Embryonic hydromyelia: cystic dilatation of the lumbosacral neural tube in human embryos.

In a large collection of human embryos (the Kyoto Collection of Human Embryos, Kyoto University), we encountered five cases with abnormal dilatation of the neural tube at the lumbosacral level. In these examples, the central canal was enlarged, and the roof plate of the neural tube was extremely thin and expanded. The mesenchymal tissue was scarce or lacking between the roof plate and the surface ectoderm. This type of anomaly was assumed to be formed after neural tube closure and may be an early form of spina bifida. In two of the cases, some abnormal cells were found ectopically between the

Myogenic specification of somites is mediated by diffusible factors.

Specification of myogenesis in early chicken somites and segmental plate was studied using transfilter explant cultures to determine if myogenic specification by axial structures is mediated by cell-cell contact. Formation of muscle fibers that express myosin heavy chain was assessed in somites transfilter from neural tube and notochord. Either the neural tube or the notochord from early chick embryos (ED 2) induces myogenesis in unspecified somites (Hamburger and Hamilton (HH) stages 11-14) when these tissues are separated by a 0.2- or a 0.05-micron pore filter. The ventral neural tube is fou

Mediolateral neural patterning during gastrulation precedes dorsoventral patterning in chick and frog.

INTRODUCTION: dorsoventral polarity of the neural tube is based on molecular patterning established during neurulation and is suggested to be induced by fine-tuned interpretation of opposing morphogen gradients in the neural tube: a ventral-to-dorsal gradient is formed by shh secreted from floor plate and notochord whereas a dorsal-to-ventral gradient is created by BMP and Wnt originating from the roof plate. Recent results revealed an unexpected early induction of Shh in the chick prospective floor plate prior to notochord formation. Hence, we addressed the questions as to whether key compone

Immunofluorescent analysis of the spinal cord in dysraphic mice.

Development of the spinal cord was analyzed immunocytochemically at 10-12 days of gestation in normal and dysraphic embryos of the loop-tail mutant mouse, using an anti-neural cell adhesion molecule (NCAM) and the lectin concanavalin A (Con A) as histological markers for evaluating neural cell organization and distribution. The normal and abnormal embryos showed similar patterns of reactivity to anti-NCAM and Con A, even though the neural folds were open and everted in the abnormal embryos, with displacement of the dorsal root ganglia. In the abnormal embryos the floor plate was similar to tha

NMDA Receptor Signaling Is Important for Neural Tube Formation and for Preventing Antiepileptic Drug-Induced Neural Tube Defects.

Failure of neural tube closure leads to neural tube defects (NTDs), which can have serious neurological consequences or be lethal. Use of antiepileptic drugs (AEDs) during pregnancy increases the incidence of NTDs in offspring by unknown mechanisms. Here we show that during Xenopus laevis neural tube formation, neural plate cells exhibit spontaneous calcium dynamics that are partially mediated by glutamate signaling. We demonstrate that NMDA receptors are important for the formation of the neural tube and that the loss of their function induces an increase in neural plate cell proliferation an

Critical period of rat development when sidedness of asymmetric body structures is determined.

We recently reported that rat embryos cultured from the presomite stage in a medium containing the alpha-1 adrenergic agonist, phenylephrine, have a high incidence of situs inversus. In the present study, we have determined more precisely the critical period of development when situs inversus is induced. Rat embryos were harvested at 8 AM on Day 9 of gestation (plug day = Day 0), and divided into different stages of development, namely, early, mid, and late primitive streak stages and early, mid, and late neural plate stages. They then were cultured in rotating bottles to which phenylephrine,

Noncanonical function of folate through folate receptor 1 during neural tube formation.

Folate supplementation reduces the occurrence of neural tube defects (NTDs), birth defects consisting in the failure of the neural tube to form and close. The mechanisms underlying NTDs and their prevention by folate remain unclear. Here we show that folate receptor 1 (FOLR1) is necessary for the formation of neural tube-like structures in human-cell derived neural organoids. FOLR1 knockdown in neural organoids and in Xenopus laevis embryos leads to NTDs that are rescued by pteroate, a folate precursor that is unable to participate in metabolism. We demonstrate that FOLR1 interacts with and op

Origin and early development of the chicken adenohypophysis.

The adenohypophysis (ADH) is an important endocrine organ involved in the regulation of many physiological processes. The late morphogenesis of this organ at neural tube stages is well known: the epithelial ADH primordium is recognized as an invagination of the stomodeal roof (Rathke's pouch), whose walls later thicken and differentiate as the primordium becomes pediculated, and then fully separated from the stomodeum. The primordium attaches to the pial surface of the basal hypothalamus, next to the neurohypophyseal field (NH; future posterior pituitary), from which it was previously separate

Ethanol impairs migration of the prechordal plate in the zebrafish embryo.

Exposure of vertebrate embryos to ethanol causes cyclopia, but little is known about the underlying mechanisms of this effect. Here we show that cyclopia can be induced in the zebrafish by a short ethanol treatment during early gastrula stages and is accompanied by loss of gene expression characteristic of the ventral aspects of the fore- and midbrain. Interestingly, defects in the expression of ventral brain markers are linked to impaired migration of the prechordal plate mesoderm indicating that the correct position of the prechordal plate mesoderm under the anterior neural plate in the zebr

Neuroepithelial cells downregulate their plasma membrane polarity prior to neural tube closure and neurogenesis.

Cell differentiation often involves changes in cell polarity. In this study we show that neuroepithelial cells, the progenitors of all neurons and macroglial cells of the vertebrate central nervous system, downregulate the polarized delivery to the apical and basolateral plasma membrane domains during development. Upon infection of the neuroepithelium of mouse embryos with fowl plague virus (FPV), polarized delivery of the viral envelope hemagglutinin, an apical marker, occurred at the neural plate stage (E8), but was downregulated at the open neural tube stage (E9). Upon infection with vesicu

Pathogenesis of neural dysraphism in the mouse mutant vacuolated lens (vl).

The pathogenesis and expression of caudal neural dysraphism were studied by means of tritiated thymidine autoradiography in the mouse mutant vacuolated lens (vl) at 11-13 days of gestation. In homozygous litters, all of the embryos microscopically exhibited spinal cord defects, even though dysraphism could be detected grossly only in the most severely affected ones. The defects ranged from extreme eversion of widely open neural folds to a dorsally expanded neurocoel with an attenuated or distorted roof plate. Incorporation of tritiated thymidine occurred in cells in the defective roof plates,

Automated Functional Screening for Modulators of Optogenetically Activated Neural Responses in Living Organisms.

All-optical methods of probing in vivo brain function are advantageous for their compatibility with automated microscopy and fast spatial targeting of neural circuit excitation and response. Recent advances in optogenetic technologies allow simultaneous light activation of specific neurons and optical readout of neural activity via fluorescent calcium reporters, providing an attractive opportunity for high-throughput screening assays that directly assess dynamic neural function in vivo. Here we describe a method to automatically record optogenetically activated neural responses in living, hydr

Convergent extension analysis in mouse whole embryo culture.

Mutations have been identified in a non-canonical Wnt signalling cascade (the planar cell polarity pathway) in several mouse genetic models of severe neural tube defects. In each of these models, neurulation fails to be initiated at the 3-4 somite stage, leading to an almost entirely open neural tube (termed craniorachischisis). Studies in whole embryo culture have identified a defect in the morphogenetic process of convergent extension during gastrulation, preceding the onset of neural tube closure. The principal defect is a failure of midline extension, both in the neural plate and axial mes

Zic2 regulates the kinetics of neurulation.

Mutation in human ZIC2, a zinc finger protein homologous to Drosophila odd-paired, causes holoprosencephaly (HPE), which is a common, severe malformation of the brain in humans. However, the pathogenesis is largely unknown. Here we show that reduced expression (knockdown) of mouse Zic2 causes neurulation delay, resulting in HPE and spina bifida. Differentiation of the most dorsal neural plate, which gives rise to both roof plate and neural crest cells, also was delayed as indicated by the expression lag of a roof plate marker, Wnt3a. In addition the development of neural crest derivatives such

Morphogenesis of the murine node and notochordal plate.

Development of the node and formation of the notochordal plate in gestational day 7-9 mice (Theiler stages 10-14) has been documented principally with scanning electron microscopy (SEM) and cell fate analyses utilizing DiI and/or CFSE as a cell label. With SEM, cells composing these two populations are initially identifiable at stage 10 at the ventral midline of the rostral half of the embryo. They can be recognized by their relatively small ventral surface area, as compared to that of the peripherally adjacent prospective gut endodermal cells, and by the presence on the ventral side of each c

Stem cell-derived models of spinal neurulation.

Neurulation is a critical step in early embryonic development, giving rise to the neural tube, the primordium of the central nervous system in amniotes. Understanding this complex, multi-scale, multi-tissue morphogenetic process is essential to provide insights into normal development and the etiology of neural tube defects. Innovations in tissue engineering have fostered the generation of pluripotent stem cell-based in vitro models, including organoids, that are emerging as unique tools for delving into neurulation mechanisms, especially in the context of human development. Each model capture

Xrx1, a novel Xenopus homeobox gene expressed during eye and pineal gland development.

We have isolated a novel Xenopus homeobox gene, Xrx1, belonging to the paired-like class of homeobox genes. Xrx1 is expressed in the anterior neural plate, and subsequently in the neural structures of the developing eye (neural retina and pigmented epithelium), and in other forebrain structures deriving from the anterior neural plate: in the pineal gland, throughout its development, in the diencephalon floor and in the hypophysis. Its rostral limit of expression corresponds to the chiasmatic ridge, which some authors consider as the anteriormost limit of the neural tube: thus, Xrx1 may represe

Formation of the cranial motor neurons in the absence of the floor plate.

The inductive signals for the differentiation of motor neurons in the spinal cord have been experimentally shown to arise from cells in the midventral region of the neural tube, often referred to as the floor plate, and from the notochord. Although the prevailing view is that a similar mechanism accounts for the differentiation of motor neurons in the brain stem, supporting experimental evidence is lacking. Here, using the formation of the trochlear nucleus in the midbrain of duck embryos as a model system, we report that the floor plate and the notochord are not necessary for the development

Periodic Activity of Gnrh-Expressing Neurons in the Ciona Brain: Insights into the Origin of Neural Crest Derived GnRH Neurons in Vertebrates.

Gonadotropin-releasing hormone (GnRH) is an important neuropeptide that regulates reproductive function in vertebrates. GnRH-secreting neurons (GnRH neurons) are derived from neurogenic placodes and the neural crest during embryonic development. In the larva of the ascidian Ciona intestinalis type A (Ciona robusta), one of the gnrh genes, gnrh2, is expressed in the protoplacodederived sensory neurons as well as the central nervous system including the brain vesicle, the motor ganglion, and the nerve cord. In this study, we identified gnrh2-expressing cells in the motor ganglion and visualized

Development of the vertebrate central nervous system: formation of the neural tube.

The developmental process of neurulation involves a series of coordinated morphological events, which result in conversion of the flat neural plate into the neural tube, the primordium of the entire central nervous system (CNS). Failure of neurulation results in neural tube defects (NTDs), severe abnormalities of the CNS, which are among the commonest of congenital malformations in humans. In order to gain insight into the embryological basis of NTDs, such as spina bifida and anencephaly, it is necessary to understand the morphogenetic processes and molecular mechanisms underlying neural tube
