Development of electrochemical microbiochip for the biological diagnosis of Neisseria gonorrhoeae.

A sexually transmitted disease is an illness that has a high probability of transmission between humans or animals who have sexual contact. Our research is based on the development of a microbiochip for Neisseria gonorrhoeae (N.G.). In our study, we have employed fusion technology between microarray technology and a microfluidic system for quantitative analysis of N.G. A great deal of attention has been focused on electrochemical detection by using a DNA probe, which is a specific DNA sequence and binds to a target biomolecule, because of high affinity, ease of usage, and fast measurement. The

Ferritin nanocages limit stable albumin and fibrinogen adsorption on gold nanoparticles under microfluidic flow.

Ferritin-encapsulated gold nanoparticles (AuFt) resist formation of a detectable stable protein corona under laminar microfluidic flow conditions. This ferritin "stealth" behaviour preserves AuFt monomers and suggests that ferritin coating improves nanoparticle biological identity for in vivo biomedical applications.

A Microfluidic Method for Simultaneous Assessment of Blood Viscosity and Red Blood Cell Aggregation During Continuous Syringe Delivery.

Accurate assessment of blood viscosity and red blood cell (RBC) aggregation under continuous flow is important for hemorheological analysis. However, simultaneous measurement remains challenging because both properties are influenced by flow conditions and RBC sedimentation. In this study, a microfluidic method is developed for the simultaneous measurement of blood viscosity and RBC aggregation index (AI) during continuous blood delivery from a driving syringe. The proposed device consists of a viscosity-sensing channel for viscosity measurement and aggregation-sensing channel for AI evaluatio

Design and validation of a flowless gradient generating microfluidic device for high-throughput drug testing.

Drug testing is a vital step in the identification of the potential efficacy of any new/existing drug and/or combinations of drugs. The conventional methods of testing the efficacy of new drugs using multiwell plates are time consuming and prone to evaporation loss and manual error. Microfluidic devices with automated generation of concentration gradients provide a promising alternative. The implementation of such microfluidic devices is still limited owing to the additional expertise and facilities required to fabricate and run these devices. Conventional microfluidic devices also need pumps,

A flexible and cost-effective manual droplet operation platform for miniaturized cell assays and single cell analysis.

Herein, we developed a flexible and cost-effective manual droplet operation system (MDOS) for performing miniaturized cell assays as well as single cell analysis. The MDOS consists of a manual x-y-z translation stage for liquid transferring and switching, a high-precision syringe pump for liquid driving and metering, a tapered capillary probe for droplet manipulation, a droplet array chip for droplet loading and reaction, sample/reagent reservoirs for storage, and a microscope for droplet observation, with a total expense of only $4,000. By using the flexible combination of three elementary op

Gravity-driven microfluidic synthesis of lipid nanoparticles via vibrating sharp-tip mixing.

Lipid nanoparticles (LNPs) are widely used in nanomedicine, yet their energy-efficient manufacture with precise control over particle uniformity remains challenging. Here we report a gravity-driven microfluidic platform that integrates acoustically actuated sharp-tip mixing to enable high-throughput LNPs synthesis without the need for active pumps. By optimizing microchannel geometry, actuation conditions, and the spatial positioning of the mixing vortex, we achieve stable gravity-driven flow and complete mixing at total flow rates (TFRs) of up to 5 ml min-1. The optimized system produces LNPs

Mechanostimulation of Multicellular Organisms Through a High-Throughput Microfluidic Compression System.

During embryogenesis, coordinated cell movement generates mechanical forces that regulate gene expression and activity. To study this process, tools such as aspiration or coverslip compression have been used to mechanically stimulate whole embryos. These approaches limit experimental design as they are imprecise, require manual handling, and can process only a couple of embryos simultaneously. Microfluidic systems have great potential for automating such experimental tasks while increasing throughput and precision. This article describes a microfluidic system developed to precisely compress wh

Direct loading of blood for plasma separation and diagnostic assays on a digital microfluidic device.

Finger-stick blood sampling is convenient for point of care diagnostics, but whole blood samples are problematic for many assays because of severe matrix effects associated with blood cells and cell debris. We introduce a new digital microfluidic (DMF) diagnostic platform with integrated porous membranes for blood-plasma separation from finger-stick blood volumes, capable of performing complex, multi-step, diagnostic assays. Importantly, the samples can be directly loaded onto the device by a finger "dab" for user-friendly operation. We characterize the platform by comparison to plasma generat

Effects of Flow-Induced Microfluidic Chip Wall Deformation on Imaging Flow Cytometry.

Imaging flow cytometry is a powerful tool by virtue of its capability for high-throughput cell analysis. The advent of high-speed optical imaging methods on a microfluidic platform has significantly improved cell throughput and brought many degrees of freedom to instrumentation and applications over the last decade, but it also poses a predicament on microfluidic chips. Specifically, as the throughput increases, the flow speed also increases (currently reaching 10 m/s): consequently, the increased hydrodynamic pressure on the microfluidic chip deforms the wall of the microchannel and produces

The Amplification of Alpha-Synuclein Amyloid Fibrils is Suppressed under Fully Quiescent Conditions.

Seed amplification assays (SAAs) are a promising avenue for the early diagnosis of neurodegenerative diseases. However, when amplifying fibrils from patient-derived samples in multiwell plates, it is currently highly challenging to accurately quantify the aggregates. It is therefore desirable to transfer such assays into a digital format in microemulsion droplets to enable direct quantification of aggregate numbers. To achieve transfer from conventional plate-based to the microfluidic digital format, effective seed amplification needs to be achieved inside the microdroplets. Therefore, we esta

Integrated Acoustofluidic Manipulation and Oscillation-Stabilized Magnetic Relaxation Biosensing for Salmonella Detection.

Reliable pathogen detection in complex suspensions requires both effective sample handling and stable signal generation. Here, we report an acoustofluidic-magnetic relaxation biosensing (MRB) platform for Salmonella detection that combines acoustofluidic particle manipulation with oscillation-stabilized magnetic readout. A cylindrical-array region enhances encounters between Salmonella and immunomagnetic nanobeads (IMNBs) through convective mixing, while a butterfly-shaped acoustic streaming vortex (BSAV) region preferentially retains micron-scale Salmonella-IMNB complexes and generates a nano

Synergistic Integration of Whole-Cell Biosensors and Droplet Microfluidics for High-Throughput Microbial Screening.

High-throughput screening (HTS) is essential for developing microbial cell factories, yet traditional droplet microfluidics often struggles to detect nonfluorescent metabolites. This perspective explores the whole-cell biosensor-encapsulated droplet reactor-based microbial high-throughput screening system (WCB-DHTS) as a versatile solution, which decouples production from sensing to reduce metabolic stress. By using engineered sensor strains to convert target metabolite concentrations into fluorescence signals, this platform enables the detection of diverse products within picoliter-scale drop

Computational Assessment of Oxygen Availability and Shear Stress in Microfluidic Cell Culture Chambers for Optimized Cell Adhesion.

Microfluidic cell culture systems provide controlled microscale environments for biomedical research; however, cell viability within closed microchambers depends on adequate oxygen availability during the adhesion phase and on the mechanical stresses generated after perfusion begins. Experimental characterization of oxygen depletion and local shear stress remains challenging due to the small dimensions involved and the complexity of transport phenomena. In this study, a computational framework was developed to assess oxygen transport and hydrodynamic shear stress in an SU-8-based microfluidic

Droplet Microfluidic Hydrogen/Deuterium Exchange for Investigating Protein Dynamics with Millisecond Precision.

Hydrogen/Deuterium eXchange (HDX) methods for studying protein dynamics would benefit from millisecond-scale incubations to probe intrinsically disordered proteins, highly dynamic regions, and conformation changes. Here, we investigate droplet microfluidics for rapid mixing to trigger D2O labeling, uniform incubations, and rapid droplet merging for acid quenching in advance of mass spectrometry. A surfactant-free merging approach combining expansion elements for synchronized droplet collision proved robust. The high diffusive flux of D2O and protons enables microsecond mixing to trigger and ar

Integrating Low-Voltage Microfluidics With On-Chip Electrospray Ionization for Miniaturized Protein Analysis.

Low-input proteomics requires analytical strategies that can handle very small volumes while minimizing loss, dilution, and handling steps. Low-voltage electrokinetic microfluidics offers a promising route to this goal by enabling controlled transport, separation, concentration, and droplet manipulation within compact chip-based platforms. This review examines the integration of electrokinetic microfluidics with on-chip electrospray ionization (ESI) for miniaturized protein analysis, with emphasis on how electric-field-driven sample handling can be combined with sensitive mass spectrometric de

Origami-Paper Microfluidics for Point-of-Need Analysis.

Point-of-care (POC) diagnostics has attracted increasing attention in biomedical research. However, it remains challenging in low resources setting. Origami-paper microfluidics has emerged as a promising approach for point-of-need (PON) analysis by leveraging the advantages of paper, including low cost, zero power, easy to access, biodegradability, and capillary-driven fluid transport. These features enable the development of PON platforms suitable for resource-limited environments without centralized instrumentation and trained personnel. This perspective highlights recent advances in origami

On-Demand Viscosity Gradients Turn Nanoliter Droplets into Programmable Sources of Enriched Picoliter Droplets.

Droplet microfluidics has revolutionized diagnostics, chemical synthesis, and biotechnology. While droplet merging and mixing are routine operations, the opposite processes─coupled enrichment and pinch-off of reagents, analytes, or products─is nontrivial. If accessed, these operations are highly valuable, enabling improved reaction kinetics, increased assay sensitivity, and purification. In this work, we present coupled enrichment and on-demand emission of picoliter-scale daughter droplets from nanoliter-scale parents. The reported approach leverages electrokinetically driven viscosity gradien

Microfluidic Platforms for Modeling Cancer-Associated Thrombosis: Current Status and Future Directions.

Cancer-associated thrombosis (CAT) is the second leading cause of death among cancer patients. Its pathogenesis is multifactorial and involves dynamic interactions among tumor cells, platelets, coagulation factors, endothelial cells, immune cells, and blood flow. Traditional in vitro 2-dimensional models fail to accurately mimic the dynamic microenvironment of CAT in humans. Although animal models can mimic complex pathological conditions, significant differences in the composition and regulation of the coagulation system between animals and humans limit their ability to accurately model CAT i
