Multimode objective lens for momentum microscopy and x-ray photoemission electron microscopy: Experiments.

A new type of objective lens has recently been proposed for use in x-ray photoemission electron microscopes (XPEEMs) and momentum microscopes. Adding a ring electrode concentric with the extractor allows the field in the gap between the sample and the extractor to be shaped. Forming a lens field in this gap reduces the field strength at the sample by up to an order of magnitude. This mitigates the risk of field emission, particularly for cleaved samples with sharp edges. A retarding field can redirect all slow electrons, thus eliminating the primary contribution to the space-charge interaction

The immotile cilia syndrome: phase contrast light microscopy, scanning and transmission electron microscopy.

In the immotile cilia syndrome, transmission electron microscopy of the cilia shows abnormalities in the arrangement of the central pairs of tubules and in the dynein arms of the peripheral tubules, or in the radial spokes, We studied four nonrelated children, 9/12, 5, 6, and 6 years old, with situs inversus and a history of chronic sinusitis and bronchitis (Kartagener's syndrome) and four children in the same age group and with the same history, but without situs inversus. Under the phase contrast microscope no motile cilia were seen in the four patients with Kartagener's syndrome and in two

[A case of colonic polypoid ganglioneuromatosis--a light microscopy, electron microscopy and immunohistochemical study].

A 68-year-old man with colonic polypoid ganglioneuromatosis is reported. He had manifested no signs and symptoms of von Recklinghausen's disease and/or a multiple endocrine neoplasia syndrome. A single polyp was found by a barium enema and an endoscopic polypectomy was performed. Light microscopy showed the propria mucosa of the polyp was composed of a proliferation of spindle cells with a fibrillar cytoplasma and large cells with a round large nucleus, a prominent nucleolus, and vast cytoplasm. Some crypts were slightly dilated. Random biopsied specimens from the colonic normal mucosa showed

Deep learning enables stochastic optical reconstruction microscopy-like superresolution image reconstruction from conventional microscopy.

Despite its remarkable potential for transforming low-resolution images, deep learning faces significant challenges in achieving high-quality superresolution microscopy imaging from wide-field (conventional) microscopy. Here, we present X-Microscopy, a computational tool comprising two deep learning subnets, UR-Net-8 and X-Net, which enables STORM-like superresolution microscopy image reconstruction from wide-field images with input-size flexibility. X-Microscopy was trained using samples of various subcellular structures, including cytoskeletal filaments, dot-like, beehive-like, and nanoclust

Complete and rapid switch from light microscopy to virtual microscopy for teaching medical histology.

During the interim between the 2003 and 2004 academic years, the cell and tissue biology and integrated medical neuroscience courses at the Medical College of Wisconsin made a complete and rapid switch from light microscopy- to virtual microscopy-based histology laboratories. This switch was prompted by the difficulties in maintaining and the cost of replacing the college's microscopes and microscope slides, and primarily by the desire to promote and streamline learning for our large classes (n > 200) of first-year medical students. A group of students who used the virtual microscope, another

Combination of a cryosectioning method and section scanning electron microscopy for immuno-scanning electron microscopy.

We describe a novel immuno-scanning electron microscopy (SEM) technique that combines both Tokuyasu's cryosectioning and section SEM methods. In this technique, semithin cryosections, cut according to the Tokuyasu method, were adhered to glass microscope slides, immunostained for bio-molecules of interest and observed by confocal laser scanning microscopy. The same sections were subsequently embedded in epoxy resin and ultrathin sections were cut on an ultramicrotome. These were then observed by SEM using a backscattered electron detector. Correlation between immunofluorescence and SEM images

Electron microscopy: a method for the diagnosis of inherited metabolic storage diseases. Electron microscopy in diagnosis.

In the diagnosis of inherited metabolic diseases, electron microscopy has become an important method complementary to clinical, histological and biochemical assays. The characteristic ultrastructure of stored material as well as the site of accumulation in the cell are shown in a number of metabolic disorders. The most prominent advantages of electron microscopical techniques as compared to alternative techniques are discussed. One of the advantages is the fact that ultrastructural investigation requires only tissue samples of very small size, and another that its results may be obtained withi

Cytochrome nanowires under electron microscopy.

Microbial nanowires are appendages that Bacteria and Archaea use to transfer electrons to external surfaces, such as minerals, electrodes, or other microbes. While initial studies suggested that nanowires were modified pili, recent advancements in cryo-EM revealed that microbial nanowires are composed of multi-heme c-type cytochromes. In this review, we discuss the discovery of microbial nanowires, advancements that allowed elucidation of their near-atomic resolution cryo-EM structures, and the impact of heme arrangement on electron transfer. We also discuss how new structural information can

Space-time light-sheet microscopy.

Light-sheet microscopy (LSM) has revolutionized bioimaging by delivering high-contrast volumetric resolution with minimal photodamage. Spatial wavefront shaping, used to generate lattice and Airy light-sheets, has been particularly effective in advancing LSM beyond the Rayleigh limit. Despite its broad adoption, most LSM implementations rely on rigid dual-objective geometries that complicate sample handling and impose a trade-off between imaging field of view (FoV) and axial resolution. Here, we introduce space-time light-sheet microscopy (ST-LSM), a single-objective strategy that exploits spa

Intravital Microscopy in Atherosclerosis Research.

Atherosclerosis is a lipid-driven inflammatory disorder that narrows the arterial lumen and can induce life-threatening complications from coronary artery disease, cerebrovascular disease, and peripheral artery disease. On a mechanistic level, the development of novel cellular-resolution intravital microscopy imaging approaches has recently enabled in vivo studies of underlying biological processes governing disease onset and progress. In particular, multiphoton microscopy has emerged as a promising intravital imaging tool utilizing two-photon-excited fluorescence and second-harmonic generatio

Auditory ossicles in ScanningElectron Microscopy.

Introduction: Knowledge about the physiology of a healthy middle ear is essential for understanding the activity and mechanics of the ear as well as the basics of ossiculoplasty. Trauma of the epithelial lining of the tympanic cavity as well as the ossicular chain may be the result of chronic inflammation and surgery. Depending on the observed changes of the middle ear lining, there are several types of distinguished chronic inflammatory changes: simple, with cholesteatoma, with the formation of inflammatory granulation tissue, in course of specific diseases.
<b
