Role of Ferroptosis in the Redox Biology of Mycotoxins.

Ferroptosis, an iron-dependent regulated cell death process driven by lipid peroxidation, has emerged as a pivotal mechanism in mycotoxin-induced redox pathology. This review synthesizes the molecular interplay between fungal secondary metabolites (e.g., aflatoxin B1, deoxynivalenol) and ferroptotic pathways, revealing how mycotoxins subvert cellular iron homeostasis through transferrin receptor 1 (TfR1) upregulation and ferritinophagy-mediated iron release, while concurrently inactivating glutathione peroxidase 4 (GPX4) and depleting reduced glutathione. Through integrative analysis of tissue

Immunosuppressive effects of the mycotoxin patulin in macrophages.

Patulin (PAT) is a fungi-derived secondary metabolite produced by numerous fungal species, especially within Aspergillus, Byssochlamys, and Penicillium genera, amongst which P. expansum is the foremost producer. Similar to other fungi-derived metabolites, PAT has been shown to have diverse biological features. Initially, PAT was used as an effective antimicrobial agent against Gram-negative and Gram-positive bacteria. Then, PAT has been shown to possess immunosuppressive properties encompassing humoral and cellular immune response, immune cell function and activation, phagocytosis, nitric oxid

The involvement of mycotoxins in the development of endemic nephropathy.

Endemic nephropathy is a human kidney disease that still escapes scientific explanation. It is accompanied by a high incidence of urothelial tumors in rural populations in endemic areas, which suggests that a natural nephrotoxic and carcinogenic compound may be involved in the etiology. The most imputed causative agent of endemic nephropathy is the mycotoxin ochratoxin A (OTA), because of its confirmed nephrotoxic and carcinogenic action. This paper presents a review of studies of OTA in food collected in the endemic areas and in blood and urine of their residents. Data on the co-occurrence of

Fumonisin B(1): a neurotoxic mycotoxin.

Fumonisin B(1) (FB(1)) is a mycotoxin produced by Fusarium spp. moulds that contaminate crop, predominantly maize, all around the world. More than 15 types of fumonisins have been indentified so far, but FB(1) is the most abundant and toxicologically the most significant one. FB(1) has a wide range of toxic effects, depending on animal species. In horses FB(1) causes equine leukoencephalomalacia (ELEM), in pigs pulmonary oedema and in experimental rodents nephrotoxicity and hepatotoxicity. In humans exposure to FB(1) is linked with higher incidence of primary liver cancer and oesophageal cance

[Mycotoxins as a cause of disease in human beings].

Secondary metabolites, toxic to macro-organisms and micro-organisms, are produced by certain molds and some plant parasitic living fungi. A risk is given for man worldwide by ingestion or apparently also be other routes always undetected. In vivo-effects of the various mycotoxins are different, but mainly the liver is affected, expecially by the intake of smaller amounts of these poisons. Accordingly cirrhosis of the liver or primary liver carcinoma are expected in man as well as in animals and were already proved outside of Europe.

Apoptotic Janus-faced mycotoxins against thoracal and breast metastases.

Abdominal organs (liver, kidney, spleen) are frequent targets of cancer cell invasion but their primary tumours are less known for their metastatic potential to other organs e.g. to the breast. Despite the known connection of the pathogenesis from breast cancer to liver metastasis, the study of the spread in the opposite direction has been neglected. The notion that breast cancer could be a metastasis besides being a primary tumour is based on rodents' tumour models upon implantation of tumour cells under the capsule of the kidney or under the Glisson's capsule of the liver of rats and mice. Association between forage mycotoxins and liver disease in horses.

BACKGROUND: Outbreaks of liver disease in horses are common but the etiology of most remains unknown. Forage mycotoxins have been suspected to be a cause. OBJECTIVES: To examine the association between outbreaks of liver disease and the presence of mycotoxins in forage stored on the same premises. ANIMALS: Premises were identified where ≥4 horses were contemporaneously affected by liver disease, and a control group was formed from premises where ≥4 horses had been examined and found to have no evidence of liver disease. METHODS: Forage was collected from 29 case and 12 control premises. The

Impact of mycotoxins on human health in developing countries.

Adverse human health effects from the consumption of mycotoxins have occurred for many centuries. Although mycotoxin contamination of agricultural products still occurs in the developed world, the application of modern agricultural practices and the presence of a legislatively regulated food processing and marketing system have greatly reduced mycotoxin exposure in these populations. At the mycotoxin contamination levels generally found in food products traded in these market economies, adverse human health effects have largely been overcome. However, in the developing world, where climatic an

Early mycotoxins exposure: A hidden driver of cardiometabolic risk.

Mycotoxins are fungal contaminants frequently detected in staple foods worldwide. While their toxic effects on growth and organ function are well recognized, their contribution to cardiometabolic disease programming during early life has received less attention. This review highlights the role of developmental exposure to major mycotoxins, including aflatoxins, ochratoxins, fumonisins, zearalenone, and deoxynivalenol, as a hidden driver of long-term cardiometabolic risk. Evidence indicates that exposure occurs during fetal life through placental transfer and after birth through breast milk, in

Environmental mycotoxins: A potential etiological factor for neurodegenerative diseases?

Mycotoxins are potential environmental risk factors for neurodegenerative diseases. These toxins penetrate the central nervous system via a compromised blood-brain barrier, which may cause oxidative stress and neuroinflammation, these can also contribute to amyloid-beta (Aβ) plaque accumulation, Tau protein hyperphosphorylation, and neurofibrillary tangle formation. Mycotoxins also activate microglia, cause neuronal apoptosis, and disrupt central nervous system function. This study examines the evidence linking mycotoxin exposure to neurodegenerative disorders like Alzheimer's and Parkinson's

Efficacy of a mycotoxin-deactivating product to reduce the impact of Fusarium mycotoxin-contaminated rations in dairy cows during early lactation.

Livestock feeds can be contaminated by several fungi. Among these, Fusarium spp. are widespread and produce mycotoxins, such as deoxynivalenol (DON), zearalenone (ZEN), and fumonisins B1 and B2 (FB1 and FB2). We investigated whether feeding a TMR contaminated with moderate to high concentrations of ZEN, DON, FB1, and FB2 to dairy cows in early lactation alters (1) their feeding behavior and rumination time; (2) milk yield, composition, and quality; (3) plasma metabolic profile and biochemical traits; and (4) postpartum uterine involution and resumption of cyclicity. We tested whether adding a

Mycotoxins in food and agriculture: from occurrence to economic consequences.

Filamentous fungus (moulds) create secondary metabolites called mycotoxins, which can be dangerous. These minuscule molecules (typically less than 1 000 Da) are almost inescapably found in nature. Aspergillus, Penicillium, Fusarium, and Alternaria are among the fungal species that produce mycotoxins. If these secondary metabolites make their way into the food chain and production, they are harmful and have a big effect. In African countries like Nigeria that rely significantly on staple foods like grains, cereals, and nuts, mycotoxin contamination is especially problematic. Mycotoxins have att

Microbial detoxification of eleven food and feed contaminating trichothecene mycotoxins.

BACKGROUND: Contamination of agricultural commodities with multiple trichothecene mycotoxins, produced by toxigenic Fusarium species, is a food safety issue, which greatly affects grain production and marketing worldwide. Importantly, exposure to multiple trichothecenes may increase toxicity in animals due to their synergistic and/or additive effects. To address the problem this study aimed to achieve a novel biological trait capable of detoxifying various food and feed contaminating trichothecenes under aerobic and anaerobic conditions and wide range of temperatures. RESULTS: A highly enrich

Mycotoxins and human disease: a largely ignored global health issue.

Aflatoxins and fumonisins (FB) are mycotoxins contaminating a large fraction of the world's food, including maize, cereals, groundnuts and tree nuts. The toxins frequently co-occur in maize. Where these commodities are dietary staples, for example, in parts of Africa, Asia and Latin America, the contamination translates to high-level chronic exposure. This is particularly true in subsistence farming communities where regulations to control exposure are either non-existent or practically unenforceable. Aflatoxins are hepatocarcinogenic in humans, particularly in conjunction with chronic hepatit

Mycotoxins in food systems in Sub Saharan Africa: A review.

Mycotoxins, toxic secondary metabolites of fungi are now recognised as major cause of food intoxications in Sub Saharan Africa (SSA). Aflatoxins, the most important of the group have been implicated in acute aflatoxicoses, carcinogenicity, growth retardation, neonatal jaundice and immunological suppression in SSA. The hot and humid tropical climate provides ideal condition for growth of toxigenicAspergillus spp, making food contamination to be widespread in SSA, with maize and groundnuts being the most contaminated. The available data suggests that cassava products (the most important African
