IgM-mediated autoimmune responses directed against multiple neoepitopes in depression: new pathways that underpin the inflammatory and neuroprogressive pathophysiology.

BACKGROUND: There is evidence that depression is accompanied by oxidative and nitrosative stress (O&NS), as indicated by increased free radical levels, lipid peroxidation, and lowered antioxidant levels. The aims of the present study are to examine whether depression is accompanied by autoimmune responses directed against a) neoepitopes that are formed following O&NS damage; and b) the major anchorage molecules, i.e. palmitic and myristic acids and S-farnesyl-L-cysteine. METHODS: We examined serum IgM antibodies to the conjugated fatty acids, palmitic and myristic acids; acetylcholine; S-farn

Cellular fatty acid composition of Kingella species, Cardiobacterium hominis, and Eikenella corrodens.

We determined the cellular fatty acid composition of reference strains and clinical isolates of each of the three Kingella species, Cardiobacterium hominis, and Eikenella corrodens by using capillary gas chromatography. Kingella denitrificans and Kingella kingae contained myristic (14:0) and palmitic (16:0) acids as major acids, whereas cis-vaccenic (18:1 omega 7c) and palmitic acids were the major acids in Kingella indologenes, C. hominis, and E. corrodens. C. hominis differed from the other four species by the absence of 3-hydroxylauric (3-OH-12:0) acid, from K. indologenes by the presence o

Salicylic acid reverses phorbol 12-myristate-13-acetate (PMA)- and tumor necrosis factor alpha (TNFalpha)-induced insulin receptor substrate 1 (IRS1) serine 307 phosphorylation and insulin resistance in human embryonic kidney 293 (HEK293) cells.

Salicylates, including aspirin, have been shown to improve insulin sensitivity both in human and animal models. Although it has been suggested that salicylates sensitize insulin action by inhibiting IkappaB kinase beta (IKKbeta), the detailed mechanisms remain unclear. Protein kinase C isoforms and tumor necrosis factor alpha (TNFalpha) signaling pathways are well described mediators of insulin resistance; they are implicated in the activation of IKKbeta and the subsequent inhibition of proximal insulin signaling via insulin receptor substrate 1 (IRS1) and Akt. This study investigated the effe

Effect of type of fatty acid attached to chitosan on walnut oil-in-water Pickering emulsion properties.

The effect of fatty acid type bonded to chitosan on the emulsifying properties of chitosan-based particles was investigated. Capric acid, myristic acid, and stearic acid were attached to chitosan chains. Longer fatty acids in the structure of chitosan lead to the better and more uniform formation of chitosan nanogels. The contact angle of chitosan, chitosan-capric acid, chitosan-myristic acid and chitosan-stearic acid were found to be 52.5°, 60.0°, 65.1° and 72.5°, respectively. Different chitosan nanogels were used to stabilize walnut oil emulsions, and the emulsion stabilized with chitosan-s

Effects of immunosuppressants, calcineurin inhibition, and blockade of endoplasmic reticulum calcium channels on free fatty acid efflux from the ischemic/reperfused rat cerebral cortex.

Elevated levels of free fatty acids (FFA) have been implicated in the pathogenesis of neuronal injury and death induced by cerebral ischemia. This study evaluated the effects of immunosuppressants agents, calcineurin inhibitors and blockade of endoplasmic reticulum (ER) calcium channels on free fatty acid formation and efflux in the ischemic/reperfused (I/R) rat brain. Changes in the extracellular levels of arachidonic, docosahexaenoic, linoleic, myristic, oleic and palmitic acids in cerebral cortical superfusates during four-vessel occlusion-elicited global cerebral ischemia were examined usi

Insulin-Sensitizing Activity of Sub-Nanoscaled Polyalkoxyvanadate Clusters.

Sub-nanoscaled polyalkoxyvanadates (PAOVs) functionalized with various aliphatic acids are evaluated for their insulin-sensitizing activity in lowering the blood glucose levels of diabetic mice in typical glucose tolerance tests. All the PAOVs can restore the blood glucose to normal levels after a single oral administration of PAOVs. Among them, the myristic acid-modified PAOVs enable the response of insulin to the repeated glucose challenges, lasting for up to 13 h. The combined administration of PAOVs exerts better glucose control over insulin alone, while the capric acid- and myristic acid-

Evaluation of an oviposition-stimulating kairomone for the yellow fever mosquito, Aedes aegypti, in Recife, Brazil.

A synthetic mixture of an oviposition-stimulating kairomone for the yellow fever mosquito, Aedes aegypti, comprising of 83% tetradecanoic acid, 16% nonanoic acid and 1% tetradecanoic acid methyl ester (NTT, in short) was tested in a dengue endemic area in Recife, Brazil. Gravid female mosquitoes confined to a cage under semi-field conditions deposited significantly higher numbers of eggs in traps baited with NTT at doses ranging from 0.6 to 600 ng/microl than in control (water) traps. When tested in homes, egg-laying in traps baited with 60 ng NTT/microl (final concentration in trap, approxima

Evidence for tetradecanal as the natural aldehyde in bacterial bioluminescence.

Dim aldehyde mutants of the luminous bacterium Beneckea harveyi emit light with exogenously added long-chain aliphatic aldehyde. In one class of these mutants, luminescence is also stimulated by myristic (tetradecanoic) acid. In such mutants the amount of light obtained by the addition of a small (limiting) amount of either tetradecanal or myristic acid may be increased 60-fold by cyanide and other agents that block respiration. This indicates that the fatty acid product of the luminescent reaction is recycled. The effect, like the stimulation by exogenous fatty acid, exhibits specificity for

Changes in plasma fatty acid composition in females with lipedema following low-carbohydrate vs low-fat diets and associations with pain reduction.

BACKGROUND: Lipedema is a chronic female disease characterized by a painful accumulation of adipose tissue in the limbs. Plasma fatty acid (FA) composition has been proposed as a potential modulator of pain. However, the pathophysiology behind lipedema pain remains uncertain. The primary objective of this secondary analysis was to compare changes in plasma concentrations of FAs between low-energy diets either low in carbohydrates or low in fat, in females with lipedema and obesity. A secondary objective was to investigate potential associations between changes in pain and changes in the concen

Red Palm Oil: A Review on Processing, Health Benefits and Its Application in Food.

This review is aimed to provide a comprehensive overview of the physicochemical properties and extraction processes of red palm oil, its nutritional properties and applications in food. Crude palm oil is firstly extracted from the fruit mesocarp and processed into red palm oil using pre-treatment of crude palm oil, with deacidification steps, and deodorization via short-path distillation. These processes help to retain β-carotene and vitamin E in red palm oil. Palmitic, stearic and myristic acids are the saturated fatty acids in red palm oil, while the unsaturated fatty acids are oleic, linole

Water soluble complexes of C14 and C16 fatty acids and alcohols in media for cultivation of leprosy-derived psychrophilic mycobacteria.

Host-grown Mycobacterium leprae cell suspensions oxidized water-soluble complexes of palmitic acid, myristic acid, cetyl alcohol, and myristyl alcohol prepared with randomly methylated-beta-cyclodextrin as host molecules. Gas chromatography analysis showed that the water-soluble complexes retained their chemical structure following sterilization in the autoclave. Bioavailability of the two long-chain fatty acids and the corresponding long-chain alcohols was confirmed by Warburg manometric techniques with host-grown M. leprae cell suspensions. A rapid gas chromatographic method for the quantitation of underivatised individual free fatty acids in plasma.

A rapid method for quantitating individual free fatty acids (FFA) in plasma has been developed. An internal standard is added to plasma, the FFA are extracted into an organic solvent, concentrated and injected into a gas chromatograph. For routine estimations, extraction and assay of a single sample takes about 30 min and up to 30 samples can be assayed in a day. Seven FFA (myristic, palmitic, palmitoleic, stearic, oleic, linoleic and linolenic acids), are routinely quantitated. This method can be used to monitor individual FFA levels, to determine the percentage composition of individual FFA

Suitability of polymer materials for production of pulmonary microparticles using a PGSS supercritical fluid technique: preparation of microparticles using PEG, fatty acids and physical or chemicals blends of PEG and fatty acids.

The production of microparticles using a supercritical carbon dioxide based PGSS technique (CriticalMix™) has been exploited to develop blended systems targeted at pulmonary delivery. Hence, PEG based polymers of different molecular weights (1000-6000 Da) were blended in situ with fatty acids (stearic, palmitic or myristic acid) or with commercially available PEG-stearates. The effect of the different thermodynamic properties of the polymers was evaluated by characterising the microparticles produced in terms of their melting temperature by conventional DSC and in the presence of high pressure

Effects of different fatty acids on the structure, physicochemical properties, and in vitro digestibility of Chinese yam resistant starch-lipid complexes.

Nine CYRS-FA complexes were prepared by resistant starch in Chinese yam (CYRS) and nine fatty acids (FAs) with different chain lengths and degrees of unsaturation. CYRS-myristic acid and CYRS-palmitic acid showed higher complexing index (CI) and relative crystallinity (RC); CYRS-myristic acid and CYRS-oleic acid exhibited lower estimated glycemic index (eGI). Chain lengths of FAs showed significantly positive correlations with CI and contact angle (CA), and yet, unsaturation degree of FAs was negative correlated with both CI and CA. The eGI exhibited positive relations with solubility, and neg

Antimicrobial and Biofilm Inhibition Activity of Novel Biobased Quaternary Ammonium Salts.

The growing threat of antibiotic-resistant bacteria continues to be one of the biggest challenges facing public health. As a result, there is an increasing focus on developing new substances with both antimicrobial and biofilm inhibition activities. One such group of compounds is surfactants, particularly quaternary ammonium salts (QASs), which are commonly used as disinfectants in healthcare. In this study, a three-step synthesis was used to prepare a range of QASs, including quaternary esters, hydroxyamides, and dihydroxyamides with alkyl chains of 12-18 carbon atoms. First, the initial step

Effects of fatty acids on the growth and composition of Malassezia pachydermatis and their relevance to canine otitis externa.

Malassezia pachydermatis is one of the many infective agents involved in canine otitis externa. Two major phenotypes (large and small colony types), which had different fatty acid compositions, were found among strains of M pachydermatis isolated from canine ears or skin. The large-colony type contained palmitic, stearic, oleic and linoleic acids, whereas the small-colony type contained myristic, palmitic, stearic, linoleic acids and five other unidentified fatty acids. Exogenous saturated fatty acids in the medium had no effect on the growth of a large colony strain but oleic and linoleic aci

Membrane-disrupting medium-chain fatty acids reduce Staphylococcus aureus persister cell survival during antibiotic treatment.

OBJECTIVE: Eliminating persister cells is essential to improve treatment of chronic infections and to limit the emergence of resistant strains. Medium-chain fatty acids (MCFAs) are widely used in cosmetics and antibiotic ointments where Staphylococcus aureus is a common commensal. This study evaluated the potential of MCFAs to eradicate S. aureus persister cells and investigated their mechanisms of action. METHODS: The bactericidal activity of MCFAs against S. aureus persisters was assessed after treatment with three antibiotics - ciprofloxacin, oxacillin, and tobramycin. Membrane permeabilit
