Sensitivity to ivermectin and pyrantel of Ancylostoma ceylanicum and Necator americanus.

Experiments were carried out in the hamster to compare the relative susceptibility of Necator americanus and Ancylostoma ceylanicum to treatment with ivermectin. A. ceylanicum was found to be 300 times more sensitive to the anthelmintic with a 50% effective dose (ED50) of the order of 10-15 micrograms kg-1 body weight whilst that for N. americanus approximated to 3-5 mg kg-1. Furthermore, whereas complete clearance of A. ceylanicum was observed with a dose of 100 micrograms kg-1, N. americanus was not totally removed after treatment with 25 mg kg-1, the highest dose tested. Both parasites prov

[Geographic distribution of Ancylostoma duodenale and Necator americanus in Taiwan].

In order to determine the geographical distribution of Ancylostoma duodenale and Necator americanus in Taiwan, 2,759 fresh stool samples were collected from 5th-grade school children of both sexes among 43 preliminary schools in 16 counties, and then examined by means of the modified test-tube filter-paper cultivation method for identification of the filariform larvae of hookworm. Both species of A. duodenale and N. americanus are widespread in Taiwan. The overall infection rate was 20.00%. The rate of A. duodenale, N. americanaus and mixed infection of both species was 9.85%, and 7.75% and 2.

Engineering Mixotrophy in the Chemolithoautotrophic Cupriavidus necator through Hydrogenase Induction.

Mixotrophy offers a promising strategy for biosynthesis by simultaneously utilizing organic carbon and CO2; however, mixotrophic microorganisms are rarely isolated outside of photoautotrophic microalgae. In this study, the chemoautotroph Cupriavidus necator H16 was found to preferentially consume fructose when coexisting with CO2 and H2, switching to utilization of only CO2 and H2 after fructose depletion. Transcriptomic analysis revealed significant differences in genes involved in energy metabolism, electron generation, and the respiratory chain. The molecular mechanism underlying the inabil

Sequence data mining in search of hookworm (Necator americanus) microRNAs.

The new world hookworm, Necator americanus is a soil-transmitted nematode responsible for Necatoriasis (a type of helminthiasis) in hosts such as humans, dogs, and cats. N. americanus genome and transcriptome has been sequenced and a draft assembly analysis has been published highlighting protein coding genes and possible drug target proteins. Hookworm microRNA identification, annotations and their public release is yet to be attempted. The same is evident from lack of hookworm miRNA information in related popular public nucleotide sequence repositories such as miRBase, GenBank, WormBase etc.

Larvicidal activity of albendazole against Necator americanus in human volunteers.

This study evaluated the efficacy and tolerance of a single oral 400-mg dose of albendazole on Necator americanus larvae, and compared its efficacy when administered between meals or with a meal. Twenty-nine healthy and hookworm-free male volunteers were exposed on the forearm to approximately 45 8-day-old N. americanus larvae. All subjects developed discrete maculopapular eruptions at the site of larval application. Following a random double-blind study design, each subject received at the end of the 6th post-infection day either the investigational drug or a placebo as follows: Group I (n =

Cross-reactivity between Necator americanus and Schistosoma mansoni in mice.

Poly-parasitism is common in endemic communities and reactivity of sera from hookworm-infected patients against schistosomular antigens has been reported. Protective cross-immunity between N. americanus and S. mansoni was investigated in NIH and BALB/c mice. Protective resistance to homologous challenge with both parasites was confirmed in this model, however, functional immunity to heterologous challenge was not demonstrated. Sera from animals which had received homologous challenge with N. americanus and from hookworm-infected mice, which had previously been exposed to radiation-attenuated S

Optimizing Cupriavidus necator H16 as a host for aerobic C1 conversion.

Biological systems capable of converting CO2 or CO2-derived, single-carbon (C1) compounds can be used to reduce or reverse carbon emissions while establishing a circular bioeconomy to provide sustainable sources of the fuels, foods, and materials humanity relies on. A robust bioeconomy will rely upon a variety of microorganisms capable of assimilating C1 compounds and converting them to valuable products at industrial scale. While anaerobic microbes are ideal hosts for production of short-chain acids and alcohols, microbes capable of aerobic respiration are well suited for biosynthesis of high

The production of Necator americanus larvae for use in experimental human infection.

BACKGROUND: Although there is unprecedented interest in experimental human hookworm infection, details of hookworm manufacture and characterisation have been sparsely reported. In this report, we detail the production and characterisation of Necator americanus larvae for use in a recently published clinical trial. METHODS: Faeces was obtained from an experimentally infected donor. Faecal hookworm DNA was determined by quantitative PCR. Paired samples were incubated in either sterile water or sterile water mixed with antimicrobials (amphotericin and gentamicin). Coproculture was performed by m

Systems metabolic effects of a necator americanus infection in Syrian hamster.

Hookworms (Ancylostoma duodenale and Necator americanus) are blood-feeding intestinal nematodes that infect approximately 700 million people worldwide. To further our understanding of the systems metabolic response of the mammalian host to hookworm infection, we employed a metabolic profiling strategy involving the combination of (1)H NMR spectroscopic analysis of urine and serum and multivariate data analysis techniques to investigate the biochemical consequences of a N. americanus infection in the hamster. The infection was characterized by altered energy metabolism, consistent with hookworm

A survey of genes expressed in adults of the human hookworm, Necator americanus.

Hookworms are gut-dwelling, blood-feeding nematodes that infect hundreds of millions of people, particularly in the tropics. As part of a program aiming to define novel drug targets and vaccine candidates for human parasitic nematodes, genes expressed in adults of the human hookworm Necator americanus were surveyed by the expressed sequence tag approach. In total 161 new hookworm genes were identified. For the majority of these, a function could be assigned by homology. The dataset includes proteases, protease inhibitors, a lipid binding protein, C-type lectins, an anti-bacterial factor, globi

Characterization of Oesophagostomum bifurcum and Necator americanus by PCR-RFLP of rDNA.

Esophagostomiasis in humans due to infection with Oesophagostomum bifurcum (nodular worm) is of major human health significance in northern Togo and Ghana, where Necator americanus (human hookworm) also exists at high prevalence. Yet, very little is known about the transmission patterns of O. bifurcum, which is in part due to the difficulties in diagnosis and in differentiating some life-cycle stages of O. bifurcum from N. americanus using morphological features. As a first step toward developing a molecular-diagnostic assay, it was evaluated whether ribosomal (r)DNA could provide genetic mark

Immunity in humans to Necator americanus: IgE, parasite weight and fecundity.

The effect of the humoral immune response on the weight and fecundity of the hookworm Necator americanus was examined in an endemically-infected human population. There was a highly significant negative correlation between total IgE levels and parasite weight and fecundity, after controlling for any effects of host age and hookworm burden. This correlation was present both at initial treatment and after 2 years' reinfection. There was a similar negative correlation between the number of eosinophils and hookworm weight and fecundity at initial treatment. Correlations with levels of specific ant

Effect of albendazole on the larvae and eggs of Necator americanus in golden hamster.

When golden hamsters infected with adult Necator americanus were treated ig with albendazole(Alb) 75 or 150 mg/kg, the efficacy rates were 83.8% and 100%, respectively. Three days before treatment, the mean numbers of larvae recorded from the incubation of feces from 2 treated groups were 29,716 +/- 3,299 and 28,256 +/- 118 per gram of feces, while in the control group, it was 8,828 +/- 2,659. Within 3 days after treatment, no larva was detected in the 2 treated groups, but in the control group the mean larva number was 9,066 +/- 7,416. Furthermore, in 2 treated groups none or only a few degen

Density-dependent effects on establishment of Necator americanus and Ancylostoma ceylanicum.

The relationships between the number of infective larvae (L3s) to which animals were exposed and the establishment of A. ceylanicum and N. americanus in hamsters were examined. There was no evidence of density-dependent constraints on the establishment of A. ceylanicum in the range 10-1000 larvae, but an experiment in which the range was extended to 1500 L3s gave a significant negative correlation between the magnitude of the infecting dose and percentage establishment. Even so the percentage reduction was relatively small, approximating to 0.89% per 100 larvae administered, and in practice de

The action of a mast cell protease on the cuticular collagens of Necator americanus.

Collagen-like proteins, thought to be responsible for maintaining the structural integrity of the nematode cuticle, were isolated from adult Necator americanus and shown to be susceptible to digestion by purified mast cell proteases. Although these collagens would appear normally to be masked by superficially expressed (surface) antigens, it is suggested that a sufficiently avid and specific immune response could remove this potentially protective coat, rendering the structurally important underlying layers open to immune attack.
