Intravesical mitomycin-C-induced interstitial pneumonia.

An 84-year-old man with no evidence of pre-existing interstitial pneumonia developed fatal interstitial pneumonia and respiratory failure that could only be explained as an adverse effect of intravesical mitomycin C chemotherapy. He had undergone transurethral resection of bladder cancer 3 times, followed by intravesical mitomycin C chemotherapy 1 month later. He had received intravesical mitomycin C chemotherapy every week for 2 months, and he had complained of dyspnea on exertion 5 days before the last intravesical mitomycin C chemotherapy session. Interstitial infiltration was detected in b

Airway complications from topical mitomycin C.

OBJECTIVE: Topical application of mitomycin C appears to be a useful adjunct in reducing cicatricial scarring of the airways. Human and animal studies have demonstrated the efficacy and safety of mitomycin C topically in the treatment of airway stenosis at concentrations ranging from 0.4 mg/mL to 10 mg/mL. Although no reports of mitomycin C toxicity have been reported in the otolaryngology literature, the ophthalmologic literature has documented serious, vision-threatening complications resulting from the use of topical mitomycin C. The purpose of this study is to report complications related

Intraoperative mitomycin C in complicated glaucomas.

Filtering surgery has been found to be less successful in certain types of glaucoma. These include young patients, those with pigmentary glaucoma, secondary glaucoma, angle recession glaucoma, aphakic or pseudophakic glaucoma, and patients requiring reoperation. This study describes the authors' attempt to evaluate the effectiveness of conventional trabeculectomy with intraoperative application of mitomycin C in such patients. Ten eyes of 8 patients were evaluated in this study. Of these cases 4 eyes (2 bilateral cases) were from the primary juvenile open angle group; 2 eyes each had pseudopha

Mitomycin C effect on Robertsonian translocations.

Centromeric breaks and dissociation of Robertsonian translocations have been suggested to be the cause of a few cases of mosaicism. One possible explanation for dissociation could be that the point of reunion of the two acrocentrics would be a structurally fragile site. Mitomycin C (MMC) treatment of lymphocyte cultures from 6 patients having a Robertsonian translocation showed that in cases 1 and 2, who were already mosaics, MMC induced a statistically significant increase of the number of cells with the dissociated translocation. In the remaining cases a preferential centromeric break on the

Clinical outcomes of PRESERFLO® Microshunt procedure with mitomycin C alone versus with mitomycin C and bevacizumab.

BACKGROUND: The PRESERFLO® Microshunt procedure offers a safe and effective alternative to trabeculectomy procedure. The adjuvant use of anti-vascular endothelial growth factor (VEGF) with trabeculectomy has been well studied, yet little is known on the effect and safety of anti-VEGF agents used as an adjunctive therapy in PRESERFLO® procedures. OBJECTIVES: To compare the outcomes of PRESERFLO Microshunt procedure using either mitomycin C (MMC) alone versus MMC augmented with bevacizumab in patients with open angle glaucoma to a follow-up time of 6 months. DESIGN: Retrospective observational

[A clinical trial of a new mitomycin C derivative, KW-2083 (7-N-(p-hydroxyphenyl)-mitomycin C)].

KW-2083 [7-n-(p-hydroxyphenyl)-mitomycin C] is a new mitomycin C (MMC) derivative. Its myelotoxicity was compared with that of MMC by using colony-forming unit-spleen (CFU-S) from the femurs of C57BL/6 mice. As a result, it was estimated that the intensity of myelotoxicity of MMC was four times greater than that of KW-2083. Based on this data, a clinical trial of KW-2083 was conducted in 24 cases with various types of advanced solid tumors. KW-2083 was administered by i.v. injection at a dose of 40 mg/body every week. Out of 15 evaluable cases, a case of ovarian cancer showed a partial respons

Inappropriate administration of mitomycin in a cataract patient.

PURPOSE: This case report describes the improper administration of intracameral mitomycin after cataract extraction in an 83-year-old male, demonstrating the need for system improvements to prevent "never events." OBSERVATIONS: An 83-year-old male presented with visually significant bilateral cataracts, (right eye (OD): 20/80, left eye (OS): 20/70). He underwent technically uncomplicated cataract surgery in the right eye. At postoperative visit one, vision was 20/250 OD, less than anticipated for unclear reasons. On subsequent visits, the patient was found to have worsening photophobia, incre

Histopathology of a functioning mitomycin-C trabeculectomy.

The ideal trabeculectomy bleb is diffuse, normally vascularized and characterized by microcystic change in the overlying conjunctiva. We compare and contrast the histopathology of a normally functioning mitomycin-C trabeculectomy site obtained from an eye enucleated for iris melanoma with abnormal blebs discussed in the literature. Representative sections of the normally functioning bleb were examined under the light microscope. The conjunctiva is composed of a uniform three-layered non-keratinizing stratified squamous epithelium overlying a single layer of oedematous basal cells. The conjunct

Effects of topical mitomycin and trimetazidine on myringosclerosis.

INTRODUCTION: Myringosclerosis, one of the most common complications of ventilation tube placement, is a kind of tympanosclerosis and is defined as subepithelial hyalinization of the tympanic membrane. There are two arguments in the development of myringosclerosis: inflammation triggering the development of myringosclerosis and free oxygen radicals released during inflammation causing myringosclerosis. OBJECTIVE: The aim of the present study was to explore the effects on the development of myringosclerosis of mitomycin, which has anti-inflammatory effects, and trimetazidine, which is believed

Uses and complications of mitomycin C in ophthalmology.

Mitomycin C is a chemotherapeutic agent that acts by inhibiting DNA synthesis. Its use and application in ophthalmology has been increasing in recent years because of its modulatory effects on wound healing. Current applications include pterygium surgery, glaucoma surgery, corneal refractive surgery, cicatricial eye disease, conjunctival neoplasia and allergic eye disease. Although it has been used successfully in these conditions, it has also been associated with significant complications. This article reviews the current trends and uses of mitomycin C in the eye and its reported complication

[Cytotoxicity of mitomycin C in rabbit eyes].

The cytotoxic effects of mitomycin C (MMC) on intact rabbit eyes were studied using light microscopy and transmission electron microscopy. A volume of 0.1 ml MMC solution (0.4 mg/ml) was given to rabbits subconjunctivally once a day. The eyes were enucleated at 1, 2, 3, and 4 weeks after subconjunctival injection. MMC showed considerable cytotoxicity in most tissues of the rabbit eyes, such as the cornea, the iris, especially to the choroid and the ciliary body. However, MMC cytotoxicity in the sclera was slight. Based on the above results it is concluded that MMC should not be used when the c
