Exercise reduces the risk of obesity-based, lifestyle-related diseases, such as metabolic abnormalities and cardiovascular diseases. The present review covers the health-promoting effects of exercise from the point of view of the physiologically active factor myokine, which is secreted by skeletal muscle, and focuses on the skeletal muscle as a new endocrine organ. Myokines have various effects, such as preventing metabolic syndrome by breaking down fat, preventing diabetes by improving glucose metabolism, and preventing progression of arteriosclerosis, dementia, and osteoporosis by enhancing

Obesity induces an imbalance in the expression and secretion of several cytokines, which contributes to the development of metabolic and cardiovascular disorders. On the contrary, skeletal muscle is known to have a role in reversing the detrimental impact of obesity. It has been established that adipose tissue acts as an endocrine organ that secretes proinflammatory and anti-inflammatory adipokines. Similarly, skeletal muscle produces secretory molecules, called myokines, from contracting muscle fibers. Myokines were recently recognized as beneficial modulators of obesity, metabolic syndrome,

Skeletal muscle, once regarded solely as a contractile tissue, is now recognized as a dynamic endocrine organ that secretes exercise-induced myokines-bioactive peptides with autocrine, paracrine, and endocrine functions. These myokines coordinate systemic energy homeostasis by regulating glucose and lipid metabolism, mitochondrial function, inflammation, and interorgan communication. Building on our previous review published in 2018, this review synthesizes major advances in exercise-induced myokines within an evidence-based framework considering mechanistic support and translational relevance

Skeletal muscle is a dynamic tissue involved not only in mechanical functions but also in the regulation of metabolic and immune processes. It secretes signaling molecules known as myokines, which act in autocrine, paracrine, and endocrine ways- affecting both muscle function and other tissues and organs. Well-known myokines include myostatin, IL-6, IL-15, FGF21, and irisin. They regulate muscle mass and strength, promote angiogenesis, maintain glucose and lipid homeostasis, and contribute to immune and anti-cancer responses. Their activity depends on physiological and pathological conditions

In the last decade, clear evidence has emerged that the cellular components of skeletal muscle are important sites for the release of proteins and peptides called "myokines", suggesting that skeletal muscle plays the role of a secretory organ. After their secretion by muscles, these factors serve many biological functions, including the exertion of complex autocrine, paracrine and/or endocrine effects. In sum, myokines affect complex multi-organ processes, such as skeletal muscle trophism, metabolism, angiogenesis and immunological response to different physiological (physical activity, aging,

Excess body weight constitutes a worldwide health problem with epidemic proportions impacting on the risk and prognosis of several disease states including malignancies. It is believed that the metabolic changes associated with weight gain, particularly visceral obesity, and physical inactivity could lead to dysfunctional adipose and muscle tissues causing insulin resistance, low-grade chronic inflammation and abnormal secretion of adipokines and myokines. The complex paracrine and endocrine interconnection between adipokines and myokines reflects a yin-yang balance with important implications

Pancreatic beta cells can adapt their mass and function to maintain normal glycemia when facing peripheral insulin resistance. To clarify the specific contribution and mechanisms of beta-cell mass adaptation in response to insulin resistance, we took advantage of genetic and pharmacologically induced insulin resistance in mice. We uncovered beta-cell expansion, via an increase in pancreatic islet density, as an adaptive mechanism triggered by mild-to-severe insulin resistance in young and older mice and documented pancreatic adaptation using 3D whole-pancreas analysis. Next, we found that insu

BACKGROUND: Persistent fatigue is one of the most common and debilitating symptoms experienced by patients recovering from COVID-19, contributing significantly to the burden of "long COVID" or post-COVID-19syndrome. However, the underlying pathophysiological mechanisms remain inadequately understood. Few studies have examined the association between fatigue and pulmonary or cardiac function, systemic biomarkers, or morphological changes in the lungs and diaphragm. Furthermore, the potential influence of vaccination on the persistence of fatigue has not been fully explored. This study aims to i

BACKGROUND: Some experimental data suggest that myokines may play an important role in developing cancer-associated cachexia (CAC), but their relevance in humans remains poorly explored. In our study, we tested the hypothesis that circulating myokines are associated with the pathogenesis of CAC in a model population of gastric cancer.
METHODS: A group of 171 treatment naïve patients with adenocarcinoma of the stomach were prospectively examined. Cachexia was defined as weight loss >5% or weight loss >2% with either BMI <20 kg/m2 or sarcopenia. A panel of 19 myokines was measured in portal and

The involvement of the hypothalamus and/or pituitary gland by granulomatous, infiltrative or autoimmune diseases is a rare condition of non-tumoral-non-vascular acquired hypothalamic dysfunction and hypopituitarism. In this paper, we present the case of a 26-year-old woman, who showed an amenorrhea-galactorrhea syndrome with hypogonadotropic hypogonadism due to an isolated hypothalamic-peduncular localization of neurosarcoidosis. Acquired GH deficiency was also demonstrated. This clinical case provided the opportunity for a review of the endocrine aspects linked to brain infiltrative diseases

BACKGROUND: Peritoneal dialysis (PD) patients are at high risk for complications such as muscle loss and peritonitis. Individual myokines and functional assessment of PD patients are inconsistently reported to associate with poor prognosis, but their complex interactions for risk stratification have not been studied.
METHODS: In 194 new PD patients, we measured the serum levels of a myokine panel, metabolic and inflammatory markers, together with physical function scores. We employed k-cluster analysis on the result and analyzed patient survival, technique survival, peritonitis-free survival,

Exercise can regulate the physiological functions of the body by inducing the secretion of myokines, which are bioactive factors mainly secreted by muscle cells. This review classifies myokines based on their functional characteristics, including metabolic regulation (such as myostatin, interleukin-6), neuroregulation (brain-derived neurotrophic factor), cell proliferation/differentiation regulation (myogenic proteins), immune regulation (tumor necrosis factor- alpha), and factors involved in angiogenesis and extracellular matrix remodeling (such as connective tissue growth factor).Cancer, as

Exercise-induced myokines, such as IL-6, irisin, and myostatin, play crucial roles in mediating systemic adaptations and modulating tumor progression. This narrative review synthesizes evidence on how regular moderate-intensity exercise affects the myokine network within the tumor microenvironment (TME) of solid tumors. Results from the 86 included studies indicate that structured training (aerobic: 45-65% VO2 max; resistance: 45-70% 1RM) enhances the expression of beneficial myokines that support immune surveillance, reduce inflammation, and improve muscle-tumor cross-talk. IL-6 demonstrates

Fatigue is a prevalent and debilitating symptom in rheumatic diseases such as rheumatoid arthritis and psoriatic arthritis. Despite advances in reducing inflammation through treatments, fatigue often persists, underscoring its multifactorial etiology. A possible link between the persistent inflammation observed in rheumatic diseases and the onset of fatigue has been suggested. The discovery that skeletal muscles also secrete cytokines and myokines, has opened new avenues for research. This narrative review explores current mechanistic insights and evidence on the dural role of myokines in exac

Over the past couple of decades, it has become apparent that skeletal muscles might be engaged in endocrine signaling, mostly as a result of exercise or physical activity in general. The importance of this phenomenon is currently studied in terms of the impact that exercise- or physical activity -induced signaling factors have, in the interaction of the "muscle-brain crosstalk." So far, skeletal muscle-derived myokines were demonstrated to intercede in the connection between muscles and a plethora of various organs such as adipose tissue, liver, or pancreas. However, the exact mechanism of mus

In the last few years, a growing number of molecules have been associated to an endocrine function of the skeletal muscle. Circulating myokine levels, in turn, have been associated with several pathophysiological conditions including the cardiovascular ones. However, data from different studies are often not completely comparable or even discordant. This would be due, at least in part, to the whole set of situations related to the preparation of the patient prior to blood sampling, blood sampling procedure, processing and/or store. This entire process constitutes the pre-analytical phase. The

INTRODUCTION: Skeletal muscles secrete myokines, including irisin and myostatin, which regulate inflammation and metabolism and may influence the severity of SARS-CoV-2 infection. This study investigated the associations between serum irisin and myostatin levels and COVID-19 severity.
METHODS: Ninety-nine adult patients hospitalized with PCR-confirmed COVID-19 were included. Serum irisin and myostatin concentrations were measured by ELISA at admission and discharge. Disease severity was evaluated using a four-point clinical scale, the RALE score for lung involvement, oxygenation indices (PaO2

With the expansion of technology and the prevalence of a sedentary lifestyle, physical inactivity and a lack of desire to strengthen muscle tissue have led to the accumulation of large forces on the joints and the spread of joint-related diseases, including osteoarthritis (OA) in humans. This disease can significantly impair the quantity and quality of life of patients and impose a large financial burden on the healthcare community. Controlling and inhibiting the progression of this disease at the cellular level is a therapeutic strategy for OA. The spread of inflammation and the destruction o
