Human Organ-on-a-Chip Microphysiological Systems to Model Musculoskeletal Pathologies and Accelerate Therapeutic Discovery.

Human Microphysiological Systems (hMPS), otherwise known as organ- and tissue-on-a-chip models, are an emerging technology with the potential to replace in vivo animal studies with in vitro models that emulate human physiology at basic levels. hMPS platforms are designed to overcome limitations of two-dimensional (2D) cell culture systems by mimicking 3D tissue organization and microenvironmental cues that are physiologically and clinically relevant. Unlike animal studies, hMPS models can be configured for high content or high throughput screening in preclinical drug development. Applications

Application of microphysiologic system to assess neutrophil extracellular trap in xenotransplantation.

Transplantation of organs, cells, or tissues from one species to another, known as xenotransplantation, has the potential to alleviate organ donor shortages and enhance the success of organ transplantation. However, the possibility of immunological rejection by the recipient is one of the biggest difficulties associated with xenotransplantation. The creation of neutrophil extracellular traps (NETs), also known as NETosis, is hypothesized as a mechanism of rejection. Innovations in microfluidics and co-culturing techniques have provided access to several classes of microengineered model systems

Modeling statin myopathy in a human skeletal muscle microphysiological system.

Statins are used to lower cholesterol and prevent cardiovascular disease. Musculoskeletal side effects known as statin associated musculoskeletal symptoms (SAMS), are reported in up to 10% of statin users, necessitating statin therapy interruption and increasing cardiovascular disease risk. We tested the hypothesis that, when exposed to statins ex vivo, engineered human skeletal myobundles derived from individuals with (n = 10) or without (n = 14) SAMS and elevated creatine-kinase levels exhibit statin-dependent muscle defects. Myoblasts were derived from muscle biopsies of individuals (median

An eighteen-organ microphysiological system coupling a vascular network and excretion system for drug discovery.

Physiological supporting systems, such as the vascular network and excretion system, are crucial for the effective functioning of organs. This study demonstrates that when a body-on-a-chip microdevice is coupled with miniaturized physiological support systems, it can create a multi-organ microphysiological system capable of more accurately mimicking the physiological complexity of a body, thereby offering potential for preclinical drug testing. To exemplify this concept, we have developed a model system comprising 18 types of microtissues interconnected by a vascular network that replicates th

Recent advances in sex-specific reproductive microphysiological systems for the systematic evaluation of assisted reproductive technology.

The rising global burden of infertility continues to increase clinical reliance on assisted reproductive technology (ART), yet overall outcomes remain constrained by limited physiological fidelity of conventional two-dimensional (2D) culture and animal models. Microphysiological systems (MPSs) offer an in vitro platform with enhanced human physiological relevance that can reconstruct reproductive microenvironments through engineered compartmentalization, quantitative fluid flow control, and stable long-term culture. Here, we systematically map recent advances in sex-specific reproductive MPS a

Placental microphysiological systems: new advances on promising platforms that mimic the microenvironment of the human placenta.

One of the most complex human physiological processes to study is pregnancy. Standard animal models, as well as two-dimensional models, lack the complexity and biological relevance required to accurately study such a physiological process. Recent studies have focused on the development of three-dimensional models based on microfluidic systems, designated as placental microphysiological systems (PMPSs). PMPS devices provide a model of the placental barrier through culturing relevant cell types in specific arrangements and media to mimic the in vivo environment of the maternal-fetal circulation.

Inversion and computational maturation of drug response using human stem cell derived cardiomyocytes in microphysiological systems.

While cardiomyocytes differentiated from human induced pluripotent stems cells (hiPSCs) hold great promise for drug screening, the electrophysiological properties of these cells can be variable and immature, producing results that are significantly different from their human adult counterparts. Here, we describe a computational framework to address this limitation, and show how in silico methods, applied to measurements on immature cardiomyocytes, can be used to both identify drug action and to predict its effect in mature cells. Our synthetic and experimental results indicate that optically o

Quercetin Alleviates Intestinal and Liver Dysfunction Induced by Hypoxia in a Microphysiological System.

Pathological hypoxia such as high-altitude exposure is harmful to the human body system and multiply organs. However, the systemic damage effects of hypoxia on the intestine and liver tissue remain unclear due to the limitations of traditional 2D cell cultures and animal models. Here, we establish a biomimetic gut-liver microphysiological system (GLMPS) as a human model of the gut-liver axis by coculturing human gut and liver cell lines interconnected under circulating fluid flow, for investigating the mechanisms of hypoxia-induced injury in the intestine and liver. Chemical Distribution Kinetics of Five Polycyclic Aromatic Hydrocarbons in a Microphysiological System.

As next-generation risk assessment accelerates the shift towards non-animal models, it also necessitates the development of tools that quantify the chemical in vitro distribution to derive an effective concentration. Although several predictive models have already been developed, none have thus far specifically focused on microphysiological systems. Unlike static well plates, microphysiological systems use (micro-) fluidic flow, consist of largely hydrophobic plastic surfaces, and often rely on protein-rich hydrogel matrices for cell scaffolding. However, these properties can alter the nominal

A microphysiological system HHT-on-a-chip platform recapitulates patient vascular lesions.

Hereditary Hemorrhagic Telangiectasia (HHT) is a rare congenital disease in which fragile vascular malformations (VM) focally develop in multiple organs. There are few treatment options and no cure. HHT patients inherit loss-of-function mutations affecting Alk1-Eng signaling; however, why this manifests as VMs remains poorly understood. Here we have developed a fully human cell-based microphysiological system of perfused vasculature in which inducible shRNA controls endogenous Alk1 in primary endothelial cells (EC). Resulting VMs develop over several days, recapitulate patient VM appearance, a

A Microphysiological System for Studying Barrier Health of Live Tissues in Real Time.

Epithelial cells create barriers that protect many different components in the body from their external environment. The gut in particular carries bacteria and other infectious agents. A healthy gut epithelial barrier prevents unwanted substances from accessing the underlying lamina propria while maintaining the ability to digest and absorb nutrients. Increased gut barrier permeability, better known as leaky gut, has been linked to several chronic inflammatory diseases. Yet understanding the cause of leaky gut and developing effective interventions are still elusive due to the lack of tools to

Pumpless, unidirectional microphysiological system for testing metabolism-dependent chemotherapeutic toxicity.

Drug development is often hindered by the failure of preclinical models to accurately assess and predict the efficacy and safety of drug candidates. Body-on-a-chip (BOC) microfluidic devices, a subset of microphysiological systems (MPS), are being created to better predict human responses to drugs. Each BOC is designed with separate organ chambers interconnected with microfluidic channels mimicking blood recirculation. Here, we describe the design of the first pumpless, unidirectional, multiorgan system and apply this design concept for testing anticancer drug treatments. HCT-116 colon cancer

Reconstructing AF-associated atrial fibrosis: Patient-specific iPSC models, fit-for-purpose atrial microphysiological systems, and nanomedicine.

Atrial fibrillation (AF)-associated fibrosis is a heterogeneous component of atrial cardiomyopathy that contributes to conduction slowing, anisotropy, reentry susceptibility, and reduced therapeutic responsiveness. However, translation of anti-fibrotic strategies remains limited by the biological heterogeneity of AF and by models that incompletely reproduce human atrial cell composition, mechanics, perfusion, and electrophysiology. This narrative review critically evaluates the convergence of patient-specific induced pluripotent stem cell (iPSC) models, biomimetic atrial-on-a-chip platforms, a

A human airway-on-a-chip microphysiological system for modeling chlorine gas toxicity.

There is a critical need to understand pathophysiological mechanisms involved in injury from acute chlorine gas (Cl2) exposure. Limited information is available regarding the time course and mechanisms of injury after acute Cl2 exposure due to a lack of human clinical data and limited fidelity of pre-clinical animal models. We designed and integrated a Cl2 exposure platform to generate and deliver precise concentrations of Cl2 to a microfluidic human airway-on-a-chip microphysiological system. Chemical, biological, structural, and functional airway-on-a-chip responses to Cl2 exposure were char

Modeling human enterovirus A71 infection using an intestinal microphysiological system.

Enterovirus A71 (EV-A71), a causative virus of hand, foot, and mouth disease, primarily infects and replicates in the intestine and, in severe cases, spreads to the central nervous system, leading to neurological complications. Therefore, suppressing viral replication in the intestine is important to prevent severe complications. However, the intestinal pathophysiological changes in EV-A71-infected patients remain poorly understood. In this study, we aimed to examine the intestinal response to EV-A71 infection using the intestinal microphysiological system (MPS) we previously developed using h

Modeling sympathetic neuro-cardiac interactions in a hiPSC-based microphysiological system.

The cardiac autonomic nervous system is a key driver of various cardiac disorders and arrhythmias. However, investigating neuronal regulation of the human heart has proven difficult due to the limited availability of reliable experimental models. Here, we present a novel microphysiological system using a compartmentalized microfluidic device (MFD) to integrate co-cultured human induced pluripotent stem cell (hiPSC)-derived cardiomyocytes (hiPSC-CMs) and sympathetic neurons (hiPSC-SNs). MFD is composed of two wide-open chambers separated by microfluidic microchannels. Patch-clamp electrophysiol

Assessing immune hepatotoxicity of troglitazone with a versatile liver-immune-microphysiological-system.

Drug-induced liver injury is a prevalent adverse event associated with pharmaceutical agents. More significantly, there are certain drugs that present severe hepatotoxicity only during the clinical phase, consequently leading to the termination of drug development during clinical trials or the withdrawal from the market after approval. The establishment of an evaluation model that can sensitively manifest such hepatotoxicity has always been a challenging aspect in drug development. In this study, we build a liver-immune-microphysiological-system (LIMPS) to fully demonstrate the liver injury tr

Biomimetic human liver MPS have evolved from simpler 2D cell models, spheroids and organoids to address the increasing need to understand patient-specific mechanisms of complex and rare diseases, the response to therapeutic treatments, and the absorption, distribution, metabolism, excret
