Metabolomics-Guided Discovery of Microginin Peptides from Cultures of the Cyanobacterium Microcystis aeruginosa.

We report a mass-spectrometry-based metabolomics study of a laboratory-cultured strain of Microcystis aeruginosa (UTEX LB2385), which has led to the discovery of five peptides (1-5) belonging to the microginin class of linear cyanopeptides. The structures and configurations of these peptides were determined by spectroscopic analyses and chemical derivitization. The microginin peptides described herein are the first reported derivatives containing N-methyl methionine (1, 5) and N-methyl methionine sulfoxide (2-4). The two tripeptide microginin analogues (4, 5) identified represent the smallest

Highly plastic genome of Microcystis aeruginosa PCC 7806, a ubiquitous toxic freshwater cyanobacterium.

BACKGROUND: The colonial cyanobacterium Microcystis proliferates in a wide range of freshwater ecosystems and is exposed to changing environmental factors during its life cycle. Microcystis blooms are often toxic, potentially fatal to animals and humans, and may cause environmental problems. There has been little investigation of the genomics of these cyanobacteria. RESULTS: Deciphering the 5,172,804 bp sequence of Microcystis aeruginosa PCC 7806 has revealed the high plasticity of its genome: 11.7% DNA repeats containing more than 1,000 bases, 6.8% putative transposases and 21 putative restr

Characteristics of toxin production and release in Microcystis aeruginosa exposed to three tetracycline antibiotics.

The application of antibiotics is remarkable globally. However, antibiotic residues in aquatic ecosystems can pose adverse effects on nontarget plants and animals. In the present study, the cell growth properties and the production and release of microcystin-LR and microcystin-RR in cyanobacterium Microcystis aeruginosa after exposure to three typical antibiotics were investigated. Results showed that the order of the degree of chronic toxicities of the three chemicals was oxytetracycline hydrochloride (OTC) > tetracycline hydrochloride (TC) > chlortetracycline hydrochloride (CTC). The target

Influence of coexisting spiramycin contaminant on the harm of Microcystis aeruginosa at different nitrogen levels.

The influence of nitrogen on the effects of the antibiotic contaminant spiramycin on Microcystis aeruginosa was studied through a 7-day exposure test. At current contamination levels of 0.5-100 mg L(-1) for nitrogen and 0.1-0.4 μg L(-1) for spiramycin, the two factors significantly interacted with each other (p<0.05) on the synthesis of chlorophyll-a and protein, the production and release of microcystins as well as the expression of ntcA gene and mcyB gene in M. aeruginosa. Nitrogen significantly affected the toxicity of spiramycin on algal growth (p<0.05) via regulation of protein synthesis.

Impact of a toxic and a non-toxic strain of Microcystis aeruginosa on the crayfish Procambarus clarkii.

The occurrence of cyanobacteria in many water bodies where crayfish such as Procambarus clarkii are abundant leads to the possibility of toxin accumulation and food chain transfer. This paper describes the accumulation and depuration of microcystins from a microcystin and a non-microcystin producing strain of Microcystis aeruginosa, on the survivorship, growth and nutritional status of P. clarkii. Crayfish larvae were resistant to cyanobacteria and their toxins, surviving cyanobacteria densities during acute exposures. Juvenile crayfish tolerated toxic cyanobacteria better than non-toxic ones.

Acclimation temperature influences phage susceptibility in a toxin-producing strain of Microcystis aeruginosa.

Microcystis aeruginosa is a freshwater cyanobacterium known for its role in the formation of harmful algal blooms. However, less well characterized are the host-phage interactions that occur in these toxic blooms. Here, we describe the relationship between a microcystin-producing strain of M. aeruginosa, NIES-298, and a lytic bacteriophage Ma-LMM01. In this study, we explored how host pre-acclimation to different temperature conditions influenced sensitivity to subsequent infection by Ma-LMM01. We focused on 19°C, a cooler temperature often associated with microcystin-producing blooms in sprin

Pharmaceutical cocktails in aquatic ecosystems: unveiling the hidden ecotoxicological threats to Microcystis aeruginosa.

Pharmaceutical compounds (PhACs), continuously released into the environment, represent a growing ecotoxicological threat to aquatic ecosystems. While the toxicity of individual pharmaceuticals has been extensively investigated, the effects of complex mixtures, the so-called "cocktail effect", remain poorly understood, particularly for primary producers such as cyanobacteria. This study evaluates the individual and combined effects of four commonly detected pharmaceuticals in aquatic environments: acetaminophen (APAP), atenolol (ATN), carbamazepine (CBZ), and diazepam (DZP), using the model cy

Multiple stressors enhance Microcystis dominance and modulate phycospheric antibiotic resistome in aquatic mesocosm.

Assessing the response of phycospheric antibiotic resistance genes (ARGs) to pollutants such as antibiotics and microplastics (MPs) under the background of climate warming is crucial for understanding ARG fate in aquatic ecosystems. In this work, we conducted mesocosm experiments to evaluate how these stressors influence phytoplankton dominance, phycospheric bacteria, and ARG evolution. Results showed that in Microcystis-dominant ecosystem, a single stressor strengthened Microcystis dominance (up to 82.37 % of total abundance), but only warming significantly enhanced phycospheric bacterial met

Toxin Release of Cyanobacterium Microcystis aeruginosa after Exposure to Typical Tetracycline Antibiotic Contaminants.

The global usage of veterinary antibiotics is significant. Antibiotics can be released into aquatic environments and elicit toxic effects on non-target organisms. In this study, the growth characteristics and toxin release of the cyanobacterium Microcystis aeruginosa (M. aeruginosa) were examined to investigate the physiological effects of tetracycline antibiotics on aquatic life. Results showed that the degree of toxicities of the following target antibiotics was TC (tetracycline hydrochloride) > CTC (chlortetracycline hydrochloride) > OTC (oxytetracycline hydrochloride) in terms of growth pa

Determination of FVIIa-sTF Inhibitors in Toxic Microcystis Cyanobacteria by LC-MS Technique.

The blood coagulation cascade involves the human coagulation factors thrombin and an activated factor VII (fVIIa). Thrombin and fVIIa are vitamin-K-dependent clotting factors associated with bleeding, bleeding complications and disorders. Thrombin and fVIIa cause excessive bleeding when treated with vitamin-K antagonists. In this research, we explored different strains of toxic Microcystis aeruginosa and cyanobacteria blooms for the probable fVIIa-soluble Tissue Factor (fVIIa-sTF) inhibitors. The algal cells were subjected to acidification, and reverse phase (ODS) chromatography-solid phase ex

Aquatic environmental safety assessment and inhibition mechanism of chemicals for targeting Microcystis aeruginosa.

Cyanobacteria are a diverse group of Gram-negative bacteria that produce an array of secondary compounds with selective bioactivity against vertebrates, invertebrates, fungi, bacteria and cell lines. Recently the main methods of controlling cyanobacteria are using chemicals, medicinal plants and microorganism but fewer involved the safety research in hydrophytic ecosystems. In search of an environmentally safe compound, 53 chemicals were screened against the developed heavy cyanobacteria bloom Microcystis aeruginosa using coexistence culture system assay. The results of the coexistence assay s

Metabolomics and genomics reveal high diversity and concentrations of cyanopeptides during a Microcystis bloom.

Cyanobacterial blooms are an immense global problem that release complex mixtures of poorly characterized biologically active cyanopeptides into freshwater. In this study, metabolomics and genomics were used to assess the diversity and concentrations of cyanopeptides during a dense Microcystis bloom during the late summer of 2023 in Lake Champlain, a large transboundary lake situated between Canada and the United States. Despite the relatively low genetic diversity of the bloom determined by 16S rRNA metabarcoding, 151 cyanopeptides were detected by non-targeted metabolomics. This represents t

Role of illumination intensity in microcystin development using Microcystis aeruginosa as the model algae.

Microcystis aeruginosa (M. aeruginosa) is one of the most common genera of cyanobacteria in algal blooms. In the present work, the impact of the illumination intensity on the growth of M. aeruginosa has been studied and a grinding method for the extraction of intracellular microcystins (MCs) was developed. The variations of algal density, pH, total phosphorus (TP), and total nitrogen (TN) have been investigated during MCs' culturing period. Results showed that the extraction efficiency of MC-YR by the grinding method was 275% higher than the sonication method, and the extraction efficiencies o

Effect of high irradiance and iron on volatile odour compounds in the cyanobacterium Microcystis aeruginosa.

The cyanobacterium, Microcystis aeruginosa was exposed to direct sunlight for 3, 6 or 9 h in media containing either low or high concentrations of iron, in order to determine any effects on the composition of volatile odour compounds (VOCs) released under photooxidative conditions. The most abundant VOCs detected included aliphatic hydrocarbons (C15-C21), naphthalene and the terpenoid compounds, beta-cyclocitral, and beta-ionone. Exposure to sunlight and low iron concentrations resulted in a decrease in beta-cyclocitral, beta-ionone, heptadecane and the total VOCs concentration after 9 h with
