BACKGROUND: The entomopathogenic fungus Metarhizium brunneum (M. brunneum) is an effective biocontrol agent against various vector arthropods such as ticks, mosquitoes, and flies. However, its virulence mechanisms remain poorly understood, which hinders its broader application. This study aims to establish an integrative framework for prioritizing virulence-related pathways in M. brunneum to aid in the development of more effective biocontrol strategies.
METHODS: A multidimensional virulence pathway scoring framework was developed using publicly available protein annotation data of M. brunneu

Beauveria bassiana and Metarhizium rileyi are entomopathogenic fungi that are widely present in nature. Due to their excellent insecticidal activity, they have been widely developed as microbial preparations and utilized in the biological control of pests. To clarify differences in pathogenicity between B. bassiana and M. rileyi against Spodoptera frugiperda, third-instar larvae were subjected to immersion treatments, whereas fifth-instar larvae were inoculated via injection with various doses of fungal strains B. bassiana AJS91881 and M. rileyi SXBN200920. Our results demonstrated differentia

With the incidence of Lyme disease increasing throughout the United States, reducing risk of exposure to the disease is of the utmost concern. In the northeastern U.S., the blacklegged tick, Ixodes scapularis, is the primary vector and the white-footed mouse, (Peromyscus leucopus), the primary reservoir for Borrelia burgdorteri, the bacterium causing Lyme disease. Targeting I. scapularis engorging on white-footed mice with an effective biological control agent, such as the fungus Metarhizium anisopliae, could be an effective and relatively safe control technique. In 2002-2003, we performed lab

This study investigated the reduction of bitterness in extensively hydrolyzed formula (EHF) for milk-allergic infants by engineering chymotrypsin CHY1 through molecular simulation and mutagenesis. Molecular docking identified key mutation sites (Ser201, Pro309, Gly345) for enhancing binding to bitter peptides. Virtual mutation analysis predicted stable interactions. Mutant enzymes were expressed in Pichia pastoris GS115, with expression levels increased to 86.24-112.17 mg/L. The P309W mutant exhibited more than twice higher activity than CHY1. Optimized enzyme combinations, particularly P309F

The indiscriminate use of synthetic acaricides in the control of Rhipicephalus microplus ticks results in the accumulation of residues in animal products and the environment, and favors the emergence of resistant strains, making it necessary to develop new control alternatives. Extracts of Metarhizium anisopliae (MA) and M. robertsii (MR) represent a promising strategy, as they contain biodegradable and specific compounds, such as enzymes. This study evaluated the effects of active and denatured extracts of MA and MR, isolated or combined with deltamethrin, on the mortality and morphology of t

Metarhizium fungi, essential for ecosystem function and commonly utilised in pest control, often occupy ecological niches contaminated by toxic compounds of both anthropogenic and microbiological origin. The present study reveals the potential of Metarhizium anisopliae for biodegradation of the Fusarium mycotoxin zearalenone (ZEN), a common contaminant of crops that poses a significant threat to human and animal health due to its oestrogenic potential and toxicity. A key aspect of the pathway described is the degradation of ZEN by cleaving the lactone bond, which results in a significant reduc

The anti-ultraviolet (UV) role of a Rad4-Rad23-Rad33 complex in budding yeast relies on nucleotide excision repair (NER), which is mechanistically distinct from photorepair of DNA lesions generated under solar UV irradiation but remains poorly known in filamentous fungi. Here, two nucleus-specific Rad4 paralogs (Rad4A and Rad4B) and nucleocytoplasmic shuttling Rad23 ortholog are functionally characterized by multiple analyses of their null mutants in Metarhizium robertsii, an entomopathogenic fungus lacking Rad33. Rad4A was proven to interact with Rad23 and contribute significantly more to con

The superoxide dismutase (SOD) family of Metarhizium robertsii, a fungal insect pathogen, comprises six members functionally unknown yet, including Cu/ZnSODs (Sod1/5/6), MnSODs (Sod2/3), and FeSOD (Sod4). Here, we show a mitochondrial localization of Sod3 and Sod4 and a requirement of either sod4 or sod6 for the fungal life as suggested by an inability to be deleted. We found remarked roles of Sod1, Sod2, and Sod3 in sustaining antioxidant activity and the fungal potential against insect pests but no role of Sod5 in all examined phenotypes. Intracellular SOD activity decreased by 49% in Δsod1

The fungus Metarhizium anisopliae IBCB425 was tested against Haemonchus contortus in several trials. Coprocultures containing approximately 10,000 H. contortus eggs were inoculated with 105, 106, 107, 108, and 109 conidia and a control. After ten days at room temperature, larvae were recovered, counted, and checked for motility. Treatment with 108 and 109 conidia reduced larval recovery, and all concentrations diminished motility compared with the control (p < 0.05). Subsequently, parcels demarcated on an animal-free paddock were artificially contaminated with 80,000 H. contortus eggs and were

Entomopathogenic fungi have been explored as a potential biopesticide to counteract the insecticide resistance issue in mosquitoes. However, little is known about the possibility that genetic resistance to fungal biopesticides could evolve in mosquito populations. Here, we detected an important genetic component underlying Anopheles coluzzii survival after exposure to the entomopathogenic fungus Metarhizium anisopliae. A familiality study detected variation for survival among wild mosquito isofemale pedigrees, and genetic mapping identified two loci that significantly influence mosquito surviv

Entomopathogenic fungi (EPF) have been widely explored for their potential in the biological control of insect pests and as an environmentally friendly alternative to acaricides for limiting tick infestation in the field. The arthropod cuticle is the main barrier against fungal infection, however, an understanding of internal defense mechanisms after EPF intrusion into the invertebrate hemocoel is still rather limited. Using an infection model of the European Lyme borreliosis vector Ixodes ricinus with the EPF Metarhizium robertsii, we demonstrated that ticks are capable of protecting themselv

This research intended to investigate if the presence of pesticides in the soil could affect the pathogenicity of Metarhizium anisopliae Metsch. (Sorokin) for Ceratitis capitata (Wied.) and assess the effect of conidia application as suspension or dry conidia. The fungicides chlorothalonyl and tebuconazol, the acaricide abamectin, the insecticide trichlorfon, and the herbicide ametrin were applied at the manufacturer-recommended doses. Soil samples were placed in glass flasks and were given the fungus as conidial suspension or dry. After pesticide application, 20 3rd-instar larvae were placed

Metarhizium anisopliae is an entomopathogenic fungus recently licensed for indoor control of cockroaches, a major source of allergens. While M. anisopliae has been shown to be non-infectious and non-toxic to mammals there has been only limited research on potential allergenicity. Using a mouse model, we previously demonstrated allergic immune and inflammatory responses to this agent. The present study was designed to determine whether these responses were associated with changes in pulmonary responses, lung pathology, and the cytokine profile in bronchoalveolar lavage fluid (BALF). Blattella germanica is the most prevalent synanthropic pest and negatively impacts human health, as do the synthetic chemical insecticides used to control them. In contrast, Metarhizium spp. is a well known fungus that can infect insects and other arthropods, causing death to their hosts. When cultivated in liquid medium it produces blastospores. This propagule has the potential to be used as a biological control agent for cockroaches. The aim of this study was to identify eleven native Metarhizium strains from Argentina through molecular taxonomy, evaluate blastospores and biomass production

Arthropod vectors are responsible for a multitude of human and animal diseases affecting poor communities in sub-Saharan Africa. Their control still relies on chemical agents, despite growing evidence of insecticide resistance and environmental health concerns. Biorational agents, such as the entomopathogenic fungus Metarhizium anisopliae, might be an alternative for vector control. Recently, the M. anisopliae isolate ICIPE 7 has been developed into a commercial product in Kenya for control of ticks on cattle. We were interested in assessing the potential of controlling not only ticks but also

LqhIT2 is an insect-specific neurotoxin from the venom of scorpion. In this study, the LqhIT2 gene was introduced into the entomopathogenic fungus, Metarhizium acridum. The virulence of the genetically modified strain MaLqhIT2 was then evaluated against locusts (Locusta migratoria manilensis). Compared with the wild-type strain, the median lethal cell density (LC50) for MaLqhIT2 was a 22.6-fold lower, and the median times to death (LT50) for MaLqhIT2 were reduced by 30.3 and 29.6 %, respectively, after topical inoculation and injection. MaLqhIT2 also grew significantly faster in the hemolymph

Ghanaian chili pepper exports are subject to stringent regulations on pesticide residues in significant foreign markets. Therefore, safer pest management options are needed to supplement current non-chemical control techniques, especially for controlling Thaumatotibia leucotreta Meyrick (False codling moth, FCM), an important phytosanitary pest. A national survey was initiated in 2023 to identify native entomopathogenic fungi (EPF) from Ghanaian farms. Seven Metarhizium isolates were recovered from soils and induced substantial mortalities of soil-residing final-instar stages of FCM in vitro.

Ribosomal proteins are considered to be involved in the immunity of different animals against pathogens. The protein level of RPL23 increased after fungal infection in termites, but how it influence active immunity in termites is unknown. The role of RPL23 gene was studied to evaluate its impact on active immunity of termite Reticulitermes chinensis against entomopathogenic fungus (EPF) Metarhizium anisopliae. The RPL23 gene fragment (414 bp) was cloned and phylogenetic analysis revealed that it's very close to termite Coptotermes formosanus. Expression of RPL23 gene was significantly higher i

Siderophores are small molecular iron chelators and participate in the multiple cellular processes in fungi. In this study, biosynthesis gene clusters of coprogens and dimerumic acids were identified by transcriptional level differences of genes related to iron deficiency conditions in Metarhizium robertsii. This leads to the characterization of new coprogen metachelin C (1) and five known siderophores metachelin A (2), metachelin A-CE (3), metachelin B (4), dimerumic acid 11-mannoside (5), and dimerumic acid (6). The structure of metachelin C (1) was elucidated by NMR spectroscopy and HR-ESI-

Species of the Metarhizium anisopliae complex are globally ubiquitous soil-inhabiting and predominantly insect-pathogenic fungi. The Metarhizium genus contains species ranging from specialists, such as Metarhizium acridum that only infects acridids, to generalists, such as M. anisopliae, Metarhizium brunneum, and Metarhizium robertsii that infect a broad range of insects and can also colonize plant roots. There is little information available about the susceptibility of Metarhizium species to clinical and non-clinical antifungal agents. We determined the susceptibility of 16 isolates comprisin
