Blisters, pruritus, and fever after bites by the Arabian tick Ornithodoros (Alectorobius) muesebecki.

A biologist was bitten by Ornithodoros (Alectorobius) muesebecki Hoogstraal, an endemic tick parasite of nesting and resting marine birds on islands in eastern Arabia. Irritating bullae developed and for four months he experienced intermittent inflammation and irritation. Two years earlier, after being bitten by the same tick species on a different island, he had experienced only irritation lasting no more than a fortnight. Petroleum-industry labourers on another island were admitted to hospital for about two weeks with bullae at numerous bite sites, intense pruritus, headache, and fever. Zirq

Tick-borne virus diseases of human interest in Europe.

Several human diseases in Europe are caused by viruses transmitted by tick bite. These viruses belong to the genus Flavivirus, and include tick-borne encephalitis virus, Omsk haemorrhagic fever virus, louping ill virus, Powassan virus, Nairovirus (Crimean-Congo haemorrhagic fever virus) and Coltivirus (Eyach virus). All of these viruses cause more or less severe neurological diseases, and some are also responsible for haemorrhagic fever. The epidemiology, clinical picture and methods for diagnosis are detailed in this review. Most of these viral pathogens are classified as Biosafety Level 3 or

Molecular screening of tick-borne pathogens in host-seeking Haemaphysalis punctata Canestrini & Fanzago, 1878 (Ixodoidea: Ixodidae) in Anatolia with the first report of Burana virus.

BACKGROUND: Haemaphysalis punctata is a widespread Palearctic tick species, yet its role in the circulation of tick-borne pathogens in Türkiye remains poorly characterized. No systematic pathogen survey has previously been conducted on host-seeking individuals of this species. This study aimed to provide the first comprehensive molecular investigation of bacterial, protozoan, and viral tick-borne pathogens (TBPs) in questing H. punctata populations in Central and Northeastern Anatolia. METHODS: A total of 96 host-seeking adult H. punctata were collected from 29 sampling sites in 11 districts

Identification of a new orthonairovirus associated with human febrile illness in China.

The genus Orthonairovirus, which is part of the family Nairoviridae, includes the important tick-transmitted pathogens Crimean-Congo hemorrhagic fever virus and Nairobi sheep disease virus, as well as many other poorly characterized viruses found in ticks, birds and mammals1,2. In this study, we identified a new orthonairovirus, Songling virus (SGLV), from patients who reported being bitten by ticks in Heilongjiang Province in northeastern China. SGLV shared similar genomic and morphological features with orthonairoviruses and phylogenetically formed a unique clade in Tamdy orthonairovirus of

Characterization of the glycoproteins of Crimean-Congo hemorrhagic fever virus.

Crimean-Congo hemorrhagic fever (CCHF) virus is the cause of an important tick-borne disease of humans throughout regions of Africa, Europe, and Asia. Like other members of the genus Nairovirus, family Bunyaviridae, the CCHF virus M genome RNA segment encodes the virus glycoproteins. Sequence analysis of the CCHF virus (Matin strain) M RNA segment revealed one major open reading frame that potentially encodes a precursor polyprotein 1,689 amino acids (aa) in length. Comparison of the deduced amino acid sequences of the M-encoded polyproteins of Nigerian, Pakistani, and Chinese CCHF virus strai

Striking tick-borne virus diversity and potential reservoirs documented during One-Health-based cross-sectional screening in Anatolia.

BACKGROUND: An expansion of recently described human pathogenic tick-borne viruses from Central Asia toward Europe has been documented. Located on important bird migration routes, Anatolia is an intercontinental crossing hub with various climactic zones and with an abundance of endemic tick species. We sought to investigate tick-borne viruses utilizing a One Health approach encompassing host-removed ticks and host samples. METHODS: We collected host-attached ticks and accompanying plasma in 2023-2024 at locations in 20 provinces representing the 7 distinct geographical regions in Anatolia. Th

[Crimean-Congo hemorrhagic fever].

Crimean-Congo hemorrhagic fever (CCHF), caused by infection with CCHF virus (CCHFV), is viral hemorrhagic fever with high case fatality rate. CCHFV is classified to Family Bunyaviridae, Genus Nairovirus. CCHF is endemic to Africa, Eastern Europe, the Middle East, central Asia, and southern Asia. CCHFV is maintained in nature in several spe- cies of ticks (Hyalomma and Ixodes species) and mammals. Humans are infected with CCHFV by tick-bite or direct contact with viremic animals such as sheep. The CCHF-endemic re- gions are relatively economically disadvantaged areas, therefore CCHF is consider

Evidence of segment reassortment in Crimean-Congo haemorrhagic fever virus.

The complete nucleotide sequences of the small (S) and medium (M) segments of three independent strains of Crimean-Congo haemorrhagic fever (CCHF) virus isolated in Uzbekistan, Iraq and Pakistan have been determined. Partial S and M segment sequences from two additional strains and partial large segment sequences from five strains of CCHF virus have also been obtained. These data have been compiled and compared with published full-length and partial sequences of other CCHF virus strains. Analysis of virus strains for which complete and partial S and M segment sequences are available reveals th

In Silico Analysis of Potential Vaccine Antigens for the Treatment of Crimean-Congo Hemorrhagic Fever Virus (Cchfv).

BACKGROUND: Crimean-Congo hemorrhagic fever virus (CCHFV), identified by the World Health Organization(WHO) as a potential epidemic threat, is a tick-borne Nairovirus primarily transmitted by Hyalomma marginatum species. Since its first detection in the 1940s, CCHFV has spread to multiple regions worldwide and remains a major public health concern due to its high fatality rate and expanding geographic distribution. The virus can be transmitted through tick bites or contact with infected individuals, and no licensed vaccine is currently available to prevent infection. METHODS: In this study, t

Identification of claramine and anidulafungin as entry inhibitors of Crimean-Congo hemorrhagic fever virus.

Crimean-Congo hemorrhagic fever virus (CCHFV) is a tick-borne enveloped virus that causes a severe disease in humans. Despite its wide geographic distribution, no approved vaccines or therapeutics exist. In this study, we achieved high-titer production of pseudotyped virus bearing CCHFV glycoproteins with a C-terminal truncation. Screening over 3,600 small compounds using this pseudotyped virus identified claramine and anidulafungin as CCHFV entry inhibitors. These hit compounds, as well as caspofungin, which is related to anidulafungin, inhibited pseudotyped viral infection in human liver cel

Structural characterization of protective non-neutralizing antibodies targeting Crimean-Congo hemorrhagic fever virus.

Crimean-Congo Hemorrhagic Fever Virus (CCHFV) causes a life-threatening disease with up to a 40% mortality rate. With no approved medical countermeasures, CCHFV is considered a public health priority agent. The non-neutralizing mouse monoclonal antibody (mAb) 13G8 targets CCHFV glycoprotein GP38 and protects mice from lethal CCHFV challenge when administered prophylactically or therapeutically. Here, we reveal the structures of GP38 bound with a human chimeric 13G8 mAb and a newly isolated CC5-17 mAb from a human survivor. These mAbs bind overlapping epitopes with a shifted angle. Crimean-Congo haemorrhagic fever virus: sequence analysis of the small RNA segments from a collection of viruses world wide.

Crimean-Congo haemorrhagic fever virus (CCHFv) is a member of the genus Nairovirus in the family Bunyaviridae. It possesses a tripartite, single stranded RNA genome of negative polarity consisting of large (L), medium (M) and small (S) segments. CCHF virus is enzootic in life stock and wild animals in many parts of the Middle East, Asia and Africa and is also recognised in Southeast Europe. Severe disease, manifest as haemorrhagic fever and high mortality rates (up to 50%), is only recognised in humans. We have determined the complete sequence of the small genomic RNA segment from several stra

Structural and functional characterisation of the Crimean-Congo haemorrhagic fever virus RNA dependent RNA polymerase.

Crimean-Congo Haemorrhagic Fever Virus (CCHFV) is found across Africa, Asia, and the Middle East where it can cause Haemorrhagic outbreaks with high case fatality rates. Central to the viral life cycle is the viral L-protein, a crucial and multifunctional protein which both transcribes and replicates the viral genome. Here, we present the cryoEM structures of an RNA free and a 5' promoter bound complex, describing the core catalytic RNA-dependent RNA polymerase (RdRp). We observe an RdRp that is substantially larger than related L-proteins and contains domain insertions unique to the nairoviru

Harnessing Hazara Virus as a Surrogate for Crimean-Congo Hemorrhagic Fever Virus Enables Inactivation Studies at a Low Biosafety Level.

Research on highly pathogenic biosafety level 4 (BSL-4) viruses that are classified as Select Agents involves transferring inactivated materials to lower containment levels for further analysis. Compliance with Select Agent and BSL-4 safety regulations necessitates the validation and verification of inactivation procedures. To streamline this process, it would be beneficial to use surrogate BSL-2 viruses for inactivation studies. This not only simplifies BSL-4 work but also enables the testing and validation of inactivation procedures in research facilities that lack access to high-containment

Tick-Borne Infectious Diseases in China, 2003-2023.

Tick-borne diseases (TBDs) pose a substantial threat to global public health, including China. This article systematically reviews the epidemiology of TBDs in Chinese mainland between 2003 and 2023. We retrieved and analyzed the incidence and distribution of notable bacterial, viral, and parasitic TBDs in China, and discussed their clinical characteristics, diagnostic approaches, and vectors. Severe fever with thrombocytopenia syndrome is the leading TBD in China, with cases rapidly increasing since its first report in 2009, and a high case fatality rate of 5%-33% in patients with severe disea

Ganjam virus (GANV), a member of genus Nairovirus of family Bunyavirdae is of considerable veterinary importance in India. Though, predominantly tick borne, GANV was also isolated from mosquitoes, man and sheep. Neutralizing and complement fixing antibodies to GANV have been detected in animal and human sera collected from different parts of the country. Thirty three strains of GANV have been isolated from India, mainly from Haemaphysalis ticks. The virus replicated in certain vertebrate and mosquito cell lines and found pathogenic to laboratory animals. One natural infection and five laborato

Current status of Crimean-Congo haemorrhagic fever in the World Health Organization Eastern Mediterranean Region: issues, challenges, and future directions.

Crimean-Congo haemorrhagic fever (CCHF) is the most widespread, tick-borne viral disease affecting humans. The disease is endemic in many regions, such as Africa, Asia, Eastern and Southern Europe, and Central Asia. Recently, the incidence of CCHF has increased rapidly in the countries of the World Health Organization Eastern Mediterranean Region (WHO EMR), with sporadic human cases and outbreaks of CCHF being reported from a number of countries in the region. Despite the rapidly growing incidence of the disease, there are currently no accurate data on the burden of the disease in the region d
