Virus Management Using Metal-Organic Framework-Based Technologies.

The eradication and isolation of viruses are two concurrent approaches to protect ourselves from viral infections and diseases. The quite versatile porous materials called metal-organic frameworks (MOFs), have recently emerged as efficient nanosized tools to manage viruses, and several strategies to accomplish these tasks have been developed. This review describes these strategies employing nanoscale MOFs against SARS-CoV-2, HIV-1, tobacco mosaic virus, etc., which include the sequestration by host-guest penetration inside pores, mineralization, design of a physical barrier, controlled deliver

Genome sequence of dahlia mosaic virus revisited: molecular characterization and genetic diversity of DMV and dahlia common mosaic virus.

Dahlia mosaic virus (DMV) and dahlia common mosaic virus (DCMV), members of the genus Caulimovirus, are associated with dahlia mosaic disease and are prevalent in dahlia plants in the USA and in several other parts of the world. To resolve a discrepancy in the publicly available genomic sequence of DMV and its taxonomic status with respect to DCMV, the complete genome sequences of several DMV and DCMV isolates from different regions of the USA were determined. Based on this sequence analysis, we resolved a discrepancy in the previously reported DMV sequence, confirmed that DMV possesses a geno

Occurrence, Genetic Variability of Tomato Yellow Ring Orthotospovirus Population and the Development of Reverse Transcription Loop-Mediated Isothermal Amplification Assay for Its Rapid Detection.

Tomato-infecting viruses have been considered as a serious threat to tomato crops in Poland. Therefore, during 2014-2021, 234 tomato samples delivered directly by greenhouse tomato growers to Plant Disease Clinic of IPP-NRI were tested. Eight virus species: pepino mosaic virus (PepMV), tomato yellow ring orthotospovirus (TYRV), tomato spotted wilt orthotospovirus (TSWV), potato virus Y (PVY), cucumber mosaic virus (CMV), tomato black ring virus (TBRV) and tomato mosaic virus (ToMV) were detected in single or mixed infection in 89 samples. The presence of TYRV was established for the first time

Cell Death Triggered by a Putative Amphipathic Helix of Radish mosaic virus Helicase Protein Is Tightly Correlated With Host Membrane Modification.

Systemic necrosis is one of the most severe symptoms caused by plant RNA viruses. Recently, systemic necrosis has been suggested to have similar features to a defense response referred to as the hypersensitive response (HR), a form of programmed cell death. In virus-infected plant cells, host intracellular membrane structures are changed dramatically for more efficient viral replication. However, little is known about whether this replication-associated membrane modification is the cause of the symptoms. In this study, we identified an amino-terminal amphipathic helix of the helicase encoded b

Cucumber mosaic virus RNA contains 7-methyl guanosine at the 5'-terminus of all four RNA species.

Purified cucumber mosaic virus (CMV) RNA was reacted with NaIO4 and then [3H]KBH4 to label ends containing exposed 2',3'-hydroxyls. The four major RNA species were then fractionated by polyacrylamide gel electrophoresis. Analysis of digests of these RNAs by ribonucleases T1 plus T2, alkaline phosphatease, snake venom diesterase and nucleotide pyrophosphatase by means of paper electrophoresis and ion-exchange chromatography indicated that the sequence m7G(5')ppp(5')N was present at the 5'-end of 75-92% of all RNA molecules. In addition, about 90% of the 3'-ends terminated in either adenosine or

Spot Inoculation of Begomovirus by Single Whitefly (Bemisia tabaci) Transmission in Mungbean Using Microcage.

The Begomovirus genus is a prominent group of viruses that are transmitted by the whitefly vector, Bemisia tabaci, and have been known to cause significant yield reductions in several economically important crops worldwide. Among these viruses, the mungbean yellow mosaic India virus is particularly problematic, causing substantial yield losses in mungbean and proving difficult to control. Therefore, developing new techniques to study the vector-virus systems involving begomoviruses is a research priority for mungbean and other crops affected by these viruses. In this chapter, we present a tech

Antiviral Properties of Alginate-Based Biomaterials: Promising Antiviral Agents against SARS-CoV-2.

The COVID-19 pandemic has made it essential to explore alternative antiviral materials. Alginate is a biodegradable, renewable, biocompatible, water-soluble and antiviral biopolymer with many potential biomedical applications. In this regard, this review shows 17 types of viruses that have been tested in contact with alginate and its related biomaterials. Most of these studies show that alginate-based materials possess little or no toxicity and are able to inhibit a wide variety of viruses affecting different organisms: in humans by the human immunodeficiency virus type 1, the hepatitis A, B,

A Reduced Starch Level in Plants at Early Stages of Infection by Viruses Can Be Considered a Broad-Range Indicator of Virus Presence.

The diagnosis of virus infection can facilitate the effective control of plant viral diseases. To date, serological and molecular methods for the detection of virus infection have been widely used, but these methods have disadvantages if applied for broad-range and large-scale detection. Here, we investigated the effect of infection of several different plant RNA and DNA viruses such as cucumber mosaic virus (CMV), tobacco mosaic virus (TMV), potato virus X (PVX), potato virus Y (PVY) and apple geminivirus on starch content in leaves of Nicotiana benthamiana. Analysis showed that virus infecti

Binding immunoglobulin 2 functions as a proviral factor for potyvirus infections in Nicotiana benthamiana.

Infection of viruses from the genera Bromovirus, Potyvirus, and Potexvirus in Nicotiana benthamiana induces significant up-regulation of the genes that encode the HSP70 family, including binding immunoglobulin protein 2 (BiP2). Three up-regulated genes were knocked down and infection assays with these knockdown lines demonstrated the importance of the BiP2 gene for potyvirus infection but not for infection by the other tested viruses. Distinct symptoms of cucumber mosaic virus (CMV) and potato virus X (PVX) were observed in the BiP2 knockdown line at 10 days postagroinfiltration. Interestingly

Genome sequencing of a novel variant of fowl adenovirus B reveals mosaicism in the pattern of homologous recombination events.

We determined the genomic sequence of a Ukrainian strain of fowl adenovirus B (FAdV-B). The isolate (D2453/1) shared 97.2% to 98.4% nucleotide sequence identity with other viruses belonging to the species Fowl aviadenovirus B. Marked genetic divergence was seen in the hexon, fiber, and ORF19 genes, and phylogenetic analysis suggested that recombination events had occurred in these regions. Our analysis revealed mosaicism in the recombination patterns, a finding that has also been described in the genomes of strains of FAdV-D and FAdV-E. The shared recombination breakpoints, affecting the same

Characterization and Construction of Full-Length cDNA Infectious Clone of a Novel BCMV Isolate in Pathogroup III.

Bean common mosaic virus (BCMV; Potyvirus phaseovulgaris) is one of the primary viruses that severely impacts the yield and quality of common beans (Phaseolus vulgaris L.) and has a worldwide distribution. Utilizing small RNA sequencing and RT-PCR validation, this study identified widespread co-infection by multiple viruses in field-collected common bean samples, with BCMV being the dominant viral species. A novel isolate, designated DY9, was obtained from these field samples. Pathotype characterization confirmed DY9 as pathotype PG-III, while previous studies reported all other PG-III members

Genome characterization and complete sequence of a new badnavirus from Pandanus amaryllifolius.

A new badnavirus was sequenced from fragrant pandan grass (Pandanus amaryllifolius) displaying mosaic and chlorosis on the leaves. The complete genome sequence was determined by high-throughput sequencing. The new badnavirus was tentatively named "pandanus mosaic associated virus" (PMaV). Similar to those of other members of the genus Badnavirus, the genome of PMaV consists of a circular DNA molecule of 7,481 bp with three open reading frames (ORF) potentially coding for three proteins. ORF3 encodes a polyprotein with conserved protein domains including zinc finger, trimeric dUTPase, aspartic

Virus Acquisition and Retention Periods for Transmission of Triticum Mosaic Virus by the Wheat Curl Mite.

The wheat curl mite (WCM), Aceria tosichella Keifer, is an important pest of winter wheat (Triticum aestivum L.) in the North American Great Plains region because of its ability to transmit three important viruses to wheat. Wheat streak mosaic virus (WSMV; Tritimovirus tritici) has been the most studied of these three viruses, but the rapid spread of Triticum mosaic virus (TriMV; Poacevirus tritici) throughout the region, along with its synergistic relationship with WSMV, has increased the need for a more complete understanding of its transmission capabilities and epidemiology. Studies were co

Mild Strain Cross-Protection in Passion Fruit.

Cross-protection is a phenomenon in which a plant prior infected with a mild viral strain can be protected from subsequent infection by severe strains of the same or closely related viruses. Since its discovery in the 1920s, this phenomenon has become an important strategy for controlling viral diseases in plants. Telosma mosaic virus (TelMV, a species of Potyvirus) is an emerging pathogen that threatens passion fruit production in subtropical and tropical regions. Here, we present a protocol for screening mild TelMV strains and their application in protecting passion fruit from severe TelMV i

Genomic Sequencing and Analysis of a Novel Human Cowpox Virus With Mosaic Sequences From North America and Old World Orthopoxvirus.

Orthopoxviruses (OPXVs) not only infect their natural hosts, but some OPXVs can also cause disease in humans. Previously, we partially characterized an OPXV isolated from an 18-year-old male living in Northern Norway. Restriction enzyme analysis and partial genome sequencing characterized this virus as an atypical cowpox virus (CPXV), which we named CPXV-No-H2. In this study, we determined the complete genome sequence of CPXV-No-H2 using Illumina and Nanopore sequencing. Our results showed that the whole CPXV-No-H2 genome is 220,276 base pairs (bp) in length, with inverted terminal repeat regi

Activation of the salicylic acid signaling pathway enhances Clover yellow vein virus virulence in susceptible pea cultivars.

The wild-type strain (Cl-WT) of Clover yellow vein virus (ClYVV) systemically induces cell death in pea cv. Plant introduction (PI) 118501 but not in PI 226564. A single incompletely dominant gene, Cyn1, controls systemic cell death in PI 118501. Here, we show that activation of the salicylic acid (SA) signaling pathway enhances ClYVV virulence in susceptible pea cultivars. The kinetics of virus accumulation was not significantly different between PI 118501 (Cyn1) and PI 226564 (cyn1); however, the SA-responsive chitinase gene (SA-CHI) and the hypersensitive response (HR)-related gene homologo

Unrecognized viral infections and chromosome abnormalities as a cause of fetal death - examination with fluorescence in situ hybridization, immunohistochemistry and polymerase chain reaction.

Fifteen to 50% of fetal deaths remain unexplained after post-mortem examination depending on inclusion criteria and classification systems. Our aim was to examine a selection of unexplained fetal deaths in order to investigate whether any common chromosome aberrations or viral infections were present. Reports from 351 fetal autopsies performed at the Department of Pathology and Medical Genetics at St. Olavs University Hospital from 2001 through 2010 were reviewed. Of these, 105 fetal deaths were classified as unexplained. Tissue samples from 30 cases were further examined with fluorescence in

Diversifying T-cell responses: safeguarding against pandemic influenza with mosaic nucleoprotein.

UNLABELLED: Pre-existing T-cell responses have been linked to reduced disease severity and better clinical outcomes during the 2009 influenza pandemic and the recent COVID-19 pandemic. We hypothesized that diversifying T-cell responses, particularly targeting conserved viral proteins such as the influenza A virus (IAV) nucleoprotein (NP), could protect against both epidemic and pandemic IAV strains. To test this, we created a mosaic nucleoprotein (MNP) by synthesizing a sequence that maximized the representation of 9-mer epitopes from 7422 NP sequences across human, swine, and avian IAVs. Nota

Transcriptional slippage in the positive-sense RNA virus family Potyviridae.

The family Potyviridae encompasses ~30% of plant viruses and is responsible for significant economic losses worldwide. Recently, a small overlapping coding sequence, termed pipo, was found to be conserved in the genomes of all potyvirids. PIPO is expressed as part of a frameshift protein, P3N-PIPO, which is essential for virus cell-to-cell movement. However, the frameshift expression mechanism has hitherto remained unknown. Here, we demonstrate that transcriptional slippage, specific to the viral RNA polymerase, results in a population of transcripts with an additional "A" inserted within a hi
