III - Prostaglandin hyperalgesia: relevance of the peripheral effect for the analgesic action of opioid-antagonists.

Morphine injected into the rat cerebral ventricles had a marked analgesic effect, while no effect was observed with pentazocine and naloxone or nalorphine caused a strong hyperalgesia. Administered systemically (IP) naloxone and nalorphine caused a transitory analgesia followed by a long lasting hyperalgesic effect; morphine and pentazocine showed only an analgesic effect. It was concluded that the site of analgesic action of opioid-antagonists is peripheral rather than central. The peptidase-resistant enkephalin-analog, BW 180c, which does not cross the blood brain barrier, caused a marked an

Localization of the central sympatho-inhibitory effect of a narcotic analgesic agent, fentanyl, in cats.

Fentanyl (10 and 30 mug/kg), a narcotic analgesic, induced in cats a decrease in blood pressure and heart rate and reduced spontaneous splanchnic nerve activity. Fentanyl reduced the pressor response to medullary stimulation, but did not change the pressor response to hypothalamic or cervical spinal cord stimulation. Fentanyl reduced the potential evoked in the splanchnic nerve by stimulation at low frequency of a pressor area of the medulla oblongata. The potentials evoked in the splanchnic nerve by hypothalamic or cervical spinal cord stimulation were only slightly changed. Nalorphine (0.5 m

On the immobilization of hartebeest and kob in Upper Volta.

24 hartebeests (Alcelaphus buselaphus major), one waterbuck (Kobus defassa) and 16 kobs (Kobus kob) were immobilized during field work in Upper Volta. The use of the newly developed piperidine derivative R 33799 at weight treated dosage levels can be strongly recommended for the immobilisation of hartebeest. In this species the drug produces a sufficient deep analgesia within a reasonable short period for all handling purposes. It is safe, has a wide therapeutic index, can be used in syringes of not more than 1 ml capacity and is quickly reversed by the antidote (nalorphine hydrobromide). No f

The effects of acute and chronic morphine treatment and of morphine withdrawal on rat brain in vivo.

1. The effects of morphine, nalorphine, acetazolamide, and 10% CO(2) on brain metabolite concentrations of 24h-starved rats were studied. 2. A single dose of morphine (20mg/kg body wt.) caused an increase in brain glucose concentration (42%) and decreased concentrations of lactate (24%), pyruvate (29%), citrate (20%), alpha-oxoglutarate (16%), malate (14%) and creatine phosphate (10%) after 30min. No changes were found in adenine nucleotide concentrations. 3. The same dose of morphine increased arterial CO(2) from 5.07 to 7.60 kN/m(2) (38 to 57 Torr), decreased the pH from 7.41 to 7.31 and dec

The pharmacology of butorphanol, a 3,14-dihydroxymorphinan narcotic antagonist analgesic.

Butorphanol, a new, totally synthetic morphinan, which is chemically related to naloxone, has been demonstrated to have both analgesic and narcotic antagonist properties. In rodent antiwrithing analgesic tests, butorphanol was 4 to 30 times more potent than morphine and dl-pentazocine, respectively. As an antagonist, butorphanol was about equivalent to nalorphine and 30 times more potent than dl-pentazocine. On the basis of the naloxone-induced mouse jumping test and the lack of substitution in withdrawn morphine-dependent mice, it is estimated that the potential for physical dependence of but

Acute opium poisoning: a report of two hundred cases in Iran.

The diagnosis and management of 200 cases of self-poisoning due to orally ingested crude opium in Iran is reported and discussed. Gastric lavage supported when necessary by nalorphine was effective treatment in the majority of cases because of the slow absorption of opium. Mechanical ventilation was required in twelve cases (6%) only. There were three deaths; two of these were due to pulmonary oedema which was apparently not the result of hypoxia.

[The antishock action of opiate antagonists].

In acute experiment on 86 adult rabbits with traumatic shock modelled according to Cannon, it was established that intravenous administration of Nalorphine at the doses of 0.5 and 2 mg/kg of weight at the early period of shock contributed to prolongation of lifetime of the majority of animals. In some of them, the small doses accelerated the occurrence of lethal outcome. In administration of 0.1 mg/kg of weight of Naloxone, almost all the animals survived. At the late period of shock, the effectiveness of these preparations is insufficient.

[Effect of synthetic analogs of enkephalins, morphine and their antagonists on the course of experimental traumatic shock].

Effects of naloxone, nalorphine, thyroliberin, morphine and two analogues of enkephalins (FK 33-824 and Tyr-D-Ala-Gly-Phe(NO2)-NH2) on the course of traumatic shock were studied in experiments on rabbits. It was found that antagonists of opioid peptides aggravated the course of traumatic shock and morphine and synthetic analogues of enkephalins exerted positive effects during its treatment. Endogenous opioid peptides are suggested to play the protective role in experimental traumatic shock.

Analysis of the inhibition of pethidine N-demethylation by monoamine oxidase inhibitors and some other drugs with special reference to drug interactions in man.

1. N-Demethylation of pethidine was studied in microsomal suspensions from unstarved male rat liver and the N-demethylase identified as belonging to the class of hepatic microsomal mixed function oxidases.2. A study of the structure/action relationships of compounds inhibiting pethidine N-demethylase revealed that hydrazine derivatives including phenylhydrazine, methylphenylhydrazine and mebanazine were all potent competitive inhibitors.3. Pethidine N-demethylase was only slightly inhibited by histamine and amphetamine but not by adrenaline and ephedrine nor by several miscellaneous compounds

[Selective effect of morphine and diazepam on aggressiveness caused by prolonged administration of apomorphine to rats].

In experiments on rats administration of apomorphine (0.5 mg/kg twice a day for 14 days) caused the intensification of aggressiveness. This exerted no substantial effect on stereotyped behavior. Haloperidol produced inhibitory effect both on aggressiveness and stereotypy. The GABA derivative phenybut did not change the effects of apomorphine. Administration of morphine and diazepam produced a selective inhibition of aggressiveness. In doses exerting antiaggressive effect these substances produced no substantial effect on stereotypy. This antiaggressive effect of morphine and diazepam was suppr

Intravenous self-administration of drugs in rats.

A standardized self-administration procedure in rats was used to determine the intravenous self-administration liability of graded doses of various drugs. Self-administration was reliably established with the tested addictive drugs (morphine, heroin, fentanyl and d-amphetamine), but not with the nonaddictive drugs (chlorpromazine and nalorphine). However, 1 out of 14 animals on nalorphine clearly demonstrated self-administering behavior. Self-administration was observed with delta1-tetrahydrocannabinol, but the percentage of animals (40% on the highest dose) that initiated this behavior and th

Effects of serine on morphine-dependent mice.

Single doses of serine (250 mg/kg i.p.) failed to elevate central glycine in mice (1 h after injection), or to compensate for losses caused by implantation of morphine base (70 mg/animal), and reduced the jumping response to nalorphine (100 mg/kg i.p.). In contrast, sustained treatment with 250-mg/kg doses of serine at 24-hour intervals produced increases in brain and medullar glycine (1 h after 4th injection: 46-50% in normal, 39-75% in morphine-dependent mice, and enhanced the jumping response (40% over controls).

The effect of morphine on the adrenergic nerves of the isolated guinea-pig jejunum.

Nerves accompanying the mesenteric blood-vessels of the isolated guinea-pig jejunum were stimulated and changes in the longitudinal muscle coat were recorded. Stimulation of the nerves led to a rapid relaxation if the intestine was contracted by a previous administration of histamine. The relaxation was inhibited by morphine and partially restored by nalorphine. Bretylium and atropine also inhibited the relaxation and cocaine increased it somewhat. Morphine, atropine, and bretylium had no significant effect on the depressant action of noradrenaline on histamine contractions, whereas cocaine sl

Apomorphine-induced pecking in pigeons.

Apomorphine produced persistent pecking in pigeons, the latent period, intensity and duration of which were related to the dose. The ED50 was estimated as 78.1+/-11.1 mug./kg. On chronic administration of apomorphine there was a significant decrease in latent period and weight which quickly returned to normal on stopping the drug. No conditioning and no tolerance were observed. The uncertain emetic effect of apomorphine in pigeons has been confirmed. Ten other centrally acting agents tested (caffeine, cocaine, 5-hydroxytryptamine, lysergic acid diethylamide, methamphetamine, morphine, nalorphi

Discriminative stimulus effects of two doses of fentanyl in rats: pharmacological selectivity and effect of training dose on agonist and antagonist effects of mu opioids.

RATIONALE: Discriminative stimulus effects of mu opioids vary systematically as a function of training dose. Differences among training doses may arise from multiple mechanisms. OBJECTIVES: In vivo apparent pA(2) analyses were used to examine the contributions of opioid mechanisms to stimulus control by low and high training doses of the mu opioid fentanyl. METHODS: Saline and one of two doses of fentanyl, administered s.c., were established as discriminative stimuli in two groups of rats (low training dose group: 0.01 mg/kg; high training dose group: 0.04 mg/kg). Generalization tests and in

Detection of O6-monoacetylmorphine in urine samples by GC/MS as evidence for heroin use.

A method for qualitative and quantitative detection of nanogram amounts of O6-monoacetylmorphine (MAM) in urine samples of heroin abusers is presented. The detection of MAM is based on the alkaline solid phase extraction of the urine samples using an octadecyl column and transformation into the pentafluoropropionyl (PFP) derivatives. PFP-MAM is separated by capillary GC and identified mass spectrometrically by selected ion monitoring (SIM). The determination of the MAM levels was carried out by quantitative GC/SIM using nalorphine as the internal standard. A positive identification of MAM allo
