Female Hsd:Athymic nude-Foxn1nu sentinel mice spontaneously develop a life-limiting protein-losing nephropathy. We aimed to characterize this disease and identify risk factors. A new model is presented for the study of the effects of supranormal temperatures on human tumors in vivo. Human tumors heterotransplanted serially in nude mice were heated in vivo by means of local radio-frequency heating. Congenitally athymic nude (nu/nu) mice and their phenotypically normal (nu/+) euthymic littermates were exposed to Coxiella burnetii administered as small-particle aerosols. After challenge, both strains of mice became infected, as characterized by rickettsemia, viable rickettsiae in the spleen, and serological conversion. The major difference noted was that euthymic animals had cleared rickettsiae from peripheral circulation and the spleen within 14 days. The central role of the thymus in immunity was assessed in nude mice. Nudes failed to reject allografts and xenografts and to respond to foreign erythrocytes but responded normally to endotoxin and pneumococcal polysaccharide. Thymus reconstitution was demonstrated in vivo and in vitro whereas reconstitution with thymic humoral factors or polyanions was not detected. Coliform overgrowth and depressed IgA levels in nudes appeared to contribute to wasting. These data emphasize the need for thymus participation in many immune phenomena.

Experimental listeriosis in phenotypically normal (nu/+) euthymic NMRI mice has a characteristic morphology and short-term course. In contrast, listeric infection in congenitally dysthymic nude (nu/nu) mice does not proceed in clear-cut phases, develops more slowly, displays a chronic tendency from the beginning, and shows a considerably different morphology. The inability of nude mice to effectively control and terminate infection by Listeria monocytogenes obviously results from the lack of T lymphocytes.

Rifampicin and rifapentine were used in the treatment of Listeria monocytogenes infections in normal mice and congenitally athymic (nude) mice. Results were compared with untreated controls and with the results obtained from a previous study of ampicillin in which this model was used.
