BACKGROUND: NAA10 is the catalytic subunit of the major N-terminal acetyltransferase complex NatA which acetylates almost half the human proteome. Over the past decade, many NAA10 missense variants have been reported as causative of genetic disease in humans. Individuals harboring NAA10 variants often display variable degrees of intellectual disability (ID), developmental delay, and cardiac anomalies. Initially, carrier females appeared to be oligo- or asymptomatic with X-inactivation pattern skewed towards the wild type allele. However, recently it has been shown that NAA10 variants can cause

N-acetyltransferase 10 (NAT10) has been considered a target for the treatment of human diseases such as cancer and laminopathies; however, its functional role in the biology of melanocytes is questionable. Using a small molecule or small interfering RNA targeting NAT10, we examined the effect of NAT10 inhibition on melanogenesis and melanoma growth in human and mouse melanoma cells. Genetic silencing or chemical inhibition of NAT10 resulted in diminished melanin synthesis through the suppression of melanogenesis-stimulating genes such as those encoding dopachrome tautomerase (DCT) and tyrosina

Resistance to platinum-based drugs represents a significant challenge in the clinical treatment of esophageal cancer. Particularly, some esophageal cancer patients exhibit primary resistance to platinum-based drugs. Our study revealed that the level of N-acetyltransferase 10 (NAT10) is inversely correlated with the efficacy of platinum-based drug neoadjuvant therapy. In the presence of oxaliplatin, NAT10 significantly reduced the cytotoxicity of oxaliplatin and enhanced esophageal cancer cells' invasive, migratory, and clonogenic abilities. Mechanistically, oxaliplatin promoted the binding of

BACKGROUND: N-terminal acetylation is a common protein modification in human cells and is catalysed by N-terminal acetyltransferases (NATs), mostly cotranslationally. The NAA10-NAA15 (NatA) protein complex is the major NAT, responsible for acetylating ~ 40% of human proteins. Recently, NAA10 germline variants were found in patients with the X-linked lethal Ogden syndrome, and in other familial or de novo cases with variable degrees of developmental delay, intellectual disability (ID) and cardiac anomalies.
METHODS: Here we report a novel NAA10 (NM_003491.3) c.248G > A, p.(R83H) missense varia

As the only known RNA N4-acetylcytidine (ac4C) acetyltransferase in mammalian cells, NAT10 has been reported to drive tumor progression, yet its role in acute myeloid leukemia (AML) chemoresistance and the underlying mechanisms remain unclear. NAT10 expression was evaluated in AML cells with acquired resistance to cytarabine (Ara-C). Integrated bioinformatic analysis combining a public NAT10-dependent ac4C profiling dataset with AML-related NAT10 knockdown transcriptomic data was conducted to screen out candidate targets. For mechanistic validation, gain- and loss-of-function assays, ac4C dot

The NatA N-acetyltransferase complex is important for cotranslational protein modification and regulation of multiple cellular processes. The NatA complex includes the core components of NAA10, the catalytic subunit, and NAA15, the auxiliary component. Both NAA10 and NAA15 have been associated with neurodevelopmental disorders with overlapping clinical features, including variable intellectual disability, dysmorphic facial features, and, less commonly, congenital anomalies such as cleft lip or palate. Cardiac arrhythmias, including long QT syndrome, ventricular tachycardia, and ventricular fib

BACKGROUND: NAA10-related and NAA15-related neurodevelopmental (ND) syndromes present with intellectual disability, hypotonia, cardiac abnormalities, and delayed development. While data exists on the clinical manifestations of these conditions, there are few reports describing the neuroanatomical abnormalities present on imaging.
OBJECTIVE: We aim to provide neuroimaging analyses for a subset of probands with NAA10- and NAA15-related neurodevelopmental symptoms and assess the severity and number of neuroanatomical anomalies and their associated functional impairments to better understand the

Intestinal fibroblast senescence contributes to gut aging and the development of related diseases, yet the regulatory mechanisms governing this process remain largely elusive. In this study, we identify the RNA N4-acetylcytidine (ac4C) modification and its cognate writer, N-acetyltransferase 10 (NAT10), as key modulators of human colonic fibroblast senescence. Through a series of loss- and gain-of-function assays, we demonstrate that NAT10-mediated mRNA ac4C modification is functionally involved in controlling intestinal fibroblast senescence. Suppression of this modification diminishes senesc

BACKGROUND: Diabetic kidney disease (DKD) is a frequent complication associated with diabetes. CREBRF was reported to be closely associated with endoplasmic reticulum stress. However, the upstream regulatory mechanism by which CREBRF modulates endoplasmic reticulum stress in DKD renal tubular injury is unknown.
METHODS: HK-2 cells were treated with high glucose to construct a cellular model of DKD. C57BL/6J mice were injected intraperitoneally with streptozotocin (STZ) to construct an animal model of DKD. Co-IP was employed to assess the interactions between BTRC or NAA10 and CREBRF, as well

OBJECTIVES: The catalytic subunit of NatA, the main component of the N-terminal acetyltransferase complex, which is involved in most of the acetylation of the human proteome, is encoded by the NAA10 gene. Mutations in the NAA10 gene lead to neurodegenerative diseases associated with disruption of acetylation. Ogden syndrome (OS) is a rare X-linked recessive or dominantly inherited disorder associated with NAA10 gene mutations, characterized by variable findings such as autism spectrum disorder, intellectual disability, and cardiac anomalies. In this article, it is aimed to clarify the function

We present a male patient born at 38-wk gestation with rhizomelic shortening of extremities, hepatomegaly, ventriculomegaly, heart failure, severely depressed left ventricular function, biventricular hypertrophy, and biatrial enlargement. Additional physical findings included anteriorly displaced anus, vertebral anomalies, and brachydactyly. The patient's cardiac malformations led to persistent hypotension, sinus tachycardia, and multiorgan failure in the absence of arrhythmias. Rapid whole-exome sequencing was ordered on day of life (DOL) 8. The patient's family elected to withdraw supportive

N-terminal acetylation is a common protein modification in eukaryotes associated with numerous cellular processes. Inherited mutations in NAA10, encoding the catalytic subunit of the major N-terminal acetylation complex NatA have been associated with diverse, syndromic X-linked recessive disorders, whereas de novo missense mutations have been reported in one male and one female individual with severe intellectual disability but otherwise unspecific phenotypes. Thus, the full genetic and clinical spectrum of NAA10 deficiency is yet to be delineated. We identified three different novel and one k

Cardiac fibrosis is featured cardiac fibroblast activation and extracellular matrix accumulation. Ac4C acetylation is an important epigenetic regulation of RNAs that has been recently discovered, and it is solely carried out by NAT10, the exclusive enzyme used for the modification. However, the potential regulatory mechanisms of ac4C acetylation in myocardial fibrosis following myocardial infarction remain poorly understood. In our study, we activated fibroblasts in vitro using TGF-β1 (20 ng/mL), followed by establishing a myocardial infarction mouse model to evaluate the impact of NAT10 on co

Glycolysis is crucial for promoting cancer progression. However, the precise mechanism underlying glycolysis regulating the angiogenic process remains to be defined. Here, we demonstrate that in human gastric cancer cells, the E3 ligase TRIM8 promotes the K63-linked ubiquitination of the glycolytic enzyme PGK1 and improves its stability, which leads to acetyltransferase ACAT1 recruitment, increased interaction of PGK1 with ACAT1, and subsequent PGK1 acetylation-dependent glycolytic activity. This activity facilitates PGK1-mediated glycolysis, lactate accumulation and triggers a significant inc

OBJECTIVE: Metabolic dysfunction-associated steatotic liver disease (MASLD) and metabolic dysfunction-associated steatohepatitis (MASH) are characterized by excessive triglyceride accumulation in the liver. However, due to an incomplete understanding of its pathogenesis, more efforts are needed to identify specific and effective treatments. N4-acetylcytidine (ac4C) is a newly discovered RNA modification to regulate mRNA. N-acetyltransferase 10 (NAT10) has not been fully explored in MASLD and MASH.
METHODS: The clinical relevance of NAT10 was evaluated based on its expression in various mouse

Most chloroplast proteins are synthesized in the cytosol as precursor proteins carrying N-terminal transit peptides that mediate post-translational import into the organelle. During this transient cytosolic phase, precursor proteins are exposed to the cellular N-terminal processing machinery, including methionine aminopeptidases and N-terminal acetyltransferases (NATs). Recent proteomic studies have provided direct evidence that plastid precursor proteins undergo N-terminal acetylation prior to import, establishing a previously unrecognized link between co-translational processing and plastid
