Neutrophil extracellular traps aggravate neuronal apoptosis and neuroinflammation via neddylation after traumatic brain injury.

Rationale: Neddylation, akin to ubiquitination, regulates various cellular processes by attaching neuronal precursor cell-expressed developmentally down-regulated protein 8 (NEDD8) to target proteins. Its specific role in traumatic brain injury (TBI) remains poorly defined. Neutrophil extracellular traps (NETs), which accumulate at injury sites in TBI models, are linked to poor outcomes, yet the connection between NETs and neddylation remains unclear. Methods: We analyzed contused brain tissues from TBI patients and mice subjected to controlled cortical impact (CCI) using Western blotting, imm

Targeting NAE1 suppresses osteoclastogenesis via dual regulation of ferritinophagy and ACSL3-mediated ferroptosis.

Neddylation regulates diverse cellular processes, yet its role in osteoclast-mediated bone resorption is poorly understood. Here, we identify NAE1 (NEDD8 activating enzyme E1 subunit 1)-mediated neddylation as a critical regulator of postmenopausal osteoporosis and osteoclast differentiation through two distinct regulatory mechanisms. Pharmacological inhibition of Nae1 or myeloid-specific genetic ablation of Nae1 attenuated osteoclastogenesis in vitro and ameliorated ovariectomy (OVX)-induced osteoporosis in vivo without impairing osteoblast function. Mechanistically, Nae1 depletion disrupted

Discovery of spirocyclic amide scaffolds as novel neddylation E1 inhibitors to suppress the growth and survival of lung cancer cells.

This study identifies and characterizes compound 24 (HA-218-6-31-80), a novel small-molecule NEDD8-activating enzyme (NAE) inhibitor. Discovered from a high-throughput screening, followed by structure-guided optimization, compound 24 possesses a distinct spirocyclic amide chemical core entirely different from the clinical-stage NAE inhibitor MLN4924. Biochemical assays confirm that compound 24 directly binds NAE and suppresses its catalytic activity, blocks the formation of E2-NEDD8 thioesters, and reduces the neddylation of CUL1 and CUL5 in purified protein systems. In human lung cancer cells

Mechanism of phospho-Ubls' specificity and conformational changes that regulate Parkin activity.

PINK1 and Parkin mutations lead to the early onset of Parkinson's disease. PINK1-mediated phosphorylation of ubiquitin (Ub), ubiquitin-like protein (NEDD8), and ubiquitin-like (Ubl) domain of Parkin activate autoinhibited Parkin E3 ligase. The mechanism of various phospho-Ubls' specificity and conformational changes leading to Parkin activation remain elusive. Herein, we show that compared to Ub, NEDD8 is a more robust binder and activator of Parkin. Structures and biophysical/biochemical data reveal specific recognition and underlying mechanisms of pUb/pNEDD8 and pUbl domain binding to the RI

Neddylation cooperates with multiple phosphorylation to enhance FGFR1 stability and drive breast cancer progression.

BACKGROUND: Fibroblast growth factor receptor 1 (FGFR1) is frequently overexpressed in breast cancer (BC), yet the mechanisms governing its protein stabilization remain unclear. This study investigates the role of neddylation, a ubiquitin-like modification, in sustaining FGFR1 levels and driving malignant progression in BC. METHODS: The interaction between FGFR1 and the neddylation pathway was explored using clinical databases and experimental models. After characterizing FGFR1 neddylation via immunofluorescence and co-immunoprecipitation, we examined protein stability and signaling pathways

Neddylation Targets and Stabilizes NLRP3 to Augment Inflammasome-Mediated Colitis and Mood Disorder.

The activation of NLRP3 inflammasome contributes to the development of numerous chronic inflammatory diseases, including ulcerative colitis and stress-induced anxiety. Recent studies have revealed that NLRP3 K48-linked ubiquitination by several E3 ligases, including Trim31, leads to degradation. Neddylation is a process highly similar to ubiquitination by covalently conjugating NEDD8 to lysines in specific substrate proteins. Neddylation of substrate proteins alters their subcellular localization, stability, and activity. The role of neddylation in the NLRP3 inflammasome remains elusive. Here,

EXOSC4 as a novel regulator of neddylation in acute myeloid leukemia.

Acute myeloid leukaemia (AML) is a common haematological malignancy with an unsatisfactory prognosis despite recent therapeutic advances. Neddylation and ferroptosis have been implicated in the regulation of malignant cell proliferation, survival and therapy response. However, the relationship between neddylation-related dysregulation and ferroptosis-associated pathways in AML remains poorly understood. Transcriptomic data from TCGA-AML and GTEx normal controls were integrated for differential expression and functional enrichment analyses. Prognostic neddylation-related genes were identified u

MLN4924 suppresses kidney renal clear cell carcinoma progression through coordinated inhibition of Cullin1 neddylation and PSMB10-associated NF-κB inflammatory signaling.

BACKGROUND: Kidney renal clear cell carcinoma (KIRC) is characterized by malignant progression, inflammatory signaling, and therapeutic resistance. This study investigated whether MLN4924, a selective NEDD8-activating enzyme inhibitor, suppresses KIRC progression through coordinated regulation of Cullin1 neddylation, PSMB10 expression, and NF-κB signaling. METHODS: KIRC cells were treated with MLN4924, followed by Western blotting, transcriptomic sequencing, pathway enrichment analysis, PSMB10 knockdown, and NF-κB pathway assessment. NEDD8, stress granules, and amyotrophic lateral sclerosis: unveiling the therapeutic potential of the NEDP1 protease.

Protein quality control (PQC) systems are crucial for maintaining cellular proteostasis, particularly under stress that promotes misfolded protein accumulation. A central component of this response is the assembly of stress granules (SGs), cytoplasmic condensates of RNA and proteins that temporarily stall translation. Aberrant SG dynamics, often linked to mutations in SG proteins, contribute to neurodegenerative diseases such as amyotrophic lateral sclerosis (ALS), where persistent protein aggregates are hallmarks. This review examines the emerging role of the ubiquitin-like modifier NEDD8 and

FBXO11 amplifies type I interferon signaling to exert antiviral effects by facilitating the assemble of TRAF3-TBK1-IRF3 complex.

As the key component of host innate antiviral immunity, type I interferons (IFN-Is) exert multiple antiviral effects by inducing hundreds of IFN-stimulated genes. However, the precise mechanism involved in host sensing of IFN-I signaling priming is particularly complex and remains incompletely resolved. This research identified F-box protein 11 (FBXO11), a component of the E3-ubiquitin ligase SKP/Cullin/F-box complex, acted as an important regulator of IFN-I signaling priming and antiviral process against several RNA/DNA viruses. FBXO11 functioned as an essential enhancer of IFN-I signaling by

Activation of MAPK/ERK signaling by Burkholderia pseudomallei cycle inhibiting factor (Cif).

Cycle inhibiting factors (Cifs) are virulence proteins secreted by the type III secretion system of some Gram-negative pathogenic bacteria including Burkholderia pseudomallei. Cif is known to function to deamidate Nedd8, leading to inhibition of Cullin E3 ubiquitin ligases (CRL) and consequently induction of cell cycle arrest. Here we show that Cif can function as a potent activator of MAPK/ERK signaling without significant activation of other signaling pathways downstream of receptor tyrosine kinases. Importantly, we found that the ability of Cif to activate ERK is dependent on its deamidase

Discovery of new 7-azaindole derivatives as CSN5 inhibitors with in vitro antitumor activity against triple-negative breast cancer.

COP9 signalosome subunit 5 (CSN5) exerts critical deneddylase activity and is frequently overexpressed in triple-negative breast cancer (TNBC), representing a compelling antitumour target. However, to date, only a limited number of inhibitors targeting CSN5 have been reported, and none of them have advanced into clinical studies. Based on lead compound 1, we designed and synthesized two series of 7-azaindole derivatives functionalized with piperazinylethylamine (2-7) and propylenediamine (8-19) side-chains. Combined enzymatic fluorescence polarization assays and cellular NEDD8-Cul1-based bioma

Ubiquitination of NKCC2 by the cullin-RING E3 ubiquitin ligase family in the thick ascending limb of the loop of Henle.

The Na+/K+/2Cl- cotransporter (NKCC2) in the thick ascending limb of the loop of Henle (TAL) mediates NaCl reabsorption. cGMP, the second messenger of nitric oxide and atrial natriuretic peptide, inhibits NKCC2 activity by stimulating NKCC2 ubiquitination and decreasing surface NKCC2 levels. Among the E3 ubiquitin ligase families, the cullin-RING E3 ubiquitin ligase (CRL) family is the largest. Cullins are molecular scaffold proteins that recruit multiple subunits to form the CRL complex. We hypothesized that a CRL complex mediates the cGMP-dependent increase in NKCC2 ubiquitination in TALs. C

Pharmacological inhibition of Epac1 protects against pulmonary fibrosis by blocking FoxO3a neddylation.

BACKGROUND: Idiopathic pulmonary fibrosis (IPF) is marked by progressive lung scarring with no existing cure, emphasising the need for new therapeutic targets. Current evidence suggests that cyclic adenosine monophosphate (cAMP) mitigates lung fibroblast proliferation via the protein kinase A pathway, but the impact of exchange proteins directly activated by cAMP 1 (Epac1) on IPF remains unexplored. OBJECTIVE: To investigate the role of Epac1 in IPF progression. METHODS: We examined lung samples from IPF patients and controls, and from a bleomycin-induced mouse model of pulmonary fibrosis. T

Neddylation and MASLD: From Pathophysiology to Therapy.

Metabolic dysfunction-associated steatotic liver disease (MASLD), formerly known as non-alcoholic fatty liver disease (NAFLD), is intimately linked to the dysregulation of key transcription factors or enzymes involved in hepatic metabolism. Neddylation, a post-translational modification, refers to the covalent attachment of neuronal precursor cell-expressed developmentally down-regulated 8 (Nedd8) to specific substrates and plays a significant role in regulating protein conformation, localisation and activity. The connection between neddylation and MASLD is a burgeoning area of research that i

Cancer-Intrinsic Protein Neddylation Modulates the Intratumoral Immune Landscape to Constrain Immune Checkpoint Blockade Therapy.

The cancer-intrinsic network controlling the response to immune checkpoint blockade therapy remains elusive. Using a published single-cell RNA sequencing (scRNA-seq) dataset from patients with breast cancer, we found that pretherapy mRNA expression of NEDD8, which encodes the regulatory protein essential for neddylation, in cancer cells was negatively associated with T-cell expansion upon pembrolizumab treatment. Genetic ablation of protein neddylation in murine tumors induced a curative response to PD-1 or PD-L1 blockade in mice. Combined analysis of neddylation-deficient murine tumors using

Targeting NAE1-mediated protein hyper-NEDDylation halts cholangiocarcinogenesis and impacts on tumor-stroma crosstalk in experimental models.

BACKGROUND & AIMS: Cholangiocarcinoma (CCA) comprises a heterogeneous group of malignant tumors associated with dismal prognosis. Alterations in post-translational modifications (PTMs), including NEDDylation, result in abnormal protein dynamics, cell disturbances and disease. Herein, we investigate the role of NEDDylation in CCA development and progression. METHODS: Levels and functions of NEDDylation, together with response to pevonedistat (NEDDylation inhibitor) or CRISPR/Cas9 against NAE1 were evaluated in vitro, in vivo and/or in patients with CCA. The development of preneoplastic lesions

Protein neddylation as a therapeutic target: challenges and opportunities.

Protein neddylation is an evolutionarily conserved posttranslational modification that conjugates NEDD8 to its substrate, catalyzed by an E1-activating enzyme, E2-conjugating enzyme, and E3 ligase. Neddylation is essential for cellular homeostasis, and its dysregulation has been implicated in diverse human diseases, including cancer, neurodegenerative diseases, and metabolic disorders, making the process a promising therapeutic target. In this Review, we systematically summarize the biochemical activity and biological functions of neddylation; its alterations in human diseases, particularly in

Identification of NUB1 as a suppressor of mutant Huntington toxicity via enhanced protein clearance.

Huntington's disease is caused by expanded CAG repeats in HTT, conferring toxic gain of function on mutant HTT (mHTT) protein. Reducing mHTT amounts is postulated as a strategy for therapeutic intervention. We conducted genome-wide RNA interference screens for genes modifying mHTT abundance and identified 13 hits. We tested 10 in vivo in a Drosophila melanogaster Huntington's disease model, and 6 exhibited activity consistent with the in vitro screening results. Among these, negative regulator of ubiquitin-like protein 1 (NUB1) overexpression lowered mHTT in neuronal models and rescued mHTT-in

Obscurin and KCTD6 regulate cullin-dependent small ankyrin-1 (sAnk1.5) protein turnover.

Protein turnover through cullin-3 is tightly regulated by posttranslational modifications, the COP9 signalosome, and BTB/POZ-domain proteins that link cullin-3 to specific substrates for ubiquitylation. In this paper, we report how potassium channel tetramerization domain containing 6 (KCTD6) represents a novel substrate adaptor for cullin-3, effectively regulating protein levels of the muscle small ankyrin-1 isoform 5 (sAnk1.5). Binding of sAnk1.5 to KCTD6, and its subsequent turnover is regulated through posttranslational modification by nedd8, ubiquitin, and acetylation of C-terminal lysine
