme oxygenase inhibitor, tin protoporphyrin, is being studied for the prevention of neonatal jaundice. This potential drug, however, is also a photosensitizer that could cause serious and unknown side effects when administered to newborns. Therefore, we have developed in vitro and in vivo procedures for the screening and further characterization of potentially safe heme oxygenase inhibitors. The ideal inhibitor: 1) contains a biocompatible metal, 2) is not degraded in tissues, 3) is a highly potent inhibitor of heme oxygenase, and 4) does not participate in photochemical reactions. Proto-

Mortality of metalloporphyrin-treated neonatal rats after light exposure.

Tin protoporphyrin (SnPP) has been used to suppress hyperbilirubinemia in human neonates through inhibition of heme oxygenase. Some of the subjects exhibited mild erythema upon receiving phototherapy. SnPP and three proposed alternatives, tin mesoporphyrin (SnMP), zinc protoporphyrin (ZnPP) and zinc mesoporphyrin (ZnMP) are potential photosensitizers. We therefore studied the phototoxic effects of these compounds in the neonatal rat model. Fed Wistar rats (24-36 h old) were injected intraperitoneally with up to 40 mumol SnPP/kg body weight, 30 mumol SnMP/kg body weight, 60 mumol ZnPP/kg body w

Optimization of Lipophilic Metalloporphyrins Modifies Disease Outcomes in a Rat Model of Parkinsonism.

Oxidative stress plays a crucial role in the pathogenesis of Parkinson disease (PD), and one strategy for neuroprotective therapy for PD is to scavenge reactive species using a catalytic antioxidant. Previous studies in our laboratory revealed that pretreatment of lipophilic metalloporphyrins showed protective effects in a mouse PD model. In this study, we optimized the formulations of these metalloporphyrins to deliver them orally and tested their efficacy on disease outcomes in a second species after initiation of an insult (i.e., disease modification). In this study, a pharmaceutical formul

Metalloporphyrins and Metallocorroles with Designed Second-Sphere Structures for Catalytic Small Molecule Activation Reactions.

Small-molecule activation reactions, including the oxygen reduction reaction (ORR), hydrogen evolution reaction (HER), and carbon dioxide reduction reaction (CO2RR), are involved in natural and artificial energy conversion processes. In enzymes, the second-sphere environment of catalytic sites can promote substrate activation, stabilize intermediates, and facilitate proton transfer. Synthetic chemists have endeavored to develop molecular catalysts with functionalized second sphere environments. However, installing functional groups at metal second-sphere sites and understanding their actual ro

Magnetic resonance imaging-histomorphologic correlation studies on paramagnetic metalloporphyrins in rat models of necrosis.

RATIONALE AND OBJECTIVES: The authors intended to confirm previous findings that paramagnetic porphyrins are avid only for intratumoral nonviable tissues, but not for viable tumor cells, and to test the hypothesis that necrosis, regardless of location and origin, can be visualized by metalloporphyrin enhanced magnetic resonance imaging (MRI). METHODS: Intravenous administrations of gadolinium mesoporphyrin (Gd-MP), manganese tetraphenylporphyrin (Mn-TPP), manganese methylpyrroporphyrin-gadopentetate dimeglumine complex (Mn-MPP-Gd) and manganese tetra(4-sulfonatophenyl)porphyrin (MnTPPS4) at 0

The use of riboflavin and metalloporphyrins in cytochrome P-450 content in Wistar rats.

Phototherapy is commonly used for the treatment of neonatal jaundice. Riboflavin is a photosensitizer that generates singlet oxygen, which promotes bilirubin photodecomposition. Metalloporphyrins are also effective photosensitizers. The effect of a combined dosing regimen of riboflavin and metalloporphyrins was studied, with the aim of increasing the efficiency of the phototherapeutic treatment of hyperbilirubinemia. It was envisaged that riboflavin and the metalloporphyrins, by promoting the photodecomposition of bilirubin, would thereby lead to a reduction of the toxic side effects associate

Inhibition of hippocampal heme oxygenase, nitric oxide synthase, and long-term potentiation by metalloporphyrins.

Four potent metalloporphyrin inhibitors of heme oxygenase were used to assess whether carbon monoxide production was required for induction of LTP in the CA1 region of the hippocampus. Although the metalloporphyrins produced a similar and substantial inhibition of heme oxygenase activity in hippocampal slices, only two compounds reduced the amount of LTP elicited by tetanic stimulation (chromium mesoporphyrin IX and zinc protoporphyrin IX). Both chromium mesoporphyrin IX and zinc protoporphyrin IX inhibited nitric oxide synthase in the hippocampus; tin mesoporphyrin IX and zinc deuteroporphyri

Metalloporphyrin-based photodynamic inactivation of multidrug-resistant environmental mastitis pathogens.

Bacteria such as Klebsiella pneumoniae, Proteus mirabilis and Streptococcus dysgalactiae are often associated with environmental and zoonotic agents causing bovine mastitis. In addition, the emergence of multidrug-resistant (MDR) strains associated with these pathogens poses a significant challenge to the effectiveness of conventional antimicrobial therapies. Thus, this study aimed to evaluate the efficacy of antimicrobial photodynamic inactivation (aPDI) as an alternative for controlling environmental pathogens related to mastitis. Four meso-Tetra-(4-pyridyl)porphyrin/palladium(II) complexes

Preparation of Metalloporphyrin-Bound Superparamagnetic Silica Particles via "Click" Reaction.

A facile approach using click chemistry is demonstrated for immobilization of metalloporphyrins onto the surface of silica-coated iron oxide particles. Oleic-acid stabilized iron oxide nanocrystals were prepared by thermal decomposition of iron(III) acetylacetonate. Their crystallinity, morphology, and superparamagnetism were determined using X-ray diffraction, transmission electron microscopy, and a superconducting quantum interference device. Monodisperse core-shell particles were produced in the silica-coating of iron oxide via microemulsion synthesis. Surface modification of these particle

Four metalloporphyrinic frameworks as heterogeneous catalysts for selective oxidation and aldol reaction.

Four porous metalloporphyrinic framework materials, [(CH3)2NH2][Zn2(HCOO)2(Mn(III)-TCPP)]·5DMF·2H2O (1; H6TCPP = tetrakis(4-carboxyphenyl)porphyrin), [(CH3)2NH2][Cd2(HCOO)2(Mn(III)-TCPP)]·5DMF·3H2O (2), [Zn2(HCOO)(Fe(III)(H2O)-TCPP)]·3DMF·H2O (3), and [Cd3(H2O)6(μ2-O)(Fe(III)-HTCPP)2]·5DMF (4) were synthesized by heating a mixture of M(III)Cl-H4TCPP (M = Mn and Fe) and M' (M' = Zn or Cd) nitrate in a mixed solvent of DMF and acetic acid. Compounds 1-3 are built up from M'2(COO)4 paddle-wheel subunits bridged by M(III)-TCPP and formate ligands to form their 3D connections. The formate pillar he

Development of sensitive metalloporphyrin probes for chemiluminescent imaging detection of serum proteins.

The development of metalloporphyrin- (ferric protoporphyrin IX chloride (FePP), cobalt (III) protoporphyrin IX chloride, copper (II) protoporphyrin IX) enhanced chemiluminescent (CL) imaging detection of serum proteins after PAGE is described in this article. The detection is based on the catalytic activity of metalloporphyrins, especially FePP, in the CL reaction of the luminol-H2O2 system. Some relatively low abundant proteins such as hemopexin (Hpx) and complement C4 are sensitively detected by FePP-enhanced CL imaging. Other proteins such as haptoglobin, apolipoprotein A-1, complement C3,

Unconventional neuroprotection against Ca2+ -dependent insults by metalloporphyrin catalytic antioxidants.

We evaluated whether both inert and catalytically active metalloporphyrin antioxidants, meso-substituted with either phenyl-based or N-alkylpyridinium-based groups, suppress Ca(2+)-dependent neurotoxicity in cell culture models of relevance to cerebral ischemia. Representatives from both metalloporphyrin classes, regardless of antioxidant strength, protected cultured cortical neurons or PC-12 cultures against the Ca(2+) ionophores ionomycin or A23187, by suppressing neurotoxic Ca(2+) influx. Some metalloporphyrins suppressed excitotoxic Ca(2+) influx indirectly induced by the Ca(2+) ionophores

Role of lipid peroxidation in metalloporphyrin-mediated phototoxic reactions in neonatal rats.

Our group previously demonstrated dose-dependent mortality in neonatal rats treated with tin protoporphyrin and light. We hypothesize that lipid peroxidation may be responsible for the toxic effects of photosensitizing metalloporphyrins. Neonatal rat blood samples with or without metalloporphyrins (40 mM) were exposed to cool white light (20 microW/cm2/nm) for 30 min at 37 degrees C. In the in vivo model, neonatal rat pups were given injections of 40 mumol of either tin protoporphyrin (4 mM), zinc protoporphyrin/kg body weight, or saline and placed over cool white light. The control animals we

Framework-Controlled Axial Coordination of Guest Molecules in Metalloporphyrin-Based MOFs.

Metalloporphyrin-based metal-organic frameworks (MOFs) offer a unique platform for probing how confined environments influence porphyrin metal site structure and reactivity. The MOF series, FeCl-PCN-222 and MnCl-PCN-222, are investigated under a series of guest environments, including acetone, imidazole, pyridine, and piperidine, to demonstrate how differences in metalloporphyrin site accessibility and reactivity influence guest molecule axial ligation patterns. Fe and Mn K-edge X-ray absorption spectroscopy (XAS) is employed to characterize the local coordination, oxidation state, and electro

Mitigation effects of solar-driven metalloporphyrins on CO2 and N2O emissions from an agricultural soil.

Developing green technologies to mitigate soil carbon dioxide (CO2) and nitrous oxide (N2O) emissions is critical for sustainable agriculture. Here, we investigated the effects of two water-soluble metalloporphyrins (FeTPPS and MnTPPS), with and without glucose addition, on CO2 and N2O emissions and the associated mechanisms in an agricultural soil under natural solar irradiation. Metalloporphyrin effects depended on metal identity and labile carbon (C) availability. FeTPPS alone significantly reduced cumulative CO2 emissions by 9.9%, and FeTPPS with glucose reduced emissions by 7.2% relative
