Methanosarcina soligelidi sp. nov., a desiccation- and freeze-thaw-resistant methanogenic archaeon from a Siberian permafrost-affected soil.

A methanogenic archaeon, strain SMA-21(T), was isolated from a permafrost-affected soil by serial dilution in liquid medium. The cells were non-motile, stained Gram-negative and grew as irregular cocci with a diameter of 1.3-2.5 µm. Optimal growth was observed at 28 °C, pH 7.8 and 0.02 M NaCl. The strain grew on H2/CO2, methanol and acetate, but not on formate, ethanol, 2-butanol, 2-propanol, monomethylamine, dimethylamine, trimethylamine or dimethyl sulfide. Major membrane lipids of strain SMA-21(T) were archaeol phosphatidylglycerol, archaeol phosphatidylethanolamine and the corresponding hy

The methanogenic archaeon Methanosarcina thermophila TM-1 possesses a high-affinity glycine betaine transporter involved in osmotic adaptation.

Methanogenic Archaea are found in a wide range of environments and use several strategies to adjust to changes in extracellular solute concentrations. One methanogenic archaeon, Methanosarcina thermophila TM-1, can adapt to various osmotic conditions by synthesis of alpha-glutamate and a newly discovered compatible solute, Ne-acetyl-beta-lysine, or by accumulation of glycine betaine (betaine) and potassium ions from the environment. Since betaine transport has not been characterized for any of the methanogenic Archaea, we examined the uptake of this solute by M. thermophila TM-1. When cells we

Disrupting the mtr-operon in Methanosarcina acetivorans enables methyl-reducing methanogenesis with hydrogen and serine as the alternative electron donors.

Methanosarcina acetivorans is a model methanogen because of its metabolic versatility and genetic tractability. This microbe is not known to natively utilize hydrogen as a catabolic electron donor, despite its genome encoding hydrogenases, and the fact that this microbe can be used to heterologously express functional hydrogenases. Its native hydrogenases are expressed at a very low level and are suggested to have a role in recycling hydrogen that is produced as a byproduct of nitrogen fixation. To explore whether hydrogen can act as a catabolic electron donor, we utilized a previously constru

Enhancement of direct interspecies electron transfer and methane production by co-culture of dual Methanosarcina species and Geobacter metallireducens.

Anaerobic digestion is a key technology for converting organic waste into methane, offering significant potential for renewable energy production and waste management. While the addition of conductive materials has been shown to improve direct interspecies electron transfer (DIET), their application faces challenges like biofouling, environmental risks, and increased operational costs. This study investigated the effects of co-culturing dual Methanosarcina (Methanosarcina barkeri and Methanosarcina acetivorans) and Geobacter metallireducens (DM-G) to enhance DIET and methane production without

Cryo-EM study and in vivo chemical mapping of the Methanosarcina acetivorans ribosome and its dimerization via a repurposed enzyme and translation factor.

Despite the overall conservation of ribosomes across all domains of life, differences in their 3D architecture, rRNA sequences, ribosomal protein composition, and translation factor requirements reflect lineage-specific adaptations to environmental niches. In the domain Archaea, structural studies have primarily focused on nonmethanogenic thermophiles and halophiles, leaving it unclear whether these represent the broader Archaea domain. Here, we report the cryo-electron microscopy (cryo-EM) structure of the ribosome from Methanosarcina acetivorans, a previously unreported high-resolution struc

Fluorescently probing anaerobic digester sludge: Measuring single-cell anabolic activity in methanogens (Methanosarcina and Methanothermobacter) with deuterium-labeled Raman analysis.

This study addresses the challenge of obtaining in situ information on substrate utilization rates for individual microbial species in complex microbial communities such as anaerobic digester sludge. To overcome this hurdle, a novel approach combining doubly-labelled deuterium, fluorescence in situ hybridization (FISH) and Raman microspectroscopy was developed. The method enables quantitative determination of anabolic heavy hydrogen incorporation into FISH-targeted, exemplified by methanogenic cells from the genera Methanosarcina and Methanothermobacter. The deuterium incorporation rates ascer

Cofactor F420 tail length diversity in specific representatives of the class Methanosarcinia during acetoclastic and methylotrophic methanogenesis: Deciphering structural variations.

OBJECTIVES: Methanogens possess a variety of coenzymes and cofactors that are involved in redox reactions that occur during methanogenesis. One essential cofactor is F420, a deazaflavin derivate with an oligoglutamate side chain which acts as a low redox potential electron carrier and participates in electron transport coupled with energy conservation. This work aimed at characterizing cofactor F420 analogues in eight distinct acetoclastic and methylotrophic pure cultures of the class Methanosarcinia. METHODS: Pure cultures of Methanothrix soehngenii, Methanothrix thermoacetophila, Methanomet

Methanogenesis from acetate: a nonmethanogenic bacterium from an anaerobic acetate enrichment.

A methanogenic acetate enrichment was initiated by inoculation of an acetate-mineral salts medium with domestic anaerobic digestor sludge and maintained by weekly transfer for 2 years. The enrichment culture contained a Methanosarcina and several obligately anaerobic nonmethanogenic bacteria. These latter organisms formed varying degrees of association with the Methanosarcina, ranging from the nutritionally fastidious gram-negative rod called the satellite bacterium to the nutritionally nonfastidious Eubacterium limosum. The satellite bacterium had growth requirements for amino acids, a peptid

Enhancement of perchloroethene dechlorination by a mixed dechlorinating culture via magnetic nanoparticle-mediated isolation method.

Highly enriched active dechlorinating cultures are important in advancing microbial remediation technology. This study attempted to enrich a rapid perchloroethene (PCE) dechlorinating culture via magnetic nanoparticle-mediated isolation (MMI). MMI is a novel method that can separate the fast-growing and slow-growing population in a microbial community without labelling. In the MMI process, PCE dechlorination was enhanced but the subsequent trichloroethene (TCE) dechlorination was inhibited, with TCE cumulative rate reached up to 80.6% within 70 days. Meanwhile, the microbial community was also

Metabolic pathway analysis of methane from methanol as substrate in microbial consortium.

Simplified anaerobic digestion (SAD) of substrates facilitates microbial methanogenic pathways. In this study, a methane-producing microbial consortium from cow dung was enriched to determine the metabolism and metabolic pathway in the SAD of methanol. The results showed that methanol as a sole substrate produced 167 mL of methane at 10 days significantly higher than 58 mL, 17.6 mL, and 4 mL generated when methanol was combined with sodium formate, sodium formate alone, or sodium acetate. The relative abundance of Methanobacterium, Candidatus_Methanomethylophilus, Methanomassiliicoccus, and Me

Trace Elements Induce Predominance among Methanogenic Activity in Anaerobic Digestion.

Trace elements (TE) play an essential role in all organisms due to their functions in enzyme complexes. In anaerobic digesters, control, and supplementation of TEs lead to stable and more efficient methane production processes while TE deficits cause process imbalances. However, the underlying metabolic mechanisms and the adaptation of the affected microbial communities to such deficits are not yet fully understood. Here, we investigated the microbial community dynamics and resulting process changes induced by TE deprivation. Two identical lab-scale continuous stirred tank reactors fed with di

Presence of an unusual methanogenic bacterium in coal gasification waste.

Methanogenic bacteria growing on a pilot-scale, anaerobic filter processing coal gasification waste were enriched in a mineral salts medium containing hydrogen and acetate as potential energy sources. Transfer of the enrichments to methanol medium resulted in the initial growth of a strain of Methanosarcina barkeri, but eventually small cocci became dominant. The cocci growing on methanol produced methane and exhibited the typical fluorescence of methanogenic bacteria. They grew in the presence of the cell wall synthesis-inhibiting antibiotics d-cycloserine, fosfomycin, penicillin G, and vanco

Strained Partnership: Biocompatibility and Transcriptomic Responses of Hydrogenotrophic Methanogens to Methyl Diethanolamine in Bio-integrated Carbon Capture and Utilization.

Biointegrated carbon capture and utilization (BICCU) represents a promising alternative to conventional carbon capture technologies, combining microbial methanogenesis with CO2 capture agents, such as methyl diethanolamine (MDEA), to produce renewable methane from dilute CO2 sources. However, the biotoxicity of these solvents poses a major challenge to the viability of the BICCU approach. This study investigates the biocompatibility and physiological responses ofMethanococcus maripaludis and Methanosarcina barkeri to MDEA, through batch experiments and transcriptomic analysis. Both methanogens

Genome-centric analysis reveals the importance of granular activated carbon on quorum sensing and glucose metabolism in anaerobic digestion.

Granular activated carbon (GAC) enhances anaerobic digestion (AD) performance, potentially via biofilm promotion, microbial enrichment, and facilitating direct interspecies electron transfer (DIET). In this work, to investigate the role of GAC, DNA was extracted from sludge in lab-scale digesters (with/without GAC) and GAC biofilm. Genome-resolved analysis revealed GAC established distinct niches enriching Methanothrix and Methanosarcina vacuolata. DIET potential would be indicated by strictly filtered pilA genes with the presence of aromatic amino acids, but not the overall pilA genes. While

Methanol-specific methyltransferase isozymes have large carbon kinetic isotope effects that impact methane isotopic signatures.

The stable carbon and hydrogen isotopic composition of methane is widely used to determine its sources. Methanogenic growth on methanol generates methane with significantly lower 13C/12C ratios relative to other substrates, which is used as a marker for this metabolism in environmental samples. The biochemical basis for the large carbon isotope effect associated with methanol growth is currently unknown. Here, we measure the change in the carbon and hydrogen stable isotopic compositions of the methane produced by Methanosarcina acetivorans during growth on methanol. We couple these results wit

Interface-Engineered Methanogenic Nanobiohybrids Redirect Hydrogen Flux for Efficient CO2 Fixation.

Nanobiohybrids that integrate synthetic materials with living microbes offer a powerful strategy for CO2 fixation, chemical production, and environmental remediation, yet their efficacy is often limited by inefficient interfacial electron delivery. Zerovalent iron (nZVI) is a potent abiotic reductant, but its rapid, nonspecific hydrogen release and inherent cytotoxicity hinder productive coupling with microbial metabolism. Here, we overcome this limitation through interfacial engineering, creating a silicate-modified nZVI (Si-nZVI) hybrid with the methanogen Methanosarcina barkeri. This tailor

The redox landscape of pyruvate:ferredoxin oxidoreductases reveals often conserved Fe-S cluster potentials.

Here, we investigate the thermodynamic driving force of internal electron transfer of pyruvate:ferredoxin oxidoreductases (PFORs), by comparing the redox properties of a series of PFORs from Chlorobaculum tepidum, Magnetococcus marinus, Methanosarcina acetivorans, as well as revisiting the single historical precedent, the enzyme from Desulfovibrio africanus. These enzymes require a thiamine pyrophosphate cofactor, three [4Fe-4S] clusters, and CoA for activity and are found within anaerobic organisms that utilize the reverse tricarboxylic acid cycle, or other reductive pathways, performing carb

Anaerobic bacteria that dechlorinate perchloroethene.

In this study, we identified specific cultures of anaerobic bacteria that dechlorinate perchlorethene (PCE). The bacteria that significantly dechlorinated PCE were strain DCB-1, an obligate anaerobe previously shown to dechlorinate chlorobenzoate, and two strains of Methanosarcina. The rate of PCE dechlorination by DCB-1 compared favorably with reported rates of trichloroethene bio-oxidation by methanotrophs. Even higher PCE dechlorination rates were achieved when DCB-1 was grown in a methanogenic consortium.

Combined inhibition of anaerobic digestion by sulfate, salinity, and ammonium: potential inhibitory factors in forward osmosis-concentrated municipal wastewater.

This study investigated the combined and interactive effects of sulfate, salinity (NaCl), and ammonium on mesophilic anaerobic digestion using synthetic wastewater simulating concentrated municipal wastewater from the forward osmosis (FO) process. Batch anaerobic digestion experiments were conducted with varying concentrations of sulfate, NaCl, and ammonium. Complete sulfate reduction was observed in all test systems, regardless of the NaCl and ammonium concentration, indicating no significant inhibitory effect on sulfate-reducing bacteria (SRB). However, the increased toxicity of hydrogen sul
