Near-infrared light is neuroprotective in a monkey model of Parkinson disease.

OBJECTIVE: To examine whether near-infrared light (NIr) treatment reduces clinical signs and/or offers neuroprotection in a subacute 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) monkey model of Parkinson disease. METHODS: We implanted an optical fiber device that delivered NIr (670 nm) to the midbrain of macaque monkeys, close to the substantia nigra of both sides. MPTP injections (1.5-2.1mg/kg) were made over a 5- to 7-day period, during which time the NIr device was turned on. This was then followed by a 3-week survival period. Monkeys were evaluated clinically (eg, posture, bradykin

[Changes of glucose in the brains of monkeys with MPTP induced Parkinson's disease].

OBJECTIVE: To examine the metabolic and behavioral changes of monkeys with MPTP induced Parkinson's disease. METHODS: Four healthy monkeys were injected with diluted MPTP solution continuously through peripheral veins. The behavioral changes of the monkeys were monitored with the Parkinson disease scale for monkeys. 18FDG-PET-CT scans for the whole brain were obtained before and 3 months after the injection of MPTP. The pathological and glucose metabolic changes of the brains were examined. RESULTS: The animal model of Parkinson's disease was successfully established in all 4 monkeys, showin

Polymyositis in an immunodeficiency disease in monkeys induced by a type D retrovirus.

Fifty percent of primates with acquired immunodeficiency caused by a well-characterized type D retrovirus (SAIDS D) developed clinical, laboratory, and histologic features of polymyositis. By use of specific antisera and immunochemical techniques, we found the virus in the lymphoid cells surrounding muscle fibers and invading the endomysia septa. SAIDS D virus was isolated from the involved muscles and infected myotubes of normal muscle in tissue culture. These results suggest that retroviruses, a group of viruses increasingly associated with human diseases, can cause polymyositis with immunod

Morphology of polymorphonuclear neutrophils during periodontal disease in the cynomolgus monkey.

The polymorphonuclear neutrophil (PMN) appears to be an important cell in the protection of the host from pathogenic periodontal micro-organisms. The purpose of the present histological studies was to observe the emigration of the PMN from the gingival vessels to the periodontal pocket during gingivitis and ligature-induced periodontitis in the cynomolgus monkey. 2 adult female monkeys were treated by application of a silk ligature around selected posterior teeth. After 9 weeks, the monkeys were perfused and block sections of both ligated (early periodontitis) and non-ligated (gingivitis) site

Ocular signs in thiamine-deficient monkeys and in Wernicke's disease in humans.

Thiamine deficiency in the monkey is the animal counterpart of Wernicke's disease in humans. In the present study, thiamine deficiency was induced in 11 monkeys while three monkeys were given paired feedings supplemented by thiamine hydrochloride and three monkeys were maintained on regular chow. The typical clinical symptoms were apathy, inattention to peripheral stimuli, ataxia, ptosis, mydriasis progressing to pupillary areflexia, nystagmus, and ophthalmoparesis progressing to total ophthalmoplegia. With thiamine treatment, recovery was prompt and complete in mild to moderate cases but dela

Experimental infection of monkeys with Herpesvirus suis (Aujeszky's-disease virus).

Monkeys were infected intranasally with Herpesvirus suis. After an incubation period of 7 to 13 days the animals became acutely ill and rapidly died. Clinical signs included salivation, incoordination, ataxia and epileptiform convulsions, but not pruritus. Histopathological changes were confined to the central nervous system, and consisted of destruction of neurones with the formation of intranuclear inclusion bodies, gliosis and perivascular cuffing. Virus was isolated from the brain and spinal cord in the later stages of the illness but neutralising antibodies were not detected in serum. The

TRANSMISSION AND CULTIVATION EXPERIMENTS WITH HUMAN TRACHOMA AND THE EXPERIMENTAL DISEASE IN MONKEYS.

1. Conjunctival tissue derived from alien and native American white persons in New York City, having trachoma in an advanced stage, has been used successfully to induce in Macacus rhesus monkeys characteristic granular conjunctivitis. The transfer of infection was effected either by a single subconjunctival injection, or by repeated swabbing with conjunctival secretions. 2. Pathogenic strains of Bacterium granulosis have been recovered from the trachomatous tissues of six out of eleven patients. In addition, the organisms have been isolated from the monkeys infected with human material. 3. Rep

Kikwit Ebola Virus Disease Progression in the Rhesus Monkey Animal Model.

Ongoing Ebola virus disease outbreaks in the Democratic Republic of the Congo follow the largest recorded outbreak in Western Africa (2013-2016). To combat outbreaks, testing of medical countermeasures (therapeutics or vaccines) requires a well-defined, reproducible, animal model. Here we present Ebola virus disease kinetics in 24 Chinese-origin rhesus monkeys exposed intramuscularly to a highly characterized, commercially available Kikwit Ebola virus Filovirus Animal Non-Clinical Group (FANG) stock. Until reaching predetermined clinical disease endpoint criteria, six animals underwent anesthe

TALEN-based generation of a cynomolgus monkey disease model for human microcephaly.

Gene editing in non-human primates may lead to valuable models for exploring the etiologies and therapeutic strategies of genetically based neurological disorders in humans. However, a monkey model of neurological disorders that closely mimics pathological and behavioral deficits in humans has not yet been successfully generated. Microcephalin 1 (MCPH1) is implicated in the evolution of the human brain, and MCPH1 mutation causes microcephaly accompanied by mental retardation. Here we generated a cynomolgus monkey (Macaca fascicularis) carrying biallelic MCPH1 mutations using transcription acti

Longitudinal transcriptomic dysregulation in the peripheral blood of transgenic Huntington's disease monkeys.

BACKGROUND: Huntington's Disease (HD) is a progressive neurodegenerative disorder caused by an expansion in the polyglutamine (polyQ) region of the Huntingtin (HTT) gene. The clinical features of HD are characterized by cognitive, psychological, and motor deficits. Molecular instability, a core component in neurological disease progression, can be comprehensively evaluated through longitudinal transcriptomic profiling. Development of animal models amenable to longitudinal examination enables distinct disease-associated mechanisms to be identified. RESULTS: Here we report the first longitudina

Lifespan profiles of Alzheimer's disease-associated genes and products in monkeys and mice.

Alzheimer's disease (AD) is characterized by plaques of amyloid-beta (Abeta) peptide, cleaved from amyloid-beta protein precursor (AbetaPP). Our hypothesis is that lifespan profiles of AD-associated mRNA and protein levels in monkeys would differ from mice and that differential lifespan expression profiles would be useful to understand human AD pathogenesis. We compared profiles of AbetaPP mRNA, AbetaPP protein, and Abeta levels in rodents and primates. We also tracked a transcriptional regulator of the AbetaPP gene, specificity protein 1 (SP1), and the beta amyloid precursor cleaving enzyme (

Type-3 von willebrand's disease in a rhesus monkey (Macaca mulatta).

Severe type-3 von Willebrand's disease (vWD) was diagnosed in a young male rhesus monkey that had excessive bleeding from minor wounds. Plasma samples from the monkey had no detectable quantitative or functional von Willebrand factor (vWF), low Factor-VIII coagulant activity, and moderate prolongation of activated partial thromboplastin time. Testing of the affected monkey's extended family revealed a likely hereditary basis for the vWD, in that the sire and a paternal half-sister had markedly reduced plasma vWF concentration. Fresh whole blood was transfused to control frequent bleeding episo

[Behaviour after transplantation of brain cells into monkey models of Parkinson's disease].

OBJECTIVE: To observe the improved degree of pathogenic behaviour in monkey models of Parkinson's disease after transplantation of substantia nigra cells of human fetus. METHOD: 1-methy-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) was used to prepare monkey models of hemiparkinson's disease, i.e., early substantia nigra cells of human fetus were stereotaxically transplanted into PD monkey's behaviour for six months, using immuno-electromicroscopy to prove the transplanted survived neuron. RESULTS: After transplantation, the model monkey's motion slow and muscle hypertonia was apparently improv

Spontaneous cardiomyopathy resembling acute rheumatic heart disease in an owl monkey.

A wild-caught male Aotus vociferans died spontaneously during quarantine. Histologic examination of the heart showed several small myocardial arteries replaced with fibrinoid necrosis and adjacent inflammation that included lymphocytes and large histiocytic cells. Less often, similar inflammatory foci were observed in the myocardium interstitium. The lesions observed in the heart of the owl monkey resembled Aschoff bodies.

The epizootics of Kyasanur Forest disease in wild monkeys during 1964 to 1973.

In the Kyasanur Forest disease area two species of wild monkeys, Presbytis entellus and Macaca radiata, succumb to the natural infection with Kyasanur Forest disease (KFD) virus (family Flaviviridae). Between October 1964 and September 1973, 1046 monkeys (860 P. entellus and 186 M. radiata) died. Of these, KFD virus was isolated from 118 P. entellus and 13 M. radiata. Maximum mortality of monkeys was reported during December through May coinciding with the season of activity of immature stages of Haemaphysalis ticks, incriminated vectors of KFD. The epizootic showed an initial spread of the di

[Characteristics of Escherichia serogroup O132:K isolated in intestinal diseases in monkeys].

The results obtained in the study of the main biological characteristics of 22 Escherichia strains, serogroup O132: K ., isolated from monkeys in the Sukhumi reserve are presented. For the first time Escherichia belonging to serovar O132: K .: H- have been detected; these organisms, in contrast to reference strain O132: K .: H28, possess a number of Shigella-like characteristics and are capable of intraepithelial parasitism in Shereny's test, which impedes their primary identification.
