Biophysical characterization of lynx-nicotinic receptor interactions using atomic force microscopy.

Nicotinic acetylcholine receptors (nAChRs) are broadly expressed in the central and peripheral nervous systems, playing essential roles in cholinergic neurotransmission. The lynx family proteins, a subset of the Ly6/uPAR superfamily expressed in multiple brain regions, have been shown to bind to nAChRs and modulate their function via allosteric regulation. The binding interactions between lynx and nAChRs, however, have not been systematically quantified and compared. In this work, we characterized the interactions between lynx1 or lynx2 and α3β4- or α7-nAChRs using single-molecule atomic force

Mapping heterogeneity of cellular mechanics by multi-harmonic atomic force microscopy.

The goal of mechanobiology is to understand the links between changes in the physical properties of living cells and normal physiology and disease. This requires mechanical measurements that have appropriate spatial and temporal resolution within a single cell. Conventional atomic force microscopy (AFM) methods that acquire force curves pointwise are used to map the heterogeneous mechanical properties of cells. However, the resulting map acquisition time is much longer than that required to study many dynamic cellular processes. Dynamic AFM (dAFM) methods using resonant microcantilevers are co

Automated setpoint adjustment for biological contact mode atomic force microscopy imaging.

Contact mode atomic force microscopy (AFM) is the most frequently used AFM imaging mode in biology. It is about 5-10 times faster than oscillating mode imaging (in conventional AFM setups), and provides topographs of biological samples with sub-molecular resolution and at a high signal-to-noise ratio. Unfortunately, contact mode imaging is sensitive to the applied force and intrinsic force drift: inappropriate force applied by the AFM tip damages the soft biological samples. We present a methodology that automatically searches for and maintains high resolution imaging forces. We found that the

Assessing mutant huntingtin fragment and polyglutamine aggregation by atomic force microscopy.

Huntington disease (HD), a neurodegenerative disorder, is caused by an expansion of more than 35-40 polyglutamine (polyQ) repeats located near the N-terminus of the huntingtin (htt) protein. The expansion of the polyQ domain results in the ordered assembly of htt fragments into fibrillar aggregates that are the main constituents of inclusion bodies, which are a hallmark of the disease. This paper describes protocols for studying the aggregation of mutant htt fragments and synthetic polyQ peptides with atomic force microscopy (AFM). Ex situ AFM is used to characterize aggregate formation in pro

Microminiaturized immunoassays using atomic force microscopy and compositionally patterned antigen arrays.

This paper combines the topographic imaging capability of the atomic force microscope (AFM) with a compositionally patterned array of immobilized antigenic rabbit IgG on gold as an approach to performing immunoassays. The substrates are composed of micrometer-sized domains of IgG that are covalently linked to a photolithographically patterned array of a monolayer-based coupling agent. The immobilized coupling agent, which is prepared by the chemisorption of dithiobis(succinimidyl undecanoate) on gold, is separated by micrometer-sized grids of a monolayer formed from octadecanethiol (ODT). Measuring of Adhesion Force in the Cell-Cell System Based on Atomic Force Microscopy Technology.

We developed and tested a method for fabrication of a biosensor chip based on native human whole blood lymphocyte and a titanium tipless cantilever. The biosensor can be used for measuring intermolecular adhesion forces in the cell-cell system by using atomic force spectroscopy. The developed biosensor chip was applied for measuring adhesion force between lymphocyte and granulocyte and between lymphocyte and erythrocyte in healthy individuals and in patients with acute lymphoblastic leukemia (before treatment, during standard treatment, and during relapse). It was found that adhesion force bet

Nanostructural analysis by atomic force microscopy followed by light microscopy on the same archival slide.

Integrated information on ultrastructural surface texture and chemistry increasingly plays a role in the biomedical sciences. Light microscopy provides access to biochemical data by the application of dyes. Ultrastructural representation of the surface structure of tissues, cells, or macromolecules can be obtained by scanning electron microscopy (SEM). However, SEM often requires gold or coal coating of biological samples, which makes a combined examination by light microscopy and SEM difficult. Conventional histochemical staining methods are not easily applicable to biological material subseq

Characterization of protein immobilization on nanoporous gold using atomic force microscopy and scanning electron microscopy.

Nanoporous gold (NPG), made by dealloying low carat gold alloys, is a relatively new nanomaterial finding application in catalysis, sensing, and as a support for biomolecules. NPG has attracted considerable interest due to its open bicontinuous structure, high surface-to-volume ratio, tunable porosity, chemical stability and biocompatibility. NPG also has the attractive feature of being able to be modified by self-assembled monolayers. Here we use scanning electron microscopy (SEM) and atomic force microscopy (AFM) to characterize a highly efficient approach for protein immobilization on NPG u

Accelerating Atomic Force Microscopy Imaging based on an Optimized Path Planning Method.

Based on compressed sensing, undersampling is a low cost and efficient way to speed up the process of atomic force microscopy (AFM) imaging. The under-sampled information obtained is important in producing high-quality reconstructed images. Different samples and dynamic measurement show different characteristic of topography, which makes it impossible to acquire acceptable AFM image with same under-sampled scanning pattern. This work aims to propose an unsampling path planning method for effective under-sampled image acquisition. The preprocess is realized by object detection and k-means metho

Cross-scale high-bandwidth atomic force microscopy with a stick-slip nanopositioner.

The core of atomic force microscopy (AFM) lies in the ultra-precise scanning between the tip and sample, which is enabled by nanopositioners. State-of-the-art AFMs generate the scanning motion using direct-drive nanopositioners, possessing either long range or high bandwidth, but not both. Here we show that with a triple-phase controller, the high-bandwidth (up to 363 Hz) nano-precision scanning can also be performed with a typical stick-slip nanopositioner. More importantly, by leveraging the displacement accumulation in the stepping mode, the same system achieved a 3 mm × 3 mm XY working ran

Interspecies Comparison of Multilayer Mechanical Properties of Synovium Using Atomic Force Microscopy.

The synovium is a loose connective tissue that separates the intra-articular (IA) joint compartments of all diarthrodial joints from the systemic circulation. It can be divided into two layers: the intima, a thin and cell-dense layer atop a more heterogeneous subintima, composed of collagen and various cell types. The subintima contains penetrating capillaries and lymphatic vessels that rapidly clear injected drugs from the joint space which may vary not only with drug size and charge but also with the microstructure and composition of the intima and subintima of the synovium. Chlamydia psittaci infected cell studies by 4Pi Raman and atomic force microscopy.

Chlamydia psittaci is an avian bacterial pathogen that can cause atypical pneumonia in humans via zoonotic transmission. It is a Gram-negative intracellular bacterium that proliferates inside membrane bound inclusions in the cytoplasm of living eukaryotic cells. The study of such cells with C. psittaci inside without destroying them poses a significant challenge. We demonstrated in this work the utility of a combined multitool approach to analyze such complex samples. Atomic force microscopy was applied to obtain high-resolution images of the surface of infected cells upon entrance of bacteria

Atomic Force Microscopy Provides New Mechanistic Insights into the Pathogenesis of Pemphigus.

Autoantibodies binding to the extracellular domains of desmoglein (Dsg) 3 and 1 are critical in the pathogenesis of pemphigus by mechanisms leading to impaired function of desmosomes and blister formation in the epidermis and mucous membranes. Desmosomes are highly organized protein complexes which provide strong intercellular adhesion. Desmosomal cadherins such as Dsgs, proteins of the cadherin superfamily which interact via their extracellular domains in Ca2+-dependent manner, are the transmembrane adhesion molecules clustered within desmosomes. Investigations on pemphigus cover a wide range

Actomyosin Cortical Mechanical Properties in Nonadherent Cells Determined by Atomic Force Microscopy.

The organization of filamentous actin and myosin II molecular motor contractility is known to modify the mechanical properties of the cell cortical actomyosin cytoskeleton. Here we describe a novel method, to our knowledge, for using force spectroscopy approach curves with tipless cantilevers to determine the actomyosin cortical tension, elastic modulus, and intracellular pressure of nonadherent cells. We validated the method by measuring the surface tension of water in oil microdrops deposited on a glass surface. We extracted an average tension of T ∼ 20.25 nN/μm, which agrees with macroscopi

Sequential adaptation in latent tuberculosis bacilli: observation by atomic force microscopy (AFM).

Mycobacterium tuberculosis (MTB) can persist within the human host for years without causing disease, in a syndrome known as latent tuberculosis. The mechanisms by which M. tuberculosis establishes a latent metabolic state is unknown, but it is hypothesized that reduced oxygen tension may trigger the bacillus to enter a state of latency. Therefore, we are studying anaerobic culture of M. tuberculosis (H37RV) as a model of latency. For the first time, the sequential adaptation of latent bacilli (every 90 days for 48 months) viewed under Atomic Force Microscopy (AFM). Two types of adaptation wer

Quantitative analysis of human keratinocyte cell elasticity using atomic force microscopy (AFM).

We present the use of atomic force microscopy (AFM) to visualize and quantify the dynamics of epithelial cell junction interactions under physiological and pathophysiological conditions at the nanoscale. Desmosomal junctions are critical cellular adhesion components within epithelial tissues and blistering skin diseases such as Pemphigus are the result in the disruption of these components. However, these structures are complex and mechanically inhomogeneous, making them difficult to study. The mechanisms of autoantibody mediated keratinocyte disassembly remain largely unknown. Here, we have u

Atomic force microscopy: a powerful tool for high-resolution imaging of spermatozoa.

Atomic force microscopy (AFM) has emerged as the only technique capable of real-time imaging of the surface of a living cell at nano-resolution. Since AFM provides the advantage of directly observing living biological cells in their native environment, this technique has found many applications in pharmacology, biotechnology, microbiology, structural and molecular biology, genetics and other biology-related fields. AFM has also proved to be a valuable tool for reproductive biologists. An exhaustive review on the various applications of AFM to sperm cells is presented. AFM has been extensively

Imaging of individual biopolymers and supramolecular assemblies using noncontact atomic force microscopy.

A variety of biopolymers is imaged using noncontact atomic force microscopy. Samples are prepared by aerosol spray deposition of aqueous solutions on freshly cleaved mica followed by air drying. The distributions of contour lengths and chain or fibril thicknesses normal to the mica substrate can be measured for individual polymer molecules or molecular assemblies. In many cases it is possible to conclude that the structures imaged and quantitatively analyzed are representative of those present in solution and not artifacts of the deposition/dessication process. Imaging of linear and cyclic tri

Humidity effects on atomic force microscopy of gold-labeled DNA on mica.

Recent work in atomic force microscopy (AFM) of deoxyribonucleic acid (DNA) has relied on immobilizing DNA molecules by drying a small volume of buffered DNA solution onto cleaved mica. When imaging in air, relative humidity has been known to affect both the resolution and measured height of the DNA strands. We present data of measured height versus humidity for DNA and attached gold labels, and we propose a model for this data based on swelling of coadsorbed buffer salts upon exposure to moisture. In this model, small particles (e.g., DNA) stay near the top of the swelling salt layer, whereas

Nanoscale insights into cellulose-imidazolium ionic liquid interactions via atomic force microscopy.

The interaction force of cellulose is a key contributing factor to the mechanical properties and functionality of cellulose-based products, while quantitative understanding the nanoscale interaction of cellulose is challenging through experimentation. Herein, atomic force microscope (AFM) force spectroscopy was innovatively used to quantitatively measure the intermolecular forces of cellulose in air/water/ionic liquids (ILs), respectively. The results showed that the intermolecular forces were reduced by 60.3% and 34.0%, when the hydroxy groups of cellulose were replaced by methyl and hydroxye
