Experimental Rickettsia typhi Infection in Monodelphis domestica: Implications for Opossums as an Amplifying Host in the Suburban Cycle of Murine Typhus.

Murine typhus is an acute undifferentiated febrile illness caused by Rickettsia typhi. In the United States, its reemergence appears to be driven by a shift from the classic rat-rat flea cycle of transmission to one involving opossums (Didelphis virginiana) and cat fleas. Little is known of the ability of opossums to act as a reservoir and amplifying host for R. typhi. Here, we use Monodelphis domestica (the laboratory opossum) as a surrogate for D. virginiana. Opossums were inoculated via the intraperitoneal (IP) or intradermal (ID) route with 1 × 106 viable R. typhi. Blood and tissues were c

ABC Efflux Transporters and Solute Carriers in the Early Developing Brain of a Marsupial Monodelphis domestica (South American Gray Short-Tailed Opossum).

This study used a marsupial Monodelphis domestica, which is born very immature and most of its development is postnatal without placental protection. RNA-sequencing (RNA-Seq) was used to identify the expression of influx and efflux transporters (ATP-binding cassettes [ABCs] and solute carriers [SLCs]) and metabolizing enzymes in brains of newborn to juvenile Monodelphis. Results were compared to published data in the developing eutherian rat. To test the functionality of these transporters at similar ages, the entry of paracetamol (acetaminophen) into the brain and cerebrospinal fluid (CSF) wa

Ultraviolet radiation-induced histopathological changes in the skin of the marsupial Monodelphis domestica. I. The effects of acute and chronic exposures and of photoreactivation treatment.

Post-ultraviolet radiation (UVR) treatment of the South American opossum, Monodelphis domestica with long-wavelength radiation (320-400 nm) suppressed the induction of histopathological alterations in the skin. This study identifies DNA as a primary chromophore involved in the induction of various photobiological responses of the skin such as hyperplasia and sunburn cell formation, and also identified pyrimidine dimers as one responsible DNA lesion. The histology of skin from opossums exposed to multiple doses of UVR showed that pre-malignant changes had occurred in the skin.

Ultraviolet radiation-induced histopathologic changes in the skin of the marsupial Monodelphis domestica. II. Quantitative studies of the photoreactivation of induced hyperplasia and sunburn cell formation.

Induction of sunburn cells (SBCs) and hyperplasia of the epidermis of the marsupial Monodelphis domestica by ultraviolet radiation (UVR) has been studied. A dose of 500 J/m2 (approximately 1 minimal erythemal dose) from an FS-40 sunlamp induced measurable numbers of SBCs with a peak number at 32-48 h post-UVR exposure of skin to photoreactivating light suppressed the induction of SBCs by approximately 75%. Pre-UVR exposure to photoreactivating light had no effect on the induction of SBCs. Induction of hyperplasia also was suppressed to a similar extent by post-UVR photoreactivation treatment.

The p53 tumor suppressor gene of the marsupial Monodelphis domestica: cloning of exons 4-11 and mutations in exons 5-8 in ultraviolet radiation-induced corneal sarcomas.

Inactivating p53 mutations are found in many ultraviolet radiation (UVR)-induced skin tumors. We examined 12 UVR-induced corneal tumors of the marsupial Monodelphis domestica for mutations in exons 5-8 of p53 and compared their mutational spectrum with that of UVR-induced skin tumors of other species. First we cloned and characterized a cDNA extending from the middle of exon 4 through exon 11 of the Monodelphis p53 gene. Based on the sequence information obtained, primers were designed to amplify introns 4-9 of the gene; intron primers to amplify individually exons 5-8 were subsequently develo

Molecular characterisation of the CD79a and CD79b subunits of the B cell receptor complex in the gray short-tailed opossum (Monodelphis domestica) and tammar wallaby (Macropus eugenii): Delayed B cell immunocompetence in marsupial neonates.

The B cell receptor (BCR) is a multiprotein complex that is pivotal to antigen recognition and signal transduction in B cells. It consists of an antigen binding component, membrane Ig (mIg), non-covalently associated with the signaling component, a disulphide-linked heterodimer of CD79a and CD79b. In this study, the gene and corresponding cDNA for CD79a and CD79b in the gray short-tailed opossum, as well as the cDNA sequences for CD79a and CD79b in the tammar wallaby, are described. Many of the structural and functional features of CD79a and CD79b were conserved in both marsupials, including t

Comparative Analyses Reveal Conserved and Modified Steps in the Testis Descent and Scrotum Development in Mouse and Opossum.

INTRODUCTION: In many mammals, the testes descend from its abdominal position on the mesonephric kidney and are housed in the scrotum. It has been speculated that metatherians and eutherians might have acquired the scrotal testis independently because metatherians have the scrotum cranially to the phallus, while eutherians, such as humans and mice, possess it caudally. Rather, recent studies based on sequence comparisons of testis-descent-related genes indicate that the metatherian-eutherian common ancestor might already possess the descent mechanisms. To further elucidate the path of scrotal

Development of olfactory glomeruli: temporal and spatial interactions between olfactory receptor axons and mitral cells in opossums and rats.

Mitral cells are the primary output neurons of the vertebrate olfactory bulb and are major recipients of sensory input from the periphery. The morphogenesis of mitral cell dendrites was followed to elucidate their early spatial and temporal interactions with olfactory receptor neurons and glia during the construction of olfactory glomeruli. Monodelphis domestica, a marsupial born at an extremely immature stage, and rats were examined. Mitral cells were retrogradely labeled by application of the lipophilic dye 1,1' dihexadecyl-3,3,3'3'-tetramethylin-docarbocyanine perchlorate (DiI) to the later

Comparative gene expression analyses reveal heterochrony for Sox9 expression in the cranial neural crest during marsupial development.

Compared to placental mammals, marsupials have short gestation period, and their neonates are relatively immature. Despite these features, marsupial neonates must travel from the birth canal to the teat, suckle and digest milk to complete development. Thus, certain organs and tissues of marsupial neonates, such as forelimbs to crawl and jaw elements to suckle, must develop early. Previous reports showed that cranial neural crest (CNC) cells, as the source of ectomesenchyme of jaw elements, are generated significantly early in gray short-tailed opossum (Monodelphis domestica) compared to other

Translational animal model for genetic predisposition to anophthalmia/microphthalmia.

Anophthalmia and microphthalmia (A/M) are severe congenital malformations that are responsible for ∼11% of childhood blindness. Despite clinical advances, approximately 70-80% of human A/M cases lack a molecular diagnosis. This study aims to characterize a spontaneous, non-syndromic model of the microphthalmia, anophthalmia, and coloboma (MAC) spectrum in the laboratory opossum (Monodelphis domestica), providing a novel experimental opportunity for embryonic developmental and ocular pathology research, as well as translational research into preventions and treatments of these conditions. Metabolic profile of the perivertebral muscles in small therian mammals: implications for the evolution of the mammalian trunk musculature.

In order to gain a better understanding of the ancestral properties of the perivertebral muscles of mammals, this study investigated the fiber type composition of these muscles in six small, extant therians (two metatherians and four eutherians) similar in body shape to early mammals. Despite a few species-specific differences, the investigated species were very similar in their overall distribution of fiber types indicating similar functional demands on the back muscles in mammals of this body size and shape. Deep and short, mono- or multisegmental muscles (i.e., mm. interspinales, intermammi

Resistance of South American opossums to vWF-binding venom C-type lectins.

Opossums in the clade Didelphini are well known to be resistant to snake venom due to endogenous circulating inhibitors which target metalloproteinases and phospholipases. However, the mechanisms through which these opossums cope with a variety of other damaging venom proteins are unknown. A protein involved in blood clotting (von Willebrand Factor) has been found to have undergone rapid adaptive evolution in venom-resistant opossums. This protein is a known target for a subset of snake venom C-type lectins (CTLs), which bind it and then induce it to bind platelets, causing hemostatic disrupti

Differential OMP expression in opossum accessory olfactory bulb.

Using immunohistochemical techniques, olfactory marker protein (OMP) was localized to the main (MOB) and accessory (AOB) olfactory bulbs of 30- and 45-day-old, and adult Brazilian opossums, Monodelphis domestica. Entire olfactory nerve and glomerular layers of the adult opossum MOB were darkly stained. In the adult AOB the rostral half of these two layers was stained more intensely than the caudal half, and both parts were less darkly stained than the MOB. This differential AOB staining was not present at 30 days of age, but was evident by postnatal day 45, although not as dramatic as in adult

[Solar radiation and melanomas--is there any doubt about the connection?].

Arguments for and against there being a connection between exposure to solar radiation and cutaneous, malignant melanoma are reviewed. Recent experiments with animals and epidemiological observations provide relatively strong arguments that solar radiation causes cutaneous, malignant melanoma. Furthermore, epidemiological data from Norway and Australia support the assumption that UVA-radiation plays a significant role in melanoma induction; this is in agreement with data from experiments with Xiphophorus and Monodelphis domestica. A new hypothesis for melanoma induction is presented: Radiation

A family of olfactory receptors uniquely expanded in marsupial and monotreme genomes are expressed by a T cell subset also unique to marsupials and monotremes.

Olfactory receptors (OR), expressed on olfactory neurons, mediate the sense of smell. Recently, OR have also been shown to be expressed in non-olfactory tissues, including cells of the immune system. An analysis of single-cell transcriptomes of splenocytes of the grey short-tailed opossum (Monodelphis domestica) found OR are expressed on a subset of T cells, the γμ T cells, that are unique to marsupials and monotremes. A majority of opossum γμ T cells transcriptomes contain OR family 14 transcripts, specifically, from the OR14C subfamily. Amongst the mammals, the OR14 gene family is expanded i

Ultrastructural study of ensheathing cells in early development of olfactory axons.

Ultrastructural observations in the grey short-tailed opossum (Monodelphis domestica) and rat revealed that ensheathing cells were intimately related to the early formation of olfactory axons. Whilst the axons were still in the olfactory epithelium, they were enveloped by ensheathing cell processes which formed a cradle-like structure on the basal side of the epithelium. Continued downgrowth of the axons towards the lamina propria resulted in an evagination with the ensheathing cell process or cell body in direct contact with the basal lamina. Subsequently the basal lamina became fragmented, a

A new developmental mechanism for the separation of the mammalian middle ear ossicles from the jaw.

Multiple mammalian lineages independently evolved a definitive mammalian middle ear (DMME) through breakdown of Meckel's cartilage (MC). However, the cellular and molecular drivers of this evolutionary transition remain unknown for most mammal groups. Here, we identify such drivers in the living marsupial opossum Monodelphis domestica, whose MC transformation during development anatomically mirrors the evolutionary transformation observed in fossils. Specifically, we link increases in cellular apoptosis and TGF-BR2 signalling to MC breakdown in opossums. We demonstrate that a simple change in

Evolutionary innovations in germline biology of placental mammals identified by transcriptomics of first-wave spermatogenesis in opossum.

Mammalian spermatogenesis is a highly stereotyped and conserved developmental process that is essential for fitness. At the same time, gene expression in spermatogenic cells is rapidly evolving. This combination of features has been suggested to drive rapid fixation of new gene expression patterns. Using a high-resolution dataset comprising bulk and single-cell data from juvenile and adult testes of the opossum Monodelphis domestica, a model marsupial, we define the developmental timing of the spermatogenic first wave in opossum and delineate conserved and divergent gene expression programs ac

Fetal inflammatory signals regulate maternal investment during marsupial pregnancy.

Marsupial pregnancy is strikingly short: placental attachment in the gray short-tailed opossum Monodelphis domestica lasts only two days. The attachment period is characterized by a spike in inflammatory signaling, development of an expanded uterine capillary network, and exponential fetal growth. This brevity has historically been attributed to a maternal immune response to fetal contact that only eutherian mammals have evolved mechanisms to tolerate. However, several inflammatory cytokines, including interleukin-1A (IL-1A) and interleukin-6 (IL-6), are produced primarily by fetal cells. We h
