In recent years, rising global protein demand and increasing awareness of sustainability, climate change, and animal welfare issues have accelerated the shift toward plant-based foods. Despite their promise as sustainable animal-protein alternatives, plant proteins are often limited by inherent astringency associated with high friction and poor lubrication. In this work, pH-responsive microgels derived from liquid-liquid phase separation (LLPS) of deamidated β-conglycinin (D-β-CG) and chitosan oligosaccharide (COS) were engineered to modulate interfacial behavior and lubrication in dairy syste

This study presents a novel approach for the synthesis of biodegradable cell-laden microgels using stop-flow lithography (SFL), addressing critical challenges in the field of tissue engineering. Traditional methods for creating 3D cell cultures often rely on non-biodegradable materials, which limit their application and raise concerns about cell viability. In this work, we successfully replace poly(ethylene glycol) diacrylate (PEGDA) with dextran-2-hydroxyethyl methacrylate (dex-HEMA), a biocompatible and biodegradable alternative. Furthermore, we introduce a technical solution for sterile cel

Functionalized hydrogel microspheres hold great promise in tissue regeneration, yet their clinical translation is often hindered by high production costs, complex fabrication processes, and reliance on external bioactive additives. To overcome these challenges, multifunctional gelatin-tea polyphenol (GTP) microgels using a simplified, cost-effective, microfluidics-inspired platform are reported, which is composed of an injection unit, a reciprocating apparatus, and a collection substrate. Using a microinjection pump, uncrosslinked microspheres with precisely controlled diameters (125-570 µm) a

Islet transplantation to treat insulin-dependent diabetes is greatly limited by the need for maintenance immunosuppression. We report a strategy through which cotransplantation of allogeneic islets and streptavidin (SA)-FasL-presenting microgels to the omentum under transient rapamycin monotherapy resulted in robust glycemic control, sustained C-peptide levels, and graft survival in diabetic nonhuman primates for >6 months. Surgical extraction of the graft resulted in prompt hyperglycemia. In contrast, animals receiving microgels without SA-FasL under the same rapamycin regimen rejected islet

Articular cartilage is comprised of two main components, the extracellular matrix (ECM) and the pericellular matrix (PCM). The PCM helps to protect chondrocytes in the cartilage from mechanical loads, but in patients with osteoarthritis, the PCM is weakened, resulting in increased chondrocyte stress. As chondrocytes are responsible for matrix synthesis and maintenance, it is important to understand how mechanical loads affect the cellular responses of chondrocytes. Many studies have examined chondrocyte responses to in vitro mechanical loading by embedding chondrocytes in 3-D hydrogels. Howeve

Microgels, a microscale variant of hydrogels (1-100 µm), exhibit high surface area and responsiveness to external stimuli while retaining the soft, viscoelastic nature of their macroscale counterparts. While microgels can be derived from both synthetic and natural polymers, protein-based microgels offer significant advantages due to their diverse function and activities. However, traditional fabrication methods, such as microfluidics and emulsion-based techniques, often involve trade-offs between scalability, structural integrity, and functionality. To overcome these limitations, liquid-liquid

Delivery of therapeutic compounds via biomaterial systems has shown promise for tissue regeneration following central nervous system (CNS) injuries. Stromal cell-derived factor-1a (SDF-1a) modulates progenitor cell recruitment to neural injury sites and may contribute to neural repair. However, SDF-1a has a short half-life and requires a delivery system to both protect and sustain its release. Here, we sought to develop a drug delivery platform capable of releasing SDF-1a in a controlled fashion while minimizing inflammation. We used modified hyaluronic acid and microfluidics to generate monod

Scars composed of fibrous connective tissues are natural consequences of injury upon incisional wound healing in soft tissues. Hydrogels that feature a sustained presentation of immunomodulatory cytokines are known to modulate wound healing. However, existing immunomodulatory hydrogels lack interconnected micropores to promote cell ingrowth. Other limitations include invasive delivery procedures and harsh synthesis conditions that are incompatible with drug molecules. Here, hybrid nanocomposite microgels containing interleukin-10 (IL-10) are reported to modulate tissue macrophage phenotype du

T cells are predominantly produced by the thymus and play a significant role in maintaining our adaptive immune system. Physiological involution of the thymus occurs gradually with age, compromising naive T cell output, which can have severe clinical complications. Also, T cells are utilized as therapeutic agents in cancer immunotherapies. Therefore, there is an increasing need for strategies aimed at generating naive T cells. The majority of in vitro T cell generation studies are performed in two-dimensional (2D) cultures, which ignore the physiological thymic microenvironment and are not sca

Microgels offer unique advantages over bulk hydrogels due to their improved diffusion limits for oxygen and nutrients. Particularly, stimuli-responsive microgels with inherently bioactive and self-supporting properties emerge as highly promising biomaterials. This study unveils the development of interparticle-crosslinked, self-supporting, ion-responsive microgels tailored for 3D and 4D (bio)printing applications. A novel strategy was proposed to develop microgels that enabled interparticle crosslinking, eliminating the need for filler hydrogels and preserving essential microscale void spaces

In this study, a one-step method is discussed for producing uniform cell-sized microgels using glass capillaries filled with a binary polymer blend of polyethylene glycol (PEG) and gelatin. Upon decreasing temperature, phase separation of the PEG/gelatin blends and gelation of gelatin occur, and then the polymer blend forms linearly aligned, uniformly sized gelatin microgels in the glass capillary. When DNA is added to the polymer solution, gelatin microgels entrapping DNA are spontaneously formed, and the DNA prevents the coalescence of the microdroplets even at temperatures above the melting

Platelets crucially facilitate wound healing but can become depleted in traumatic injury or chronic wounds. Previously, our group developed injectable platelet-like particles (PLPs) comprised of highly deformable, ultralow crosslinked pNIPAm microgels (ULCs) coupled to fibrin binding antibodies to treat post-trauma bleeding. PLP fibrin-binding facilitates homing to sites of injury, promotes clot formation, and, due to high particle deformability, induces clot retraction. Clot retraction augments healing by increasing clot stability, enhancing clot stiffness, and promoting cell migration into t

Hemostatic immaturity in neonates presents critical challenges, especially during surgery, where bleeding and thrombosis risks are elevated. Current treatments rely on transfusing adult blood products, which may cause complications resulting from structural and functional differences between neonatal and adult fibrinogen. To address this, we developed hemostatic B-knob-triggered microgels (BK-TriGs) that target and bridge fibrinogen hole b sites. Functionalized with a fibrin hole b-specific peptide, BK-TriGs enhance clot density and resistance to degradation. In vitro studies using neonatal pl

Over the past two decades, microfluidic approaches have been extensively explored for the production of monodisperse alginate hydrogel microparticles, particularly for applications such as 3D in vitro cell culture in tissue engineering. However, conventional methods often suffer from satellite droplet contamination and inefficient oil-to-water phase transfer, limiting purity and scalability. Here, we present an integrated microfluidic platform for the continuous, on-chip production, gelation, extraction, and purification of monodisperse alginate hydrogel microparticles. The device combines a f

Intestinal Salmonella infections are primarily treated with antibiotics, yet non-selective bactericidal activity often disrupts the gut microbiota and limits therapeutic efficacy. Here, we introduce a precision anti-infective strategy based on spatial reprogramming, which redefines where antibacterial activity occurs rather than increasing antimicrobial potency. By functionally integrating Salmonella-specific nanobodies anchored on oral hydrogel microgels with meropenem-loaded bacterial membrane vesicles, we construct a targeted delivery system, termed HAMA-Nb@BM. Surface-anchored nanobodies s

Helicobacter pylori (H. pylori) is a prevalent global pathogen responsible for gastritis and the potential development of gastric cancer. Sulforaphane (SFN), a foodborne compound, exhibits notable antibiotic properties against H. pylori. However, its utility is limited by poor stability and susceptibility to environmental degradation. Here, we developed a gastric-responsive-release microgel for delivering anti-H. pylori SFN. The microgels were prepared by cross-linking α-lactalbumin nanotubes then coated with chitosan (CTS-MGs). SFN was loaded into microgels with a loading rate of 10.73 ± 0.25

HYPOTHESIS: Recently, the anomalous shrinkage of surface-supported hyaluronate/poly-l-lysine (HA/PLL) microgels (µ-gels), which exceeds that reported for any other multilayer-based systems, has been reported [1]. The current study investigates the capability of these unique µ-gels for the encapsulation and retention of macromolecules, and proposes the shrinkage-driven assembly of biopolymer-based µ-gels as a novel tool for one-step surface biofunctionalization.

The complexation of cationic antimicrobials with polyanionic microgels on a biomaterial surface can render that surface self-defensive against bacteria by killing those bacteria which physically contact the antimicrobial-loaded microgels. This killing has been attributed to the contact-driven transfer of antimicrobial from a microgel to a challenging bacterium, though much remains unknown about this process. Here we use a combination of experiments and computational modeling to identify key aspects of the complexation phenomena which influence the self-defensive properties. We synthesize poly(

The generation of foams using particle-like objects has gained momentum in the past years as such Pickering foams are relevant in many industrial applications. In this context, we investigate the foamability of microgels, which are soft polymeric particles which architecture can be finely tailored during synthesis. Herein, foams were produced by continuously bubbling air into dispersions of poly(N-isopropylacrylamide) (PNIPAM) microgels. The foaming ability of the microgel dispersions has been assessed by combining observations at both the macroscopic and the local scales. Increased microgel c
