Cranial neural crest extirpation and cyclopian development.

Extirpation of cranial neural crest cells in presomite to 5 somite chick embryos produces monsters which in all essential respects are identical to human cyclopias. The primary lesion involves damage to the forebrain and consequently the inhibition of the nasal placode and nasal organ development as well as the destruction of the anterior neural crest cells, with the resultant absence of the nasal and interobital septa and of the facial dermal skeleton.

Role of DNMT3B in the regulation of early neural and neural crest specifiers.

The de novo DNA methyltransferase DNMT3B functions in establishing DNA methylation patterns during development. DNMT3B missense mutations cause immunodeficiency, centromere instability and facial anomalies (ICF) syndrome. The restriction of Dnmt3b expression to neural progenitor cells, as well as the mild cognitive defects observed in ICF patients, suggests that DNMT3B may play an important role in early neurogenesis. We performed RNAi knockdown of DNMT3B in human embryonic stem cells (hESCs) in order to investigate the mechanistic contribution of DNMT3B to DNA methylation and early neuronal d

Infantile hemangiomas exhibit neural crest and pericyte markers.

Infantile hemangiomas (IHs) are the most common benign tumors of infancy and occur with greater than 60% prevalence on the head and neck. Despite their prevalence, little is known about the pathogenesis of this disease. Given the predilection of hemangioma incidence on the face and its nonrandom distribution on embryological fusion plates, we postulated that IHs are derived from pericytes of the neural crest. We performed an analysis on 15 specimens at various stages of the IH progression. Experiments performed included immunohistochemical staining, immunofluorescent staining, quantitative rea

A neural crest origin for cohesinopathy heart defects.

Mutations in subunits or regulators of cohesin cause a spectrum of disorders in humans known as the 'cohesinopathies'. Cohesinopathies, including the best known example Cornelia de Lange syndrome (CdLS), are characterized by broad spectrum, multifactorial developmental anomalies. Heart defects occur at high frequency and can reach up to 30% in CdLS. The mechanisms by which heart defects occur are enigmatic, but assumed to be developmental in origin. In this study, we depleted cohesin subunit Rad21 by 70-80% in a zebrafish cohesinopathy model. The hearts of Rad21-depleted animals were smaller,

Pax7 lineage contributions to the mammalian neural crest.

BACKGROUND: Neural crest cells are vertebrate-specific multipotent cells that contribute to a variety of tissues including the peripheral nervous system, melanocytes, and craniofacial bones and cartilage. Abnormal development of the neural crest is associated with several human maladies including cleft/lip palate, aggressive cancers such as melanoma and neuroblastoma, and rare syndromes, like Waardenburg syndrome, a complex disorder involving hearing loss and pigment defects. We previously identified the transcription factor Pax7 as an early marker, and required component for neural crest deve

Signals controlling neural crest contributions to the heart.

Cardiac neural crest cells represent a unique subpopulation of cranial neural crest cells that are specified, delaminate and migrate from the developing neural tube to the caudal pharynx where they support aortic arch artery development. From the caudal pharynx, a subset of these cells migrates into the cardiac outflow tract where they are needed for outflow septation. Many signaling factors are known to be involved in specifying and triggering the migration of neural crest cells. These factors have not been specifically studied in cardiac crest but are assumed to be the same as for the other

Taking neural crest stem cells to new heights.

The carotid body is an organ of the peripheral nervous system that senses oxygen concentration in the blood and responds to changes by regulating breathing. Pardal et al. (2007) now report the discovery of carotid body stem cells, which proliferate in response to hypoxia and generate neurons that secrete dopamine. This new source of adult stem cells may be useful in therapies for treating Parkinson's disease.

Corneal keratocytes retain neural crest progenitor cell properties.

Corneal keratocytes have a remarkable ability to heal the cornea throughout life. Given their developmental origin from the cranial neural crest, we asked whether this regenerative ability was related to the stem cell-like properties of their neural crest precursors. To this end, we challenged corneal stromal keratocytes by injecting them into a new environment along cranial neural crest migratory pathways. The results show that injected stromal keratocytes change their phenotype, proliferate and migrate ventrally adjacent to host neural crest cells. They then contribute to the corneal endothe

Neural crest-derived defects in experimental esophageal atresia.

Esophageal atresia (EA) is often associated with cardiovascular and other malformations that are likely neural crest derived. The present study tests the hypothesis that the heart and great vessels and the thymus and parathyroids may be abnormal in the rat model of EA as a result of disturbed neural crest development. Time-mated pregnant rats received intraperitoneally on d 8 and 9 of gestation either 2 mg/kg adriamycin or vehicle. Esophageal, heart, and thymic malformations were sought under the microscope in term fetuses. The parathyroids were histologically investigated. Control fetuses had

Neural crest and normal development: a new perspective.

Several clinical syndromes, including the DiGeorge syndrome, are characterized by clusters of developmental defects of the heart and great vessels with structures derived from the embryonic pharyngeal apparatus including thymus and parathyroids. The connective tissue derivatives of neural crest are necessary for the normal development of these structures, and there is new experimental evidence that depletion of neural crest causes defects similar to these clinical syndromes. Therefore it is proposed that many of these syndromes are due to inappropriate development of neural crest. The implicat

The Ocular Neural Crest: Specification, Migration, and Then What?

During vertebrate embryonic development, a population of dorsal neural tube-derived stem cells, termed the neural crest (NC), undergo a series of morphogenetic changes and extensive migration to become a diverse array of cell types. Around the developing eye, this multipotent ocular NC cell population, called the periocular mesenchyme (POM), comprises migratory mesenchymal cells that eventually give rise to many of the elements in the anterior of the eye, such as the cornea, sclera, trabecular meshwork, and iris. Molecular cell biology and genetic analyses of congenital eye diseases have provi

Neural crest and the origin of species-specific pattern.

For well over half of the 150 years since the discovery of the neural crest, the special ability of these cells to function as a source of species-specific pattern has been clearly recognized. Initially, this observation arose in association with chimeric transplant experiments among differentially pigmented amphibians, where the neural crest origin for melanocytes had been duly noted. Shortly thereafter, the role of cranial neural crest cells in transmitting species-specific information on size and shape to the pharyngeal arch skeleton as well as in regulating the timing of its differentiatio

Aebp2 as an epigenetic regulator for neural crest cells.

Aebp2 is a potential targeting protein for the mammalian Polycomb Repression Complex 2 (PRC2). We generated a mutant mouse line disrupting the transcription of Aebp2 to investigate its in vivo roles. Aebp2-mutant homozygotes were embryonic lethal while heterozygotes survived to adulthood with fertility. In developing mouse embryos, Aebp2 is expressed mainly within cells of neural crest origin. In addition, many heterozygotes display a set of phenotypes, enlarged colon and hypopigmentation, similar to those observed in human patients with Hirschsprung's disease and Waardenburg syndrome. These p

Targeting of endothelin receptor-B to the neural crest.

Endothelin receptor B (Ednrb) plays a critical role in the development of melanocytes and neurons and glia of the enteric nervous system. These distinct neural crest-derived cell types express Ednrb and share the property of intercalating into tissues, such as the intestine whose muscle precursor cells also express Ednrb. Such widespread Ednrb expression has been a significant obstacle in establishing precise roles for Ednrb in development. We describe here the production of an Ednrb allele floxed at exon 3 and its use in excising the receptor from mouse neural crest cells by use of Cre-recomb

Ocular and facial maldevelopment: the role of neural crest.

Three children are described who show a spectrum of clinical abnormalities affecting their eyes and non ocular tissues. These entities are best explained as disorders of neural crest migration and once recognised should lead to a search for other systemic developmental disorders. The mechanism of combined facial skeleton and eye malformation is discussed in the light of our current understanding of ocular embryology, and the justification for considering these and other entities as examples of Neurocristopathies is explored.

[Rieger syndrome as an expression of neural crest dysgenesis].

Malformations of the anterior segment of the eye can be associated with different systemic findings. The regular combination of aplasia of the permanent central and lateral incisors, typical ocular changes, and umbilical dysmorphology is known as the Rieger syndrome. Aplasia of the permanent incisors can serve as a diagnostic symptom. We report on three siblings who had characteristical symptoms of Rieger syndrome. The tissue involved derives from the neural crest cells. Mesenchymal dysgenesis is discussed as the etiology of this disorder.

[Role of the neural crest in the eye development of birds].

By means of the method for formation of quail-hen transplantation chimeras , contribution of the neural crest cells in the eye development has been studied in early avian embryos. In 6-9 day-old chimeras the neural crest cells participate in formation of the eye sclera and ciliary body, and they also form corneal endothelium and stroma.

Neural crest interactions in the development of the immune system.

The thymus is formed by contributions from different sources which must interact in a precisely timed sequence for proper development. Pharyngeal pouch epithelium interacts with embryonic connective tissue in preparation for migration of blood-borne lymphoid stem cells into the thymic primordium. The embryonic connective tissue in this region is derived from neural crest. Ablation of small portions of neural crest prevents or alters the development of the thymus. Other pharyngeal pouch derivatives such as thyroids and parathyroids are affected. Furthermore, defects in the heart and great vesse
