Naked mole rats are among the most hypoxia-tolerant mammals identified and live in chronic hypoxia throughout their lives. The physiological mechanisms underlying this tolerance, however, are poorly understood. Most vertebrates hyperventilate in acute hypoxia and exhibit an enhanced hyperventilation following acclimatization to chronic sustained hypoxia (CSH). Conversely, naked mole rats do not hyperventilate in acute hypoxia and their response to CSH has not been examined. In this study, we explored mechanisms of plasticity in the control of the hypoxic ventilatory response (HVR) and hypoxic

Over the previous several decades, many non-traditional research models have offered new avenues of exploration for biomedical research. The promise of these animals is primarily derived from adaptations to unique or challenging environments that share key factors with a disease or pathology of interest (e.g., hypoxemia or hypercarbia are clinically relevant and are also in vivo consequences of environmental hypoxia and hypercapnia, respectively). Animals adapted to such environments allow us to ask the question: how has nature solved a particular problem and what can we learn to inform novel

Reproductive suppression is a defining feature of cooperative breeding, yet the hormonal mechanisms regulating infertility in non-breeding individuals remain poorly understood. This study examines associations between circulating prolactin (PRL) and faecal metabolites of androgens (fAM), glucocorticoids (fGCM), and progesterone (fPM) in relation to socially induced infertility in two closely related cooperatively breeding mole-rat subspecies: the highveld (Cryptomys hottentotus pretoriae) and Natal mole-rat (C. hottentotus natalensis). Our results reveal a fundamental dichotomy in reproductive

Naked mole-rats are extremely long-living rodents with a maximum lifespan of 37 years, and their cellular aging and tissue aging are almost nonexistent. Therefore, in this study, we aim to analyze the extracellular matrix of the temporomandibular joint (TMJ) of naked mole-rats at the molecular level and explore the molecules involved in anti-aging and their localization. Micro-computed tomography (CT) scans revealed increased mineral density and wear of the mandibular condyle in aged mice. Conversely, CT scans did not reveal wear of the mandibular condyle in naked mole-rats, and histological a

Naked mole-rats (NMR, Heterocephalus glaber) defy Gompertzian rules of mortality and age-dependent senescence to lead healthy lives well into their fourth decade. The NMR immune system likely contributes to this phenotype by protecting against infections, regulating tissue repair, and eradicating neoplastic cells. In keeping with their many other paedomorphic traits, adult NMRs retain functional thymic tissue in the submandibular triangle. Despite this additional thymic tissue, perinatal NMR thymi are half the size of mouse thymi, yet both species show similar thymic involution trajectories. S

The naked mole-rat (NMR) Heterocephalus glaber (from the Greek/latin words ἕτερος, heteros = divergent, κεφαλή, kephalē = head and glabra = hairless) was first described by Rüppell (Fig. 1) and belongs to the Hystricognath (from the Greek words ὕστριξ, hystrix = porcupine and γνάθος, gnathos = jaw) as a suborder of rodents. NMR are characterized by the highest longevity among rodents and reveal a profound cancer resistance. Details of its skin-specific protective and resistance mechanisms against aging and carcinogenesis have so far not been adequately characterized. Recently, our knowledge of

The parotid gland of a female African mole-rat, Tachyoryctes splendens, was examined by light and electron microscopy. A pure seromucous gland of conventional histology, its striated and excretory ducts contain prominent cytoplasmic inclusions that are lightly stained with toluidine blue. At the ultrastructural level, the inclusions are seen to consist of a light to moderately dense structureless matrix in which are suspended scattered, randomly oriented filaments that either are 4-7 nm or approx 14 nm thick. A few multivesicular bodies or putative lipid droplets are sparsely distributed in th

The naked mole-rat (Heterocephalus glaber) is a fascinating model organism which challenges conventional paradigms in evolutionary developmental biology. As one of the two known eusocial mammals with a reproductive hierarchy akin to social insects, the naked mole-rat presents an exceptional system for studying the interplay between social structure, environmental adaptation, and developmental plasticity. This chapter explores how the species' unique reproductive strategies-including lifelong fertility, postnatal oogenesis, and social suppression of reproduction-reshape our understanding of mam

BACKGROUND: Human aging is characterized by endothelial dysfunction that drives a systemic prothrombotic shift. In contrast, the long-lived naked mole rat (NMR) represents a unique model of delayed aging, exhibiting a notable resistance to age-related pathologies. However, while its cardiovascular stability is well-documented, the NMR hemostatic profile across its lifespan remains unexplored.
OBJECTIVES: To assess whether NMRs undergo age-related hypercoagulability and to compare their hemostatic trajectory with that of humans.
METHODS: We compared young (2-year-old) and aged (20-year-old) N

Lacking fur, living in eusocial colonies and having the longest lifespan of any rodent, makes naked mole-rats (NMRs) rather peculiar mammals. Although they exhibit a high degree of polymorphism, skeletal plasticity and are considered a novel model to assess the effects of delayed puberty on the skeletal system, scarce information on their morphogenesis exists. Here, we examined a large ontogenetic sample (n = 76) of subordinate individuals to assess the pattern of bone growth and bone microstructure of fore- and hindlimb bones by using histomorphological techniques. The naked mole rat (Heterocephalus glaber, Rüppell, 1842) is a unique eusocial rodent with unusually long lifespan. Therefore, the study of spontaneous and experimentally induced pathologies in these animals is one of the most important tasks of gerontology. Various infections, noninfectious pathologies (including age-dependent changes), and tumors have been described in the naked mole rat. The most frequent pathologies are traumas (bite wounds), purulent and septic complications of traumatic injuries, renal tubular calcinosis, chronic progressive nephropathy, hepatic hemosiderosis, testicular

The aim of this study was to elucidate the ultrastructural changes in the liver cells of mole rats (Spalax leucodon) exposed to ultraviolet radiation (UVR). Thirteen mole rats used in this study were caught from nature. They were divided into four groups. The first group was separated as a control and was not given any radiation. The rest were exposed to ultraviolet C (UVC) radiation for 7, 14, and 21 days. The electron microscopic examinations revealed that significant ultrastructural changes occurred in the liver tissue. These changes were the reduction in cytoplasmic organelles, dilatation

The rudimentary eyes of the mole rat (Spalax ehrenbergi) are located under the skin and do not respond to light stimuli. However, removal of the eyes disturbs photoperiod perception in these animals. To help clarify the possibly remaining function of the eyes in this species, the authors studied their development and ultrastructure. In the early embryos the presumptive eye regions--the epithelium, lens vesicle, and optic cup--appear initially normal. As development progresses, the iris-ciliary body complex originates prematurely from the margin of the optic cup and shows a very rapid and massi

Evaporation is one of the most important pathways of water and heat loss for terrestrial vertebrates and can be partitioned into cutaneous (CEWL) and respiratory (REWL) components of total evaporative water loss (TEWL). Subterranean mammals inhabiting burrows with high humidity and relatively high ambient temperatures (Ta) face significant thermoregulatory challenges. We examined the effects different Tas (27.5, 32 and 37.5 °C) on the partitioning of TEWL in two social African mole-rats that differ in pelage. In the naked mole-rat (Heterocephalus glaber), TEWL increases ∼33 % with increasing T

Cellular senescence is an important anticancer mechanism that restricts proliferation of damaged or premalignant cells. Cellular senescence also plays an important role in tissue remodeling during development. However, there is a trade-off associated with cellular senescence as senescent cells contribute to aging pathologies. The naked mole rat (NMR) (Heterocephalus glaber) is the longest-lived rodent that is resistant to a variety of age-related diseases. Remarkably, NMRs do not show aging phenotypes until very late stages of their lives. Here, we tested whether NMR cells undergo cellular sen

The subterranean niche is a specialised and particularly challenging environment to obtain direct physiological and behavioural measurements from free-living animals. Rhythmicity has been examined in many mole-rat species in the laboratory, but field reports are relatively scarce. We implanted Natal mole-rats with temperature loggers in summer and winter to record core body temperature (Tb), before releasing the animals again. Animals were recaptured after two months to recover the loggers. Natal mole-rats can maintain rhythmicity of their Tb in their natural habitat and display seasonal diffe

Fossorial giant Zambian mole-rats are believed to live in a hypoxic and hypercapnic subterranean environment but their physiological responses to these challenges are entirely unknown. To investigate this, we exposed awake and freely-behaving animals to i) 6 h of normoxia, ii) acute graded normocapnic hypoxia (21, 18, 15, 12, 8, and 5% O2, 0% CO2, balance N2; 1 h each), or iii) acute graded normoxic hypercapnia (0, 2, 5, 7, 9, and 10% CO2, 21% O2, balance N2; 1 h each), followed by a 1 h normoxic normocapnic recovery period, while non-invasively measuring ventilation, metabolic rate, and body

Naked mole-rats (NMRs) are unique long-lived mammals that possess marked resistance to cancer including lack of any reported spontaneous skin cancer incidences. Here, we show that in comparison with mice, homeostatic epidermal keratinocytes in NMRs exhibit elevated expression of 324 tumor suppressor genes of different classes including 55 genes regulating DNA damage/repair. Furthermore, NMR skin grafted onto nude mice exhibits complete resistance to 7,12-dimethylbenz[a]-anthracene/12-O-tetradecanoylphorbol-13-acetate (DMBA/TPA)-induced skin carcinogenesis in tissue-autonomous manner. Background/Objectives: Subterranean mole-rats live in an intricate system of underground tunnels, a unique acoustic environment that has led to adaptations to their hearing. Most experimenters have concluded that mole-rats have poor hearing thresholds, perhaps 20-40 dB less sensitive than rodents living on the surface. The potential problem identified here is that mole-rat thresholds have all been measured in air, whereas there is some evidence-theoretical and observational-to suggest that these animals may hear more sensitively via bone conduction. Methods: A wide-ranging review of the litera

Naked mole-rats (Heterocephalus glaber) are very unusual among subterranean mammals in that they live in large colonies and are extremely social, spending large amounts of time gathered together in underground nests more than a meter below the surface. Many respiring individuals resting in deep, poorly ventilated nests deplete the oxygen supply and increase the concentration of carbon dioxide. Consistent with living in that atmosphere, naked mole-rats tolerate levels of low oxygen and high carbon dioxide that are deadly to most surface-dwelling mammals. Naked mole-rats appear to have evolved a
