We have studied the growth-inhibitory potency of methotrexate and methotrexate-gamma-aspartate encapsulated in liposomes conjugated to ligands of ecto-NAD+-glycohydrolase (Salord, J. et al., Biochim. CD38 is a 42-kDa type II transmembrane glycoprotein that has been shown to catalyze the synthesis and hydrolysis of cyclic ADP-ribose, a metabolite with well-known calcium mobilizing properties independent of IP3. Inhibitors of poly(ADP-ribose) polymerase show a synergistic potentiation of cytotoxicity with certain DNA-damaging agents. The NADase activity of the ectoenzyme CD38 on intact cells has been measured by a fluorometric assay using epsilon-NAD as a substrate.
