Towards single molecule biosensors using super-resolution fluorescence microscopy.

Conventional immunosensors require many binding events to give a single transducer output which represents the concentration of the analyte in the sample. Because of the requirements to selectively detect species in complex samples, immunosensing interfaces must allow immobilisation of antibodies while repelling nonspecific adsorption of other species. These requirements lead to quite sophisticated interfacial design, often with molecular level control, but we have no tools to characterise how well these interfaces work at the molecular level. The work reported herein is an initial feasibility

Reliability of acridine orange fluorescence microscopy in oral cytodiagnosis.

CONTEXT AND AIMS: The oral cavity is the most predominant location in the head and neck region for primary malignant epithelial tumors. Oral cancer is estimated to be the sixth most common malignancy. Early recognition is imperative for successful treatment and good prognosis. Exfoliative cytology is a simple and reasonably effective technique for rapid initial evaluation of a suspicious oral lesion. The present study was conducted to determine the reliability of acridine orange fluorescence microscopy for cytodiagnosis as a more rapid and easier method for the final evaluation of the cytologi

Fluorescence microscopy of the avian (Gallus domesticus) carotid body.

Sterba's technique to detect neurosecretory substances was applied to carotid body tissue, surrounding branchial derivatives, ganglion nodosum and the carotid arterial wall. Mainly a yellowish-green, a yellowish-red and a bright yellow fluorescence was observed as well as greenish-yellow fibres with varicosities and terminal knobs. The metachromatic reaction of the technique was briefly discussed.

Mapping of protein transduction pathways with fluorescent microscopy.

The number of various cargo delivered into cells by CPPs demonstrates the effective transport abilities of these short-peptidic sequences. Over the years of research, the translocation process of CPP-cargo complexes has been mapped to being of mostly endocytic nature, however, there is still no consensus on which of the endocytic routes is prevalent and to which extent the interplay between different modes of endocytosis is taking place. The intracellular trafficking of CPPs attached to a cargo molecule is even less understood. Therefore, the internalization and the subsequent intracellular ta

[Fluorescent microscopy in the diagnosis of Dictyocaulus infection in sheep].

Fluorescence microscopy was employed to study Dictyocaulus infection in 15 experimentally infected lambs each given 2000 D. filaria larvae. Studied were also the sera of 10 control lambs and of 20 sheep and goats infected with other helminths. Antibodies were established after the 14th day in 53 per cent of the lambs, the 20th day in 86.6 per cent, and the 27th day in all animals infected with the parasite. Antigens were found mainly in the wall of the digestive and reproductive organs, and in the cuticle of the larvae. As a method of early diagnosis fluorescence microscopy is simple, precise,

Mapping ALA-induced PPIX fluorescence in normal brain and brain tumour using confocal fluorescence microscopy.

We have mapped the microscopic distribution of aminolevulinic acid (ALA) induced protoporphyrin IX (PPIX) in normal rabbit brain and in brain tumour at various time intervals after i.v. injection of ALA. In normal brain, regions without a blood-brain barrier (BBB) such as the choroid plexus and meninges and also structures associated with the CSF synthesized the highest amounts of PPIX. We also detected ALA in the CSF within 2 h after ALA administration when the serum concentration was high. Our fluorescence studies have shown that ALA is able to cross an intact BBB and be metabolized in regio

Fluorescence confocal microscopy for margin assessment in prostatectomy: IP8-FLUORESCE study protocol.

BACKGROUND: Radical prostatectomy (RP) represents the cornerstone of surgical treatment for prostate cancer. Assessing surgical margin status intraoperatively with current techniques remains challenging due to high costs in the context of an already stretched pathology workforce. Fluorescence confocal microscopy (FCM) is a promising technique to detect margins in prostate cancer surgery not bound by such limitations. STUDY DESIGN: The Imperial Prostate 8 - Fluorescence Confocal Microscopy for Rapid Evaluation of Surgical Cancer Excision (IP8-FLUORESCE) study is a multicentre, prospective, ex

Correlative X-ray imaging and fluorescence microscopy in hydrated environments.

Imaging cellular organization in its native state is essential for understanding how the molecular architecture governs biological function. Fluorescence microscopy is one of the pivotal techniques that provides molecular specificity for imaging in real space, but is limited to labeled components. In contrast, X-rays are sensitive to electron density variations, enabling label-free structural characterization in reciprocal space. In particular, scanning small-angle X-ray scattering (SAXS) combines information from real and reciprocal space and enables access to intact cells owing to the high p

Asynchronous-spectral fusion fluorescence microscopy for microsecond-scale behavioral dynamics.

Event-based image sensors provide microsecond temporal resolution but lack spectral discrimination, whereas diffractive spectral imagers encode wavelength information at conventional frame rates. We introduce a fluorescence microscopy architecture that fuses asynchronous event streams with diffraction-encoded CMOS measurements to decouple temporal and spectral sampling. The system achieves ~2.2 µm spatial resolution over a 0.5 mm field of view and effective temporal resolution down to 10 µs and differentiates fluorophores whose emission peaks are separated by only 23 nm. By synchronizing and c

2-photon fluorescence microscopy of brain cells beyond 2 mm.

3-photon microscopy (3 PM) has been the dominant technology for in vivo imaging of deep brain cells down to the hippocampus. However, tissue attenuation and rapid 3 PM signal decay limit the imaging depth to 1.5 mm. Here, we demonstrate non-invasive 2-photon fluorescence (2PF) imaging to a depth of 2 mm in the mouse brain. Excitation in the 1700-nm window minimizes brain attenuation, while 2-photon excitation reduces signal decay compared with other multiphoton modalities. IRDye 800CW 2-DG enables brain-cell labeling and 2PF excitation in this window. Its 2PF emission extends into the NIR-II r

Maximising imaging volumes of expanded tissues for inverted fluorescence microscopy.

Expansion microscopy (ExM) has enabled nanoscale imaging of tissues by physically enlarging biological samples in a swellable hydrogel. However, the increased sample size and water-based environment pose challenges for deep imaging using conventional inverted confocal microscopes, particularly due to the limited working distance of high-numerical-aperture (NA) water immersion objectives. Here, we introduce a practical imaging alternative that utilizes an inverted water-dipping objective and a refractive-index-matched optical path using fluorinated ethylene propylene (FEP) film. Through point s

De novo designed bright, hyperstable rhodamine binders for fluorescence microscopy.

De novo protein design has emerged as a powerful strategy with the promise to create new tools. The practical performance of designed fluorophore binders, however, has remained far from meeting fluorescence microscopy demands. Here, we design de novo Rhodamine Binder (Rhobin) tags that combine ideal properties including size, brightness, and now adding hyperstability. Rhobin allows live and fixed cell imaging of a wide range of subcellular targets in mammalian cells. Its reversible fluorophore binding further enables live super-resolution STED microscopy with low photobleaching, as well as PAI

Fluorescence Lifetime Imaging Microscopy of Porphyrins in Helicobacter pylori Biofilms.

Bacterial biofilm constitutes a strong barrier against the penetration of drugs and against the action of the host immune system causing persistent infections hardly treatable by antibiotic therapy. Helicobacter pylori (Hp), the main causative agent for gastritis, peptic ulcer and gastric adenocarcinoma, can form a biofilm composed by an exopolysaccharide matrix layer covering the gastric surface where the bacterial cells become resistant and tolerant to the commonly used antibiotics clarithromycin, amoxicillin and metronidazole. Antimicrobial PhotoDynamic Therapy (aPDT) was proposed as an alt

Demonstration of early lesions of muscle by infrared fluorescence microscopy.

Previous investigations demonstrated decrease or loss of IR fluorescence in certain lesions of smooth and striated muscle. These investigations were extended to diseases of skeletal muscle. Sections were stained and photographed as described (Puchtler et al. 1980). Various diseases caused significant changes of IR fluorescence. Often, IR fluorescence demonstrated lesions in muscle fibers which appeared unremarkable in direct light and/or conventional darkfield microscopy. The patterns of IR fluorescence varied widely, even within groups of diseases, e.g. muscular dystrophies. However, owing to

Fluorescence microscopy test in porphyrias, photodermatoses and lead exposed persons.

Fluorescence microscopy tests were carried out in different groups of patients Peripheral blood diluted with saline was used and 200 high power fields were inspected in every case. The results were presented as the number of fluorescing erythrocytes (FE) per 100000 red blood cells (or 200 fields). In the controls, porphyria cutanea tarda patients and patients with photodermatoses other than erythopoietic protoporphyria and pellagra almost no FE were detected. In erythropoietic protoporphyria the mean value was 10600, in lead poisoning 1032, in patients exposed to lead 48.2, in sideropenic anae
