Localization of CORO1A in the macrophages containing Mycobacterium leprae.

Mycobacteria have acquired an intracellular lifestyle within the macrophage, which is best exemplified by the enlarged infected histiocytes seen in lepromatous leprosy. To survive within the cell, mycobacteria must escape intracellular bactericidal mechanisms. In a study of Mycobacterium bovis Bacille Calmette-Guérin (M. bovis BCG) infection, it was shown that the host protein, CORO1A, also known as tryptophan aspartate-containing coat protein (TACO), accumulates on the phagosomal membrane, resulting in inhibition of phagosome-lysosome fusion, and thus augmenting intracellular survival. In thi

The distribution of Mycobacterium leprae in the hair follicle of the eyebrow.

Longitudinal sections of hair follicles from the eyebrows of patients with active, untreated, lepromatous leprosy were studied. Mycobacterium leprae were found in the dermal papilla and outer root sheath of both anagen and telogen hair follicles. Bacilli were rarely found in those cell lines continuous with the environment, i.e., the hair shaft and the inner root sheath. The biochemical fate of these cell lines is such that they would not provide a suitable medium for the survival and transport of bacilli into the environment. We conclude that it is unlikely that the hair follicle plays a sign

In vitro activation of neutrophils by suspensions of Mycobacterium leprae.

Activation, defined as an increase in the proportion of cells that reduce nitroblue-tetrazolium in vitro, is present in neutrophils from patients with reactional lepromatous leprosy but not in neutrophils from patients with non-reactional lepromatous leprosy. Neutrophils from patients with all forms of leprosy are equally well activated by endotoxin in vitro. We have now shown that in vitro activation induced by Mycobacterium leprae suspensions is of comparable magnitude in neutrophils from patients with all forms of leprosy (including lepromatous and reactional lepromatous leprosy). There is

Antibody response in rabbits to immunization with Mycobacterium leprae.

Mycobacterium leprae purified from liver tissue of an infected armadillo (the A/10 preparation) was tested for antigenic composition by immunization of rabbits and characterization of the antibody response by crossed immunoelectrophoresis. The rabbit antisera detected seven distinct components in the M. leprae preparation. This number is far lower than in similar experiments with other mycobacteria. The M. leprae sonic extract gave far fewer lines after polyacrylamide gel electrophoresis and staining with Coomassie brillant blue than sonic extracts prepared from BCG, M. smegmatis, and M. phlei

Unprecedented in vivo activity of telacebec against Mycobacterium leprae.

BACKGROUND: New drugs targeting the electron transport chain (ETC) seem to be a promising advance in leprosy treatment. In this study, we evaluated the bactericidal activity of telacebec (TCB), a phase 2 drug candidate for tuberculosis, alongside known ETC-targeting antibiotics, bedaquiline (BDQ) and clofazimine (CFZ), as monotherapy or in combination. METHODOLOGY/ PRINCIPAL FINDINGS: We used the reference leprosy proportional bactericidal mouse footpad model. Four hundred and ten mice were inoculated in the footpads with 5x104 to 5x100 bacilli of M. leprae strain THAI53 for the untreated con

Mycobacterium leprae in Nine-Banded Armadillos (Dasypus novemcinctus), Ecuador.

We found Mycobacterium leprae, the most common etiologic agent of Hansen disease or leprosy, in tissues from 9 (18.75%) of 48 nine-banded armadillos (Dasypus novemcinctus) collected across continental Ecuador. Finding evidence of a wildlife reservoir is the first step to recognizing leprosy zoonotic transmission pathway in Ecuador or elsewhere.

Mycobacterium leprae-induced nerve damage: direct and indirect mechanisms.

Leprosy is a chronic infectious disease caused by Mycobacterium leprae. This disease is characterized by skin and peripheral nerve trunk damage. The mechanisms responsible for the observed nerve damage in leprosy could be directly related to the ability of M. leprae to infect Schwann cells, leading to triggering of signaling events. Therefore, we hypothesize that in response to M. leprae infection, activation of the Notch signaling pathway in Schwann cells could play a crucial role in glial cell dedifferentiation. On the other hand, nerve damage evidenced in this disease may be additionally ex

Bilateral iridocyclitis caused by Mycobacterium leprae diagnosed through paracentesis.

The authors conducted an anterior chamber paracentesis in a patient with lepromatous leprosy showing bilateral iridocyclitis. The paracentesis was performed in the outpatients department. The aqueous humor was studied by Ziehl-Nielsen staining method and the result was the isolation of the M. leprae in the anterior chamber. This study shows that M. leprae can promote uveitis in leprosy patients. Therefore, it should be looked for in patients having this type of disease.

Activity of two doses of rifampin against Mycobacterium leprae.

In the course of a clinical trial designed to re-examine the bactericidal efficiency of 600-mg doses of rifampin (RMP) against Mycobacterium leprae, two doses of RMP, either 600 mg or 1200 mg, were administered 28 days apart to 29 previously untreated patients with lepromatous or borderline leprosy. Seven, 28, and 35 days after the start of the trial, skin biopsies were performed and immunologically normal mice were inoculated with 5 x 10(3) or 10(4) M. leprae in each hind foot pad. The patients assigned to the two regimens did not differ significantly in terms of sex, age, disease classificat

An Id reaction to Mycobacterium leprae: first documented case.

We present a case of generalized sarcoidal dermatitis in a patient with tuberculoid leprosy. After careful consideration of the reactional states in leprosy and recognizing that id reactions occur in other mycobacterial infections, we concluded that the patient's eruption was likely an id reaction to his Mycobacterium leprae infection. It was most reminiscent of the tuberculids: lichenoid tuberculid and lichen scrofulosorum. However, its histomorphologic appearance was distinctive, showing elongated granulomata along with foci of incipient perineural granuloma formation, similar to the histomo

A species-specific repetitive sequence in Mycobacterium leprae DNA.

A 2.2-kilobase Mycobacterium leprae DNA insert fragment from a recombinant genomic library (pYA1065) was found to hybridize to at least 19 fragments of chromosomal M. leprae DNA by Southern hybridizations. The probe hybridized to identical fragments of chromosomal DNA from four M. leprae isolates (two from patients with leprosy, one from a naturally infected armadillo, and one from a naturally infected Mangabey monkey) whether the chromosomal DNA was digested with BamHI, BstEII, PstI, or SacI. The pYA1065 probe is specific for M. leprae; it did not hybridize to chromosomal DNA from 14 cultivab

Evaluation of Mycobacterium leprae immunogenicity via adoptive transfer studies.

The immune response of mice to live, heat-killed, or autoclaved Mycobacterium leprae was investigated. After sensitization with 10(7) organisms in each group, recipient mice were transfused with the sensitized splenocytes 28 days later. A selected number of these mice were infected with 5 X 10(3) M. leprae, and the remaining animals were sacrificed at scheduled intervals for evidence of cell-mediated immunity to the M. leprae cell extract. Data from these and the bacteriological assays showed that all three materials induce cell-mediated immunity and also extend protection against the M. lepra

Adenosine triphosphate content of Mycobacterium leprae from leprosy patients.

Mycobacterium leprae obtained from randomly selected lepromatous leprosy patients were used to evaluate the ATP assay technique for detecting viability of these cells. The findings were further confirmed by the standard mouse foot pad technique. While the latter takes about 8-12 months to obtain any valid information on the status of M. leprae, the ATP data can be generated within hours and at much lower cost. It is hoped that the ATP data could also instantaneously identify viable bacilli from patients taking dapsone and thereby identify dapsone-resistant patients so that alternative treatmen

Cell-mediated immune response in Mycobacterium leprae infected mice.

Positive skin reactions to homologous antigens in mice infected with Mycobacterium leprae were observed at the time that the bacillary multiplication was nearing the plateua level. Once developed, the skin reactivity persisted for a considerable length of time. Lymphocyte transformation studies indicated that the lymphocytes from infected animals were unable to respond to specific antigenic stimuli in the early phase of infection. Possible explanation for this failure could be that the lymphocytes although sensitized, were not capable of responding to the stimuli due to a rather low level of s

In vivo activities of novel benzoxazinorifamycins against Mycobacterium leprae.

Among the four newly synthesized benzoxazinorifamycin derivatives, KRM-1648 and KRM-2312 completely inhibited the multiplication of rifampicin-sensitive as well as rifampicin-resistant strains of M. leprae in the foot-pads of mice. Both were found to be more potent than rifampicin and were bactericidal. In combination with ofloxacin, another potent bactericidal drug against M. leprae, both KRM-1648 and KRM-2312 exhibited synergism. Thus, combination of one of these benzoxazinorifamycin derivatives and ofloxacin in multidrug regimens is worth evaluating in clinical trials.

Delipidified cell components of Mycobacterium leprae and its applications.

Delipidified cell components (DCC) of Mycobacterium leprae obtained as an insoluble material consist of several proteins. This preparation, DCC, has ability to differentially bind to sera from lepromatous leprosy patients and antibodies to this complex get reduced as patients improve under chemotherapy. The antigenic complex has no ability to bind to proteins of sera from normal healthy individuals or tuberculoid leprosy patients. The DCC is antigenic and is recognised by immune deficient cells of lepromatous leprosy patients, leading to lymphocyte proliferation, production of Interleukin II a

Lack of Mycobacterium leprae-specific uptake in Schwann cells.

Among mycobacteria, Mycobacterium leprae have a unique property to infect peripheral nerves, which is the cause of a variety of debilities seen in leprosy. The possibility of selective uptake of M. leprae by Schwann cells was studied using a rat Schwannoma cell line 33B and rat sciatic nerve-derived Schwann cells. M. leprae were phagocytosed by 33B cells but so also were seven other mycobacteria ("Mycobacterium w," BCG, M. tuberculosis H37Rv, M. nonchromogenicum, M. vaccae, ICRC bacillus, and M. smegmatis) which do not involve peripheral nerves. All three mycobacteria tested (M. leprae, M. tub

Delayed hypersensitivity tests with Mycobacterium leprae purified protein derivative.

Skin tests were conducted on 3 lepromatous, 3 dimorphous and 6 tuberculoid leprosy patients and 3 others not suffering from leprosy with lepromin, purified protein derivative from M. leprae of armadillo origin and tuberculin. Results show that a delayed hypersensitivity reaction could be produced with PPD in 72 hours on all Mitsuda positive cases, with one anomalous exception, without cross reaction to tuberculin. The results were promising from the point of view of substituting lepromin with PPD in usual tests.

IFN-γ-Driven macrophage responses in the immunity to Mycobacterium tuberculosis and Mycobacterium leprae.

Interferon-gamma (IFN-γ) is a key stimulator of macrophage defense against Mycobacterium tuberculosis (Mtb) and Mycobacterium leprae (M. leprae). Both pathogens adopt measures to circumvent the effects of the immune system, weakening the impact of IFN-γ and enabling them to survive in the cells. This review synthesizes how IFN-γ overdose transacts the JAK/STAT1-IRF1-transmitter to encourage maturation of phagolysosomes, reactive oxygen and nitrogen product generation, LC3-associated phagocytosis (LAP), autophagy, and improved antigen presentation and juxtaposes these pathways in tuberculosis a
