Lysosome-Targeted Naphthalimide-Based Fluorescence for the Detection of Fe(III) and Monitoring of Iron Metabolism.

Iron is crucial for numerous biological processes, and lysosomes play an essential role in iron metabolism by regulating Fe3+ levels. Disruptions of this regulation can lead to Fe3+ accumulation, resulting in membrane damage and ferroptosis. Here, we have developed a water-soluble fluorescent probe BiNIT that specifically targets lysosomes for the selective detection of Fe3+. BiNIT features a bis-naphthalimide structure linked by a thiophene moiety and incorporates two quaternary ammonium groups, which enhance its ability to target lysosomes and its solubility in aqueous environments. The prob

Design and Enzyme-Targeted Assessment of 1,2,4-Triazole-1,8-Naphthalimide Hybrids in Drug Discovery.

This study reports the design, synthesis, and biological evaluation of novel hybrid 1,2,4-triazole-1,8-naphthalimide derivatives using both classical and environmentally friendly synthetic routes. The synthetic strategy involved a multistep process starting with the preparation of triazolylthioacetic acid esters, followed by electrophilic cyclization-employing both conventional bromine and a green, bromine-free method-and culminating in amidation reactions to yield the target compounds. Structural modifications, including the incorporation of pyridinyl and benzoate moieties, were introduced to

Structure-Function Analysis of Hydroxy-1,8-Naphthalimide Photoacids for ESPT-Driven H2S Probes.

Hydroxy-1,8-naphthalimides are a class of fluorophores known for their strong photoacidity, which facilitates excited-state proton transfer (ESPT) and results in red-shifted fluorescence emission. Based on this unique photophysical behavior, we synthesized and evaluated four reaction-based fluorescent probes (L1-L4) for the selective detection of hydrogen sulfide (H2S). Structure-function analysis revealed that the position of the hydroxyl substituent plays a critical role in modulating ESPT efficiency, emission wavelength, and overall probe sensitivity. Specifically, L1 (3-hydroxy) and L3 (4-

Theoretical Study of Three Ratiometric 1,8-naphthalimide Fluorescent Probes for Hydrogen Sulfide Detection.

Three ratiometric 1,8-naphthalimide fluorescent probes for hydrogen sulfide detection have been studied theoretically by using density functional theory and time-dependent functional theory. From the detailed comparison of the optimized geometries, it is found that the change of substituents has a slight effect on the structure of 1,8-naphthalimide fluorophore, and the effect is mainly located in the part attached to the changed substituent. The change of the electron-withdrawing or electron-donating ability of the substituents on the 1,8-naphthalimide has an effect on the electronic spectra.

Spectroscopic and Biological Properties of the 3-Imino-1,8-naphthalimide Derivatives as Fluorophores for Cellular Imaging.

This paper presents the photophysical and biological properties of eight 3-imino-1,8-naphthalimides. The optical properties of the compounds were investigated in the solvents that differed in their polarity (dichloromethane, acetonitrile, and methanol), including three methods of sample preparation using different pre-dissolving solvents such as dimethyl sulfoxide or chloroform. In the course of the research, it was found that there are strong interactions between the tested compounds and DMSO, which was visible as a change in the maximum emission band (λem) of the neat 3-imino-1,8-naphthalimi

Synthesis, G-Quadruplex DNA binding and cytotoxic properties of naphthalimide substituted styryl dyes.

G-Quadruplex DNAs, formed by G-rich DNA sequences in human genes, are promising targets for design of cancer drugs. In this study, two naphthalimide substituted styryl dyes with different sizes of aromatic groups were synthesized. The spectral analysis showed that the dye X-2 with a large aromatic group formed aggregates in buffer solution displaying very weak fluorescence intensity, and disaggregated in the presence of G-Quadruplex DNAs with large intensity enhancements (up to ~1800 fold). Moreover, X-2 displayed good selectivity to G-Quadruplex DNAs. In contrast, dye X-3 with the smaller aro

A dual-function naphthalimide derivative for Cr(VI) detection and supramolecular room-temperature phosphorescence.

A functional fluorescent molecule, 4-bromo-1,8-naphthalimide derivative (BN1B) was designed and synthesized and used as a fluorescent probe for detecting Cr(VI) in aqueous media. The fluorescence intensity of BN1B decreased with increasing Cr(VI) concentration, and the detection limits were 3.64 × 10⁻⁶ mol·L⁻1 for CrO₄2⁻ and 2.98 × 10⁻⁶ mol·L⁻1 for Cr₂O₇2⁻. Moreover, a distinct fluorescence color change was observed for BN1B solutions containing CrO₄2⁻ or Cr₂O₇2⁻ under 356 nm UV irradiation, producing pale yellow and orange yellow emissions, respectively. The probe features simple preparation,

Rational design of a 1,8-naphthalimide-based fluorescent probe for water detection in organic solvents.

Generally, trace amounts of water in organic solvents are deadly to chemical production and laboratory chemical reactions. Thus, a novel derivative of 1,8-naphthalimide (NPP-N+) was designed and synthesized for detecting water content in water-miscible organic solvents. NPP-N+ features a donor-π-acceptor (D-π-A) structure, where strong intramolecular charge transfer (ICT) and suppressed twisted intramolecular charge transfer (TICT) in aqueous media lead to stronger fluorescence in water than in organic solvents. The experimental results show that the detection limits of NPP-N+ for water in eth

A Golgi Apparatus-Targeting, Naphthalimide-Based Fluorescent Molecular Probe for the Selective Sensing of Formaldehyde.

Formaldehyde (FA) is a colorless, flammable, foul-smelling chemical used in building materials and in the production of numerous household chemical goods. Herein, a fluorescent chemosensor for FA is designed and prepared using a selective organ-targeting probe containing naphthalimide as a fluorophore and hydrazine as a FA-binding site. The amine group of the hydrazine reacts with FA to form a double bond and this condensation reaction is accompanied by a shift in the absorption band of the probe from 438 nm to 443 nm upon the addition of FA. Further, the addition of FA is shown to enhance the

Structural effects of naphthalimide-based fluorescent sensor for hydrogen sulfide and imaging in live zebrafish.

Hydrogen sulfide (H2S) is an important biological messenger, but few biologically-compatible methods are available for its detection in aqueous solution. Herein, we report a highly water-soluble naphthalimide-based fluorescent probe (L1), which is a highly versatile building unit that absorbs and emits at long wavelengths and is selective for hydrogen sulfide over cysteine, glutathione, and other reactive sulfur, nitrogen, and oxygen species in aqueous solution. We describe turn-on fluorescent probes based on azide group reduction on the fluorogenic 'naphthalene' moiety to fluorescent amines a

A naphthalimide-based bifunctional fluorescent probe for the differential detection of Hg²⁺ and Cu²⁺ in aqueous solution.

A novel fluorescent probe NPM based on naphthalimide was designed and synthesized. Interestingly, NPM exhibited highly selective fluorescence turn-on for Hg(2+) and turn-off for Cu(2+) in aqueous solution (10 mM HEPES, pH 7.5). Its fluorescence intensity enhanced in a linear fashion with the concentration of Hg(2+) and decreased in a nearly linear fashion with the concentration of Cu(2+). Thus NPM could be potentially used for the quantification of Hg(2+) and Cu(2+) in aqueous solution. A series of model compounds were rationally designed and synthesized in order to explore the sensing mechani

Synthesis of Naphthalimides through Tandem Pd(II)-Catalyzed C(sp3)-H Oxidation and Diels-Alder Reaction Using a Transient Directing Group Strategy.

Naphthalimides have found extensive applications in materials science and pharmaceuticals. It is still highly desirable to develop efficient methods for the synthesis of naphthalimides with structural diversity. In this work, we developed a new approach for the synthesis of naphthalimides via a tandem reaction of o-methylbenzaldehydes and maleimides. The tandem reaction involves Pd(II)-catalyzed benzylic C(sp3)-H oxidation using an amino acid as the transient directing group and Diels-Alder reaction. The subsequent dehydration forms naphthalimides. The reaction introduces the imide moiety and

Thionated donor-acceptor naphthalimides as efficient heavy-atom-free triplet photosensitizers for type I/II ROS generation in hypoxia-immune photodynamic therapy.

Photodynamic therapy (PDT) is a promising non-invasive cancer treatment but faces challenges from the hypoxic tumor environment and inefficient reactive oxygen species (ROS) generation. To address this, we designed a series of donor-acceptor naphthalimide derivatives via synergistic integration of electron-donating groups and stepwise thionation of the NI core, achieving ROS generation even under hypoxia. The donor-acceptor structure and stepwise thionation synergistically extend absorption to the 500-700 nm red light region and reduce the singlet-triplet energy gap (ΔEST), thereby enhancing t

Column Chromatography-Free Synthesis of Spirooxindole and Spiroindanone-Based Naphthalimides as Potent c-MYC G4 Stabilizers and HSA Binders for Elevating Anticancer Potential.

G-quadruplex (G4) DNA plays a pivotal regulatory role in fundamental biological processes, integral for governing cellular functions such as replication, transcription, and repair in living cells. Within cancer cells, G4 DNA exerts an impact on the expression of crucial genes such as c-MYC, effectively repressing its activity when structured within its promoter region. Therefore, employing molecular scaffolds to target these structures offers an attractive strategy for altering their functions. In our pursuit of potent and selective G-quadruplex binders, herein we report a series of spironapht

Novel Water-Soluble, Heavy Atom-Free Thioxanthene-Fused Naphthalimide-Polyamine Phototheranostic Conjugates.

Photodynamic therapy (PDT) is a promising therapeutic modality based on the combined use of light and photosensitizers (PSs) and approved for treating several types of cancers. Thio-heterocyclic naphthalimide-based PSs are particularly attractive as phototheranostic agents, combining fluorescence imaging for diagnosis with reactive oxygen species (ROS)-mediated cytotoxicity for treatment. However, no effective conjugation strategy for simultaneously endowing naphthalimides with water solubility and efficient ROS production ability currently exists. Herein, novel water-soluble, heavy atom-free

An ICT-FRET Fluorescent Probe Derived from Coumarin-Naphthalimide for Ratiometric Detection of Copper Ions.

This research presents a new ratiometric fluorescent probe designed for Cu²⁺ detection, utilizing a dual mechanism of intramolecular charge transfer (ICT) and fluorescence resonance energy transfer (FRET). In this probe, coumarin acts as the energy donor while naphthalimide serves as the acceptor, with a 2-picolinic ester moiety functioning as the Cu²⁺ binding site. In the absence of Cu²⁺, the 2-picolinic ester effectively suppresses electron transfer, inhibiting both ICT and FRET, resulting in strong blue fluorescence (λem = 478 nm) from the coumarin moiety. However, upon Cu²⁺ binding, the Cu

Naphthalimide-mediated multitargeting antibacterial actions to enhance efficacy and broaden antibacterial spectrum of nitroimidazolyl ethylalcohols.

To combat the escalating challenge of bacterial resistance, a novel class of naphthalimide-mediated nitroimidazolyl ethylalcohols (NNEs) was developed. Most of the prepared NNEs showed potent antibacterial activities. Notably, nitrophenylamino NNE 37 g enhanced the activity of metronidazole against anaerobic bacteria and expanded its antibacterial spectrum to aerobic bacteria (MICs = 0.25-4 μg/mL), being superior to ciprofloxacin and ceftizoxime. Structure-activity relationship analysis suggested that the nitroimidazolyl ethylalcohol fragment was essential for activity, whereas chirality showe
